1MiR-122-5p suppresses the proliferation, migration, and invasion of gastric cancer cells by targeting LYN显示文摘Gastric cancer(GC)is one of malignant tumors with high mortality and morbidity in the world.MicroRNA-122(miR-122)acts as a tumor suppressor in a variety of cancers and has been found to be dominant in gastric adenocarcinoma.However,the specific biological function of miR-122-5p in GC is not completely clear.In this study,we found that miR-122-5p was low-expressed in GC tissues and cell lines by using qRT-PCR.Overexpression of miR-122-5p inhibited the proliferation,migration,and invasion of GC cells by using CCK-8 and transwell assays.On the contrary,downregulation of miR-122-5p promoted the proliferation,migration,and invasion of GC cells.In addition,we found that the expression of LYN,an Src family tyrosine kinase,was inversely correlated with miR-122-5p expression in GC tissues by using western blot analysis,immunohistochemistry,and qRT-PCR assays.Meanwhile,luciferase assay results indicated that LYN is a direct target of miR-122-5p in GC cells.Moreover,silencing LYN expression by its siRNA inhibited the proliferation,migration,and invasion of GC cells.Importantly,overexpression of LYN restored miR-122-5p-mediated inhibition of the proliferation,migration,and invasion of GC cells.Taken together,our results indicated miR-122-5p inhibited the proliferation,migration,and invasion by targeting LYN in GC.Lei Meng Zhangming Chen Zhe Jiang Ting Huang Jie Hu Panquan Luo Hongli Zhang Mengqi Huang Lei Huang Yu Chen Ming Lu A-man Xu Songcheng Ying 2020Acta Biochimica et Biophysica Sinica2020,52,1:12
2Robustness analysis of underground powerhouse construction simulation based on Markov Chain Monte Carlo method显示文摘Scheduling is a major concern in construction planning and management, and current construction simulation research typically targets the shortest total duration. However, uncertainties are inevitable in actual construction, which may lead to discrepancies between the actual and planned schedules and increase the risk of total duration delay. Therefore, developing a robust construction scheduling technique is of vital importance for mitigating disturbance and improving completion probability. In the present study, the authors propose a robustness analysis method that involves underground powerhouse construction simulation based on the Markov Chain Monte Carlo(MCMC) method. Specifically, the MCMC method samples construction disturbances by considering the interrelationship between the states of parameters through a Markov state transition probability matrix, which is more robust and efficient than traditional sampling methods such as the Monte Carlo(MC) method. Additionally, a hierarchical simulation model coupling critical path method(CPM) and a cycle operation network(CYCLONE) is built, using which construction duration and robustness criteria can be calculated. Furthermore, a detailed measurement method is presented to quantize the robustness of underground powerhouse construction, and the setting model of the time buffer is proposed based on the MCMC method. The application of this methodology not only considers duration but also robustness, providing scientific guidance for engineering decision making. 3Nonvolatile memristor based on heterostructure of 2D room-temperature ferroelectric α-In2Se3 and WSe2显示文摘Two-dimensional(2D) ferroelectricity is considered to have significant potential for information storage in the future. Semiconducting ferroelectrics that are stable at room temperature afford many possibilities for the assembly of various high-performance heterostructures and fabricating multifuntional devices.Herein, we report the synthesis of a stable van der Waals(vd W) single-crystal semiconductor α-In2 Se3.Piezoresponse force microscopy(PFM) measurements demonstrated the out-of-plane ferroelectricity in ~15 layers α-In2 Se3 at room temperature. Both ferroelectric domains with opposite polarization and the tested amplitude and phase curve proved that this semiconductor exhibits hysteresis behavior during polarization.In the α-In2Se3/WSe2 vertical heterostructure device, the switchable diode effect and nonvolatile memory phenomenon showed a high on/off ratio and a small switching voltage. The distinct resistance switches were further analyzed by band alignment of the heterostructure under different polarizations by first principle calculations. 