4Effect of Salinity on Hemolymph Osmotic Pressure,Sodium Concentration and Na^+-K^+-ATPase Activity of Gill of Chinese Crab,Eriocheir sinensis显示文摘The effects of salinity on hemolymph osmotic pressure, Na+ concentration and Na+-K+-ATPase activity of gill of Chinese crab Eriocheir sinensis were studied. The results showed that hemolymph osmotic pressure and Na+ concentration increased signifi- cantly (P<0.05), and the Na+-K+-ATPase activity of gills decreased significantly (P<0.05) when salinity increased from 0 to 16. The hemolymph osmotic pressure and Na+ concentration in each treatment group rose remarkably at 0.125 d or 0.25 d, while the Na+-K+-ATPase activity of gill reduced gradually with increased experiment time in 3 d. Then the three parameters remained at a constant level after 0.25 d, 0.125 d and 3 d, respectively, and higher hemolymph osmotic pressure, higher Na+ concentration and lower Na+-K+-ATPase activity of gill occurred at higher salinity. 5Effect of Salinity on the Biosynthesis of Amines in Litopenaeus vannamei and the Expression of Gill Related Ion Transporter Genes显示文摘This study examined the effect of salinity on the expression of Na+/K+-ATPase(NKA) α-subunit and vacuolar-type H+-ATPase(V-ATPase) β-subunit gene in the gill of Litopenaeus vannamei. Semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) assay showed that the expression of NKA α-subunit and V-ATPase β-subunit gene was significantly influenced by salinity. It was found that the NKA activity significantly varied with salinity in time and dose dependent manner; whereas the V-ATPase activity did not. The abundance of NKA α-subunit gene transcript increased rapidly when the salinity decreased from 26 b to 21, and slowly when the salinity decreased from 26 to 31 within the first 24 h. When the salinity decreased from 26 to 21, the transcription of NKA α-subunit gene in gill epithelium was higher at 12 h than that at 0 h, which was consistent with the result of immunoblotting assay of NKA α-subunit. In addition, salinity had a significant time- and dose-dependent effect on the concentration of biogenic amines in both hemolymph and gill. As compared to other parameters, the concentration of dopamine(DA) and 5-hydroxytryptamine(5-HT) varied in different patterns when the salinity decreased from 26 to 21 or increased from 26 to 31, suggesting that DA and 5-HT played different regulatory roles in osmotic adaption and modulation of shrimp when salinity varies.PAN Luqing LIU Hongyu ZHAO Qun 2014Journal of Ocean University of China2014,13,3:5
6Levels of Metabolic Enzymes and Nitrogenous Compounds in the Swimming Crab Portunus trituberculatus Exposed to Elevated Ambient Ammonia-N显示文摘This study investigated the effects of ammonia-N(control and 1 and 5 mg L^(-1)) on the changes of metabolic enzymes activities and nitrogenous compounds levels in the gills, hepatopancreas, muscles, and hemolymph of the swimming crab Portunus trituberculatus. Results showed that the levels of hemolymph ammonia increased significantly at 6 h and then decreased to control level at 12 h. The activities of key enzymes involved in biosynthesis pathways of glutamine(Gln) and urea, such as glutamate dehydrogenase(GDH), glutamine synthetase(GS), and arginase(ARG), increased remarkably in different tissues of P. trituberculatus. Gln level was the highest in muscles, while urea content was the largest in hepatopancreas. The m RNA expression level of xanthine dehydrogenase(XDH) was downregulated sharply in gills but upregulated in the hepatopancreas and muscles. A small amount of uric acid was detected in the hemolymph. This result suggests that Gln in muscles may be used as an intermediary nitrogen repository, while urea may be primarily synthesized in the hepatopancreas and then transferred to other tissues via the hemolymph. Uric acid could be synthesized by the anabolism of purine nucleotides, in additon to the inhibitory activities of gills. In addition, the levels of Gln, urea and uric acid exhibited dose-and time-dependent effects with ammonia content. 