1Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The tumorigenic potential of CD133 + ,CD44 + ,CD44 + CD133 + cells and CD44 + CD133 + CD24 + cells was stronger than that of CD133 - ,CD44 - ,CD44 + CD133 - cells and CD44 + CD133 + CD24 - cells in mice,respectively.In conclusion,the CD44 + CD133 + CD24 + cells have some biological properties of cancer stem-like cells or are highly similar to the characteristics of cancer stem cells(CSC).These results provide an important method for identifying cancer stem-like cells in B16F10 cells and for further cancer target therapy.Jun Dou Meng Pan Ping Wen Yating Li Quan Tang Lili Chu Fengshu Zhao Chuilian Jiang Weihua Hu Kai Hu Ning Gu 2007Cellular & Molecular Immunology2007,4,6:25

3Are humic substances soil microbial residues or unique synthesized compounds?A perspective on their distinctiveness显示文摘Humic substances(HS),which are defined as a series of highly acidic,relatively high-molecular-weight,and yellow to black colored substances formed during the decay and transformation of plant and microbial remains,ubiquitously occur in nature.Humic substances represent the largest stable organic carbon pool in terrestrial environments and are the central characteristic of the soil.However,the validity of the HS concept and the justification of their extraction procedure have been recently debated.Here,we argue that the traditional humic paradigm is still relevant.Humic substances are distinctive and complex because the extracted HS formed during the humification are chemically distinct from their precursors and are heterogeneous among soils.By reviewing the concept,formation pathways,and stabilization of HS,we propose that the key question facing soil scientists is whether HS are soil microbial residues or unique synthesized compounds.Without revealing the distinctiveness of HS,it is impossible to address this question,as the structure,composition,and reactivity of HS are still poorly known owing to the heterogeneity and geographical variability of HS and the limits of the currently available analytical techniques.In our view,the distinctiveness of HS is fundamental to the soil,and thus further studies should be focused on revealing the distinctiveness of HS and explaining why HS hold this distinctiveness.Sen DOU Jun SHAN Xiangyun SONG Rui CAO Meng WU Chenglin LI Song GUAN 2020Pedosphere2020,30,2:12
4Cordycepin protects renal ischemia/reperfusion injury through regulating inflammation,apoptosis,and oxidative stress显示文摘Cordycepin(3-deoxyadenosine)is a naturally occurring adenosine analog and one of the bioactive constituents isolated from Cordyceps sinensis,species of the fungal genus Cordyceps.It has traditionally been a prized Chinese folk medicine for the human well-being.However,the actions of cordycepin against renal ischemia/reperfusion injury(/R)are still unknown.In the present study,rats were subject to VR and cordycepin was intragastrically administered for seven consecutive days before surgery to investigate the effects and mechanisms of cordycepin against renal/R injury.The test results of kidney and peripheral blood samples of experimental animals showed that cordycepin significantly decreased serum blood urea nitrogen and creatinine levels and markedly attenuated cell injury.Mechanistic studies showed that cordycepin significantly regulated inflammation,apoptosis,and oxidative stress.These data provide new insights for investigating the natural product with the nephroprotective effect against I/R,which should be developed as a new therapeutic agent for the treatment of I/R in the future.Feng Han Meng Dou Yuxiang Wang Cuixiang Xu Yang Li Xiaoming Ding Wujun Xue Jin Zheng Puxun Tian Chenguang Ding 2020Acta Biochimica et Biophysica Sinica2020,52,2:9
