3Effect of high-intensity interval training in adolescents with asthma:The eXercise for Asthma with Commando Joe’s^(■)(X4ACJ)trial显示文摘Background:Higher levels of cardiorespiratory fitness are associated with reduced asthma severity and increased quality of life in those with asthma.Therefore,the purpose of this study was to evaluate the effectiveness of a 6-month high-intensity interval training(HIIT)intervention in adolescents with and without asthma.Methods:A total of 616 adolescents(334 boys;13.0±1.1 years,1.57±0.10 m,52.6±12.9 kg,mean±SD),including 155 with asthma(78 boys),were recruited as part of a randomized controlled trial from 5 schools(4 control and 1 intervention).The 221 intervention participants(116 boys;47 asthma)completed 6 months of school-based HIIT(30 min,3 times per week,10-30 s bouts at>90%age-predicted maximum heart rate with equal rest).At baseline,mid-intervention,post-intervention,and 3-month follow-up,measurements for 20-m shuttle run,body mass index(BMI),lung function,Pediatric Quality of Life Inventory,Paediatric Asthma Quality of Life Questionnaire,and Asthma Control Questionnaire were collected.Additionally,69 adolescents(39 boys(of the 36 with asthma there were 21 boys))also completed an incremental ramp test.For analysis,each group’s data(intervention and control)were divided into those with and without asthma.Results:Participants with asthma did not differ from their peers in any parameter of aerobic fitness,at any time-point,but were characterized by a greater BMI.The intervention elicited a significant improvement in maximal aerobic fitness but no change in sub-maximal parameters of aerobic fitness,lung function,or quality of life irrespective of asthma status.Those in the intervention group maintained their BMI,whereas BMI significantly increased in the control group throughout the 6-month period.Conclusion:HIIT represents an effective tool for improving aerobic fitness and maintaining BMI in adolescents,irrespective of asthma status.HIIT was well-tolerated by those with asthma,who evidenced a similar aerobic fitness to their healthy peers and responded equally well to a HIIT program.Charles O.N.Winn Kelly A.Mackintosh William T.B.Eddolls Gareth Stratton Andrew M.Wilson Melitta A.McNarry Gwyneth A.Davies 2021Journal of Sport and Health Science2021,10,4:1

6Non-small cell lung carcinoma(NSCLC):Implications on molecular pathology and advances in early diagnostics and therapeutics显示文摘Continuous revision of the histologic and stage-wise classification of lung cancer by the World Health Organization (WHO) provides the foundation for therapeutic advances by promoting molecular targeted and immunotherapies and ensuring accurate diagnosis. Cancer epidemiologic data provide helpful information for cancer prevention, diagnosis, and management, supporting health-care interventions. Global cancer mortality projections from 2016 to 2060 show that cancer will overtake ischemic heart diseases (IHD) as the leading cause of death (18.9 million) immediately after 2030, surpassing non-small cell lung cancer (NSCLC), which accounts for 85 percent of lung cancers. The clinical stage at the diagnosis is the main prognostic factor in NSCLC therapies. Advanced early diagnostic methods are essential as the initial stages of cancer show reduced mortality compared to the advanced stages. Sophisticated approaches to proper histological classification and NSCLC management have improved clinical efficiency. Although immune checkpoint inhibitors (ICIs) and targeted molecular therapies have refined the therapeutic management of late-stage NSCLC, the specificity and sensitivity of cancer biomarkers should be improved by focusing on prospective studies, followed by their use as therapeutic tools. 10Quantitative trait loci controlling Phytophthora cactorum resistance in the cultivated octoploid strawberry(Fragaria×ananassa)显示文摘The cultivated strawberry,Fragaria×ananassa(Fragaria spp.)is the most economically important global soft fruit.Phytophthora cactorum,a water-borne oomycete causes economic losses in strawberry production globally.A biparental cross of octoploid cultivated strawberry segregating for resistance to P.cactorum,the causative agent of crown rot disease,was screened using artificial inoculation.Multiple putative resistance quantitative trait loci(QTL)were identified and mapped.Three major effect QTL(FaRPc6C,FaRPc6D and FaRPc7D)explained 37%of the variation observed.There were no epistatic interactions detected between the three major QTLs.Testing a subset of the mapping population progeny against a range of P.cactorum isolates revealed no significant interaction(p=0.0593).However,some lines showed higher susceptibility than predicted,indicating that additional undetected factors may affect the expression of some quantitative resistance loci.Using historic crown rot disease score data from strawberry accessions,a preliminary genome-wide association study(GWAS)of 114 individuals revealed an additional locus associated with resistance to P.cactorum.Mining of the Fragaria vesca Hawaii 4 v1.1 genome revealed candidate resistance genes in the QTL regions.Charlotte F.Nellist Robert J.Vickerstaff Maria K.Sobczyk César Marina-Montes Fiona M.Wilson David W.Simpson Adam B.Whitehouse Richard J.Harrison 2019Horticulture Research2019,6,1:1