7Effects of Mercuric Chloride on Antioxidant System and DNA Integrity of the Crab Charybdis japonica显示文摘Mercury (Hg) is one of the commonly encountered heavy metals, which is widespread in inshore sediments of China. In order to investigate the toxicity of Hg on marine invertebrates, we studied the effects of the divalent mercuricion (Hg2+) (at two final concentrations of 0.0025 and 0.0050 mg L-1, prepared with HgCl2) on metallothionein (MT) content, DNA integrity (DNA strand breaks) and catalase (CAT) in the gills and hepatopancreas, antioxidant enzyme activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), in the hemolymph, gills and hepatopancreas of the portunid crab Charybdis japonica for an experiment period up to 15 d. The results indicated that MT was significantly induced after 3 d, with a positive correlation with Hg2+ dose and time in the hepatopancreas and a negative correlation with Hg2+ dose and time in the gills. While CAT in the hemolymph was not detected, it increased in the hepatopancreas during the entire experiment; SOD and GPx in the three tissues were stimulated after 12 h, both attained peak value and then reduced during the experimental period. Meanwhile, DNA strand breaks were all induced significantly after 12 h. 8Effects of Heavy Metal Ions (Cu^(2+), Pb^(2+) and Cd^(2+)) on DNA Damage of the Gills, Hemocytes and Hepatopancreas of Marine Crab, Charybdis japonica显示文摘There are rising concerns about the hazardous effects of heavy metals on the environment. In this study, comet assay and DNA alkaline unwinding assay were conducted on the tissues (gills, hepatopancreas, and hemocytes) of Charybdis japonica in order to illustrate genotoxicity of three heavy metal ions (Cu2+, Pb2+, and Cd2+) on the marine crabs C. japonica. The crabs were exposed to Cu2+ (10, 50, and 100 ?g.L?1), Pb2+ (50, 250, and 500 ?g L?1) and Cd2+ (5, 25, and 50 ?g L?1), and the tissues were sampled at days 0.5, 1, 3, 6, 9, and 15. DNA alkaline unwinding assay was used for testing the DNA single strand break in gills and hepatopancreas and comet assay was employed for testing the DNA damage in hemocytes. The results showed that the DNA damage (F-value) of gills in the crabs exposed to the three heavy metals was decreased gradually during the exposure periods and there was a dose-time response relationship in certain time, suggesting that the levels of DNA single strand break in all the experimental groups increased significantly compared to the controls. Changes of F-value in hepatopancreas of the crabs exposed to the three heavy metals were similar to those in gills except that the peak values were found in the 500 ?g L?1 Pb2+ treatment group at day 3 and the 50 ?g L?1 Cd2+ treatment group at day 9. The ranks of DNA damage in gills and hepatopancreas induced by the three heavy metal ions (50 ?g L?1, day 15) were Cd2+ >Pb2+ >Cu2+ and Pb2+ >Cu2+ >Cd2+. The levels of DNA damage in gills were higher than those in hepatopancreas in the same experimental group. 9Effects of Hypoxia on Dopamine Concentration and the Immune Response of White Shrimp (Litopenaeus vannamei)显示文摘Effects of hypoxia on the dopamine concentration and the immune response of White Shrimp Litopenaeus vannamei were studied. The results showed that hypoxia had significant effects on the concentration of dopamine (DA) in the haemolymph, haemocyte count, phenoloxidase activity, phagocytic activity of haemocytes and bacteriolytic and antibacterial activity in the haemolymph (P<0.05). The concentration of the dopamine in haemolymph reached its maximum in the 3.0 and 1.5 mg L-1 DO groups at 12 h and 6 h, and then returned to normal after 24 h and 12 h, respectively. All immune parameters decreased with the reduction of dissolved oxygen. Total haemocyte count (THC), the hyaline cells and semi-granular cells in the 3.0 mg L-1 DO group became stable after 12 h, while granular cells did so after 24 h. The THC and different haemocyte count (DHC) in the 1.5 mg L-1 DO group became stable after 24 h. Phenoloxidase activity and bacteriolytic activity in the 3.0 and 1.5 mg L-1 DO groups reached their stable levels after 24 h and 12 h respectively, while phagocytic activity and antibacterial activity became stable after 24 and 12, and 36 and 24 h, respectively. 10Effect of Benzo[a]pyrene on Detoxification and the Activity of Antioxidant Enzymes of Marine Microalgae显示文摘The objective of this study was to examine the effect of benzo[a]pyrene(Ba P) on the detoxification and antioxidant systems of two microalgae,Isochrysis zhanjiangensis and Platymonas subcordiformis.In our study,these two algae were exposed to Ba P for 4 days at three different concentrations including 0.5 μg^(L-1)(low),3 μg^(L-1)(mid) and 18 μg^(L-1)(high).The activity of detoxification enzymes,ethoxyresorufin O-deethylase(EROD) and glutathione S-transferase(GST) increased in P.subcordiformis in all Ba P-treated groups.In I.zhanjiangensis,the activity of these two enzymes increased at the beginning of exposure,and then decreased in the groups treated with mid-and high Ba P.The activity of antioxidant enzyme superoxide dismutase(SOD) increased in I.zhanjiangensis in all Ba P-treated groups,and then decreased in high Ba P-treated group,while no significant change was observed in P.subcordiformis.The activity of antioxidant enzyme catalase(CAT) increased in I.zhanjiangensis and P.subcordiformis in all Ba Ptreated groups.The content of malondialdehyde(MDA) in Isochrysis zhanjiangensis increased first,and then decreased in high Ba P-treated group,while no change occurred in P.subcordiformis.These results demonstrated that Ba P significantly influenced the activity of detoxifying and antioxidant enzymes in microalgae.The metabolic related enzymes(EROD,GST and CAT) may serve as sensitive biomarkers of measuring the contamination level of Ba P in marine water.SHEN Chen MIAO Jingjing LI Yun PAN Luqing 2016Journal of Ocean University of China2016,15,2:4
