| 2 | Local Proinflammatory Effects of Repeated Skin Exposure to Warfarin, An Anticoagulant Rodenticide in Rats显示文摘Objective: To evaluate the effects of epicutaneous application of anticoagulant warfarin, by examining the presence of tissue injury and immune/inflammatory activity in exposed skin. Methods: Rats were exposed to warfarin by applying 10 μg of warfarin‐sodium to 10‐12 cm 2 skin (range 0.8‐1 μg per 1 cm 2 ) for 3 consecutive days. Tissue injury was evaluated by lipid peroxidation, histomorphological changes and signs of reparative activity in skin. T cell infiltration and selected aspects of epidermal cell activity were examined as indicators of immune/inflammatory skin response to warfarin application. Results: Repeated warfarin application exerted no effect on skin metabolic viability, but resulted in tissue injury (increased malondialdehyde, MDA, production, evident histo‐morphological changes in epidermis and dermis depicting cell injury and death). Increased numbers of proliferating cell nuclear antigen (PCNA + ) cells indicated reparative processes in injured skin. Infiltration of CD3 + cells (T lymphocytes) along with the increased production of tumor necrosis factor‐α (TNF‐α) by epidermal cells from warfarin‐treated skin and their co‐stimulatory effect in an in vitro T‐cell activation assay demonstrated immunomodulatory effects of epicutaneous warfarin. Conclusion: Presented data have documented tissue damage associated with immune/ inflammatory activity in skin exposed to warfarin. | 4 | Preventive effect of a pectic polysaccharide of the common cranberry Vaccinium oxycoccos L. on acetic acid-induced colitis in mice显示文摘AIM: To study isolation and chemical characterization of pectin derived from the common cranberry Vaccinium oxycoccos L. (oxycoccusan OP) and the testing of its preventive effect on experimental colitis.METHODS: Mice were administrated orally with OP two days prior to a rectal injection of 5% acetic acid and examined for colonic damage 24 h later. Colonic inflammation was characterized by macroscopical injury and enhanced levels of myeloperoxidase activity measured spectrophotometrically with o-phenylene diamine as the substrate. The mucus contents of the colon were determined by the Alcian blue dye binding method. Vascular permeability was estimated using 4% Evans blue passage after i.p. injection of 0.05 mol/L acetic acid. RESULTS: In the mice treated with OP, colonic macroscopic scores (1.1 ± 0.4 vs 2.7, P < 0.01) and the total square area of damage (10 ± 2 vs 21 ± 7, P < 0.01) were significantly reduced when compared with the vehicle-treated colitis group. OP was shown to decrease the tissue myeloperoxidase activity in colons (42 ± 11 vs 112 ± 40, P < 0.01) and enhance the amount of mucus of colitis mice (0.9 ± 0.1 vs 0.4 ± 0.1, P < 0.01). The level of colonic malondialdehyde was noted to decrease in OP-pretreated mice (3.6 ± 0.7 vs 5.1 ± 0.8, P < 0.01). OP was found to decrease the inflammatory status of mice as was determined by reduction of vascular permeability (161 ± 34 vs 241 ± 21, P < 0.01). Adhesionof peritoneal neutrophils and macrophages was also shown to decrease after administration of OP (141 ± 50 vs 235 ± 37, P < 0.05).CONCLUSION: Thus, a preventive effect of pectin from the common cranberry, namely oxycoccusan OP, on acetic acid-induced colitis in mice was detected. | 5 | Treatment of rectal prolapse in children with cow milk injection sclerotherapy:30-year experience显示文摘AIM:To evaluate the role and our experience of injecti-on sclerotherapy with cow milk in the treatment of rectal prolapse in children. METHODS:In the last 30 years (1976-2006) we made 100 injections of sclerotherapy with cow milk in 86 chil-dren. In this study we included children who failed to respond to conservative treatment and we perform ope-rative treatment. RESULTS:In our study we included 86 children and in all of the patients we perform cow milk injection sclerot-herapy. In 95.3% (82 children) of