1Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. 2A Study on Planning Strategies for Urban Housing Block Development显示文摘As a city is the carrier of human society and housing is an important part of a citizen's life and survival, the citizens' choice of their housing mode will inf luence the material and spiritual life of the individuals, families, and society. In view of the diversification of values and investments, people are eager for a harmonious relationship between the community and the city. As a kind of compact and efficient housing mode, the housing block highlights the organic link of the community within the city in an open and shared living environment. This paper reviews the development of housing blocks in various countries and summarizes the characteristics of housing blocks through a comparison with traditional gated residential quarters and urban blocks. 3Culturing adequate CAR-T cells from less peripheral blood to treat B-cell malignancies显示文摘Objective:Chimeric antigen receptor-modified T(CAR-T)cells have shown impressive results against relapsed/refractory B cell malignancies.However,the traditional manufacture of CAR-T cells requires leukapheresis to isolate large amounts of peripheral blood T cells,thus making some patients ineligible for the procedure.Methods:We developed a simple method for CAR-T cell preparation requiring small volumes of peripheral blood.First,CD3+T cells isolated from 50 mL peripheral blood from patients(B-cell malignancies)were stimulated with immobilized anti-CD3/RetroNectin in 6-well plates and then transduced with CAR-expressing lentiviral vector.After 4 d,the T cells were transferred to culture bags for large-scale CAR-T cell expansion.In vitro and animal experiments were performed to evaluate the activity of the manufactured CAR-T cells.Finally,29 patients with B-cell acute lymphoblastic leukemia(B-ALL)and 9 patients with B-cell lymphoma were treated with the CAR-T cells.Results:The CAR-T cells were expanded to 1–3×10^(8) cells in 8–10 d and successfully killed B cell-derived malignant tumor cells in vitro and in vivo.For patients with B-ALL,the complete remission rate was 93%1 month after CAR-T cell infusion;after 12 months,the overall survival(OS)and leukemia-free survival rates were 69%and 31%,respectively.For patients with lymphoma,the objective response rate(including complete and partial remission)was 78%2 months after CAR-T cell infusion,and after 12 months,the OS and progression-free survival rates were 71%and 43%,respectively.Cytokine-release syndrome(CRS)occurred in 65.51%and 55.56%of patients with B-ALL and B-cell lymphoma,respectively;severe CRS developed in 20.69%of patients with B-ALL and in no patients with lymphoma.Conclusions:Our novel method can generate sufficient numbers of CAR-T cells for clinical use from 50–100 mL peripheral blood,thus providing an alternative means of CAR-T cell generation for patients ineligible for leukapheresis.Lu Han Jian Zhou Linlin Li Keshu Zhou Lingdi Zhao Xinghu Zhu Qingsong Yin Yufu Li Hongqin You Jishuai Zhang Yongping Song Quanli Gao 2021Cancer Biology & Medicine2021,18,4:2
4The Effects of Therapeutic Horseback Riding Program on Motor Skills inChildren with Autism Spectrum Disorder显示文摘Therapeutic horseback riding(THR)as an animal-assisted intervention is one of the innovative approaches emer-ging in the treatment for children with autism spectrum disorder(ASD).The current study was designed to inves-tigate the effects of a 12-week,twice a week THR program on motor skills in sixty-eight children with ASD aged 5–10 years old.All participants selected met the DSM-V criteria for ASD,and a total offifty-three participants completed the study.A randomized controlled trial design was utilized for the study.Data was collected via a pre-THR test,interim-THR test,and post-THR test to investigate the possible changes in motor skills throughout the 12-week THR program.Results showed that the THR program significantly improved overall motor skills across time points(p<0.05)and sub-skills of run,gallop and two-hand catch(as compared to the control group,p<0.05).In conclusion,the THR program may be an effective option for improving motor skills in children with ASD and further investigation with a longer period of intervention is warranted.Mengxian Zhao Yonghao You Jinming Li Sean Healy Alyx Taylor Zhihao Zhang Linlin Li Liye Zou 2022International Journal of Mental Health Promotion2022,24,4:1
5Molecular level understanding of CO_(2) capture in ionic liquid/polyimide composite membrane显示文摘Ionic liquid(IL)/polyimide(PI)composite membranes demonstrate promise for use in CO_(2)separation applications.However,few studies have focused on the microscopic mechanism of CO_(2)in these composite systems,which is important information for designing new membranes.In this work,a series of systems of CO_(2)in 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide composited with 4,4-(hexafluoroisopropylidene)diphthalic anhydride(6FDA)-based PI,6FDA-2,3,5,6-tetramethyl-1,4-phenylene-diamine,at different IL concentrations were investigated by all-atom molecular dynamics simulation.The formation of IL regions in PI was found,and the IL regions gradually became continuous channels with increasing IL concentrations.The analysis of the radial distribution functions