2Combined hepatic resection with fenestration for highly symptomatic polycystic liver disease:A report on seven patients显示文摘AIM: To evaluate the immediate and long-term results in a series of patients with highly symptomatic polycystic liver disease (PLD) treated by combined hepatic resection with cystic fenestration.METHODS: We reviewed our recent experience with a combined hepatic resection-fenestration procedure in seven highly symptomatic patients with PLD. Clinical data, liver manifestation of computed tomography (CT), and morbidity were recorded pre- and post-operation. Follow-up was made by clinical and CT examinations in all patients.RESULTS: Symptomatic relief and reduction in abdominal girth were obtained in all patients during an average followup period of 20.4 mo. CT scans confirmed post-resection hypertrophy of the spared liver and lack of significant cyst progression. All patients had mild to severe ascites. Two patients were complicated with pleural effusion.CONCLUSION: Some highly symptomatic patients with massive PLD may benefit from combined hepatic resection and fenestration at acceptable risk. 4Design and synthesis of novel water-soluble amino acid derivatives of chlorin p6 ethers as photosensitizer显示文摘Eight new water-soluble amino acid conjugates 6 a-h of chlorin p6 ethers(5 a-d) were synthesized and preliminarily investigated for their in vitro PDT antitumor activity and structure-activity relationship(SAR). The results showed that all compounds exhibited much higher phototoxicity against tumor cells than talaporfin. SAR analysis indicated that PDT antitumor effect enhanced with the increase of carbon chain length of alkoxyl ether bonds at 3~1-position, and L-aspartic acid was superior to L-glutamic acid. 5Differential CI^-and HCO_3^- mediated anion secretion by different colonic cell types in response to tetromethylpyrazine显示文摘AIM: Colonic epithelium is known to secrete both CI- and HCO3, but the secretory mechanisms of different colonic cell types are not fully understood. The present study aimed to investigate the differential activation of Cl-and HCO3 secretion by tetramethylpyrazine (TMP) in human crypt-like cell line, T84, and villus-like cell line, Caco-2, in comparison to the TMP-induced secretory response in freshly isolated rat colonic mucosa. METHODS: Colonic epithelial anion secretion was studied by using the short circuit current (Isc) technique. RT-PCR was used to examine the expression of Na^+-HCO3-cotranspoter in different epithelial cell types. RESULTS: TMP produced a concentration-dependent Isc which was increase in both T84 and Caco-2 cells. When extracellular CI was removed, TMP-induced Isc was abolished by 76.6% in T84 cells, but not in Caco-2 cells. However, after both CI and HCO3- were removed, TMP-induced Isc in Caco-2 cells was reduced to 10%. Bumetanide, an inhibitor of Na^+-K^+-Cl-cotranspoter, inhibited the TMP-induced Isc by 96.7% in T84 cells, but only 47.9% in Caco-2 cells. In the presence of bumetanide and 4, 4'-diisothiocyanostilbene-2, 2'-disulfonic acid, an inhibitor of Na^+-HCO3- cotransporter, inhibited the TMP-induced current in Caco-2 cells by 93.3% .In freshly isolated rat colonic mucosa, TMP stimulated distinct Isc responses similar to that observed in T84 and Caco-2 cells depending on the concentration used. RT-PCR revealed that the expression of Na^+-HCO3- cotransporter in Caco-2 cells was 4-fold more greater than that in T84 cells. 6CpG ODN Enhances Immunization Effects of Hepatitis B Vaccine in Aged Mice显示文摘Oligodeoxynucleotides (ODN) containing unmethylated CpG dinucleotides in contexts of unique sequence (CpG motifs) is active as adjuvant in induction of cellular and humoral immune responses in young mice. To date, there are only limited reports about effect of CpG ODN on immune responses against hepatitis B (HB) infection in aged mice. Our studies demonstrated there were significant increases in secreting of total anti-HB IgG, IgG1 and IgG2a, as well as of IL-12 and IFN-γ, when CpG ODNs were injected together with hepatitis B antigen in aged mice. Moreover, CpG ODN could stimulate proliferation of spleen lymphocytes in a dose-dependent manner. 