| 1 | Soybean AP1 homologs control flowering time and plant height显示文摘Flowering time and plant height are key agronomic traits that directly affect soybean(Glycine max)yield.APETALA1(AP1)functions as a class A gene in the ABCE model for floral organ development,helping to specify carpel,stamen,petal,and sepal identities.There are four AP1 homologs in soybean,all of which are mainly expressed in the shoot apex.Here,we used clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein 9 technology to generate a homozygous quadruple mutant,gmap1,with loss-of-function mutations in all four GmAP1 genes.Under short-day(SD)conditions,the gmap1 quadruple mutant exhibited delayed flowering,changes in flower morphology,and increased node number and internode length,resulting in plants that were taller than the wild type.Conversely,overexpression of GmAP1a resulted in early flowering and reduced plant height compared to the wild type under SD conditions.The gmap1 mutant and the overexpression lines also exhibited altered expression of several genes related to flowering and gibberellic acid metabolism,thereby providing insight into the role of GmAP1 in the regulatory networks controlling flowering time and plant height in soybean.Increased node number is the trait with the most promise for enhancing soybean pod number and grain yield.Therefore,the mutant alleles of the four AP1 homologs described here will be invaluable for molecular breeding of improved soybean yield. | Liyu Chen Haiyang Nan Lingping Kong Lin Yue Hui Yang Qingsong Zhao Chao Fang Haiyang Li Qun Cheng Sijia Lu Fanjiang Kong Baohui Liu Lidong Dong | 2020 | Journal of Integrative Plant Biology2020,62,12: | 9 |
| 2 | Independent Component Analysis and Graph Theoretical Analysis in Patients with Narcolepsy显示文摘The present study was aimed to evaluate restingstate functional connectivity and topological properties of brain networks in narcolepsy patients compared with healthy controls.Resting-state fMRI was performed in 26 adult narcolepsy patients and 30 matched healthy controls.MRI data were first analyzed by group independent component analysis,then a graph theoretical method was applied to evaluate the topological properties in the whole brain.Small-world network parameters and nodal topological properties were measured.Altered topological properties in brain areas between groups were selected as regionof-interest seeds,then the functional connectivity among these seeds was compared between groups.Partial correlation analysis was performed to evaluate the relationship between the severity of sleepiness and functional connectivity or topological properties in the narcolepsy patients.Twenty-one independent components out of 48 were obtained.Compared with healthy controls,the narcolepsy patients exhibited significantly decreased functional connectivity within the executive and salience networks,along with increased functional connectivity in the bilateral frontal lobes within the executive network.There were no differences in small-world network properties between patients and controls.The altered brain areas in nodal topological properties between groups were mainly in the inferior frontal cortex,basal ganglia,anterior cingulate,sensory cortex,supplementary motor cortex,and visual cortex.In the partial correlation analysis,nodal topological properties in the putamen,anterior cingulate,and sensory cortex as well as functional connectivity between these regions were correlated with the severity of sleepiness(sleep latency,REM sleep latency,and Epworth sleepiness score)among narcolepsy patients.Altered connectivity within the executive and salience networks was found in narcolepsy patients.Functional connection changes between the left frontal cortex and left caudate nucleus may be one of the parameters describing the severity of narcolepsy.Changes in the nodal topological properties in the left putamen and left posterior cingulate,changes in functional connectivity between the left supplementary motor area and right occipital as well as in functional connectivity between the left anterior cingulate gyrus and bilateral postcentral gyrus can be considered as a specific indicator for evaluating the severity of narcolepsy. | Fulong Xiao Chao Lu Dianjiang Zhao Qihong Zou Liyue Xu Jing Li Jun Zhang Fang Han | 2019 | Neuroscience Bulletin2019,35,4: | 7 |
| 3 | FT5a interferes with the Dt1-AP1 feedback loop to control flowering time and shoot determinacy in soybean显示文摘Flowering time and stem growth habit determine inflorescence architecture in soybean, which in turn influences seed yield. Dt1, a homolog of Arabidopsis TERMINAL FLOWER 1(TFL1), is a major controller of stem growth habit, but its underlying molecular mechanisms remain unclear.Here, we demonstrate that Dt1 affects node number and plant height, as well as flowering time,in soybean under long-day conditions. The b ZIP transcription factor FDc1 physically interacts with Dt1, and the FDc1-Dt1 complex directly represses the expression of APETALA1(AP1). We propose that FT5 a inhibits Dt1 activity via a competitive interaction with FDc1 and directly upregulates AP1. Moreover, AP1 represses Dt1 expression by directly binding to the Dt1 promoter, suggesting that AP1 and Dt1 form a suppressive regulatory feedback loop to determine the fate of the shoot apical meristem. | 4 | A review and research on comprehensive characterization of microscopic shale gas