| 1 | Determination of Arctic melt pond fraction and sea ice roughness from Unmanned Aerial Vehicle (UAV) imagery显示文摘Melt ponds on Arctic sea ice are of great significance in the study of the heat balance in the ocean mixed layer, mass and salt balances of Arctic sea ice, and other aspects of the earth-atmosphere system. During the 7 th Chinese National Arctic Research Expedition, aerial photographs were taken from an Unmanned Aerial Vehicle over an ice floe in the Canada Basin. Using threshold discrimination and three-dimensional modeling, we estimated a melt pond fraction of 1.63% and a regionally averaged surface roughness of 0.12 for the study area. In view of the particularly foggy environment of the Arctic, aerial images were defogged using an improved dark channel prior based image defog algorithm, especially adapted for the special conditions of sea ice images. An aerial photo mosaic was generated, melt ponds were identified from the mosaic image and melt pond fractions were calculated. Three-dimensional modeling techniques were used to generate a digital elevation model allowing relative elevation and roughness of the sea ice surface to be estimated. | 8 | Putative functions of EpsK in teichuronic acid synthesis and phosphate starvation in Bacillus licheniformis显示文摘Extracellular polymeric substances(EPSs)are extracellular macromolecules in bacteria,which function in cell growth and show potential for mechanism study and biosynthesis application.However,the biosynthesis mechanism of EPS is still not clear.We herein chose Bacillus licheniformis CGMCC 2876 as a target strain to investigate the EPS biosynthesis.epsK,a member of eps cluster,the predicted polysaccharide synthesis cluster,was overexpressed and showed that the overexpression of epsK led to a 26.54%decrease in the production of EPS and resulted in slenderer cell shape.Transcriptome analysis combined with protein-protein interactions analysis and protein modeling revealed that epsK was likely responsible for the synthesis of teichuronic acid,a substitute cell wall component of teichoic acid when the strain was suffering phosphate limitation.Further cell cultivation showed that either phosphate limitation or the overexpression of teichuronic acid synthesis genes,tuaB and tuaE could similarly lead to EPS reduction.The enhanced production of teichuronic acid induced by epsK overexpression triggered the endogenous phosphate starvation,resulting in the decreased EPS synthesis and biomass,and the enhanced bacterial chemotaxis.This study presents an insight into the mechanism of EPS synthesis and offers the potential in controllable synthesis of target products. | Yiyuan Xu Lijie Yang Haiyan Wang Xiaoyu Wei Yanyan Shi Dafeng Liang Mingfeng Cao Ning He | 2022 | Synthetic and Systems Biotechnology2022,7,2: | 1 |

| 13 | Oceanic vertical mixing of the lower halocline water in the Chukchi Borderland and Mendeleyev Ridge显示文摘Oceanic vertical mixing of the lower halocline water(LHW)in the Chukchi Borderland and Mendeleyev Ridge was studied based on in situ hydrographic and turbulent observations.The depth-averaged turbulent dissipation rate of LHW demonstrates a clear topographic dependence,with a mean value of 1.2×10^(-9) W/kg in the southwest of Canada Basin,1.5×10^(-9) W/kg in the Mendeleyev Abyssal Plain,2.4×10^(-9) W/kg on the Mendeleyev Ridge,and2.7×10^(-9) W/kg on the Chukchi Cap.Correspondingly,the mean depth-averaged vertical heat flux of the LHW is0.21 W/m^(2) in the southwest Canada Basin,0.30 W/m^(2) in the Mendeleyev Abyssal Plain,0.39 W/m^(2) on the Mendeleyev Ridge,and 0.46 W/m^(2) on the Chukchi Cap.However,in the presence of Pacific Winter Water,the upward heat released from Atlantic Water through the lower halocline can hardly contribute to the surface ocean.Further,the underlying mechanisms of diapycnal mixing in LHW—double diffusion and shear instability—was investigated.The mixing in LHW where double diffusion were observed is always relatively weaker,with corresponding dissipation rate ranging from 1.01×10^(-9) W/kg to 1.57×10^(-9) W/kg.The results also show a strong correlation between the depth-average dissipation rate and strain variance in the LHW,which indicates a close physical linkage between the turbulent mixing and internal wave activities.In addition,both surface wind forcing and semidiurnal tides significantly contribute to the turbulent mixing in the LHW. | Long Lin Hailun He Yong Cao Tao Li Yilin Liu Mingfeng Wang | 2021 | Acta Oceanologica Sinica2021,40,11: | 0 |