5A phaseⅠtrial of an oral subtype-selective histone deacetylase inhibitor,chidamide,in combination with paclitaxel and carboplatin in patients with advanced non-small cell lung cancer显示文摘Objective: This phase Ⅰ study was to evaluate safety, maximum tolerated dose, pharmacokinetics and preliminary antitumor activity of chidamide, a novel subtype-selective histone deacetylase(HDAC) inhibitor, in combination with paclitaxel and carboplatin in patients with advanced non-small cell lung cancer(NSCLC).Methods: Ten patients received oral chidamide 20, 25, or 30 mg twice per week continuously with paclitaxel(175 mg/m^2) and carboplatin [area under the curve(AUC) 5 mg/m L/min] administered in a 3-week cycle. Patients with response and stable disease after four cycles maintained chidamide monotherapy until disease progression or unacceptable toxicity. Blood samples were collected for pharmacokinetic analysis after the first single oral of chidamide and first combination treatment in cycle 1 from all patients.Results: Two dose-limiting toxicities were recorded in the 30 mg cohort, including thrombocytopenia and prolonged neutropenia in the first cycle. Grade 3/4 neutropenia in any cycle was observed in all patients, but was not associated with significant complications. Other grade 3/4 hematologic toxicities included thrombocytopenia and leucopenia. No significant changes were observed in pharmacokinetic parameters for both chidamide and paclitaxel. One patient in the 20 mg cohort had confirmed partial response(PR). Two out of 5 patients with brain metastases had intracranial complete remission after 4-cycle treatment.Conclusions: Chidamide combined with paclitaxel and carboplatin was generally tolerated without unanticipated toxicities or clinically relevant pharmacokinetic interactions. 6Hsa-miR-202-3p,up-regulated in type 1 gastric neuroendocrine neoplasms,may target DUSP1显示文摘AIM To detect abnormal micro RNA(miRNA) expression in type 1 gastric neuroendocrine neoplasms(g-NENs) and find potential target genes.METHODS Tumour tissues from patients with type 1 g-NENs were used as experimental samples, and gastric mucosal tissues from the same patients obtained during gastroscopy review after several months were used as control samples. miRNA expression was examined with Agilent human miRNA chips and validated via RTPCR. Three types of target gene prediction software(TargetScan, PITA, and microRNAorg) were used to predict potential target genes of the differentially expressed miRNAs, and a dual-luciferase reporter assay system was used for verification. RESULTS Six miRNAs were significantly upregulated or downregulated in the tumours compared to the control samples. Among them, miR-202-3 p was extraordinarily upregulated. RT-PCR of seven sample sets confirmed that miR-202-3 p was upregulated in tumour tissues. In total, 215 target genes were predicted to be associated with miR-202-3 p. Among them, dual-specificity phosphatase 1(DUSP1) was reported to be closely related to tumour occurrence and development. The dual-luciferase reporter assay showed that miR-202-3 p directly regulated DUSP1 in 293 T cells. 8miR-124/IRE-1α affects renal ischemia/reperfusion injury by regulating endoplasmic reticulum stress in renal tubular epithelial cells显示文摘Acute kidney injury(AKI)refers to a clinical syndrome that occurs as a result of a rapid decline in renal function caused by multiple factors.Renal ischemia/reperfusion(/R)injury is one of the main causes of AKI and has a high incidence and mortality,However,the specific pathogenesis of renal I/R injury is still unclear.In recent years,a major breakthrough has been made in the study of endoplasmic reticulum stress(ERS)-mediated apoptosis in I/R injury.It has been reported that miRNAs play protective roles in ischemic/reperfused organs,but the molecular mechanisms have not been investigated deeply.In this study,the renal VR mouse model was used to explore the roles of miR-124 in ERS and in renal I/R injury.The western blot results showed that the expression levels of ERS-related proteins IRE-1α,XBP-1,and glucose-regulated protein 78(GRP78)were significantly increased in the IVR model group when compared with those in the control group.Meanwhile,qPCR results showed that miR-124 expression was decreased in the I/R injury model,and overexpression of miR-124 using miR-124 mimics effectively reduced the expression of ERS-related proteins and alleviated renal LR injury.In addition,luciferase reporter assay was performed,and the results showed that IRE 1αand miR-124 may have direct interaction.In conclusion,our data indicated that miR-124 was a negative regulator of ERS via binding to IRE-1α,ultimately conferring its protective effect on the kidney,which demonstrates the regulatory mechanism of miR-124 in renal I/R injury and provides new ideas and methods for the prevention and treatment of renal I/R injury.Chenguang Ding Meng Dou Yuxiang Wang Yang Li Ying Wang Jin Zheng Xiao Li Wujun Xue Xiaoming Ding Puxun Tian 2020Acta Biochimica et Biophysica Sinica2020,52,2:4