patients sclerotherapy was successful. In 4 (4.7%) patients we had recurrent rectal prolapse where we performed operative treatment. Below 4 years we had 62 children (72%) and 24 older children (28%). In children who needed operative trea-tment we performed Thiersch operation and without any complications. It is found that the magnets solution-treated at 1219℃ possess a single 1:7 H phase, exhibiting the homogeneous cellular structure during further aging treatment, leading to the optimum magnetic properties. However, for the magnets solution-treated at 1211 and 1223℃,2:17 H or 1:5 H secondary phase will also form besides 1:7 H main phase, which cannot transform into cellular structure,thus deteriorating the magnetic properties greatly. The irreversible magnetization investigations with recoil loops also propose a non-uniform pinning in the magnets induced by the secondary precipitates. At proper solution temperature, Zr is supposed to occupy the Fe-Fe dumbbell sites in the form of Zr-vacancy pairs, leading to the minimum c/a ratio and thus stabilizing the 1:7 H phase. | 7 | The pathologic relevance of metabolic criteria in patients with biopsy-proven nonalcoholic fatty liver disease and metabolic dysfunction associated fatty liver disease:A multicenter cross-sectional study in China显示文摘Background:This study aimed to assess the association between metabolic syndrome(Met S)and severity of nonalcoholic fatty liver disease(NAFLD),and to discuss the pathological relevance of the diagnostic criteria in metabolic(dysfunction)associated fatty liver disease(MAFLD).Methods:This was a multicenter,cross-sectional study.Patients with NAFLD confirmed by liver biopsy were enrolled between July 2016 and December 2018 from 14 centers across the mainland of China.Anthropometric and metabolic parameters were collected to assess the pathological relevance.Results:Of 246 enrolled patients with NAFLD,150(61.0%)had the comorbidity of Met S.With the increase of metabolic components,the proportions of nonalcoholic steatohepatitis(NASH)and significant fibrosis were notably increased.The comorbid three metabolic components significantly increased the proportion of NASH,and further increase of metabolic components did not increase the proportion of NASH.However,the increase of metabolic components was parallel to the increase of the proportion of liver fibrosis.Among the 246 patients,239(97.2%)met the diagnostic criteria of MAFLD.Although non-MAFLD patients had less NASH,they present with similar proportion of significant fibrosis and cirrhosis.In the diagnostic criteria of MAFLD,BMI≥23 kg/m2 was related to NASH(Mantel-Haenszel Common Estimate OR:2.975;95%CI:1.037–8.538;P=0.043),and T2 DM was related to significant fibrosis(Mantel-Haenszel Common Estimate OR:2.531;95%CI:1.388–4.613;P=0.002).The homeostasis model assessment of insulin resistance(HOMA-IR)≥2.5 was the most significant factor for NASH(OR:4.100;95%CI:1.772–9.487;P=0.001)and significant factor for liver fibrosis(OR:2.947;95%CI:1.398–6.210;P=0.004)after the adjustments of the BMI and diabetes.Conclusions:Metabolic dysregulations are important risk factors in NAFLD progression.The insulin resistance status may play a predominant role in the progression in MAFLD patients. | Rui-Xu Yang Zheng-Sheng Zou Bi-Hui Zhong Hong Deng Fang-Ping He Jun-Ping Shi Cai-Yan Zhao Yu-Qiang Mi Yong-Jian Zhou Fu-Sheng Di Rui-Dan Zheng Qin Du Jia Shang Branko Popovic Jin Jun Chen Jian-Gao Fan | 2021 | Hepatobiliary & Pancreatic Diseases International2021,20,5: | 5 |
