| 1 | Glucocorticoid receptor expression in neonatal rat cortex following recurrent seizures The role in developing brain injury显示文摘BACKGROUND: Studies have explored changes in neonatal rat glucocorticoid receptor (GR) expression changes following mature brain injury.OBJECTIVE: To investigate the temporal and special changes of GR during brain development in rats with recurrent seizures.DESIGN,TIME AND SETTING: A randomized,controlled animal experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central South University,from February 2008 to March 2009.MATERIALS: Rabbit anti-rat GR monoclonal antibody was purchased from Santa Cruz Biotechnology,USA;goat anti-rabbit IgG was purchased from Zhongshan Goldenbridge Biotechnology,China.METHODS: A total of 48 Sprague-Dawley rats,7 days old,were randomly assigned to control and seizure groups,with 24 animals in each group.Seizures were induced by inhalant flurothyl.MAIN OUTCOME MEASURES: Changes in GR protein expression in the rat cerebral cortex were detected by Western blotting analysis and immunohistochemistry.RESULTS: GR expression in the cerebral cortex of control rats significantly increased with aging (P < 0.05),and varied in the frontal lobe,temporal lobe,and parietal lobe.GR was predominantly expressed in the cytoplasm early and rapidly increased in the nuclei.GR protein expression in the cerebral cortex after seizure was lower in the cytoplasm at 15 days and in nuclear protein at 19 days.CONCLUSION: GR expression displayed temporal and spatial changes in brain development.Recurrent seizures in neonatal rats cause abnormal GR expression and might play an important role in developing brain injury. | Tao Bo Lu Yi Tuanmei Wang Jian Li Xingfang Li Dingan Mao | 2010 | Neural Regeneration Research2010,5,2: | 2 |

| 5 | Gamma-aminobutyric acid A receptor gamma 2 subunit following Mg-free-induced seizures in cultured developing neurons显示文摘BACKGROUND:Studies have demonstrated that in vitro cultured cortical neurons from embryonic rats can produce spontaneous recurrent epileptiform discharges following transient Mg2+-free extracellular solution culture. OBJECTIVE: To explore gamma-aminobutyric acid A receptor (GABAAR) γ2 subunit expression follow-ing Mg2+-free-induced seizures in cultured developing neurons. DESIGN,TIME AND SETTING: Cellular and molecular biology. The in vitro experiment was performed at the Department of Pediatrics,Second Xiangya Hospital of Central Southern University between January 2007 and February 2008. MATERIALS: Cortical neurons of Wistar rats on gestational days 16-17 were used. Normal extracellular solution (pH 7.3) consisted of NaCl 145 mmol/L,KCl 2.5 mmol/L,HEPES 10 mmol/L,MgCl2 1 mmol/L,CaCl2 2 mmol/L,glucose 10 mmol/L,and glycine 0.01 mmol/L. In addition,there was no MgCl2 in the Mg2+-free extracellular solution. METHODS: Cortical neurons cultured for 6 days were exposed to normal extracellular solution (control group) and Mg2+-free media (Mg2+-free group) respectively for 3 hours,followed by continuous culture in DMEM solution. MAIN OUTCOME MEASURES: On days 1,7 and 12 after Mg2+-free treatment,real-time RT-PCR,im-munochemistry,and flow cytometry were used to detect GABAAR γ2 subunit expression. RESULTS: Compared with the control group,GABAAR γ2-positive cells decreased significantly on days 1 and 7 after Mg2+-free treatment (P<0.01),but significantly increased on day 12 (P<0.01). GABAAR γ2 subunit mRNA expression decreased significantly at 7 days Mg2+-free treatment when measured by real-time RT-PCR compared with the control group (P<0.05). CONCLUSION: GABAAR γ2 subunit expression is modified following Mg-free-induced seizures in cul-tured developing neurons. | 6 | Labor Cost Analysis for Pome Production in Different Cultivation Modes in Hebei Province显示文摘Taking the traditional fruit pear as the example, this paper analyzes labor cost for pome production in Hebei Province, based on the representative cases and the research of pome production in different cultivation modes. Firstly, it conducts cost analysis for medium-density pome production in Xinji City, focusing on the comparison of the costs for the main production labor in standard thin planting mode and dwarf close planting mode. According to the research results, labor cost has a great influence on the total production cost of pome.
