3Lower Bifidobacteria counts in both duodenal mucosa-associated and fecal microbiota in irritable bowel syndrome patients显示文摘AIM: To determine the composition of both fecal and duodenal mucosa-associated microbiota in irritable bowel syndrome (IBS) patients and healthy subjects using molecular-based techniques. METHODS: Fecal and duodenal mucosa brush samples were obtained from 41 IBS patients and 26 healthy subjects. Fecal samples were analyzed for the composition of the total microbiota using fluorescent in situ hybridization (FISH) and both fecal and duodenal brush samples were analyzed for the composition of bif idobacteria using real-time polymerase chain reaction. RESULTS: The FISH analysis of fecal samples revealed a 2-fold decrease in the level of bifidobacteria (4.2 ± 1.3 vs 8.3 ± 1.9, P < 0.01) in IBS patients compared to healthy subjects, whereas no major differences in other bacterial groups were observed. At the species level, Bifidobacterium catenulatum levels were significantly lower (6 ± 0.6 vs 19 ± 2.5, P < 0.001) in the IBS patients in both fecal and duodenal brush samples than in healthy subjects.CONCLUSION: Decreased bifidobacteria levels in both fecal and duodenal brush samples of IBS patients compared to healthy subjects indicate a role for microbiotic composition in IBS pathophysiology.Angèle PM Kerckhoffs Melvin Samsom Michel E van der Rest Joris de Vogel Jan Knol Kaouther Ben-Amor Louis MA Akkermans 2009World Journal of Gastroenterology2009,15,23:33

8Applicability of five models to simulate water infiltration into soil with added biochar显示文摘As a soil amendment, biochar can reduce soil bulk density, increase soil porosity, and alter soil aggregates and thus affect the infiltration. Researchers have proposed and revised several theoretical models to describe the process of soil infiltration. Although these models have been successfully used to evaluate the soil infiltration in different scenarios in agricultural fields, little effort has been devoted to assess their performances in arid and semi-arid soils after the addition of biochar. A laboratory experiment was performed to study the infiltration characteristics of two typical Loess Plateau soils at three particle sizes(2–1, 1–0.25, and <0.25 mm) and five biochar application amounts(0, 10, 50, 100, and 150 g/kg). The performance of five models(i.e., the Philip model, Kostiakov model, Mezencev model, USDA-NRCS model, and Horton model) in simulating the infiltration process was then evaluated based on the adjusted coefficient of determination and a reduced Chi-Square test. Results indicated that the Horton model best simulated the water-infiltration process in an aeolian sandy soil with added biochar. However, the Mezencev model best simulated the infiltration process in a loamy clay soil(Eum-Orthic Anthrosol). The three-parameter model, i.e., Mezencev and Horton models can better describe the relationship between cumulative infiltration and infiltration time. 9Sol-gel derived HA/TiO_2 double coatings on Ti scaffolds for orthopaedic applications显示文摘Hydroxyapatite/titania (HA/TiO2) double layers were coated onto Ti scaffolds throughout for orthopaedic applications by sol-gel method. Differential scanning calorimetry (DSC), thermogravimetric analysis (TG) and X-ray diffractometry (XRD) were used for the characterisation of the phase transformations of the dried gels and coated surface structures. Scanning electron microscope (SEM) equipped with energy dispersive spectrometry (EDS) was used for the observation and evaluation of the morphology and phases of the surface layers and for the assessment of the in vitro tests. The in vitro assessments were performed by soaking the HA/TiO2 double coated samples into the simulated body fluid (SBF) for various periods. The TiO2 dipping-coating method at a speed of 12 cm/min, followed by a heat treatment at 600 ℃ for 20 min. The HA la lyaeyre wr wasa ssu cbosaetqeude bnytl ya dipping-coated on the outer surface at the same speed and then heat-treated at difference temperatures. The results indicat that the HA phase begins to crystallize after a heat treatment at 560 ℃. The crystallinity increases obviously at 760 ℃. SEM observations find no delamination or crack at the interfaces of HA/TiO2 and TiO2/Ti. The HA/TiO2 coated Ti scaffolds displays excellent bone-like apatite forming ability when it is soaked into SBF. 11^(226)Ra evidence for the ecosystem shift over the past 40 years in the North Pacific Subtropical Gyre显示文摘Surface seawater was collected for ^(226)Ra measurement in the North Pacific Subtropical Gyre from July to October, 1999 and October to December, 2003. Combined with the historical data reported for this sea area, a declined trend of surface ^(226)Ra concentrations was observed since 1960s, indicating the ecosystem shift in response to global warming. On one side, the enhanced stratification of the upper water column resulting from global warming reduced the ^(226)Ra input from the depth, on the other, the temporal increase of biological production resulting from the climate-related ecosystem structure change strengthened the ^(226)Ra removal from the surface ocean. Both the physical and biological processes resulted in the decrease of surface ^(226)Ra concentrations in the North Pacific Subtropical Gyre. The temporal trend of surface ^(226)Ra concentrations was consistent with the trends of chlorophyll a, silicate, phosphate and primary production previously reported. 