7Improvement of barrier function and stimulation of colonic epithelial anion secretion by Menoease Pills显示文摘AIM: Menoease Pills(MP)1 a Chinese medicine-based new formula for postmenopausal women, has been shown to modulate the endocrine and immune systems. The present study investigated the effects of MP and one of its active ingredients, ligustrazine, on epithelial barrier and ion transport function in a human colonic cell line, T84.METHODS: Colonic transepithelial electrophysiological characteristics and colonic anion secretion were studied using the short circuit current (Isc) technique. RT-PCR was used to examine the expression of cytoplasmic proteins associated with the tight junctions, ZO-l(zonula occludens-1) and ZO-2 (zonula occludens-2).RESULTS: Pretreatment of T84 cells with MP (15 μg/mL) for 72 h significantly increased basal potential difference,transepithelial resistance and basal ISC. RT-PCR results showed that the expressions of ZO-land ZO-2 were significantly increased after MP treatment, consistent with improved epithelial barrier function. Results of acute stimulation showed that apical addition of MP produced a concentrationdependent (10-5 000 μg/mL, EC50 = 293.9 μg/mL) increase in ISC. MP-induced ISC was inhibited by basolateral treatment with bumetanide (100 μmol/L), an inhibitor of the Na^+-K^+-2Cl^- cotransporter, apical addition of Cl^- channel blockers, diphenylamine-2, 2'-dicarboxylic acid (1mmol/L) or glibenclamide (1 retool/L), but not 4, 4'-diisothiocyanostilbene-2, 2'-disulfonic acid or epithelial Na+ channel blocker,amiloride. The effect of MP on ZOo1 and ZO-2was mimicked by Ligustrazine and the ligustrazine-induced ISC was also blocked by basolateral application of bumetanide and apical addition of diphenylamine-2, 2'-dicarboxylic acid or glibenclamide, and reduced by a removal of extracellular Cl^-.CONCLUSION: The results of the present study suggest that MP and lligustrazine may improve epithelial barrier function and exert a stimulatory effect on colonic anion secretion, indicating the potential use of MP and its active ingredients for improvement of GI tract host defense and alleviation of constipation often seen in the elderly.Jin-XiaZhu NingYang Gui-HongZhang Lai-UngTsang Yu-LinGou Hau-YanConnieWong Yiu-WaChung Hsiao-ChangChan 2004World Journal of Gastroenterology2004,10,17:2

10MODELING OF MESO-SCALE STRUCTURES IN PARTICLE-FLUID SYSTEMS: THE EMMS/CFD APPROACH显示文摘Meso-scale structures existing in the form of particle-rich clusters, streamers or strands in circulating fluidized beds, and of ascending bubble plumes and descending liquid-rich vortices in bubble columns and slurry-bed reactors, as commonly observed, have played an important role in the macro-scale behavior of particle-fluid systems. These meso-scale structures span a wide range of length and time scales, and their origin, evolution and influence are still far from being well understood. Recent decades have witnessed the emergence of computer simulation of particle-fluid systems based on computa-ional fluid dynamic (CFD) models. However, strictly speaking these models are far from mature and the complex nature of particle-fluid systems arising from the meso-scale structures has been posing great challenges to investigators. The reason may be that the current two-fluid models (TFM) are derived either from continuum mechanics by using different kinds of averaging techniques for the conservation equations of single-phase flow, or from the kinetic theory of gases in which the assumption of molecular chaos is employed, thereby losing sight of the meso-scale heterogeneity at the scale of computational cells and leading to inaccurate calculation of the interaction force between particles and fluids. For example, the overall drag force for particles in a cell is usually calculated from the empirical Wen & Yu/Ergun correlations,which should be suspected since these correlations were originally derived from homogeneous systems. Schemes to solve this problem for gas-particles systems may be classified into four categories. First, one could capture the detailed meso-scale structure information at the cell scale by employing the so-called direct numerical simulation (DNS) (Hu, 1996), the pseudo-particle modeling (PPM) (Ge & Li, 2003), or the Lattice-Boltzmann method (LBM) to track the interface between gas and particles. Second, refinement of the computational meshes may reduce the heterogeneity to some extent and may be capable of capturing some meso-scale heterogeneity though there still exists some argument about the physical rationality'of this approach such as the treatment of particle phase as a continuum while fining the meshes. Third, it is generally agreed that