| 2 | In-Situ Synthesis of CQDs/BiOBr Material via Mechanical Ball Milling with Enhanced Photocatalytic Performances显示文摘Designing simple, efficient, and environmentally friendly methods to construct high-efficient photocatalysts is an important strategy to promote the further development of the field of photocatalysis. Herein, flower-like carbon quantum dots(CQDs)/Bi OBr composite photocatalysts have been prepared via in-situ synthesis by mechanical ball milling in the existence of ionic liquid. The CQDs/Bi OBr composites exhibit higher photo-degradation performance for tetracycline(TC) than Bi OBr monomer and the commercial Bi_(2)O_(3) under visible light irradiation. For comparison, the different Br sources and synthetic methods are chosen to prepare Bi OBr and CQDs/Bi OBr composites. Photocatalysts prepared by ball milling and ionic liquid present significantly enhanced photocatalytic performance for removing TC. In addition, the introduction of CQDs could distinctly enhance the photocatalytic performances of pure Bi OBr. The reason is that CQDs as electron acceptor effectively separate electrons and holes and inhibit their recombination. The intermediates during photocatalytic degradation were tested using liquid chromatography-mass spectrometry(LC-MS) and possible degradation pathways were given. During degradation, ·OH, O_(2)^(·-)and h^(+) were identified to be the main active species based on electron spin resonance(ESR) spectra and free radical trapping experiments. | 5 | In situ formation of fluorescent silicon-containing polymer dots for alkaline phosphatase activity detection and immunoassay显示文摘The discovery and application of analyte-triggered fluorophore generation or fluorogenic reaction are significant and beneficial to the development of novel fluorescence(FL) analysis method. In this study, for the first time, we have reported a fluorogenic reaction to prepare fluorescent silicon-containing polymer dots(Si-PDs) by simply mixing N-[3-(trimethoxysilyl)propyl]ethylenediamine(DAMO) and hydroquinone(HQ) in aqueous solution at ambient temperature. Inspired by the alkaline phosphatase(ALP)-catalyzed hydrolysis of the substrate sodium 4-hydroxyphenyl phosphate(4-HPP) into HQ and the resultant HQcontrolled intense green Si-PDs generation, we have established a straightforward ALP activity assay by innovatively employing commercially available 4-HPP as the substrate. More significantly, the specific preparation method, clear formation mechanism and excellent performance enable the Si-PDs as well as its generation process to develop facile and attractive FL immunoassay.With the help of the universal ALP-based enzyme-linked immunosorbent assay(ELISA) platform and corresponding antibody, a convenient and conceptual ALP-based fluorescent ELISA has been constructed and applied in sensing cardiac troponin Ⅰ(cTnI),a well-known biomarker of acute myocardial infarction. | 7 | Cytoplasmic YAP1-mediated ESCRT-III assembly promotes autophagic cell death and is ubiquitinated by NEDD4L in breast cancer显示文摘Background:Nuclear Yes1-associated transcriptional regulator(YAP1)promotes tumor progression.However,the function of cytoplasmic YAP1 in breast cancer cells and its impact on the survival of breast cancer patients remain unclear.Our research aimed to explore the biological function of cytoplasmic YAP1 in breast cancer cells and the possibility of cytoplasmic YAP1 as a predictive marker of breast cancer survival.Methods:We constructed cell mutant models,including NLS-YAP15SA(nuclear localized),YAP1S94A(incapable of binding to the TEA domain transcription factor family)and YAP1S127D(cytoplasmic localized),and used Cell Counting Kit-8(CCK-8)assays,5-ethynyl-2’-deoxyuridine(EdU)incorporation assays,and Western blotting(WB)analysis to detect cell proliferation and apoptosis.The specific mechanism of cytoplasmic YAP1-mediated endosomal sorting complexes required for transport III(ESCRT-III)assembly was studied by co-immunoprecipitation,immunofluorescence staining,and WB analysis.Epigallocatechin gallate(EGCG)was used to simulate YAP1 retention in the cytoplasm in in vitro and in vivo experiments to study the function of cytoplasmic YAP1.YAP1 binding to NEDD4-like E3 ubiquitin protein ligase(NEDD4L)was identified using mass spectrometry and was verified in vitro.Breast tissue microarrays were used to analyze the relationship between cytoplasmic YAP1 expression and the survival of breast cancer patients.Results:YAP1 was mainly expressed in the cytoplasm in breast cancer cells.Cytoplasmic YAP1 promoted autophagic death of breast cancer cells.Cytoplasmic YAP1 bound to the ESCRT-III complex subunits charged multivesicular body protein 2B(CHMP2B)and vacuolar protein sorting 4 homolog B(VPS4B),promoting assembly of CHMP2B-VPS4B and activating autophagosome formation.EGCG retained YAP1 in the cytoplasm,promoting the assembly of CHMP2B-VPS4B to promote autophagic death of breast cancer cells.YAP1 bound to NEDD4L,and NEDD4L mediated ubiquitination and degradation of YAP1.Breast tissue microarrays revealed that high levels of cytoplasmic YAP1 were beneficial to the survival of breast cancer patients.Conclusions:Cytoplasmic YAP1 mediated autophagic death of breast cancer cells by promoting assembly of the ESCRT-III complex;furthermore,we established a new breast cancer survival prediction model based on cytoplasmic YAP1 expression. | Yan Guo Yuqing Cui Yangyang Li Xiaoying Jin Dandan Wang Mengxia Lei Fengzhi Chen Yali Liu Jinwen Xu Guanyu Yao Guangchun Zeng Xuesong Chen | 2023 | Cancer Communications2023,43,5: | 0 |
| 8 | Characteristics of MBR Oily Sludge and Its Influence on Membrane Fouling under Two Aeration Modes显示文摘A membrane bioreactor(MBR)with ordinary aeration(reactor R1)and a MBR with microporous aeration(reactor R2)are conducted in parallel to investigate the characteristics of oily sludge and its effect on membrane fouling.The results indicate that the order of membrane fouling rate from high to low is:reactor R1 dissolved oxygen(DO)(1 mg/L)>reactor R2 DO(1 mg/L)>reactor R1 DO(4 mg/L)>reactor R2 DO(4 mg/L).Membrane fouling rate is not related to oily sludge concentration but to oily sludge sedimentation performance and the small particle oily sludge is the key factor to affect the membrane fouling.The soluble microbial products(SMP)are examined by three-dimensional excitation-emission matrix(3DEEM)fluorescence spectra.3DEEM spectra demonstrate that the main organic substances of the SMP in two reactors are tyrosine aromatic protein,fulvic acid-like substances and soluble microbial products under DO are1 mg/L and 4 mg/L,respectively.The proportion sum of fulvic acid-like substances and soluble microbial products is the key factor affecting membrane fouling,and membrane fouling accelerates as the ratio increases. | Yuansha Xie Huiying Liu Yan Wang Jun Yi Xianwei Wu Mengxia Wu Jie Dai | 2019 | Open Journal of Yangtze Oil and Gas2019,4,2: | 0 |
