1Breeding better cultivars,faster:applications of new technologies for the rapid deployment of superior horticultural tree crops显示文摘Woody perennial plants,including trees that produce fruits and nuts of horticultural value,typically have long breeding cycles,and development and introduction of improved cultivars by plant breeders may require many breeding cycles and dozens of years.However,recent advances in biotechnologies and genomics have the potential to accelerate cultivar development greatly in all crops.This mini-review summarizes approaches to reduce the number and the duration of breeding cycles for horticultural tree crops,and outlines the challenges that remain to implement these into efficient breeding pipelines.Steve van Nocker Susan E Gardiner 2014Horticulture Research2014,1,1:19
2Transcriptional dynamics of the developing sweet cherry(Prunus avium L.)fruit:sequencing,annotation and expression profiling of exocarp-associated genes显示文摘The exocarp,or skin,of fleshy fruit is a specialized tissue that protects the fruit,attracts seed dispersing fruit eaters,and has large economical relevance for fruit quality.Development of the exocarp involves regulated activities of many genes.This research analyzed global gene expression in the exocarp of developing sweet cherry(Prunus avium L.,‘Regina’),a fruit crop species with little public genomic resources.A catalog of transcript models(contigs)representing expressed genes was constructed from de novo assembled short complementary DNA(cDNA)sequences generated from developing fruit between flowering and maturity at 14 time points.Expression levels in each sample were estimated for 34695 contigs from numbers of reads mapping to each contig.Contigs were annotated functionally based on BLAST,gene ontology and InterProScan analyses.Coregulated genes were detected using partitional clustering of expression patterns.The results are discussed with emphasis on genes putatively involved in cuticle deposition,cell wall metabolism and sugar transport.The high temporal resolution of the expression patterns presented here reveals finely tuned developmental specialization of individual members of gene families.Moreover,the de novo assembled sweet cherry fruit transcriptome with 7760 full-length protein coding sequences and over 20000 other,annotated cDNA sequences together with their developmental expression patterns is expected to accelerate molecular research on this important tree fruit crop.Merianne Alkio Uwe Jonas Myriam Declercq Steven Van Nocker Moritz Knoche 2014Horticulture Research2014,1,1:4
3Apple whole genome sequences:recent advances and new prospects显示文摘In 2010,a major scientific milestone was achieved for tree fruit crops:publication of the first draft whole genome sequence(WGS)for apple(Malus domestica).This WGS,v1.0,was valuable as the initial reference for sequence information,fine mapping,gene discovery,variant discovery,and tool development.A new,high quality apple WGS,GDDH13 v1.1,was released in 2017 and now serves as the reference genome for apple.Over the past decade,these apple WGSs have had an enormous impact on our understanding of apple biological functioning,trait physiology and inheritance,leading to practical applications for improving this highly valued crop.Causal gene identities for phenotypes of fundamental and practical interest can today be discovered much more rapidly.Genome-wide polymorphisms at high genetic resolution are screened efficiently over hundreds to thousands of individuals with new insights into genetic relationships and pedigrees.High-density genetic maps are constructed efficiently and quantitative trait loci for valuable traits are readily associated with positional candidate genes and/or converted into diagnostic tests for breeders.We understand the species,geographical,and genomic origins of domesticated apple more precisely,as well as its relationship to wild relatives.The WGS has turbo-charged application of these classical research steps to crop improvement and drives innovative methods to achieve more durable,environmentally sound,productive,and consumer-desirable apple production.This review includes examples of basic and practical breakthroughs and challenges in using the apple WGSs.Recommendations for“what’s next”focus on necessary upgrades to the genome sequence data pool,as well as for use of the data,to reach new frontiers in genomics-based scientific understanding of apple.Cameron P.Peace Luca Bianco Michela Troggio Eric van de Weg Nicholas P.Howard Amandine Cornille Charles-Eric Durel Sean Myles ZoëMigicovsky Robert J.Schaffer Evelyne Costes Gennaro Fazio Hisayo Yamane Steve van Nocker Chris Gottschalk Fabrizio Costa David Chagné Xinzhong Zhang Andrea Patocchi Susan E.Gardiner Craig Hardner Satish Kumar Francois Laurens Etienne Bucher Dorrie Main Sook Jung Stijn Vanderzande 2019Horticulture Research2019,6,1:3
4Physiological evaluation of nitrogen use efficiency of different apple cultivars under various nitrogen and water supply conditions显示文摘Nitrogen(N) deficiency is a common problem for apple(Malus×domestica) production in arid regions of China.However,N utilization efficiency(NUE) of different apple cultivars grown under low N conditions in arid regions has not been evaluated.In this study,NUE was assessed for one-year-old seedlings of six apple cultivars,Golden Delicious,Qinguan,Jonagold,Honeycrisp,Fuji and Pink Lady,grafted onto Malus hupehensis Rehd.rootstocks.Four treatments were used,including control water with control N(CWCN),limited water with control N(LWCN),control water with low N(CWLN) and limited water with low N(LWLN).Our results showed that growth indices such as biomass,plant height and stem diameter,and photosynthetic rate of all cultivars decreased in the order CWCN>CWLN>LWCN>LWLN.When subjected to LWLN treatment,Qinguan showed better growth and photosynthetic characters than other tested cultivars.Additionally,Qinguan and Pink Lady had higher NUE,while Honeycrisp and Jonagold had lower NUE,based on the determination of biomass,photosynthetic parameters,chlorophyll content,the maximal photochemical efficiency of PSII(Fv/Fm),15 N and N contents.WANG Qian LIU Chang-hai HUANG Dong DONG Qing-long LI Peng-min Steve van NOCKER MA Feng-wang 2020Journal of Integrative Agriculture2020,19,3:3
