| 2 | Targeting ERK,an Achilles' Heel of the MAPK pathway,in cancer therapy显示文摘The mitogen-activated protein kinases(MAPK) pathway, often known as the RAS-RAFMEK-ERK signal cascade, functions to transmit upstream signals to its downstream effectors to regulate physiological process such as cell proliferation, differentiation, survival and death. As the most frequently mutated signaling pathway in human cancer, targeting the MAPK pathway has long been considered a promising strategy for cancer therapy. Substantial efforts in the past decades have led to the clinical success of BRAF and MEK inhibitors. However, the clinical benefits of these inhibitors are compromised by the frequently occurring acquired resistance due to cancer heterogeneity and genomic instability. This review briefly introduces the key protein kinases involved in this pathway as well as their activation mechanisms. We also generalize the correlations between mutations of MAPK members and human cancers, followed by a summarization of progress made on the development of small molecule MAPK kinases inhibitors. | 3 | Interleukin 17-Producing γδ T Cells Increased in Patients with Active Pulmonary Tuberculosis显示文摘Although it has been known that γδ T cells may play an important role in the immune response to infection of Mycobacterium tuberculosis (M. tb), the mechanisms by which the γδ T cells participate in the innate and/or acquired immunity to tuberculosis (TB) have not been full elucidated. In the present study, 27 patients with active pulmonary TB and 16 healthy donors (HD) were performed. We found that proportion of IL-17-producing cells among lymphocyte was similar between TB patients and HD, whereas the proportions of γδ T cells in IL-17-producing cells (59.2%) and IL-17-producing cells in γδ T cells (19.4%) in peripheral blood were markedly increased in TB patients when compared to those in HD (43.9% and 7.7%, respectively). In addition, the proportions of IFN-γ-producing γδ T cells in TB patients were obviously lower than that in HD. Upon re-stimulated with M. tb heat-treated antigen (M. tb-HAg) in vitro, fewer IL-17-producing γδ T cells were generated from HD and TB patients, whereas IFN-γ-producing γδ T cells were increased in TB patients compared to that in HD. | 4 | Effects of rhubarb on isolated gastric muscle strips of guinea pigs显示文摘AIM: To study the effects of rhubarb (dried root of Rheum officinale Baill.) on contractile activity of isolated gastric muscle strips of guinea pigs and its possible mechanism.METHODS: A total of 48 guinea pigs were killed to remove the whole stomach. Then, the stomach was opened and the mucosal layer was removed. Parallel to the circular fibers, muscle strips were cut from the body. Each isolated gastric muscle strip was suspended in a tissue chamber containing 5 mL Krebs solution, constantly warmed by water jacket at 37 ℃ and bubbled continuously with a mixed gas of 950 mL/L O2 and 50 mL/L CO2. After being incubated for 1 h with 1 g tension, rhubarb of varied concentrations (1%, 2%, 7%, 20% and 70%) was added cumulatively into the tissue chamber at intervals of 2 min. Atropine (10-6 mol/L) or isoptin (5×10-8 mol/L) orhexamethonium(10-5 mol/L) was given 2 min before the administration of rhubarb. The isometrical response was measured with an ink-writing recorder.RESULTS: Rhubarb dose dependently increased the resting tension of gastric body circular muscle(CM)(r = 0.726, P<0.05). Atropine (r= 0.829, P<0.05), isoptin (r = 0.764,P<0.05) and hexamethonium (r = 0.797, P<0.05) did notaffect its action in a dose-related manner. Atropine apparently reduced the increasing action of 1%, 3%, 10%, 30% and 100% rhubarb on the resting tension of gastric body CM. Isoptin inhibited the effect of 10%, 30% and 100% rhubarb on the resting tension of gastric body CM. Hexamethonium reduced the increasing action of 1%, 10%, 30% and 100% rhubarb on the resting tension of gastric body CM. Rhubarb increased the contractile frequency of CM of body. While atropine, isoptin and hexamethonium did not inhibit the contractile frequency of gastric body CM in comparison with rhubarb at the same concentration, rhubarb at the highest concentration (100%) decreased the meancontractile amplitude of gastric body CM. Atropine, isoptin and hexamethonium did not affect the mean contractile amplitude of gastric body CM compared to rhubarb at the same concentration.CONCLUSION: Rhubarb has exciting actions on isolated gastric smooth muscle strips of guinea pig. | 7 | Effect of rhubarb on contractile response of gallbladder smooth muscle strips isolated from guinea pigs显示文摘AIM: To investigate the effect of rhubarb on contractile response of isolated