18AB018.Ocular hypertension promotes early mitochondrial fragmentation in retinal endothelial cells in a mouse model of glaucoma显示文摘Background:Retinal endothelial cells are very active and contribute to the integrity of the neurovascular unit.Vascular dysfunction has been proposed to contribute to the pathogenesis of glaucoma.Here,we evaluated the hypothesis that ocular hypertension triggers mitochondrial alterations in endothelial cells impairing the integrity of the blood retinal barrier(BRB).Methods:Ocular hypertension was induced by injection of magnetic microbeads into the anterior chamber of EndoMito-EGFP mice,a strain expressing green fluorescent protein selectively in the mitochondria of endothelial cells.Capillary density,mitochondrial volume,and the number of mitochondrial components were quantified in 3D-reconstructed images from whole-mounted retinas using Imaris software.Dynamin-related protein(DRP-1),mitofusin-2(MFN-2)and optic atrophy-1(OPA-1)expression were assessed by western blot analysis of enriched endothelial cells.Mitochondrial structure was evaluated by transmission electron microscopy(TEM)and oxygen consumption rate was monitored by Seahorse analysis.The integrity of the BRB was evaluated by quantifying Evans blue leakage.Results:Our data demonstrate that two and three weeks after ocular hypertension induction,the total mitochondria volume in endothelial cells decreased from 0.140±0.002µm3 from non-injured retinas to 0.108±0.005 and 0.093±0.007µm3,respectively in glaucomatous eyes(mean±S.E.M,ANOVA,P<0.001;N=6/group).Frequency distribution showed a substantial increase of smaller mitochondria complexes(<0.5µm3)in endothelial cells from glaucomatous retinas.Significant upregulation of DRP-1 was found in vessels isolated from glaucomatous retinas compared to the intact retinas,while MFN-2 and OPA-1 expression was not affected.Structural alteration in endothelial cell mitochondria was confirmed by TEM,which were accompanied by a 1.93-fold reduction in the oxygen consumption rate as well as 2.6-fold increase in vasculature leakage in glaucomatous retinas(n=3-6/group).In addition,this model did not trigger changes in the density of the vascular network,suggesting that mitochondrial fragmentation was not due to endothelial cell loss.Conclusions:This study shows that ocular hypertension leads to early alterations in the dynamic of endothelial cell mitochondria,contributing to vascular dysfunction in glaucoma.Jorge L.Cueva Vargas Yoko Ito Khalil Miloudi Ariel M.Wilson Christine Vande Velde Przemyslaw Sapieha Adriana Di Polo 2018Annals of Eye Science2018,,1:0

In vivo laser-mediated retinal ganglion cell optoporation using Kv1.1 conjugated gold nanoparticles显示文摘Background:There is a current void in efficient,cell-specific,retinal drug delivery systems,thus developing a safe,effective,selective drug delivery system would open novel therapeutic avenues.We previously demonstrated that femtosecond(fs)laser irradiation can transfect DNA plasmids into cultured cells in the presence of gold nanoparticles(AuNPs).These AuNPs locally amplify laser energy at a submicron range creating transient pores allowing exogenous genetic material or cell impermeable dyes to enter the cell.Here,we sought out to selectively optoporate retinal cells in vivo with functionalized AuNPs and a 800 nm femtosecond(fs)laser.Methods:The cell-surface Kv1.1 voltage-gated channel was chosen to selectively target retinal ganglion cells(RGC)in the rat retina.Citrate-capped spherical 100 nm AuNPs functionalized with orthopyridyl-disulfide-poly(ethylene glycol)(5 kDa)-N-hydroxysuccinimide(OPSS-PEG-NHS)conjugated to a Kv1.1 monoclonal antibody were injected intravitreally in Sprague Dawley rats 3 hours prior to irradiation,concomitantly to a FITC-dextran dye to detect optoporation.The eyes of anesthetized rats were placed in the beam path of a laser system consisting of an 800 nm,100 fs laser and a Heidelberg Spectralis HRA ophthalmoscope for fundus visualization.The rat retina was irradiated at powers ranging from 20-750 mW,the eyes fixed in 4%paraformaldehyde,dissected,rinsed,mounted and imaged by confocal microscopy.Results:Our novel laser system coupled to a Heidelberg ophthalmoscope allowed for a clear visualisation of the rat ocular fundus.A timecourse of AuNP intravitreal injections revealed that optimal nanoparticle dispersion on the retinal surface occurred at 3 hours post injection.Following Kv1.1-AuNP and FITC-dextran intravitreal injection and incubation,irradiation at 120-750 mW resulted in FITC uptake by retinal cells.Conclusions:Since living biological tissues absorb energy very weakly at 800 nm,this non-invasive tool may provide a safe,cost effective clinically relevant approach to selectively target retinal cells and limit complications associated with surgical interventions,and potential biological hazards associated with viral-based gene therapy.Ariel M.Wilson Javier Mazzaferri Éric Bergeron Sergiy Patskovsky Santiago Costantino Michel Meunier Przemyslaw Sapieha 2018Annals of Eye Science2018,,1:0
