| 5 | Lamivudine resistance mutations in patients infected with hepatitis B virus genotype D显示文摘AIM: To determine the distribution of viral genotypes for primary or acquired lamivudine resistance. METHODS: A total of 283 patients with chronic hepatitis B virus (HBV) infection (245 patients with chronic hepatitis B and 38 inactive hepatitis B surface antigen carriers) were included in the study. The HBV geno-type was determined by using quantitative real-time polymerase chain reaction and sequence analysis, and tyrosine-methionine-aspartate-aspartate (YMDD) motif mutations were determined using the reverse transcriptase hybridization method. RESULTS: Lamivudine resistance was determined in a total of 25 (10.7%) chronic hepatitis B patients. Eight subjects (4%) had primary resistance to lamivudine, and 17 (53.1%) had secondary resistance to lamivudine. Genotype D, which was isolated from 267 of the patients with chronic HBV infection, was the dominant genotype in Turkey. | 6 | Laser Remelting of Plasma Sprayed NiCrAlY and NiCrAlY-Al_2O_3 Coatings显示文摘Two types of plasma sprayed coatings (NiCrAlY and NiCrAlY-A12O3) were remelted by a 5 kW cw CO2 laser. With increasing laser power and decreasing traverse speed in the ranges of 200-700 W and 5-30 mm/s respectively, the melted track grew in width and depth. In the optimum range of laser parameters, a homogeneous remelted layer without voids, cavities, unmelted particles and microcracks was formed. On the surface of remelted layers, Al203 and YAIO3 were detected. As a result of isothermal oxidation tests, weight gains of laser remelted coatings were obviously lower than that only plasma sprayed, especially laser remelted NiCrAlY-Al2O3 coatings. | 8 | Agrobacterium-mediated transformation and assessment of factors influ-encing transgene expression in loblolly pine (Pinus taeda L.)显示文摘This investigation reports a protocol for transfer and expression of foreign chimeric genes in loblolly pine (Pinus taeda L.). Transformation was achieved by co-cultivation of mature zygotic embryos with Agrobacterium tumefaciens strain LBA4404 which harbored a binary vector (pBI121) including genes for β-glucuronidase (GUS) and neomycin phosphotransferase (NPTII). Factors influencing transgene expression including seed sources of loblolly pine, concentration of bacteria, and the wounding procedures of target explants were investigated. The expression of foreign gene was confirmed by the ability of mature zygotic embryos to produce calli in the presence of kanamycin, by histochemical assays of GUS activity, by PCR analysis, and by Southern blot.
