1Anti-hepatocarcinoma effects of 5-fluorouracil encapsulated by galactosylceramide liposomes in vivo and in vitro显示文摘AIM: To study the anti-hepatocarcinoma effects of 5-fluorouracil (5-Fu) encapsulated by galactosylceramide liposomes (5-Fu-GCL) in vivo and in vitro.METHODS: Tumor-bearing animal model and HepA cell line were respectively adopted to evaluate the anti-tumor effects of 5-Fu-GCL in vivoand in vitro. Tumor cell growth inhibition effects of 5-Fu-GCL in vitro were assessed by cell viability assay and MTT assay, In vivo experiment,the inhibitory effects on tumor growth were evaluated by tumor inhibition rate and animal survival days. High performance liquid chromatography was used to detect the concentration-time course of 5-Fu-GCL in intracellular fluid in vitro and the distribution of 5-Fu-GCL in liver tumor tissues in vivo. Apoptosis and cell cycle of tumor cells were demonstrated by flow cytometry.RESULTS: In vitro experiment, 5-Fu-GCL (6.25-100μmol/L) and free 5-Fu significantly inhibited HepA cell growth.Furthermore, IC50 of 5-Fu-GCL (34.5μmol/L) was lower than that of free 5-Fu (51.2μmol/L). In vivo experiment,5-Fu-GCL (20, 40, 80mg/kg) significantly suppressed the tumor growth in HepA bearing mice model. Compared with free 5-Fu, the area under curve of 5-Fu-GCL in intracellular fluid increased 2.6 times. Similarly, the distribution of 5-Fu-GCL in liver tumor tissues was significantly higher than that of free 5-Fu. After being treated with 5-Fu-GCL, the apoptotic rate and the proportion of HepA cells in the S phase increased, while the proportion in the G0/G1 and G2/M phases decreased.CONCLUSION: 5-Fu-GCL appears to have anti-hepato-carcinoma effects and its drug action is better than free 5-Fu. its mechanism is partly related to increased drug concentrations in intracallular fluid and liver tumor tissues,enhanced tumor cell apoptotic Fate and arrest of call cycle in S phase.YongJin JunLi Long-FuRong Yuan-HaiLi LinGuo Shu-YunXu 2005World Journal of Gastroenterology2005,11,17:8

3Novel derivative of Danshensu acts as anti-thrombotic agent for modulation of protein disulfide isomerase family member ERp57显示文摘OBJECTIVE To identify the specific targets of a novel derivative of Danshensu ADTM as the protein disulfide isomerase(PDI)family proteins including ERp57.To further investigate the underlying mechanism of ADTM to modulate ERp57 to regulate platelet function with direct interaction withαⅡbβ3 integrin.METHODS To isolate the protein targets that bound to ADTM,a biotin-conjugated ADTM analogue(BAA)was designed and synthesized.BAA(300μmol·L-1)was incubated with rat blood platelet lysates and the BAA-protein complexes were pulled down with NeutrAvidin-agarose followed by protein profiling using LC-MS/MS.To determine platelet aggregation in vitro,rabbit platelets were incubated with the indicated concentrations of compounds and aggregation was induced by ADP(10μmol·L-1)or AA(200μmol·L-1)and measured using a platelet aggregometer.To determine platelet aggregation-induced by ADP in rat in vivo,ADTM(5-20mg·kg-1)in comparison with DSS(10mg·kg-1)and clopidogrel(18mg·kg-1)were administered daily by i.v.injection for 5d,respectively.To determine the action of ADTM on the ERp57/αⅡbβ3 interaction,it was examined by immunoprecipitation with anti-αⅡbβ3antibody,followed by detection of ERp57 immunoreactivity using immunoblotting.RESULTS BAA could bind to various proteins involved in platelet function.In particular,platelet aggregation-associated proteins were identified with>95% protein identification probability including ERp72,ERp57ERp5 and PDI,which are members of the protein disulfide isomerase(PDI)family related to platelet function and redox homeostasis.ADTM exhibited potent inhibition on the redox activity of ERp57 in a concentration-dependent manner(IC50=100 300μmol·L-1).In in vitro studies,ADTM exhibited concentration-dependent inhibition on ADP-induced and AA-induced platelet aggregation with comparable effects to aspirin and clopidogrel.In vivo study showed that ADP-induced platelet