9Molecular Clone, Expression, and Prediction of Construction and Function to Key Genes of Interleukin Family of Porcine显示文摘This research was to clone, express, and analyze the structure and function of major molecules of porcine interleukin family. Genes of porcine interleukin family were cloned by RT-PCR from stimulated porcine PBMC by LPS and PHA, and then expressed in E. coli, and the structure and function of these molecules were predicted by ExPASY. The results showed that genes of IL-4, IL-6, and IL-18 were successfully cloned and expressed. Furthermore, the expression products of recombinant IL-4 and IL-6 both have multiple biological activities. By analyzing these genes with the NCBI/GenBank data, the homologies of the nucleotide acid sequence are 99.25, 99.21, and 100%, respectively, and have great species differences when compared with other animal species. The results of the prediction showed that all these molecules contain several phosphorylation, glycosylation, protein kinase, and signal transduction bonding sites in secondary structure, and all are compact globularity protein in space configuration. These characteristics of structure are the basis for their multiple biological functions. 10Adjuvant Effects of Recombinant Plasmids of Porcine IL-4 and IFN-γ to Cysticercosis cellulosae Vaccines in Mice and Pigs显示文摘To investigate the adjuvant potential of porcine IL-4 and IFN-γ in mice and pigs,the genes of porcine IL-4 and IFN-γ were cloned and the recombinant mammalian expression plasmids were constructed for in vivo expression of the cytokines.Adjuvant effects of recombinant expression plasmids of IL-4 and IFN-γ(pcDNA-IL-4,pcDNA-IFN-γ) co-administrated with Cysticercus cellulosae crude antigen or TSOL18 recombinant protein antigen have been carried out in mice and pigs,respectively.We have demonstrated that recombinant plasmids of the cytokines as an adjuvant could induce stronger immune response in mice and pigs.With the C.cellulosae parasite antigen,porcine pcDNA-IL-4 induced higher specific antibody of immunized mice than pcDNA-IFN-γ.But pcDNA-IFN-γ is significantly stronger than that of no adjuvant or empty plasmids with the antigen control group.For the TSOL18 recombinant protein antigen vaccine,pcDNA-IL-4 still had a stronger ability to enhance specific antibody in swine than pcDNA-IFN-γ(P < 0.01),but the immune protective rate was lower in challenged pigs(only 68.7%).Although pcDNA-IFN-γ showed lower specific antibody,the protection rate was very high(91%) than other group(P < 0.01).This study indicated that the recombinant expression plasmids of porcine IL-4 and IFN-γ display stronger adjuvant effects to C.cellulosae vaccine,further research should be carried out for understanding of the interaction mechanism.JING Zhi-zhong DOU Yong-xi MENG Xue-lian CHEN Guo-hua WANG Pei-ya LUO Qi-hui YUAN Gai-ling ZHENG Ya-dong CAI Xue-peng 2010Agricultural Sciences in China2010,9,1:4
11Synthesis, characteristics of hierarchical EU-1 zeolite for xylene isomerization probe reaction显示文摘Introduced a method of synthesizing hierarchical EU-1 zeolite with organosilanes as additive, and studied the influences of following different kinds of organosilanes on the synthesis of hierarchical EU-1 zeolite: γ-glycidoxy propyl trimethoxy silane(GPTMS), N-β-(aminoethyl)-γ-aminopropyl methyl dimethoxyl silane(APAEDMS),and N-(β-aminoethyl)-γ-aminopropyl dimethoxyl(ethyoxyl) silane(TMPED). The hierarchical EU-1 samples were characterized by XRD, SEM, N_2 adsorption, FT-IR and NH_3-TPD to analyze the crystallinity, morphology, surface area, pore size distribution and acidity. The results showed that hierarchical EU-1 zeolites were successfully synthesized; organosilanes have great influence on crystal morphology of EU-1 zeolites; the exterior surface area of hierarchical EU-1 zeolite, which synthesized with organosilanes(APAEDMS) adding into synthesis system, increased by 62.1% and mesopore volume increased by 129.1% compared with conventional EU-1 zeolites, thus can reduce the diffusional restriction markedly in catalytic reaction. The catalytic performance of hierarchical EU-1zeolites were evaluated in m-xylene isomerization on fixed bed reactor. The catalytic data showed that the isomerization activity PX/X of the hierarchical EU-1 zeolites reached around 24.09% in theoretical thermodynamic equilibrium from 23.83%, and the selectivity of C_8 aromatic hydrocarbon increased from 75.16% to 84.87%. 