11Effect of Dopamine Injection on the Hemocyte Count and Prophenoloxidase System of the White Shrimp Litopenaeus vannamei显示文摘Effects of dopamine injection on the hemocyte count, phenoloxidase activity, serine proteinase activity, proteinase in-hibitor activity and α2-macroglobulin-like activity in L. vannamei were studied. Results showed that dopamine injection resulted in a significant effect on the parameters measured (P < 0.05), while no significant difference was observed in the control group (0.85% NaCl). In the experimental groups,the hemocyte count reached the minimum in 3 h;granular and semi-granular cells became stable after 12 h and hyaline cells and the total hemocyte count became stable after 18 h. Phenoloxidase activity reached the minimum in 6 h, and then became stable after 9 h. Serine protease activity and proteinase inhibitor activity reached the minimum in 3 h, and α2-macro-globulin-like activity reached the maximum in 3 h,and all the three parameters became stable after 12 h. 12Effects of Qizhukangxian granules on idiopathic pulmonary fibrosis:a randomized,double blind,placebo-controlled and multicenter clinical pilot trial显示文摘OBJECTIVE:To evaluate the effects of Qizhukangxian granules(QG)on idiopathic pulmonary fibrosis(IPF).METHODS:This is a randomized,double blind,placebo-controlled and multicenter clinical pilot trial.Six medical centers in Tianjin,China,participated in the study.A total of 120 IPF patients were enrolled and randomized into two groups,with 60 patients in each group.The treatment group was treated with QG,while the control group received a Qizhukangxian placebo.The pharmacological treatment lasted for 48 weeks from the enrollment date.The indexes of patients were recorded on the admission day and at the end of the 24th and 48th weeks.Data were analyzed to study the effects of QG;forced vital capacity,change in forced vital capacity and maximal 6-min walk test(6MWT)distance were the primary endpoints.Secondary endpoints were percentage of patients with episodes of acute exacerbation of IPF,pulmonary function,changes in pulse oxygen saturation during the 6MWT,dyspnea score,St.George's respiratory questionnaire score,arterial blood gas analyses and the total Traditional Chinese Medicine symptom pattern score.RESULTS:After 24 weeks of treatment,QG showed greater efficacy than the placebo in certain parameters,including the dyspnea score,Traditional Chinese Medicine symptom pattern score and some indicators in the St.George's respiratory questionnaire score.Analysis of the indexes obtained from all patients at the end of the 48th week showed that the therapeutic effects in the treatment group were significantly better than those in the control group because remarkable differences were observed in most of the primary and secondary endpoints between the two groups,except for the maximal distance of the 6MWT and arterial blood gas analyses.No adverse reaction was observed in either group during the 48-week trial treatment period.CONCLUSION:QG could effectively treat IPF patients by ameliorating pulmonary function,improving the quality of life and lowering the percentage of acute exacerbations.Guo Sijia Song Yalin Feng Jihong Liu Shuang Li Yuechuan Liu Min Wei Luqing Zhang Xian Xie Hui Sun Zengtao 2020Journal of Traditional Chinese Medicine2020,40,4:3
13Immunotoxicity Effect of Benzo[α]Pyrene on Scallop Chlamys farreri显示文摘The toxic effects of benzo[α]pyrene (B[α]P) at different concentrations (0.1, 0.5, 1, 2.5 and 7.5 μgL-1) on scallop (Chlamys farreri) immune system were studied. The results showed that B[α]P had significant toxic effects on the haemocyte counts, neutral red uptake, phagocytosis, bacteriolytic and antibacterial activity (P<0.05), while the seawater control and acetone control had no significant differences. The haemocyte counts, neutral red uptake, phagocytosis and bacteriolytic activity in all B[α]P treatment groups as well as antibacterial activity in groups of 0.5, 1, 2.5 and 7.5 μgL-1 B[α]P decreased significantly (P<0.05). Some of these indices tended to be stable on the sixth day and others on the ninth day, and the indices showed clear time-and concentra- tion-response to B[α]P. Bacteriolytic activity in 0.1μgL-1 B[α]P treatment group and antibacterial activity in 0.1 μgL-1 and 0.5 μgL-1 B[α]P treatment groups increased at the beginning of exposure and reached their peaks on day 1 and day 6, respectively. Following that, both activities decreased gradually and became stable after day 9. When all the indices reached stability, they were significantly lower than those in control group (P<0.05), except for antibacterial activity in 0.1 μgL-1 B[α]P treatment group (P>0.05). 