| 8 | Mechanism of diarrhea in microscopic colitis显示文摘AIM: To search the pathophysiological mechanism of diarrhea based on daily stool weights, fecal electrolytes,osmotic gap and pH.METHODS: Seventy-six patients were included: 51 with microscopic colitis (MC) [40 with lymphocytic colitis (LC);11 with collagenous colitis (CC)]; 7 with MC without diarrhea and 18 as a control group (CG). They collected stool for 3 d. Sodium and potassium concentration were determined by flame photometry and chloride concentration by titration method of Schales. Fecal osmotic gap was calculated from the difference of osmolarity of fecal fluid and double sum of sodium and potassium concentration.RESULTS: Fecal fluid sodium concentration was significantly increased in LC 58.11±5.38 mmol/L (P<0.01)and CC 54.14±8.42 mmol/L (P<0.05) than in CG 34.28±2.98 mmol/L. Potassium concentration in LC 74.65±5.29 mmol/L (P<0.01) and CC 75.53±8.78 mmol/L (P<0.05) was significantly less compared to CG 92.67±2.99 mmol/L.Chloride concentration in CC 36.07±7.29 mmol/L was significantly higher than in CG 24.11±2.05 mmol/L (P<0.05).Forty-four (86.7%) patients had a secretory diarrhea compared to fecal osmotic gap. Seven (13.3%) patients had osmotic diarrhea.CONCLUSION: Diarrhea in MC mostly belongs to the secretory type. The major pathophysiological mechanism in LC could be explained by a decrease of active sodium absorption. | 12 | Multiscale Modeling and Simulations of Flows in Naturally Fractured Karst Reservoirs显示文摘Modeling and numerical simulations of fractured,vuggy,porus media is a challenging problem which occurs frequently in reservoir engineering.The problem is especially relevant in flow simulations of karst reservoirs where vugs and caves are embedded in a porous rock and are connected via fracture networks at multiple scales.In this paper we propose a unified approach to this problem by using the StokesBrinkman equations at the fine scale.These equations are capable of representing porous media such as rock as well as free flow regions(fractures,vugs,caves)in a single system of equations.We then consider upscaling these equations to a coarser scale.The cell problems,needed to compute coarse-scale permeability of Representative Element of Volume(REV)are discussed.A mixed finite element method is then used to solve the Stokes-Brinkman equation at the fine scale for a number of flow problems,representative for different types of vuggy reservoirs.Upscaling is also performed by numerical solutions of Stokes-Brinkman cell problems in selected REVs.Both isolated vugs in porous matrix as well as vugs connected by fracture networks are analyzed by comparing fine-scale and coarse-scale flow fields.Several different types of fracture networks,representative of short-and long-range fractures are studied numerically.It is also shown that the Stokes-Brinkman equations can naturally be used to model additional physical effects pertaining to vugular media such as partial fracture with fill-in by some material and/or fluids with suspended solid particles. | Peter Popov Yalchin Efendiev Guan Qin | 2009 | Communications in Computational Physics2009,6,6: | 4 |
| 13 | Biochemical markers for non-invasive assessment of disease stage in patients with primary biliary cirrhosis显示文摘AIM: To evaluate different biochemical markers and their ratios in the assessment of primary biliary cirrhosis (PBC) stages. METHODS: This study included 112 patients with PBC who underwent a complete clinical investigation. We analyzed the correlation (Spearman's test) between ten biochemical markers and their ratios with different stages of PBC. The discriminative values were compared using areas under receiver operating characteristic (ROC) curves. RESULTS: The mean age of patients included in the study was 53.88 ± 10.59 years, including 104 females and 8 males. We found a statistically significant correlation between PBC stage and Aspartate aminotransferase (AST), Alanine aminotransferase (ALT) to platelet ratio (APRI), ALT/platelet count, AST/ALT, ALT/AST and ALT/Cholesterol ratios, with the values of Spearman's rho of 0.338, 0.476, 0.404, 0.356, 0.351 and 0.325, respectively. The best sensitivity and specificity was shown for AST/ALT, with an area under ROC of 0.660. CONCLUSION: Biochemical markers and their ratios do correlate with different sensitivity to and specificity of PBC disease stage. | 15 | Fecal microbial transplant for the treatment of pediatric