4Development and validation of an HPLC–MS/MS method to determine clopidogrel in human plasma显示文摘A quantitative method for clopidogrel using online-SPE tandem LC–MS/MS was developed and fully validated according to the well-established FDA guidelines.The method achieves adequate sensitivity for pharmacokinetic studies,with lower limit of quantifications(LLOQs)as low as 10 pg/m L.Chromatographic separations were performed on reversed phase columns Kromasil Eternity-2.5-C18-UHPLC for both methods.Positive electrospray ionization in multiple reaction monitoring(MRM)mode was employed for signal detection and a deuterated analogue(clopidogrel-d_4)was used as internal standard(IS).Adjustments in sample preparation,including introduction of an online-SPE system proved to be the most effective method to solve the analyte back-conversion in clinical samples.Pooled clinical samples(two levels)were prepared and successfully used as real-sample quality control(QC)in the validation of back-conversion testing under different conditions.The result showed that the real samples were stable in room temperature for 24 h.Linearity,precision,extraction recovery,matrix effect on spiked QC samples and stability tests on both spiked QCs and real sample QCs stored in different conditions met the acceptance criteria.This online-SPE method was successfully applied to a bioequivalence study of75 mg single dose clopidogrel tablets in 48 healthy male subjects.Gangyi Liu Chunxia Dong Weiwei Shen Xiaopei Lu Mengqi Zhang Yuzhou Gui Qinyi Zhou Chen Yu 2016Acta Pharmaceutica Sinica B2016,6,1:3
5The prognostic value of programmed cell death ligand 1expression in non-Hodgkin lymphoma:a meta-analysis显示文摘Objective: Although the prognostic value of programmed cell death-ligand 1(PD-L1) expression in non-Hodgkin lymphoma(NHL) has been evaluated in many studies, the results remain controversial. To investigate the prognostic role of PD-L1 expression and the association between PD-L1 expression and clinicopathological features of NHL, we performed a meta-analysis.Methods: The Pub Med, EMBASE, and Cochrane Library databases were searched up to November 30, 2017. The hazard ratio(HR), 95% confidence interval(CI), and odds ratios(OR) with 95% CIs were combined to evaluate the association of PD-L1 expression with overall survival(OS) and clinicopathological features. Review manager 5.3 and STATA 12.0 were used in this meta-analysis.Results: A total of 2,005 patients across nine studies were enrolled in our meta-analysis, and the pooled results showed that high PD-L1 expression was associated with a poor prognosis(HR=2.04, 95% CI: 1.18–3.54, P=0.01). In the subgroup analysis according to histology types, pooled results demonstrated that an increased PD-L1 expression was an unfavorable prognostic factor for diffuse large B-cell lymphoma(HR=1.92, 95% CI: 1.06–3.48, P=0.03) but not for natural killer/T-cell lymphoma(HR=2.41, 95%CI: 0.47–12.22, P=0.29). Pooled ORs indicated that PD-L1 expression was higher in NHL with international prognostic indices of≥3. However, PD-L1 expression had no correlation with gender, age, disease stage, lactate dehydrogenase level, B symptoms, and germinal center B-cell-like lymphoma.Conclusions: High PD-L1 expression was a poor prognostic biomarker in patients with NHL. 6SETD8 induces stemness and epithelial–mesenchymal transition of pancreatic cancer cells by regulating ROR1 expression显示文摘Pancreatic cancer(PC)is one of the most deadly diseases,and its incidence is increasing year by year.The methyltransferase SETD8 has been demonstrated to play an important role in tumor cell proliferation and metastasis.However,little is known about whether SETD8 could affect the invasion and metastasis of PC and the mechanism underlying the regulation.Based on our previous report,here,we further found that SETD8 could promote the invasion and migration of PC cells by inducing the expression of receptor tyrosine kinase-like orphan receptor 1(ROR1).ROR1 was predominantly upregulated in PC tissues and was correlated with lymph node metastasis and worse prognosis.Mechanistically,SETD8 mediated ROR1 activity and regulated PC cells invasion and migration,although promoting the expression of stemness and epithelial–mesenchymal transition-related molecules.This promotion effect disappeared when the catalytically inactive mutant SETD8 was overexpressed,which could be counteracted by the SETD8-specific methyltransferase inhibitor UNC0379.Collectively,our results demonstrate that SETD8 may be a novel prognostic factor and a therapeutic target of PC.Mengqi Liu Yihua Shi Qiangsheng Hu Yi Qin Shunrong Ji Wensheng Liu Qifeng Zhuo Guixiong Fan Zeng Ye Changfeng Song Xianjun Yu Xiaowu Xu Wenyan Xu 2021Acta Biochimica et Biophysica Sinica2021,53,12:2