reservoir space显示文摘In this paper,substantial domestic and foreign research results of microscopic shale reservoir space were systemically reviewed,the research history consisting of simple observation and qualitative classification,quantitative research,the combination of qualitative and quantitative research successively as well as the characteristics of each research stage were summarized.In addition,the current problems existing in the characterization methods of shale reservoir space were also analyzed.Furthermore,based on massive actual detection of typical core samples obtained from more than 50 global shale gas wells and relevant practical experience,a comprehensive characterization method of combining qualitative with the semiquantitative characterization was put forward.In detail,the indicators of the qualitative characterization include pore combination type and organic-matter microscopic morphology type,while the core elements of the semi-quantitative characterization include the percentage of the organic-matter area and the plane porosity of the pores of different types.Based on the reference of the naming and classification of rocks,the three-end-member diagram method was used to characterize microscopic shale reservoir space.This is achieved by plotting the three end-member diagram of 3 kinds of first-order critical reservoir spaces,i.e.,organic-matter pores,matrix pores,and micro-fractures,in order to intuitively present the features of the microscopic pore combination.Meanwhile,statistic histograms of organic-matter microscopic morphology type and the plane porosity of different types of pores were adopted to characterize the development degree of second-order pores quantitatively.By this comprehensive characterization method,the importance of both pore combination and the microscopic morphology of organic matter were emphasized,revealing the control of organic-matter microscopic morphology over the organic-matter pores.What is more,high-resolution FE-SEM was adopted to obtain semi-quantitative statistics results.In this way,the features of pore development and pore combination were quantified,not only reflecting the types and storage capacity of the microscopic shale reservoir space,but also presenting the hydrocarbongenerating potential of organic matter in shale.Therefore,the results of this research are capable of providing in-depth microscopic information for the assessment and exploration and development of shale gas resources. | Yu-ru Yang Xiao-chen Liu Hui Zhang Gang-yi Zhai Jiao-dong Zhang Zhi-fang Hu Shu-jing Bao Cong Zhang Xiang-hua Wang Xiao Yang Zheng-zhuang Liu Ting Xie Juan Chen Liyu Fang Li-juan Qin | 2019 | China Geology2019,2,4: | 3 |
| 5 | Encapsulation of ultrafine Pd nanoparticles within the shallow layers of UiO-67 for highly efficient hydrogenation reactions显示文摘Metal-organic frameworks(MOFs)have been used to encapsulate active metal nanoparticles(MNPs)to fabricate MNPs@MOFs composites with high catalytic efficiencies.However,the diffusion of reactants and the accessibility of MNPs located in the center of MOFs may be hindered due to the inherent microporous structures of MOFs,which would affect the catalytic activities of MNPs.Herein,we report a solvent assisted ligand exchange-hydrogen reduction(SALE-HR)strategy to selectively encapsulate ultrafine MNPs(Pd or Pt)within the shallow layers of a MOF,i.e.,UiO-67.The particle sizes of the encapsulated MNPs and the thickness of the MNPs-embedded layers can be adjusted easily by controlling the SALE conditions(e.g.time and temperature).Crucially,the LE-Pd@UiO-80-0.5 composite with the thinnest Pd-embedded layers displays remarkable catalytic efficiency with a high turnover frequency(TOF)value of 600 h^-1towards hydrogenation of nitrobenzene under 1 atm H_2at room temperature.The results indicate that the catalytic efficiency and the utilization of MNPs can be enhanced by compactly encapsulating MNPs within the shallow layers of MOFs as close to their outer surfaces as possible,owing to the short masstransfer distance and enhanced accessibility of overall MNPs. | Zhiming Cui Ting Fan Liyu Chen Ruiqi Fang Chuanmao Li Yingwei Li | 2021 | Science China Chemistry2021,64,1: | 1 |

| 10 | Multiplex CRISPR/Cas9-mediated knockout of soybean LNK2 advances flowering time显示文摘Flowering time is an important agronomic trait for soybean yield and adaptation. However, the genetic basis of soybean adaptation to diverse latitudes is still not clear. Four NIGHT LIGHT-INDUCIBLE AND CLOCK-REGULATED 2(LNK2) homeologs of Arabidopsis thaliana LNK2 were identified in soybean. Three single-guide RNAs were designed for editing the four LNK2 genes. A transgene-free homozygous quadruple mutant of the LNK2 genes was developed using the CRISPR(clustered regularly interspaced short palindromic repeats)/Cas9(CRISPR-associated protein 9). Under long-day(LD) conditions, the quadruple mutant flowered significantly earlier than the wild-type(WT). Quantitative real-time PCR(q RT-PCR)revealed that transcript levels of LNK2 were significantly lower in the quadruple mutant than in the WT under LD conditions. LNK2 promoted the expression of the legume-specific E1 gene and repressed the expression of FT2 a. Genetic markers were developed to identify LNK2 mutants for soybean breeding.These results indicate that CRISPR/Cas9-mediated targeted mutagenesis of four LNK2 genes shortens flowering time in soybean.