and hydrogen bond numbers demonstrated that PI had a stronger interaction with cations than anions.However,the hydrogen bonds among PI chains were destroyed by the addition of IL,which was favorable for transporting CO_(2).Furthermore,the self-diffusion coefficient and free energy barrier suggested that the diffusion coefficient of CO_(2)decreased with increasing IL concentrations up to 35 wt-%due to the decrease of the fractional free volume of the composite membrane.However,the CO_(2)selfdiffusion coefficients increased when the IL contents were higher than 35 wt-%,which was attributed to the formation of continuous IL domain that benefitted the transportation of CO_(2).Linlin You Yandong Guo Yanjing He Feng Huo Shaojuan Zeng Chunshan Li Xiangping Zhang Xiaochun Zhang 2022Frontiers of Chemical Science and Engineering2022,16,2:1

7Common susceptibility variants of KDR and IGF-1R are associated with poststroke depression in the Chinese population显示文摘Background Depression,one of the most frequent complications after stroke,increases the disease’s burden and physical disability.Poststroke depression(PSD)is a multifactorial disease with genetic,environmental and biological factors involved in its occurrence.Genetic studies on PSD to date have mainly focused on the monoamine system and brain-derived neurotrophic factors.However,understanding is still limited about the influence of the single nucleotide polymorphism(SNP)of other neurotrophic factors on PSD.Aims The present study aimed to investigate the relationship between seven vascular endothelial growth factor(VEGF)family gene variants that occur with PSD.Methods A multicentre candidate gene study from five hospitals in Jiangsu Province from June 2013 to December 2014 involved 121 patients with PSD and 131 patients with non-PSD.Demographic characteristics and neuropsychological assessments were collected.Theχ^(2)test was used to evaluate categorical variables,while the independent t-test was applied to continuous variables.SNPs in seven genes(VEGFA,VEGFB,KDR,FLT-1,IGF-1,IGF-1R and PlGF)were genotyped.Single-marker association for PSD was analysed byχ^(2)tests and logistic regression using SPSS and PLINK software.Results Patients with PSD included more women and those with lower education levels,lower body mass indexes,lower Mini-Mental State Examination scores,and higher scores on the 17-item Hamilton Depression Rating Scale than non-PSD patients.Ninety-two SNPs with seven genes were genotyped and passed quality control.The rs7692791 CC genotypes,the C allele of KDR and the rs9282715 T allele of IGF-1R increased the risk for PSD(χ^(2)=7.881,p=0.019;χ^(2)=4.259,p=0.039;χ^(2)=4.222,p=0.040,respectively).In addition,the SNP rs7692791 of KDR was significantly associated with PSD by the logistic regression of an additive model(p=0.015,OR=9.584,95%CI:1.549 to 59.31).Conclusions Patients with rs7692791 C allele carriers or the CC genotype of KDR and the rs9282715 T allele of IGF-1R may have PSD susceptibility.Findings such as these may help clinicians to identify the high-risk population for PSD earlier and,thus,enable them to provide more timely interventions.Yingying Yue Linlin You Fuying Zhao Kezhong Zhang Yanyan Shi Hua Tang Jianxin Lu Shenghua Li Jinxia Cao Deqin Geng Aiqin Wu Yonggui Yuan 2023General Psychiatry2023,36,1:1
8A neural network-based production process modeling and variable importance analysis approach in corn to sugar factory显示文摘Corn to sugar process has long faced the risks of high energy consumption and thin profits.However,it’s hard to upgrade or optimize the process based on mechanism unit operation models due to the high complexity of the related processes.Big data technology provides a promising solution as its ability to turn huge amounts of data into insights for operational decisions.In this paper,a neural network-based production process modeling and variable importance analysis approach is proposed for corn to sugar processes,which contains data preprocessing,dimensionality reduction,multilayer perceptron/convolutional neural network/recurrent neural network based modeling and extended weights connection method.In the established model,dextrose equivalent value is selected as the output,and 654 sites from the DCS system are selected as the inputs.LASSO analysis is first applied to reduce the data dimension to 155,then the inputs are dimensionalized to 50 by means of genetic algorithm optimization.Ultimately,variable importance analysis is carried out by the extended weight connection method,and 20 of the most important sites are selected for each neural network.The results indicate that the multilayer perceptron and recurrent neural network models have a relative error of less than 0.1%,which have a better prediction result than other models,and the 20 most important sites selected have better explicable performance.The major contributions derived from this work are of significant aid in process simulation model with high accuracy and process optimization based on the selected most important sites to maintain high quality and stable production for corn to sugar processes.Yi Tong Mou Shu Mingxin Li Yingwei Liu Ran Tao Congcong Zhou You Zhao Guoxing Zhao Yi Li Yachao Dong Lei Zhang Linlin Liu Jian Du 2023Frontiers of Chemical Science and Engineering2023,17,3:1