12The effects of soil moisture and salinity as functions of groundwater depth on wheat growth and yield in coastal saline soils显示文摘In the coastal saline soils,moisture and salinity are the functions of groundwater depth affecting crop growth and yield.Accordingly,the objectives of this study were to:1)investigate the combined effects of moisture and salinity stresses on wheat growth as affected by groundwater depth,and 2)find the optimal groundwater depth for wheat growth in coastal saline soils.The groundwater depths(0.7,1.1,1.5,1.9,2.3,and 2.7 m during 2013-2014(Y1)and 0.6,1.0,1.4,1.8,2.2,and 2.6 m during 2014-2015(Y2))of the field experiment were maintained by soil columns.There was a positive correlation between soil moisture and salinity.Water logging with high salinity(groundwater depth at 0.7 m in Y1 and 0.6 m in Y2)showed a greater decline towards wheat growth than that of slight drought with medium(2.3 m in Y1)or low salinity(2.7 m in Y1,2.2 and 2.6 m in Y2).The booting stage was the most sensitive stage of wheat crop under moisture and salinity stresses.Data showed the most optimal rate of photosynthesis,grain yield,and flour quality were obtained under the groundwater depth(ditch depth)of 1.9 m(standard soil moisture with medium salinity)and 2.3 m(slight drought with medium salinity)in Y1 and 1.8 m(standard soil moisture with medium salinity)and 2.2 m(slight drought with low salinity)in Y2.The corresponding optimal soil relative moisture content and conductivity with the 1:5 distilled water/soil dilution,in the depth of 0-20 cm and 20-40 cm in coastal saline soils,were equal to 58.67-63.07%and 65.51-72.66%in Y1,63.09-66.70%and 69.75-74.72%in Y2;0.86-1.01 dS m^-1 and 0.63-0.77 dS m^-1 in Y1,0.57-0.93 dS m^-1 and 0.40-0.63 dS m^-1 in Y2,respectively.ZHANG He LI Yan MENG Ya-li CAO Nan LI Duan-sheng ZHOU Zhi-guo CHEN Bing-lin DOU Fu-gen 2019Journal of Integrative Agriculture2019,18,11:3
13Rapid Urban Land Expansion in Earthquake-Prone Areas of China显示文摘A timely understanding of urban expansion in earthquake-prone areas is crucial for earthquake risk assessment and urban planning for earthquake mitigation.However, a comprehensive evaluation of urban expansion in earthquake-prone areas is lacking in China, especially in the context of rapid urbanization. Based on time series urban land data and seismic ground-motion parameter zonation maps, this study analyzed urban expansion in the most seismically hazardous areas(MSHAs) of China from1992 to 2015 on the national, regional, and city scales. The results show that urban land area in the MSHAs expanded by 6767 km^2 from 1992 to 2015, with a gain of 350%.Specifically, the increase in urban land area of small cities in the MSHAs of western China during this period was the fastest, 6.24 times greater than that at the national level. In terms of spatial patterns, the urban land patches in the MSHAs in 2015 were more fragmented than those in 1992 on all scales. The percentage of change in the number of patches and the landscape shape index of the urban land patches of small cities in the MSHAs of western China were the highest across all cities. Therefore, we believe that special attention should be paid to the cities in the MSHAs that exhibit the most rapid increases in both urban land area and fragmentation, especially the small cities in western China. 