| 1 | Effects of stem structure andcell wall components on bending strength in wheat显示文摘Morphological traits, anatomical features, chemical components and bending stress in the stems of three genotypes of wheat (Triticum aestivum L.), namely Xiaoyan54, 8602 and Xiaoyan81, were ex- amined by means of light microscopy coupled with Fourier transform infrared spectroscopy (FTIR). No- ticeable changes in morphological and anatomical traits were observed, including outer radius of stem, the ratio of stem outer radius to stem wall thickness, various tissue proportions and variations among dif- ferent types of vascular bundles. The results of chemical analysis revealed that Xiaoyan81 had the highest cellulose content in comparison with Xiao- yan54 and 8602, whereas lignin level in Xiaoyan81 was lower than that in 8602 but higher that that in Xiaoyan54. Bending stress analysis demonstrated that Xiaoyan81 may be the main target for identifica- tion, for it had the highest bending stress among the stems of three genotypes. Associated with bending stress, all the results presented here suggested that the ratio of stem wall thickness to its outer radius, schlerenchyma tissue proportion, the average num- ber of big VB per unit and the cellulose content are four important factors affecting the mechanical strength of Xiaoyan81 wheat stems, which can be considered as the key parameters for selecting varie- ties with bending stress. | 2 | Cytological and Molecular Characteristics of Pollen Abortion in Lily with Dysplastic Tapetum显示文摘Lily was grown worldwide as a fresh cutting flower because of its colorful petals, but its anther contained a large number of pollen grains that cause serious pollen contamination, however, pollen abortion can effectively reduce the level of pollen pollution. Our analysis aims to use cytological observation to detect the critical stage when pollen abortion occurs and to provide comprehensive gene expression information at the transcriptional level. The result showed that pollen abortion in ‘Little Kiss’ began at the mononuclear stage and the callose that covers the microspores failed to degenerate when young pollens were released from the tetrads. In addition, compared with the normally developed one,the tapetum of ‘Little Kiss’ degraded in advance while the degradation of callose was delayed. Furthermore, 103 differentially expressed genes(DEGs) related to the advance degeneration of tapetum cells and callose were found in the expression levels, including 22 transcription factors(TFs). In particular, two β-glucanase genes(endo-1,3(4)-β-glucanase, exo-β-glucanase) responsible for callose degeneration were significantly down-regulated. These results suggested that pollen abortion may occur at mononuclear stage and that early degeneration of tapetum cells resulted in a significant down-regulation of β-glucanase genes. | 4 | Photocatalytic and Electrocatalytic Generation of Hydrogen Peroxide: Principles, Catalyst Design and Performance显示文摘Hydrogen peroxide(H_(2)O_(2)) is a high-demand organic chemical reagent and has been widely used in various modern industrial applications. Currently,the prominent method for the preparation of H_(2)O_(2) is the anthraquinone oxidation.Unfortunately, it is not conducive to economic and sustainable development since it is a complex process and involves unfriendly environment and potential hazards. In this context, numerous approaches have been developed to synthesize H_(2)O_(2). Among them, photo/electro-catalytic ones are considered as two of the most promising manners for on-site synthesis of H_(2)O_(2). These alternatives are sustainable in that only water or O_(2) is required. Namely, water oxidation(WOR) or oxygen reduction(ORR)reactions can be further coupled with clean and sustainable energy. For photo/electro-catalytic reactions for H_(2)O_(2) generation, the design of the catalysts is extremely important and has been extensively conducted with an aim to obtain ultimate catalytic performance. This article overviews the basic principles of WOR and ORR,followed by the summary of recent progresses and achievements on the design and performance of various photo/electro-catalysts for H_(2)O_(2) generation. The related mechanisms for these approaches are highlighted from theoretical and experimental aspects. | 6 | A new approach to promote astaxanthin accumulation via Na_2WO_4 in Haematococcus pluvialis显示文摘The optimum concentration of Na_2 WO_4 was