inflammatory bowel disease显示文摘The role of fecal microbial transplant(FMT) in the treatment of pediatric gastrointestinal disease has become increasingly popular among pediatric practitioners, patients, and parents. The success of FMT for the treatment of recurrent Clostridium difficile infection(RCDI) has bolstered interest in its potential application to other disease states, such as inflammatory bowel disease(IBD). FMT has particular interest in pediatrics, given the concerns of patients and parents about rates of adverse events with existing therapeutic options, and the greater cumulative medication burden associated with childhoodonset disease. Published literature on the use of FMT in pediatrics is sparse. Only 45 pediatric patients treated for RCDI have been reported, and only 27 pediatric patients with pediatric IBD. The pediatric microbiome may uniquely respond to microbial-based therapies. This review will provide a comprehensive overview of fecal microbial transplant and its potential role in the treatment of pediatric inflammatory bowel disease. We will discuss the microbiome in pediatric inflammatory bowel disease, existing adult and pediatric literature on the use of FMT in IBD treatment, and pediatric FMT trials that are currently recruiting patients. | 17 | Subchronic Oral Cadmium Exposure Exerts both Stimulatory and Suppressive Effects on Pulmonary Inflammation/Immune Reactivity in Rats显示文摘Objective The aim of this study is to investigate the effects of oral cadmium(Cd) ingestion on the pulmonary immune response. Methods Determination of Cd content in lungs and histopathological evaluation of the tissue was performed in rats following 30-day oral Cd administration(5 and 50 mg/L). Antioxidant enzyme defense(superoxide dismutase and catalase), cell infiltration, and production of tumor necrosis factor(TNF) and interferon(IFN)-γ, as well as the activity of myeloperoxidase(MPO), nitric oxide(NO), and various cytokines [interleukin(IL)-1β, IL-6, IL-10, and IL-17] were investigated. Results Cd caused tissue damage and cell infiltration in the lungs, and this damage was more pronounced at higher doses. Cd deposition resulted in lung inflammation characterized by a dose-dependent IL-1β increase in lung homogenates, increased TNF levels at both doses, and IL-6 stimulation at low doses with inhibition observed at higher doses. Cd exerted differential effects on lung leukocytes isolated by enzyme digestion, and these effects were characterized by a lack of change in the production of reactive oxygen and nitrogen species, an inhibition of IL-1β and TNF, and stimulation of MPO and IFN-γ. The higher capacity of Cd-exposed lung cells to respond to the opportunistic pathogen Staphylococcus epidermidis was demonstrated in vitro. | 20 | Unique Marine Olenekian-Anisian Boundary Section from South Primorye, Russian Far East显示文摘To show paleontological characteristics of the Olenekian-Anisian boundary beds in the Russian Far East, a review of new data on the Upper Olenekian and Lower Anisian biostratigraphy of South Primorye is given on the basis of five sections: Golyi Cape, Petrovka River, Zhitkov Peninsula, Tchernyschew Bay and Atlasov Cape, using new ammonoid, brachiopod and conodont findings. The most representative ammonoid assemblage at the base of the Anisian was discovered in the Ussuriphyllites amurensis Zone (10.6 m thick) of the Atlasov Cape Section: Parasageceras sp. nov., Prionitidae gen. et sp. nov., Ussuriphyllites amurensis (Kiparisova) (dominant), Megaphyllites atlasoviensis Zakharov, Leiophyllites praematurus Kiparisova, Leiophyllites sp., Ussurites sp., Paradanubites sp. indet., Paracrochordiceras sp. nov., Prohungarites popowi Kiparisova, Arctohungarites primoriensis Zakharov, A. solimani (Toula), Salterites sp. indet. (gigantic shell), and Tropigastrites sublachontanus Zakharov. Conodonts Neospathodus cf. homeri (Bender) were found in the lower part of the Ussuriphyllites amurensis Zone of the Atlasov Cape.
