| 1 | Construction of a full bacterial artificial chromosome(BAC) library of Oryza sativa genome显示文摘We have constructed a full BAC library for the superior eaxly indica variety of OryZa sativa, Guang Lu Ai4. The MAX Efficiency DHlOB with increased stabilltyof inserts was used as BAC host cells. The potent pBelo BACII with double selection markers was used as cloning vector. The cloning efficiency we have reached was as high as 98%, and the transformation efficiency was raised up to 1Oo transformants / pg of large fragment DNA. The BAC recombinant transformants were picked at random and analyzed for the size of inserts, which turned out to be of 120 kb in length on average. We have obtained more than 20,000 such BAC clones. According to conventional probabillty equation, they covered the entire rice genome of 420,000 kb in length. The entire length of inserts of the library obtained has the 5- to 6- fold coverage of the genome. | 2 | Ternary geochemical-tracing system in natural gas accumulation显示文摘The establishment of geochemical-tracing system of gas generation and accumulation is helpful to re-elucidating the gas migration and accumulation in time and space. To deduce the complex process of gas accumulation, a ternary geochemical-tracing system is set up, according to stable isotope inheritance of source rocks, kinetic fractionation of stable isotopes, time-accumulating effect of noble gas isotopes, mantle-derived volatile inheritance, and organic molecule inheritance of light hydrocarbons and thermally kinetic fractionation in their generation, in combination with the previous achievements of gas geochemistry and geochemical parameters of gas-source correlation. There are tight interactions for the geochemical parameters with much information about parent inheritance and special biomarkers, in which they are confirmed each other, reciprocally associated and preferentially used for the requirement so that we can use these geochemical parameters to effectively demonstrate the sources of natural gas, sedimentary environments and thermal evolution of source rocks, migration and accumulation of natural gas, and rearrangement of natural gas reservoirs. | 7 | Construction of Oryza sativa genome contigs by fingerprint strategy显示文摘We described the construction of BAC contigs of the genome of a indica variety of Oryza sativa, Guang Lu Ai 4.An entire representative (sixfold coverage of rice chromosomes) and genetically stable BAC library of rice genome constructed in this lab has been systematically analysed by restriction enzyme fragmentation and polyacrylamide gel electrophoresis. And all the images thus obtained were subject to image-processing, which consisted of preliminary location of bands, cooperative tracking of lanesby correlation of adjacent bands, a precise densitometric pass, alignment at the marker bands with the standard,optional interactive editing, and normalization of the accepted bands. The contigs were generated based on the Computer Software specially designed for genome map ping. The number of contigs with 600 kb in length on average was 464; of contigs with 1000 kb in length on average was 107; of contigs with 1500 kb in length on average was 23. Therefore, all the contigs we have obtained amounted up to 420 megabases in length. Considering the size of rice genome (430 megabased), the contigs generated in this lab have covered nearly 98% of the rice genome. | 15 | Three-dimensional electric field simulation and flashover path analysis of ice-covered suspension insulators显示文摘Flashover of ice-covered insulators seriously affects the safe operation of transmission lines.It is necessary to study the electric field distribution for flashover analysis of ice-covered insulators.The electric field of ice-covered insulators was mostly calculated by a two-dimensional(2D)axisymmetric model that cannot be well corresponded to actual icing situations.In this study,a 3D electric field simulation model of ice-covered suspension insulators under moderate icing condition with applied DC voltage was established.Two characteristic parameters,E_(av)and E_(max)were proposed to measure electric field distortion degree.Based on the simulation results,the effects of water film conductivity,icicle length,icicle deviation angle,and icicle distribution on Eav and Emax were studied.The results showed that icicles have a significant influence on the electric field distribution of insulators.E_(av)increases with the increase of icicle length and decreases with the increase of icicle deviation angle.E_(max)increases with the increase of the icicle length,icicle spacing and adjacent icicle length difference.When the icicle deviation angle is 45°,E_(max)is the largest.Lastly,the possible flashover paths were analysed based on the simulation results,which would provide a theoretical basis for building a flashover model of ice-covered insulators. | Lichun Shu Yanqing Liu Xingliang Jiang Qin Hu Gaohui He Zhou Yu Longfang Xiao | 2020 | High Voltage2020，5，3： | 1 |
| 16 | Multiple resonant excitations of surface plasmons in a graphene stratified slab by Otto configuration and their independent tuning显示文摘Multiple resonant excitations of surface plasmons in a graphene stratified slab are realized by Otto configuration at terahertz frequencies. The proposed graphene stratified slab consists of alternating dielectric layers and graphene sheets, and is sandwiched between a prism and another semi-infinite medium. Optical response and field distribution are determined by the transfer matrix method with the surface current density boundary condition.Multiple resonant excitations appear on the angular reflection spectrum, and are analyzed theoretically via the phase-matching condition. Furthermore, the effects of the system parameters are investigated. Among them, the Fermi levels can tune the corresponding resonances independently.