aggregation was significantly compromised(>40%reduction)in rats treated with ADTM(20mg·kg-1).Similarly,ADTM also exhibited significant anti-thrombotic effect in vivo as shown in the ferric chloride(FeCl3)-induced venous thrombosis.Immunoprecipitation with anti-αⅡbβ3antibody,followed by detection of ERp57 immunoreactivity using immunoblotting showed that ADTM disrupted the interaction of ERp57 with αⅡ bβ3.CONCLUSION These results demonstrated that ADTM exhibited broad-spectrum anti-platelet activities and ERp57 is a potential therapeutic target for anti-platelet therapy.Guo-zhenCUI Lu-chenSHAN LinGUO IvanKeungCHU Guo-huiLI QuanQUAN YunZHAO Chenong-mengCHONG ZaijunZHANG PeiYU Pui-manHOI Ye-weiSUN Yu-qiangWANG Simonming-yuenLEE 2015中国药理学与毒理学杂志2015,29,S1:3
4Cerium oxide nanoparticles coated by surfactant sodium bis(2—ethylhexyl) sulphosuccinate(AOT):local atomic structures and x—ray absorption spectroscopic studies显示文摘Cerium oxide nanoparticles coated by sodium bis(2-ethylexyl) sulphosuccinate(AOT) were prepared by using a microemulsion method.Transmission electron microscopy revealed an average particle siae of 2-3nm.X-ray diffraction showed that the cerium oxide nanoparticles retain the CeF2-type cubic structures like the bulk crystal.The intermediate valence offormally tetravalent compounds had been detected by x-ray-absorption near-edge structetravalent compounds had been detected by x-ray-absorption near-dege structure(XANES) spectra of Ce LIII absorption in bulk CeO2 and the cerium oxide nanoparticles.Two well resoved white lines can be assigned to the electron configurations of 4f^0L and 4f^1L,respectively,where L denotes a ligand hole.At the same time,the cerium oxide nanoparticles also showed the structural featres of trivalent compounds,in comparison to the trivalent Ce(NO3)3·6H2O.Fuor Lorentzian functions and two arctan functions were used to fit the normalized XANES spectra.The extended x-ray-absorption fine-structure(EXAFS) technique was used to probe the local atomic structures around the absorber Ce.The multielectrorn excitation effect on the EXAFS spectra was eliminated.A cor-shell model was used to deduce the near-neighbour structural parameters around cerium.Bulk CeO2 with eight oxygen atoms located at 2.343A was used as the reference sample to extract the backscattering amplitude and phase shift of the Ce-O bond.One half of the atome locate at the core part with the CeF2-type cubic structures(eight oxyens at 2.343A around Ce),the other half of the atoms are amorphous phase located in the shell part (surface of the nanoparticles) with approximately Ce2O3 structural features (averageed seven oxygens at 2.50A around Ce).ZhonghuaWu RobertEBenfield LinGuo HuanjunLi QinglinYang Didier 2001Beijing Synchrotron Radiation Facility2001,,2:2

6Evaluation of a Rat Model of Experimental Autoimmune Encephalomyelitis with Human MBP as Antigen显示文摘Experimental autoimmune encephalomyelitis (EAE) is a good model for human multiple sclerosis (MS)research. However, there are some defects in the traditional models. Here, we improved the model by using the human myelin basic protein (MBP) as antigen. EAE was induced by immunization of female Wistar rats with human MBP. Compared with the traditional models, the new model was evaluated by clinical signs to pathological changes. The immune state of the model was assessed by the lymphocyte infiltrative response and levels of TNF-α,IFN-γ, IL-10. It was found that most of rats exhibited tail tone loss and hind-limb paralysis,also there were demyelination, infiltrative lymphocyte foci, 'Neuronophagia' in the cortex of cerebra and the white matter of spinal cords. PBMC and spleen lymphocytes were strongly response to the stimulation of MBP and PHA. The levels of TNF-α,IFN-γ were altered with the severity of EAE. In the remitting phase, IL-10 was increased significantly. This study demonstrate that the animal model of EAE induced by human MBP bears resemblance to the features of human multiple sclerosis and promises to be a better model than ever before