a cascade description, viz. extracting the closure correlations for TFM from microscopic simulations such as PPM and LBM (van der Hoef et al., 2004), can suggest a practical way to explore the multi-scale heterogeneity. Although the above three schemes are logical and fundamental, they are generally difficult to implement at present due to the complexity of the models or the enormous computational cost. The fourth scheme we adopted in this study is the so-called energy-minimization multi-scale (EMMS) model which seems to be a simple yet reasonable approach at the moment. In the present approach, a “structure” model is established to describe the meso-scale heterogeneity through the definition of eight“structure parameters” and the resolution of structure involving a particle-rich dense cluster phase and a gas-rich dilute phase. Gas-solid interaction is also resolved into that between gas and particles inside both the dense cluster phase and the dilute phase, and that between the cluster phase and the dilute phase. This means that the drag force for the dense cluster phase includes two parts, namely, bypassing drag (ki) and permeating drag (kc) as depicted in Fig.1. We found that the absolute value of the difference (Ak) between kc and ki could be employed to evaluate the extent of the system heterogeneity. On the basis of this structure model, the average acceleration (a) induced by gas-solid interactions can be obtained, and then the average drag coefficient (β) for the two-fluid model can be calculated. Calculation results show that the computed value of β with the EMMS model is much less than that with the Wen & Yu/Ergun correlations, which is in reasonable agreement with conclusions derived from experiments. We further simplified this model by assuming that the dense phase voidage (εc) is a constant because the direct incorporation of this new model into the two-fluid model is difficult at present due to computational cost resulting from the iterative process and certain limitations of this model stemming from its original derivation from the global fluidized bed system. With the Ug and Gs for the global system as the input parameters, we can calculate β for each specified ε and thereby obtain the correlation of correction factor (ω(ε) as a function of ε. Implanting this correlation into each cell and employing the local slip velocity and voidage,β for each cell can thus be obtained. The simulation is performed for a FCC riser by combining the two-fluid model with this EMMS-based drag model and the Wen & Yu/Ergun correlations respectively. Comparison of the simulation results shows that the EMMS-based drag model produces more reasonable results than the Wen & Yu/Ergun correlations. The former shows its improvement in predicting the solids entrainment rate, the meso-scale heterogeneous structure involving clusters or strands, and the radial and axial voidage distributions. For the latter, the simulated flow structure is homogeneous and no clusters are observed, and the predicted solid entrainment rate is too much larger than experimental measurements. We also employ different approaches to predict the occurrence of choking, indicating that this EMMS-based drag model has the ability to capture this important phenomenon in CFB systems. 11Semiotic Resourcefulness in Crisis Risk Communication: The Case of COVID-19 Posters显示文摘The unprecedented outbreak of COVID-19 presents a public health crisis on a global scale.Various measures have been taken to communicate crisis risks to the general public.These measures are meant to keep the public well informed,stay alert,and take precautionary measures to help curb the spread of the virus.The current study is part of an ongoing project aimed at exploring patterns of communication in the COVID-19 crisis discourse.Based on a collection of posters designed for public use during the outbreak,this paper analyses the richness of semiotic resources that combine to construct and convey the intended message of the posters.Drawing from scholarly insights into understanding the situatedness of meaning-making,the paper revisits some of the classical concerns about the relationship between text and image in semiotic artefacts and reveals the meaningmaking patterns in the semiotic designs of risk communication posters.The patterns are found to rest upon a host of textual and graphic features that contribute to the essential semiotic encoding of entity,condition,action,and sentiment.The findings are summarized by conceptualizing the assemblages of resources in the poster as a semiotic ensemble where the coordination and collaboration among semiotic resources can work to reduce potential ambiguities and amplify the communicative effect.Ningyang Chen 2020Language and Semiotic Studies2020,6,3:1

