1Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable.Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun 2020Cellular & Molecular Immunology2020,17,6:13
2Simultaneous determination of ivabradine and N-desmethylivabradine in human plasma and urine using a LC-MS/MS method: application to a pharmacokinetic study显示文摘A sensitive and specific liquid-chromatography tandem mass spectrometry(LC-MS/MS)assay has been developed and validated for the simultaneous quantification of ivabradine and its active metabolite N-desmethylivabradine in human plasma and urine.The assay employed a single liquid–liquid extraction of the analytes from plasma and urine samples,and diazepam was used as internal standard(IS).The chromatographic separation was achieved on a Diamonsil C18 column(150 mm4.6 mm,5 mm,Dikma)using a mixture of methanol and aqueous 5 mM ammonium acetate buffer containing 0.2%formic acid(80:20,v/v)as mobile phase.The assay for ivabradine and N-desmethylivabradine in plasma showed good linearity(r≥0.99)over the ranges 0.1013–101.3 ng/mL and 0.085–25.5 ng/mL,respectively.The assay for ivabradine and N-desmethylivabradine in urine showed good linearity(r≥0.99)over the ranges 10.13–6078 ng/mL and 8.5–850 ng/mL,respectively.The intra-and inter-day accuracy and precision values were found to be within the assay variability limits(RSD<15%)in accordance with FDA guidelines.The methods were successfully used for evaluating the pharmacokinetic properties of ivabradine and N-desmethylivabradine in human plasma and urine in Chinese healthy volunteers.Chengtao Lu Yanyan Jia Jing Yang Xin Jin Ying Songa Wenxing Liu Yi Ding Xiaoli Sun Aidong Wen 2012Acta Pharmaceutica Sinica B2012,2,2:3
3Determination of ifenprodil by LC–MS/MS and its application to a pharmacokinetic study in healthy Chinese volunteers显示文摘This paper reports the development and validation of an assay for ifenprodil based on liquid chromatography–tandem mass spectrometry(LC–MS/MS)and its application to a pharmacokinetic study involving single and multiple intravenous infusions to healthy Chinese volunteers.After sample preparation of plasma by liquid–liquid extraction with ethyl acetate,the analyte and internal standard,urapidil,were separated by reversed phase chromatography in a run time of 4 min and detected by positive ion electrospray ionization followed by multiple reaction monitoring of the precursor-to-product ion transitions at m/z 326.2-308.1 for ifenprodil and m/z 388.4-205.3 for IS.The assay was linear in the concentration range 0.2–50.0 ng/mL with recovery 476.4%.In the pharmacokinetic study of single intravenous infusions of 5,10 and 15 mg ifenprodil,peak plasma concentrations and areas under the plasma concentration–time curve were both linearly related to dose.In the pharmacokinetic study of multiple once daily intravenous infusions of 10 mg ifenprodil for 7 days,pharmacokinetic parameters were similar to those after the single dose showing that ifenprodil does not accumulate on repeated administration.Jing Yang Chengtao Lu Wei Song Jiankang Li Yi Ding Yanrong Zhu Jinyi Cao Likun Ding Yanyan Jia Aidong Wen 2013Acta Pharmaceutica Sinica B2013,3,3:2

7Sulfur vs. tellurium: the heteroatom effects on the nonfullerene acceptors显示文摘The effect of chalcogen heteroatom variation on donor materials has been systematically investigated. However, this effect on acceptors has rarely been explored. Herein, nonfullerene acceptors BFPSP and BFPTP were reported by simply changing the chalcogen atoms from S to Te. The differences between BFPSP and BFPTP in light absorption, energy levels, excited-state lifetimes, energy loss, charge mobilities, morphology, and photovoltaic properties were systematically investigated to understand the heteroatom effects. 10Haploidentical hematopoietic stem cell transplantation may improve long-term survival for children with high-risk T-cell acute lymphoblastic leukemia in first complete remission显示文摘Background: The role of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in children with high-risk (HR) T-cell acute lymphoblastic leukemia (T-ALL) in first complete remission (CR1) is still under evaluation. Moreover, relapse is the main factor affecting survival. This study aimed to explore the effect of allo-HSCT (especially haploidentical HSCT [haplo-HSCT]) on improving survival and reducing relapse for HR childhood T-ALL in CR1 and the prognostic factors of childhood T-ALL in order to identify who could benefit from HSCT.Methods: A total of 74 newly diagnosed pediatric T-ALL patients between January 1, 2012 and June 30, 2018 were enrolled in this retrospective study. Patients were stratified into the low-risk