7FGFBPI-mediated crosstalk between fibroblasts and pancreatic cancer cells via FGF22/FGFR2 promotes invasion and metastasis of pancreatic cancer显示文摘Fibroblast growth factor-binding protein 1(FGFBP1)promotes fibroblast growth factor(FGF)activity by releasing FGFs from extracellular matrix storage.We previously reported that the tumor suppressor F-box and WD repeat domain-containing 7 suppresses FGFBP1 by reducing expression of c-Myc,which inhibits the proliferation and migration of pancreatic cancer cells.However,the potential mechanism by which FGFBP1 facilitates pancreatic ductal adenocarcinoma(PDAC)remains unexplored.In this study,we focused on the function of FGFBP1 in the interplay between cancer-associated fibroblasts(CAFs)and pancreatic cancer cells(PCCs).Decreased FGF22 expression was detected in CAFs co-cultured with PCCs with FGFBP1 abrogation,which was verified in the cell culture medium by enzyme-linked immunosorbent assay.Active cytokine FGF22 significantly facilitated the migration and invasion of PANC-1 and Mia PaCa-2 cells.The number of penetrating PCCs cocultured with CAFs with FGF22 abrogation was significantly less than that of the control group.Interestingly,higher expressions of FGF22 and fibroblast growth factor receptor 2(FGFR2)were associated with worse prognosis of patients with PDAC and FGFR2,an independent prognostic marker of PDAC.The PANC-1 and Mia PaCa-2 cells with silenced FGFR2 showed weaker invasion and metastasis,even if these cells were simultaneously treated with cytokine FGF22.These results revealed that FGFBPI-mediated interaction between CAFs and PCCs via FGF22/FGFR2 facilitates the migration and invasion of PCCs.FGFR2 could act as a prognostic marker for patients with PDAC.Zheng Zhang Yi Qin Shunrong Ji Wenyan Xu Mengqi Liu Qiangsheng Hu Zeng Ye Guixiong Fan Xianjun Yu Wensheng Liu Xiaowu Xu 2021Acta Biochimica et Biophysica Sinica2021,53,8:2
8Composition control and temperature inferential control of dividing wall column based on model predictive control and PI strategies显示文摘The dividing wall column(DWC) is considered as a major breakthrough in distillation technology and has good prospect of industrialization. Model predictive control(MPC) is an advanced control strategy that has acquired extensive applications in various industries. In this study, MPC is applied to the process for separating ethanol,n-propanol, and n-butanol ternary mixture in a fully thermally coupled DWC. Both composition control and temperature inferential control are considered. The multiobjective genetic algorithm function 'gamultiobj' in Matlab is used for the weight tuning of MPC. Comparisons are made between the control performances of MPC and PI strategies. 10Room-temperature conversion of ethane and the mechanism understanding over single iron atoms confined in graphene显示文摘The catalytic conversion of ethane to high value-added chemicals is significantly important for utilization of hydrocarbon resources.However, it is a great challenge due to the typically required high temperature(> 400 ℃) conditions.Herein, a highly active catalytic conversion process of ethane at room temperature(25 ℃) is reported on single iron atoms confined in graphene via the porphyrin-like N4-coordination structures.Combining with the operando time of flight mass spectrometer and density functional theory calculations, the reaction is identified as a radical mechanism, in which the C–H bonds of the same C atom are preferentially and sequentially activated, generating the value-added C2 chemicals, simultaneously avoiding the over-oxidation of the products to CO2.The in-situ formed O–FeN4–O structure at the single iron atom serves as the active center for the reaction and facilitates the formation of ethyl radicals.This work deepens the understanding of alkane C–H activation on the FeN4 center and provides the reference in development of efficient catalyst for selective oxidation of light alkane.Suheng Wang Haobo Li Mengqi He Xiaoju Cui Lei Hua Haiyang Li Jianping Xiao Liang Yu N.Pethan Rajan Zhaoxiong Xie Dehui Deng 2019Journal of Energy Chemistry2019,28,9:1