explored in relation to the cell density and astaxanthin content in Haematococcus pluvialis. Then, the cellular morphology, nitrate reductase(NR) activity, soluble sugar and protein contents, and chlorophyll ?uorescence were measured, and the transcriptional expression of carotenogenic genes was determined by quantitative real-time PCR. The results showed that 3.0 mmol/L of Na_2 WO_4 was the optimum concentration to induce astaxanthin accumulation, with a maximum content of 49.41±0.13 pg/cell reached on the tenth day. The NR activity decreased signi?cantly and continually after Na_2 WO_4 treatment. The soluble sugar content increased gradually during the experimental period and was eventually signi?cantly higher than that in the control. The soluble protein content increased rapidly,reached a maximum in day 0.5 and day 1 and then decreased. The ef fective photochemical effciency of PSII( F v'/F m') and light saturation( E k) ?rst decreased and then tended to stabilize, and NADP +-glyceraldehyde-3-phosphate dehydrogenase(GAPDH) gene expression was correlated with photosynthesis. The transcriptional expression of ipi, psy and bkt was signi?cantly increased compared with that in the control after application of Na_2 WO_4, and the relative expression of ipi reached the highest level on the ?fth day, with a 98.03±1.92-fold increase. | 7 | Changes in Growth, Carbon and Nitrogen Enzyme Activity and mRNA Accumulation in the Halophilic Microalga Dunaliella viridis in Response to NaCl Stress显示文摘Many species of microalga Dunaliella exhibit a remarkable tolerance to salinity and are therefore ideal for probing the effects of salinity. In this work, we assessed the effects of NaCl stress on the growth, activity and m RNA level of carbon and nitrogen metabolism enzymes of D. viridis. The alga could grow over a salinity range of 0.44 mol L^(-1) to 3.00 mol L^(-1) NaCl, but the most rapid growth was observed at 1.00 mol L^(-1) NaCl, followed by 2.00 mol L^(-1) NaCl. Paralleling these growth patterns, the highest initial and total Rubisco activities were detected in the presence of 1.00 mol L^(-1) NaCl, decreasing to 37.33% and 26.39% of those values, respectively, in the presence of 3.00 mol L^(-1) Na Cl, respectively. However, the highest extracellular carbonic anhydrase(CA) activity was measured in the presence of 2.00 mol L^(-1) NaCl, followed by 1.00 mol L^(-1) NaCl. Different from the two carbon enzymes, nitrate reductase(NR) activity showed a slight change under different NaCl concentrations. At the transcriptional level, the m RNAs of Rubisco large subunit(rbcL), and small subunit(rbc S), attained their highest abundances in the presence of 1.00 and 2.00 mol L^(-1)NaCl, respectively. The CA mRNA accumulation was induced from 0.44 mol L^(-1) to 3.00 mol L^(-1) NaCl, but the NR m RNA showed the decreasing tendency with the increasing salinity. | 16 | Species Composition and Diversity of Macrobenthos in the Intertidal Zone of Xiangshan Bay,China显示文摘Xiangshan bay is a narrow semi-closed bay and situated on the northwestern coast of the East China Sea. Over past decades, it has become to a major bay with intensive human activities, dense urbanized area, and poor water quality. The aim of this paper was to reveal the ecological status through the elucidation of the species composition, abundance, biomass and diversity of macrobenthos in this bay. Six intertidal sections were surveyed from January 2007 to November 2008 quarterly. Sections TG, HD and XH are located in the three inner bays, sections QJ and WS are located near the thermal power plants, and section XX is located at the outer part of Xiangshan Bay. Great variations in macrobenthos community were indentified, and the species composition of the community in the present study showed the dominance in the order of molluscs(bivalves and gastropods), crustaceans and others, and only few Polychaeta were recorded. Only three dominant species, Littorina brevicula, Ilyplax tansuiensis, and Cerithidea cingulata were collected in all the sections, and a total of 19 dominant species were recorded only in one section. Two-way ANOVA analyses of abundance indicated that there were significant differences among sections or seasons. Shannon-Wiener diversity index(H') had its maximum(2.45) in section QJ, and minimum(1.76) in section TG. Multiple irregular k-dominance plots clearly showed that the study area was polluted and the macrobenthos community was under stress.