chemotherapy cohort (n = 16), HR chemotherapy cohort (n = 31), and HR transplant cohort (n = 27). Characteristics, survival outcomes, and prognostic factors of all patients were then analyzed.Results: Patient prognosis in the HR chemotherapy cohort was significantly worse than that in the low-risk chemotherapy cohort (5-year overall survival [OS]: 58.5%vs. 100%,P = 0.003;5-year event-free survival [EFS]: 54.1%vs. 83.4%,P = 0.010;5-year cumulative incidence of relapse [CIR]: 45.2%vs. 6.3%,P = 0.011). In HR patients, allo-HSCT improved the 5-year EFS and CIR compared to that of chemotherapy (5-year EFS: 80.1%vs. 54.1%,P = 0.041;5-year CIR: 11.6%vs. 45.2%,P = 0.006). The 5-year OS was higher in the HR transplant cohort than that in the HR chemotherapy cohort (81.0%vs. 58.5%,P = 0.084). 13To Construct High Level Secure Communication System: CTMI Is Not Enough显示文摘Public key cryptographic(PKC) algorithms, such as the RSA, elliptic curve digital signature algorithm(ECDSA) etc., are widely used in the secure communication systems, such as OpenSSL, and a variety of information security systems. If designer do not securely implement them, the secret key will be easily extracted by side-channel attacks(SCAs) or combinational SCA thus mitigating the security of the entire communication system. Previous countermeasures of PKC implementations focused on the core part of the algorithms and ignored the modular inversion which is widely used in various PKC schemes. Many researchers believe that instead of straightforward implementation, constant time modular inversion(CTMI) is enough to resist the attack of simple power analysis combined with lattice analysis. However, we find that the CTMI security can be reduced to a hidden t-bit multiplier problem. Based on this feature, we firstly obtain Hamming weight of intermediate data through side-channel leakage. Then, we propose a heuristic algorithm to solve the problem by revealing the secret(partial and full) base of CTMI. Comparing previous nec-essary input message for masking filtering, our procedure need not any information about the secret base of the inversion. 14Polydopamine functionalized mesoporous silica as ROS-sensitive drug delivery vehicles for periodontitis treatment by modulating macrophage polarization显示文摘Periodontitis is recognized as the major cause of tooth loss in adults, posing an adverse impact on systemic health. In periodontitis, excessive production of reactive oxygen species (ROS) at the inflamed site culminates in periodontal destruction. In this study, a novel ROS-responsive drug delivery system based on polydopamine (PDA) functionalized mesoporous silica nanoparticles was developed for delivering minocycline hydrochloride (MH) to treat periodontitis. The outer PDA layer and the inner MH of the nanoparticles acted as ROS scavengers and anti-inflammatory agents, respectively. Under the synergistic action of PDA and MH, macrophages were polarized from the pro-inflammatory M1 to the anti-inflammatory M2 phenotype. The in vitro experiments provided convincing evidence that PDA could scavenge ROS effectively, and the expression of pro-inflammatory cytokines was attenuated and the secretion of anti-inflammatory cytokines was enhanced through M1 to M2 polarization of macrophages with the cooperation of MH. In addition, the results obtained from the periodontitis rat models demonstrated that the synergetic effect of PDA and MH prevented alveolar bone loss without causing any adverse effect. 16Feature Preserving Milli-Scaling of Large Format Visualizations显示文摘Ultra-scale data analysis has created many new challenges for visualization.For example,in climate research with two-dimensional time-varying data,scientists find it crucial to study the hidden temporal relationships from a set of large scale images,whose resolutions are much higher than that of general computer monitors.When scientists can only visualize a small portion(< 1=1000) of a time step at one time,it is extremely challenging to analyze the temporal features from multiple time steps.As this problem cannot be simply solved with interaction or display technologies,this paper presents a milli-scaling approach by designing downscaling algorithms with significant ratios.Our approach can produce readable-sized images of multiple ultra-scale visualizations,while preserving important data features and temporal relationships.Using the climate visualization as the testing application,we demonstrate that our approach provides a new tool for users to effectively make sense of multiple,large-format visualizations.Yunwei Zhang Aidong Lu Jian Huang 2012Tsinghua Science and Technology2012,17,4:0