11Gelation of Hole Transport Layer to Improve the Stability of Perovskite Solar Cells显示文摘To achieve high power conversion efficiency(PCE) and long-term stability of perovskite solar cells(PSCs), a hole transport layer(HTL) with persistently high conductivity, good moisture/oxygen barrier ability, and adequate passivation capability is important. To achieve enough conductivity and effective hole extraction, spiro-OMe TAD, one of the most frequently used HTL in optoelectronic devices, often needs chemical doping with a lithium compound(LiTFSI). However, the lithium salt dopant induces crystallization and has a negative impact on the performance and lifetime of the device due to its hygroscopic nature. Here, we provide an easy method for creating a gel by mixing a natural small molecule additive(thioctic acid, TA) with spiro-OMe TAD. We discover that gelation effectively improves the compactness of resultant HTL and prevents moisture and oxygen infiltration. Moreover, the gelation of HTL improves not only the conductivity of spiro-OMe TAD, but also the operational robustness of the devices in the atmospheric environment. In addition, TA passivates the perovskite defects and facilitates the charge transfer from the perovskite layer to HTL. 13Quick Phase Identification for Dense Seismic Array with Aid from Long Term Phase Records of Co-located Sparse Permanent Stations显示文摘The phase identification and travel time picking are critical for seismic tomography,yet it will be challenging when the numbers of stations and earthquakes are huge.We here present a method to quickly obtain P and S travel times of pre-determined earthquakes from mobile dense array with the aid from long term phase records from co-located permanent stations.The records for 1768 M≥2.0 events from 2011 to 2013 recorded by 350 ChinArray stations deployed in Yunnan Province are processed with an improved AR-AIC method utilizing cumulative envelope and rectilinearity.The reference arrivals are predicted based on phase records from 88 permanent stations with similar spatial coverage,which are further refined with AR-AIC.Totally,718573 P picks and 512035 S picks are obtained from mobile stations,which are 28 and 22 times of those from permanent stations,respectively.By comparing the automatic picks with manual picks from 88 permanent stations,for M≥3.0 events,81.5%of the P-pick errors are smaller than 0.5 second and 70.5%of S-pick errors are smaller than1 second.For events with a lower magnitude,76.5%P-pick errors fall into 0.5 second and 69.5%S-pick errors are smaller than 1 second.Moreover,the Pn and Sn phases are easily discriminated from directly P/S,indicating the necessity of combining traditional auto picking and integrating machine learning method.CHEN Yini LI Jun WANG Zhenjie ZHAO Mengqi YU Tiehong LI Danning YU Junyi WANG Weitao 2020Earthquake Research in China2020,34,3:1

15Interleukin-13 promotes cellular senescence through inducing mitochondrial dysfunction in IgG4-related sialadenitis显示文摘Immunoglobulin G4-related sialadenitis(IgG4-RS)is an immune-mediated fibro-inflammatory disease and the pathogenesis is still not fully understood.The aim of this study was to explore the role and mechanism of interleukin-13(IL-13)in the cellular senescence during the progress of IgG4-RS.We found that the expression of IL-13 and IL-13 receptorα1(IL-13Rα1)as well as the number of senescent cells were significantly higher in the submandibular glands(SMGs)of IgG4-RS patients.IL-13 directly induced senescence as shown by the elevated activity of senescence-associatedβ-galactosidase(SA-β-gal),the decreased cell proliferation,and the upregulation of senescence markers(p53 and p16)and senescence-associated secretory phenotype(SASP)factors(IL-1βand IL-6)in SMG-C6 cells.Mechanistically,IL-13 increased the level of phosphorylated signal transducer and activator of transcription 6(p-STAT6)and mitochondrial-reactive oxygen species(mt ROS),while decreased the mitochondrial membrane potential,ATP level,and the expression and activity of superoxide dismutase 2(SOD2).Notably,the IL-13-induced cellular senescence and mitochondrial dysfunction could be inhibited by pretreatment with either STAT6 inhibitor AS1517499 or mitochondria-targeted ROS scavenger Mito TEMPO.Moreover,IL-13 increased the interaction between p-STAT6 and c AMP-response element binding protein(CREB)-binding protein(CBP)and decreased the transcriptional activity of CREB on SOD2.Taken together,our findings revealed a critical role of IL-13 in the induction of salivary gland epithelial cell senescence through the elevated mitochondrial oxidative stress in a STAT6–CREB–SOD2-dependent pathway in IgG4-RS.Mengqi Zhu Sainan Min Xiangdi Mao Yuan Zhou Yan Zhang Wei Li Li Li Liling Wu Xin Cong Guangyan Yu 2022International Journal of Oral Science2022,14,3:1