14Effects of Na^+/K^+ Ratio of Groundwaters on the Gill Ion-Transport Enzyme Activity,Plasma Osmolality and Growth of Cynoglossus semilaevis Juveniles显示文摘The effects of environmental Na+/K+ ratio on the gill ion-transport enzyme activity,plasma osmolality and growth of Cynoglossus semilaevis juveniles were investigated. The results showed that,plasma osmolality was similar among flounder adapted to different Na+/K+ ratios of saline groundwaters (P>0.05),while the growth,gill Na+,K+-ATPase and HCO3--ATPase activities were affected by Na+/K+ ratio significantly (P<0.05). The gill Na+,K+-ATPase activity reached its maximum on day 3,then decreased gradually from day 3 to day 9 and remained constant from day 9 to day 15. The peak values of gill Na+,K+-ATPase activity were detected on day 3 for all Na+/K+ ratios of saline groundwaters,then the enzyme activities descended,and on day 9 the enzyme activi-ties achieved steady state,and the gill HCO3--ATPase activity increased rapidly and achieved steady state after one day. During steady state,the gill Na+,K+-ATPase and HCO3--ATPase activity of Na+/K+ ratios 20 and 40 treatments were significantly higher than those in the control group (Na+/K+ ratio 27.5),while there were no significant differences between the Na+/K+ ratio 30 treatment and the control group; the gill Na+,K+-ATPase activity of Na+/K+ ratio 20 and 40 treatments were significantly higher than that for ratio 30 treatment,but there were no significant differences of gill HCO3--ATPase activity among these treatments. At the end of the 15-day experiment,the weight gain (%) and specific growth rate (SGR) of flounders maintained in seawater were significantly higher than those in groundwaters; significant differences also occurred among the treatments; Na+/K+ ratio 30 treatment had the highest values (33.7% and 1.94 respectively),which were significantly higher than those under Na+/K+ ratios 20 and 40 treatments. 15The effect of oxidation on physicochemical properties and aqueous stabilization of multiwalled carbon nanotubes:comparison of multiple analysis methods显示文摘Surface oxidation can alter physicochemical properties of multiwalled carbon nanotubes(MWCNTs) and influence their aqueous stabilization.Many techniques have been used to characterize the physicochemical properties and aqueous stabilization of MWCNTs.However,the relationship between the change in physicochemical property and the aqueous stabilization of MWCNTs merits more studies,and the multiple characterization techniques have not been well compared.This study systematically and comparatively investigated the effect of oxidation on the physicochemical properties and aqueous stabilization of MWCNTs using multiple analysis methods.Increased surface area,disclosed tube ends,defects on the sidewalls,disruption of the electronic structure,and removal of metal catalysts and amorphous carbon were observed for the oxidized MWCNTs(o-MWCNTs) using the multipoint Brunauer-Emmett-Teller(BET) method,transmission electron microscope observation,Raman spectroscopy,UV-Vis spectroscopy,and thermogravimetric analysis.An oxidation-time-dependent increase in oxygen content of the MWCNTs was verified by the methods of elemental analysis,mass difference calculation,and X-ray photoelectron spectroscopy(XPS).Fourier transform infrared spectroscopy,XPS,and the Boehm titration were employed to study the functionalities on the MWCNT surfaces.Despite the limitations of these techniques,the results indicated that the dramatic increase in carboxyl groups was mainly responsible for the significant increase in oxygen content after the oxidation.The dissociation of the grafted functional groups increased electronegativity of the o-MWCNTs and facilitated the aqueous stabilization of o-MWCNTs through electrostatic repulsions.The oxidation affected the UV-Vis absorbance of MWCNT suspensions.The absorbances at 800 nm of the stabilized MWCNT suspensions had a good correlation with the MWCNT concentrations and could be used to quantify the MWCNT suspensions.The findings of this work are expected to boost the research on carbon nanotubes and their environmental behaviors.Luqing Zhang Minhao Wang Jing Fang Kun Yang Daohui Lin 2016Science China Chemistry2016,59,11:2