16Effects of a Multivitamin/multimineral Supplement on Young Males with Physical Overtraining: A Placebo-controlled, Randomized, Double-blinded Cross-over Trial显示文摘Objective To investigate the effects of vitamin-mineral supplement on young males with physical overtraining. Methods Two hundred and forty male Chinese field artillery personnel who undertook large scale and endurance military training and were on ordinary Chinese diet were randomized to receive a multivitamin/multimineral supplement or a placebo for 1 week. After a 1-week wash-out period, a cross-over with 1 week course of a placebo or multivitamin/multimineral supplement was conducted. Blood and urine samples were analyzed for adrenal, gonadal and thyroid hormones. In addition, cellular immune parameters (CD3+, CD3+CD4+, CD3+CD8+, CD4/CD8, CD3-CD56+, CD3-CD19+) were examined and psychological tests were performed before and after the training program and nutrition intervention. Results After a large scale and endurance military training, the participants showed significantly increased thyroid function, decreased adrenal cortex, testosterone and immunological function, and significantly increased somatization, anger and tension. Compared to placebo, multivitamin/ multimineral intervention showed significant effects on functional recovery of the pituitary-adrenal axis, pituitary-gonadal axis, pituitary-thyroid axis and immune system as well as psychological parameters. Conclusion High-intensity military operations have significant impacts on the psychology, physical ability and neuroendocrine-immune system in young males. 17A novel phenotype with splicing mutation identified in a Chinese family with desminopathy显示文摘Background:Desminopathy, a hereditary myofibrillar myopathy, mainly results from the desmin gene (DES) mutations.Desminopathy involves various phenotypes, mainly including different cardiomyopathies, skeletal myopathy, and arrhythmia.Combined with genotype, it helps us precisely diagnose and treat for desminopathy.Methods:Sanger sequencing was used to characterize DES variation, and then a minigene assay was used to verify the effect of splice-site mutation on pre-mRNA splicing.Phenotypes were analyzed based on clinical characteristics associated with desminopathy.Results:A splicing mutation (c.735+1G>T) in DES was detected in the proband.A minigene assay revealed skipping of the whole exon 3 and transcription of abnormal pre-mRNA lacking 32 codons.Another affected family member who carried the identical mutation, was identified with a novel phenotype of desminopathy, non-compaction of ventricular myocardium.There were 2 different phenotypes varied in cardiomyopathy and skeletal myopathy among the 2 patients, but no significant correlation between genotype and phenotype was identified.Conclusions:We reported a novel phenotype with a splicing mutation in DES, enlarging the spectrum of phenotype in desminopathy.Molecular studies of desminopathy should promote our understanding of its pathogenesis and provide a precise molecular diagnosis of this disorder, facilitating clinical prevention and treatment at an early stage.Peng Fan Chao-Xia Lu Xue-Qi Dong Di Zhu Kun-Qi Yang Ke-Qiang Liu Di Zhang Ying Zhang Xu Meng Hui-Qiong Tan Li-Tian YU Ke-Fei DOU Ya-Xin Liu Xue Zhang Xian-Liang Zhou 2019Chinese Medical Journal2019,,2:2
18Interface Engineering of CoS/CoO@N‑Doped Graphene Nanocomposite for High‑Performance Rechargeable Zn–Air Batteries显示文摘Low cost and green fabrication of high-performance electrocatalysts with earth-abundant resources for oxygen reduction reaction(ORR)and oxygen evolution reaction(OER)are crucial for the large-scale application of rechargeable Zn-air batteries(ZABs).In this work,our density functional theory calculations on the electrocatalyst suggest that the rational construction of interfacial structure can induce local charge redistribution,improve the electronic conductivity and enhance the catalyst stability.In order to realize such a structure,we spatially immobilize heterogeneous CoS/CoO nanocrystals onto N-doped graphene to synthesize a bifunctional electrocatalyst(CoS/CoO@NGNs).The optimization of the composition,interfacial structure and conductivity of the electrocatalyst is conducted to achieve bifunctional catalytic activity and deliver outstanding efficiency and stability for both ORR and OER.The aqueous ZAB with the as-prepared CoS/CoO@NGNs cathode displays a high maximum power density of 137.8 mW cm^−2,a specific capacity of 723.9 mAh g^−1 and excellent cycling stability(continuous operating for 100 h)with a high round-trip efficiency.In addition,the assembled quasi-solid-state ZAB also exhibits outstanding mechanical flexibility besides high battery performances,showing great potential for applications in flexible and wearable electronic devices.Yuhui Tian Li Xu Meng Li Ding Yuan Xianhu Liu Junchao Qian Yuhai Dou Jingxia Qiu Shanqing Zhang 2021Nano-Micro Letters2021,13,1:2