17Multi-parametric MRI Diagnoses Cerebellar Hemangioblastoma:A Case Report显示文摘We performed contrast-enhanced T2 fluid-attenuated inversion recovery(T2-FLAIR)and dynamic contrast enhanced MRI to illustrate the imaging characteristics of one case of hemangioblastoma.T2-FLAIR showed a large cyst located in the right cerebellum with mural nodule.The intensely enhancing cyst wall was observed on enhanced T2-FLAIR images acquired from 5.6 to 23 minutes after contrast administration,and quantitative dynamic contrast enhanced-MRI demonstrated that both the cyst wall and mural nodule presented high Ktrans,Kep and Ve values compared with the contralateral normal cerebellar tissues.The cyst showed gradual enhancement and reached the highest signal intensity at 67 minutes after contrast administration on enhanced T2-FLAIR images.In conclusion,early enhancement of cyst wall on T2-FLAIR might be the characteristic imaging findings for cystic hemangioblastoma,which may assist in the diagnosis of hemangioblastoma preoperatively.Zhiye Chen Mengqi Liu Shengyuan Yu Lin Ma 2018Chinese Medical Sciences Journal2018,33,3:0
184–λ hybrid InGaAsP-Si evanescent laser array with low power consumption for on-chip optical interconnects显示文摘A 4 –λ hybrid InGaAsP-Si evanescent laser array is obtained by bonding III–V distributed feedback lasers to a silicon on insulator(SOI) substrate using a selective area metal bonding technique. Multiple wavelengths are realized by varying the width of the III–V ridge waveguide. The threshold current is less than 10 mA for all wavelength channels under continuous-wave(CW) operation at room temperature, and the lowest threshold current density is 0.76 kA∕cm2. The side mode suppression ratio(SMSR) is higher than 40 dB for all wavelength channels when the injection current is between 20 mA and 70 mA at room temperature, and the highest SMSR is up to 51 dB. A characteristic temperature of 51 K and thermal impedance of 144°C/W are achieved on average.The 4 –λ hybrid InGaAsP-Si evanescent laser array exhibits a low threshold and high SMSR under CW operation at room temperature. 19Low-temperature synthesis of sp^2 carbon nanomaterials显示文摘sp^2 carbon nanomaterials are mainly composed of sp^2-hybridized carbon atoms in the form of a hexagonal network. Due to the p bonds formed by unpaired electrons, sp^2 carbon nanomaterials possess excellent electronic, mechanical, and optical properties, which have attracted great attention in recent years.As the advanced sp^2 carbon nanomaterials, graphene and carbon nanotubes(CNTs) have great potential in electronics, sensors, energy storage and conversion devices, etc. The low-temperature synthesis of graphene and CNTs are indispensable to promote the practical industrial application. Furthermore, graphene and CNTs can even be expected to directly grow on the flexible plastic that cannot bear high temperature,expanding bright prospects for applications in emerging flexible nanotechnology. An in-depth understanding of the formation mechanism of sp^2 carbon nanomaterials is beneficial for reducing the growth temperature and satisfying the demands of industrial production in an economical and low-cost way. In this review, we discuss the main strategies and the related mechanisms in low-temperature synthesis of graphene and CNTs, including the selection of precursors with high reactivity, the design of catalyst, and the introduction of additional energy for the pre-decomposition of precursors. 20Low fabrication cost wavelength tunable WG-FP hybrid-cavity laser working over 1.7μm显示文摘A wide wavelength tuning range and single-mode hybrid cavity laser consists of a square Whispering-Gallery(WG)microcavity and a Fabry–Pérot(FP)was introduced and demonstrated.A wavelength tuning range over 12.5 nm from 1760.87 to 1773.39 nm which was single-mode emitting was obtained with the side-mode suppression ratio over 30 dB.The hybrid cavity laser does not need grating etching and special epitaxial structure,which reduces the fabrication difficulty and cost,and shows the potential for gas sensing with absorption lines in this range.Fangyuan Meng Hongyan Yu Xuliang Zhou Mengqi Wang Yejin Zhang Wenyu Yang Jiaoqing Pan 2022Journal of Semiconductors2022,43,6:0