14Disseminated tumor cells homing into rats' liver:A new possible mechanism of HCC recurrence显示文摘AIM:To detect the origin of hepatocellular carcinoma (HCC)recurring and attempt to propose a new recurrent mechanism.METHODS:Orthotopic liver allotransplantation was performed on male rats with HCC- induced by diethylnitrosamine using female donors.Metastatic tumors in transplanted livers were obtained.A DNA probe that exhibits specificity for the rat Y chromosome was generated by using a set of primers specificto murine srygene.In situ hybridization (ISH) for Y chromosome was used to detected the origin of HCC recurring. Male HCC tissue was designed to be positive control. ISH on female tissue and using non-labeled with DIG probe was thought to be negative control.RESULTS: Positive marks were seen through ISH for Y chromosome in recurrent tumor tissue and positive control.No signal was detected in both negative controls.CONCLUSION: Recurrent HCC after liver transplantation originated from disseminated tumor cells in recipients.Extrahepatic cells homing into liver may be a new HCC recurrence mechanism. 16Metabolic engineering of Bacillus amyloliquefaciens for efficient production ofα-glucosidase inhibitor1-deoxynojirimycin显示文摘Owing to the feature of strongα-glucosidase inhibitory activity,1-deoxynojirimycin(1-DNJ)has broad application prospects in areas of functional food,biomedicine,etc.,and this research wants to construct an efficient strain for 1-DNJ production,basing on Bacillus amyloliquefaciens HZ-12.Firstly,using the temperature-sensitive shuttle plasmid T2(2)-Ori,gene ptsG in phosphotransferase system(PTS)was weakened by homologous recombination,and non-PTS pathway was strengthened by deleting its repressor gene iolR,and 1-DNJ yield of resultant strain HZ-S2 was increased by 4.27-fold,reached 110.72 mg/L.Then,to increase precursor fructose-6-phosphate(F-6-P)supply,phosphofructokinase was weaken,fructose phosphatase GlpX and 6-phosphate glucose isomerase Pgi were strengthened by promoter replacement,moreover,regulator gene nanR was deleted,1-DNJ yield was further increased to 267.37 mg/L by 2.41-fold.Subsequently,promoter of 1-DNJ synthetase cluster was optimized,as well as 5′-UTRs of downstream genes in synthetase cluster,and 1-DNJ produced by the final strain reached 478.62 mg/L.Last but not the least,1-DNJ yield of 1632.50 mg/L was attained in 3 L fermenter,which was the highest yield of 1-DNJ reported to date.Taken together,our results demonstrated that metabolic engineering was an effective strategy for 1-DNJ synthesis,this research laid a foundation for industrialization of functional food and drugs based on 1-DNJ.Xujie Li Meng Zhang Yu Lu Ningyang Wu Jian’gang Chen Zhixia Ji Yangyang Zhan Xin Ma Junyong Chen Dongbo Cai Shouwen Chen 2023Synthetic and Systems Biotechnology2023,8,3:0

20Study of Ni80Fe20/Fe50Mn50 superlattice microstructures by transmission electron microscopy and x—ray diffraction显示文摘The detailed microstructures of Ni80Fe20/Fe50Mn50 superlattices have been characterized using both x-ray diffraction techniques and transmission electron microscopy.The obrivous layered structure,typical column structure and twins which exist in Ni80Fe20/Fe50Mn50 superlattices were observed through performing transmission microscopy.By combining the technique of lowangle x-ray reflectivity(specular and off-specular scans)with the anomalous scattering effect and high-angle x-ray diffraction(using conventional x-ray),wequantitatively analysed the microstructural variation as a function of annealing temperature.It is found that the lateral correlation length,the(111)peak intensity of the superlattices and the average multilayer coherence length all increase with a rise in annealing temperature annealing can decrease the rootmean-square roughness at the interfaces of Ni80Fe20/Fe50Mn50 superlattices.the obtained microstructural knowledge will be helpful in understanding the magnetic properties of the ni80Fe20/Fe50Mn50 exchange bias system.MingXu TaoYang GuangmingLuo ZhengqiLu CuixiuLiu NingYang Zhenh 2001Beijing Synchrotron Radiation Facility2001,,2:0
