2Dlg1 Knockout Inhibits Microglial Activation and Alleviates Lipopolysaccharide-Induced Depression-Like Behavior in Mice显示文摘Microglia-mediated neuroinflammation is widely perceived as a contributor to numerous neurological diseases and mental disorders including depression.Discs large homolog 1(Dlg1),an adaptor protein,regulates cell polarization and the function of K?channels,which are reported to regulate the activation of microglia.However,little is known about the role of Dlg1 in microglia and the maintenance of central nervous system homeostasis.In this study,we found that Dlg1 knockdown suppressed lipopolysaccharide(LPS)-induced inflammation by downregulating the activation of nuclear factor-jB signaling and the mitogen-activated protein kinase pathway in microglia.Moreover,using an inducible Dlg1 microglia-specific knockout(Dlg1flox/flox;CX3CR1CreER)mouse line,we found that microglial Dlg1 knockout reduced the activation of microglia and alleviated the LPS-induced depressionlike behavior.In summary,our results demonstrated that Dlg1 plays a critical role in microglial activation and thus provides a potential therapeutic target for the clinical treatment of depression.Zhixin Peng Xiaoheng Li Jun Li Yuan Dong Yuhao Gao Yajin Liao Meichen Yan Zengqiang Yuan Jinbo Cheng 2021Neuroscience Bulletin2021,37,12:2
3Sealing reliability modeling of aviation seal based on interval uncertainty method and multidimensional response surface显示文摘Many variables affect the sealing performance, and their distribution characteristics are difficult to obtain with probabilistic methods owing to the high cost involved. Numerous problems in engineering are similar due to the appearance of small-sample parameters. In this study, the sealing reliability of an aviation seal was defined as the research object, and an interval uncertainty method and multidimensional response surface were proposed to calculate the sealing reliability.Based on this, we first analyzed the failure mechanism of the aviation seal and established a leakage rate model. Then, based on the non-probabilistic interval model, an interval uncertainty method was proposed to construct the analytical model. With reference to the limit state equation from the structural reliability theory, the multidimensional response surface was used for fast calculation.Then, we chose the single-cylinder gas steering gear used in aircraft as the case study, its sealing reliability in working and non-working statuses were calculated, and the results were verified with the actual maintenance records. 4The Dynamic Analysis of Agro-ecological Economic System on the Basis of Emergy: A Case Study of Wu'an City in Hebei Province显示文摘Using the method of emergy analysis, we analyze the input and output of agroecological economic system, and select five indicators (net emergy yield ratio, emergy investment ratio, environmental loading ratio, emergy sustainability index, and dominance of emergy yield system) for assessment. The results show that the emergy input-output in Wu'an City is in general on the rise; the emergy investment ratio rises constantly, but the net emergy yield ratio decreases, and the comparative advantage in the prices of agricultural products is gradually lost. At the same time, with increase in the non-renewable industrial support emergy, the environmental pressures are also mounting. 5Regulation of hydrogen evolution performance of titanium oxide-carbon composites at high current density with a Ti-O hybrid orbital显示文摘Rational design and controllable synthesis of practical electrodes with high sta bility and activity at high current density for a hydrogen evolution reaction(HER)are critical for renewable and sustainable energy conversion.However,high-performance TiO_(2)-based electrocatalysts for HER are quite limited,and the cat alytic active centers still remain elusive.Herein,a simple strategy is demonstrated for the synthesis of TiO_(2)-carbon composite(TiO_(2)/C)with high HER performance and stability.The remarkable HER performance of TiO_(2)/C can be ascribed to the doping of carbon atoms,which leads to stronger hybridization of Ti 3d and O 2p orbitals,thus substantially improving the electrocatalytic efficiency.This study elucidates that the hydrogen evolution activity of oxide electrocatalysts can be largely improved by regulating their electronic structures by doping carbon atoms and also provides an effective strategy for designing heterostructured electro catalysts with high catalytic activity and stability at high current density for HER.Meichen Liu Zhiping Lei Qingping Ke Peixin Cui Jiancheng Wang Jingchong Yan Zhanku Li Hengfu Shui Shibiao Ren Zhicai Wang Ying Kong Shigang Kang 2022Carbon Energy2022,4,3:0
6Magnesium Demethylcantharidate induces apoptosis in hepatocellular carcinoma cells via ER stress显示文摘Cantharidin(CTD)is a bioactive ingredient isolated from Cantharis vesicatoria(blister beetles),which has potential therapeutic value as an anticancer agent.Magnesium Demethylcantharidate(MDC)is a recently developed derivative of Cantharidin(CTD),and previous studies have illustrated its excellent anticancer activity on HCC cells.However,the effect and mechanism of MDC remains unclear and need to be further studied.In particular,whether MDC can cause ER stress in HCC is still unknown.In this study,we demonstrated that endoplasmic reticulum stress(ERS)-related proteins were changed in SMMC-7721 and Bel-7402 cells after being exposed to MDC.Moreover,we found that MDC could significantly inhibit the growth of xenograft tumor in nude mice.In summary,we confirmed that MDC could induce ERS in HCC cells and thus induce apoptosis.XINTING ZHU MENG YE KELAN FANG FANG LIU JING HUI MEICHEN LIU XIAOFEI LI RONG YAN YUN LIU 2022BIOCELL2022,46,12:0
7Metabolomics analysis of poly(L-lactic acid)nanofibers’performance on PC12 cell differentiation显示文摘The aim of this article is to reveal the influence of aligned/random poly(L-lactic acid)(PLLA)nanofibers on PC12 cell differentiation from the perspective of metabolic level.First,three materials-PLLA aligned nanofibers(PLLA AF),PLLA random nanofibers(PLLA RF)and PLLA films(control)-were prepared by electrospinning and spin coating.Their surface morphologies were characterized.Subsequently,the cell viability,cell morphology and neurite length of PC12 cells on the surface of the three materials were evaluated,indicating more neurites in the PLLA RF groups but the longer average neurite length in the PLLA AF groups.Next,the metabolite profiles of PC12 cells cultured on the surface of the three nanofibers after 12 h,24 h and 36 h showed that,compared with the control,51,48 and 31 types of differential metabolites were detected at the three time points among the AF groups,respectively;and 56,45 and 41 types among the RF groups,respectively.Furthermore,the bioinformatics analysis of differential metabolites identified two pathways and three metabolites critical to PC12 cell differentiation influenced by the nanofibers.In addition,the verification experiment on critical metabolites and metabolic pathways were performed.The integrative analysis combining cytology,metabolomics and bioinformatics approaches revealed that though both PLLA AF and RF were capable of stimulating the synthesis of neurotransmitters,the PLLA AF were more beneficial for PC12 cell differentiation,whereas the PLLA RF were less effective.Xiaoman Su Yan Huang Rong Chen Yiwen Zhang Meichen He Xiaoying Lu 2021Regenerative Biomaterials2021,8,4:0