gallbladder muscle strips from guinea pigs and its mechanism.METHODS: Guinea pigs were killed to remove the whole gallbladder. Two or three smooth muscle strips (8 mm×3mm) were cut along the longitudinal direction. The mucosa on each strip was carefully removed. Each longitudinal muscle strip was suspended in a tissue chamber containing 5 mL Krebs solution (37 ℃), bubbled continuously with 950 mL/L O2 and 50 mL/L CO2. The resting tension (g), mean contractile amplitude (mm),and contractile frequency (waves/min) were simultaneously recorded on recorders. After 2-h equilibration, rhubarb (10, 20, 70, 200, 700, 1 000 g/L) was added cumulatively to the tissue chamber in turns every 2 min to observe their effects on gallbladder.Antagonists were given 3 min before administration of rhubarb to investigate the possible mechanism.RESULTS: Rhubarb increased the resting tension (from 0 to 0.40±0.02, P<0.001), and decreased the mean contractile amplitude (from 5.22±0.71 to 2.73±0.41,P<0.001). It also increased the contractile frequency of the gallbladder muscle strips in guinea pigs (from 4.09±0.46to 6.08±0.35, P<0.001). The stimulation of rhubarb on the resting tension decreased from 3.98±0.22 to 1.58±0.12by atropine (P<0.001), from3.98±0.22 to 2.09±0.19 by verapamil (P<0.001) and from 3.98±0.22 to 2.67±0.43by phentolamine (P<0.005). But the effect was not inhibited by hexamethonium (P>0.05). In addition, the action of mean amplitude and frequency was not inhibited by the above antagonists.CONCLUSION: Rhubarb can stimulate the motility of isolated gallbladder muscle strips from guinea pigs. S1 P either activates S1 P receptors(S1PR1-5) through 'inside-out signaling' or acts directly on intracellular targets to regulate various cellular processes. In the past two decades, much progress has been made in exploring S1 P signaling and its pathogenic roles in diseases as well as in developing modulators of S1 P signaling, including S1 P agonists, S1 P antagonists and sphingosine kinase(SphK) inhibitors.Ceramide and S1 P have been defined as reciprocal regulators of cell fate, and S1 P signaling has been shown to be crucial for the pathogenesis of various diseases, including autoimmune diseases, inflammation and cancer; therefore, targeting S1 P signaling may curtail the process of pathogenesis and serve as a potential therapeutic target for the treatment of these diseases. | 15 | Antidepressant-like effect of active fraction of Polyrhachisvicina Roger in a rat depression model显示文摘OBJECTIVE: To investigate the antidepressant-likeeffect of active fraction of Polyrhachis vicina Roger(AFPR) in a rat depression model, and to elucidate the underlying mechanism.METHODS: AFPR was extracted with ethanol followed by petroleum ether. Its antidepressant-like effect was investigated in mice by tail suspension test(TST), forced swimming test(FST) and open field test(OPT). A repeated dose of reserpine(0.5 mg/kg, daily for 14 d) was used to establish a rat depression model. Fluoxetine was used as positive control agent. The effect of AFPR on reserpine-induced ptosis, hypothermia and akinesia, the levels of monoamines and their metabolites, and the activity of monoamine oxidase(MAO) in hippocampus and prefrontal cortex were determined.RESULTS: Administration of AFPR by gavage at 160 and 320 mg/kg significantly reduced the duration of immobility in the FST and TST, and did not affect locomotor activity in the OPT. In the reserpine-induced depression model, AFPR attenuated anhedonia, demonstrated by reversing hypothermia, akinesia and sucrose consumption. AFPR significantly increased the concentration of monoamines, including dopamine, serotonin, noradrenaline and acetylcholine.CONCLUSION: AFPR normalized the metabolism rates of noradrenaline, serotonin and dopamine,and the activity of MAO, which were altered by chronic reserpine exposure. | 18 | Self-adaptive spatial image denoising model based on scale correlation and SURE-LET in the nonsubsampled contourlet transform domain显示文摘A novel self-adaptive image denoising model based on scale correlation and Stein’s unbiased risk estimate-linear expansion of thresholds(SURE-LET)in the nonsubsampled contourlet transform domain is proposed in this paper.First we implement the multidimensional and translation invariant decomposition for spatial images by the nonsubsampled contourlet transform,and establish the image cross-scale description structure.Then combining the scale correlation,we make improvements for the existing SURE-LET denoising idea and establish the self-adaptive denoising mechanism.The scale correlation calculation is needed for the coefficients at different scales and sub-bands to determine whether the coefficients are retained or processed with the adaptive SURE-LET threshold shrinkage.And meanwhile a new local context self-adaptive threshold strategy is proposed in the process of scale correlation calculation.Experimental results both on spatial images and standard images demonstrate that the proposed algorithm performs significantly better in terms of both the visual subjective evaluation and the quantitative objective evaluation.The method can achieve better noise suppression,and effectively retain image edge details. | LIANG MeiYu DU JunPing LIU HongGang | 2014 | Science China(Information Sciences)2014,57,9: | 4 |