13Influence of particle composition on thorium scavenging in the marginal China seas显示文摘Thorium-234 and particle composition (organic matter, biogenic silica, carbonate and lithogenic component) were examined in the East China Sea (ECS) and the northern South China Sea (NSCS) in order to constrain the particle types scavenging thorium isotopes. Good positive correlations be-tween particulate organic matter (POM) or carbonate and thorium-234 in suspended particulate matter (SPM) indicates that POM and carbonate are efficient to scavenge thorium-234. No relation-ship between biogenic silica and thorium-234 suggests that geochemical behavior of thorium-234 may be not influenced by biogenic silica. A simple model was used to evaluate the affinity of thorium-234 to different particle components. The results show that POM is the most efficient scavenger for thorium-234 in the ECS and the NSCS, followed by carbonate. The authors’ re-sults lend support to the utility of thorium-234 as a proxy of POC and carbonate in the upper layer. 184-Phenylbutyric acid accelerates rehabilitation of barrier function in IPEC-J2 cell monolayer model显示文摘As the first line of defence against pathogens and endotoxins crossing the intestine-blood barrier,the intestinal epithelial barrier plays a determinant role in pigs’health and growth.4-Phenylbutyric acid(4-PBA),an aromatic fatty acid,was reported to benefit homeostasis of endoplasmic reticulum and protein synthesis.However,whether 4-PBA affects intestinal epithelial barrier function in pigs is unknown.This study aimed to explore the effects of 4-PBA on the intestinal barrier function,using in vitro models of well-differentiated intestinal porcine epithelial cell(IPEC-J2)monolayers in the transwell plates.Cell monolayers with or without 4-PBA(1.0 mmol/L)treatment were challenged with physical scratch,deoxynivalenol(DON,2.0μg/mL,48 h),and lipopolysaccharide(LPS,5.0μg/mL,48 h),respectively.Transepithelial electrical resistance(TEER)and fluorescein isothiocyanate-dextran(FD-4)permeability were measured to indicate barrier integrity and permeability.Real-time PCR and Western blot were conducted to determine relative gene and protein expressions of tight junction proteins.As expected,physical scratch,DON,and LPS challenges decreased TEER and increased FD-4 permeability.4-PBA treatment accelerated cell mitigation and rehabilitation of the physical scratch-damaged intestinal epithelial barrier but did not alleviate DON or LPS induced barrier damage.However,once 48-h DON and LPS challenges were removed,rehabilitation of the epithelial barrier function of IPEC-J2 monolayer was accelerated by the 4-PBA treatment.Also,the relative gene and protein expressions of zonula occludens-1(ZO-1),occludin,and claudin-1 were further upregulated by the 4-PBA treatment during the barrier rehabilitation.Taken together,4-PBA accelerated the IPEC-J2 cell monolayer barrier recovering from physical scratch,DON-,and LPS-induced damage,via enhancing cell mitigation and expressions of tight junction proteins.Qian Jiang Jie Yin Jiashun Chen Xiaokang Ma Miaomiao Wu Xilong Li Kang Yao Bi'e Tan Yulong Yin 2021Animal Nutrition2021,,4:2

20Finite element modeling assumptions impact on seismic response demands of MRF-buildings显示文摘Recent seismic events have raised concerns over the safety and vulnerability of reinforced concrete moment resisting frame 'RC-MRF' buildings. The seismic response of such buildings is greatly dependent on the computational tools used and the inherent assumptions in the modelling process. Thus, it is essential to investigate the sensitivity of the response demands to the corresponding modelling assumption. Many parameters and assumptions are justified to generate effective structural finite element(FE) models of buildings to simulate lateral behaviour and evaluate seismic design demands. As such, the present study focuses on the development of reliable FE models with various levels of refinement. The effects of the FE modelling assumptions on the seismic response demands on the design of buildings are investigated. the predictive ability of a FE model is tied to the accuracy of numerical analysis; a numerical analysis is performed for a series of symmetric buildings in active seismic zones. The results of the seismic response demands are presented in a comparative format to confirm drift and strength limits requirements.