| 14 | Comparative cost analysis of three different anesthesia methods in gynecological laparoscopic surgery显示文摘In the current study,we assessed and evaluated the costs and benefits of three popular methods of general anesthesia practiced in our department for gynecological laparoscopic surgery in recent years.Sixty adult female patients who underwent elective gynecological laparoscopic surgery under general anesthesia were randomly divided into three groups:group Ⅴ,group Ⅰ and group C.In group Ⅴ,anesthesia was induced intravenously with midazolam,remifentanil,propofol and vecuronium,and maintained with continuous infusion of propofol and remifentanil.In group Ⅰ,anesthesia was intravenously induced with midazolam,fentanyl,propofol and vecuronium,and maintained with inhaled isoflurane and intravenous bonus of fentanyl.In group C,anesthesia was induced as in group Ⅰ,but maintained with isoflurane inhalation combined with propofolremifentanil infusion.All patients received vecuronium for muscle relaxation.Perioperative incidences of complications and total anesthesia costs for patients in all groups were recorded.In addition,postoperative satisfaction of the patients was also noted,and similar outcomes of the satisfaction were reported in all 60 patients.Although there was no statistical significance among groups,the incidence of postoperative nausea and vomiting were higher in group C,and the rates of shivering and the needs for analgesics were higher in group Ⅴ.Anesthesia costs in group Ⅰ were the lowest.Therefore,it is concluded that the costs of anesthesia induced with midazolam,fentanyl,propofol,vecuronium,and maintained with isoflurane,fentanyl and vecuronium are cheapest,and there is no significant difference in patients’satisfaction and safety among the three above-mentioned methods of anesthesia in our department. | Xiaohui Chi Yeling Chen Mingfeng Liao Fei Cao Yuke Tian Xueren Wang | 2012 | Frontiers of Medicine2012,6,3: | 0 |
| 15 | Genomic and metabolomic analysis of Bacillus licheniformis with enhanced poly-γ-glutamic acid production through atmospheric and room temperature plasma mutagenesis显示文摘Poly-γ-glutamic acid is an extracellular polymeric substance with various applications owing to its valuable properties of biodegradability,flocculating activity,water solubility,and nontoxicity.However,the ability of natural strains to produce poly-γ-glutamic acid is low.Atmospheric and room temperature plasma was applied in this study to conduct mutation breeding of Bacillus licheniformis CGMCC 2876,and a mutant strain M32 with an 11%increase in poly-γ-glutamic acid was obtained.Genome resequencing analysis identified 7 nonsynonymous mutations of ppsC encoding lipopeptide synthetase associated with poly-γ-glutamic acid metabolic pathways.From molecular docking,more binding sites and higher binding energy were speculated between the mutated plipastatin synthase subunit C and glutamate,which might contribute to the higher poly-γ-glutamic acid production.Moreover,the metabolic mechanism analysis revealed that the upregulated amino acids of M32 provided substrates for glutamate and promoted the conversion between L-and D-glutamate acids.In addition,the glycolytic pathway is enhanced,leading to a better capacity for using glucose.The maximum poly-γ-glutamic acid yield of 14.08 g·L^(–1)was finally reached with 30 g·L^(–1)glutamate. | Xiaoyu Wei Lijie Yang Haiyan Wang Zhen Chen Yiyuan Xu Yue Weng Mingfeng Cao Qingbiao Li Ning He | 2022 | Frontiers of Chemical Science and Engineering2022,16,12: | 0 |