| 19 | Efficacy of Lidan Tang on high-fat-diet induced gallstone in mice and possible mechanism显示文摘OBJECTIVE:To investigate efficacy of Lidan Tang(LDT)on gallstone induced by high fat diet in mice,and to study its underlying mechanism.METHODS:Mice were fed with high fat diet every day and treated with LDT(9.01 times of human clinic dosage).Mice were randomly divided into 6 groups as control group,gallstone model group(high-fat diet),positive control ursodeoxycholic acid(UDCA)group(80 mg·kg^-1·d^-1,i.g.),LDT low dose group(6 kg/d,i.g.),LDT middle dose group(12 kg/d,i.g.),and LDT high dose group(24 kg/d,i.g.).The whole experiment was lasted for 4 weeks.The levels of ALT,AST,LDH,CHO,HDL-C and LDL-C in serum were measured,the pathological sections were observed by hematoxylin-eosin staining,the activities of antioxidant enzymes were measured by kits,and the proteins related to oxidative stress and lipid transport were detected by Western blot analysis.RESULTS:LDT could significantly reduce the contents of ALT and AST in serum and improve the pathological tissue of liver.LDT could significantly reduce the content of MDA and LPO,and increase the level of GSH and GSH-PX in liver tissue.The data of Western blot showed that LDT had antioxidant effect promoting Keap1/Nrf2 pathway and regulated the process of lipid transport,which was statistically significant.In addition,LDT treatment inhibited the expression of ATP-binding cassette transports ABCG5/8 in liver,and reduced cholesterol transport from the hepatocytes to the gallbladder.CONCLUSION:LDT has protective effect on gallstones induced by high fat diet in mice,which might be based on the protective effect on liver,including enhancing the antioxidant capacity of liver and reducing the production of lipid peroxides. | Gao Yan Liu Caipin Li Juntong Zhai Yingying Lin Meiyu Wu Qinglin Chu Shifeng Zhang Zhao Li Jianping Zhou Xin Li Yueting Chen Naihong | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 4 |
| 20 | Spatial Analysis of Commuting Mode Choice in Guangzhou, China显示文摘Metropolitan cities in China are commonly confronted with unresolved traffic congestion issues, primarily due to rapidly increasing traffic demand. Group disparity between commuting mode choice and its spatial distribution on road networks has enabled us to examine the factors that give rise to the discrepancies and the fundamental spatial causes of traffic congestion. In recent years, micro-perspective, individual, and behavior-based spatial analysis have mushroomed and been facilitated with effective tools such as temporal geographic information systems (T-GIS). It is difficult to study the interrelations between transport and space on the basis of commuting mode choice since the mode choice data are invisible in a specific space such as a particular road network. Therefore, in the field of transport, the classical origin destination (OD) four-stage model (FSM) is usually employed to calculate data when studying commuting mode choice. Based on the relative principles of T-GIS and the platform of ArcGIS, this paper considers Guangzhou as a case study and develops a spatio-temporal tool to examine the daily activities of residents. Meanwhile, the traffic volume distribution in rush hours, which was analyzed according to commuting modes and how they were reflected in the road network, was scrutinized with data extracted from travel diaries. Moreover, efforts were made to explain the relationship between traffic demand and urban spatial structure. Based on the investigation, this research indicates that traffic volumes in divergent groups and on the road networks is driven by: 1) the socio-economic characteristics of travelers; 2) a jobs-housing imbalance under suburbanization; 3) differences in the spatial supply of transport modes; 4) the remains of the Danwei (work unit) system and market development in China; and 5) the transition of urban spatial structure and other factors. | ZHOU Suhong DENG Lifang HUANG Meiyu | 2013 | Chinese Geographical Science2013,23,3: | 4 |