16Preparation of slightly crosslinked monodisperse poly(maleic anhydride-cyclohexyl vinyl ether-divinylbenzene)functional microspheres with anhydride groups via precipitation polymerization显示文摘Slightly crosslinked monodisperse poly(maleic anhydride-cyclohexyl vinyl ether-divinylbenzene)(MA-CHVE-DVB) microspheres were prepared via precipitation polymerization while using 2,2-azobisisobutyronitrile as an initiator in a mixture of methyl ethyl ketone and n-heptane without any stabilizer.The number-average diameter of the resultant poly(MA-CHVE-DVB) microspheres ranged from 0.478 to 1.386 μm with a polydispersity index of 1.00 to 1.02 that depended on the feed ratios of the MA/CHVE/DVB monomers.The introduction of one electron donor monomer cyclohexyl vinyl ether strongly affected the yield,size,and morphology of these slightly crosslinked microspheres.Quinolinetype chelating resins were obtained after combining the poly(MA-CHVE-DVB) with 8-hydroxyquinoline;the adsorption properties of these materials were measured through their ability to remove Cu^(2+) ions from water.The poly(MA-CHVE-DVB) microspheres with low degrees of crosslinldng provided more effective functional groups and therefore better ion removal capabilities.These slightly crosslinked microspheres may have applications in water treatment as well as in sensing and drug delivery.Cen Yin Anhou Xu Li Gong Luqing Zhang Bing Geng Shuxiang Zhang 2015Particuology2015,13,2:2
17Reactive oxygen species in paraventricular nucleus involved in cardiac sympathetic afferent reflex in rats显示文摘Objective: The present study was designed to determine if reactive oxygen species (ROS) in the paraventricular nucleus (PVN) were involved in modulating cardiac sympathetic afferent reflex (CSAR) in anesthetized rats. Methods: Malondialdehyde (MDA), the end product of lipid peroxidation, in the PVN, was determined by thiobarbituric acid (TBA) spectrometric method. Renal sympathetic nerve activity (RSNA) and arterial pressure were recorded in sinoaortic-denervated and cervical-vagotomized rats. The CSAR was evaluated by the response of the RSNA evoked by epicardial application of bradykinin (BK, 0.4 μg). Results: The MDA in the PVN was significantly increased after epicardial application of BK compared with control (2.0±0.3 vs 0.8±0.1 nmol/mg protein, P<0.01). Microinjection of a superoxide anion scavenger, tiron (20 nmol) into the PVN significantly inhibited the CSAR evoked by BK (12.3±1.9 vs 4.2±1.2%, P<0.01) and decreased MDA level (1.9±0.3 vs 0.6±0.1 nmol/mg protein, P<0.01) compared with control. 18A hypothesis for crack free interior surfaces of Longyou caverns caved in argillaceous siltstone 2000 years ago显示文摘Five complete caverns were discovered in Longyou in 1992.They were manually caved in argillaceous siltstone at shallow depths more than 2000 years ago.When they were un-watered,their integrity was maintained completely,and their interior rock surfaces were free of old cracks.Since then,however,the rock's interior faces have initiated and propagated more and more cracks.This paper attempts to address the question of why the rock interior faces were free of old cracks once they were unearthed.To address this question,this paper proposes a hypothesis that the argillaceous siltstone has the ability of self-healing its cracks over a short period of time under weak acid water environment.Data and evidence are presented herewith to prove the hypothesis.They include observations and measurements in the field and test results in the laboratory.Specifically,a three-point bending test is used to form a tensile crack in a rectangular rock specimen and a deadload test for the specimen immersed in initially weak acid water is used for self-healing its crack.The results have shown that the argillaceous siltstone is in a state of weak alkalinity and the rain water at the site is in a state of weak acidity.Therefore,when it is immersed in weak acid water for some time,the argillaceous siltstone would be able to make chemical reactions to generate new minerals such as calcite.The new minerals would be able to infill the cracks and then heal the crack within a few years.Once the crack is self-healed,the rock can regain its strength and integrity.Consequently,the rock interior surfaces could be free of old cracks when the water was pumped out of the caverns.Zhong Qi YUE Shaopeng FAN Zhifa YANG Lihui LI Luqing ZHANG Zhongjian ZHANG 2010Frontiers of Structural and Civil Engineering2010,4,2:2