for the study of MS. 12Fuzzy Hybrid Coyote Optimization Algorithm for Image Thresholding显示文摘In order to address the problems of Coyote Optimization Algorithm in image thresholding,such as easily falling into local optimum,and slow convergence speed,a Fuzzy Hybrid Coyote Optimization Algorithm(here-inafter referred to as FHCOA)based on chaotic initialization and reverse learning strategy is proposed,and its effect on image thresholding is verified.Through chaotic initialization,the random number initialization mode in the standard coyote optimization algorithm(COA)is replaced by chaotic sequence.Such sequence is nonlinear and long-term unpredictable,these characteristics can effectively improve the diversity of the population in the optimization algorithm.Therefore,in this paper we first perform chaotic initialization,using chaotic sequence to replace random number initialization in standard COA.By combining the lens imaging reverse learning strategy and the optimal worst reverse learning strategy,a hybrid reverse learning strategy is then formed.In the process of algorithm traversal,the best coyote and the worst coyote in the pack are selected for reverse learning operation respectively,which prevents the algorithm falling into local optimum to a certain extent and also solves the problem of premature convergence.Based on the above improvements,the coyote optimization algorithm has better global convergence and computational robustness.The simulation results show that the algorithmhas better thresholding effect than the five commonly used optimization algorithms in image thresholding when multiple images are selected and different threshold numbers are set.Linguo Li Xuwen Huang Shunqiang Qian Zhangfei Li Shujing Li Romany F.Mansour 2022Computers, Materials & Continua2022,,8:0

14Extraction and biodegradation of ginkgolic acids from Ginkgo biloba sarcotestae显示文摘Ginkgolic acids are unwanted constituents in standard Ginkgo biloba leaves extracts. Thus, for the quality control of ginkgo extracts, it is important to establish an effective degradation method, with high catalytic efficiency and safety, to remove ginkgolic acids.Laccases are oxidases with potential for application in elimination of hazardous phenolic compounds. In this study, single-factor and orthogonal experiments were used to optimize extraction of ginkgolic acid from G. biloba sarcotestae. The results showed that ethanol was the best solvent, with the highest extraction rate for ginkgolic acid at 85% ethanol. On this basis, we measured ethanol volume fraction, extraction time, temperature and solidliquid ratio using an orthogonal experiment. By using absorbance of 310 nm as standard, the optimal extraction conditions were 85% ethanol with, solid-liquid ratio of1:14 at 40°C for 12 h. These conditions gave a ginkgolic acid yield of 73.1 mg·g^(–1). Subsequently, recombinant laccase was used to degrade the ginkgolic acid in several laccase/mediator systems, of which Lac C was the best. At50°C, p H 4.5, enzyme concentration of 0.01 U·m L^(–1),0.5 mmol·L^(–1) mediator ABTS and reaction time of 3 h, the degradation rate of ginkgolic acid reached 100%. 16A Multi-Category Brain Tumor Classification Method Bases on Improved ResNet50显示文摘Brain tumor is one of the most common tumors with high mortality.Early detection is of great significance for the treatment and rehabilitation of patients.The single channel convolution layer and pool layer of traditional convolutional neural network(CNN)structure can only accept limited local context information.And most of the current methods only focus on the classification of benign and malignant brain tumors,multi classification of brain tumors is not common.In response to these shortcomings,considering that convolution kernels of different sizes can extract more comprehensive features,we put forward the multi-size convolutional kernel module.And considering that the combination of average-pooling with max-pooling can realize the complementary of the high-dimensional information extracted by the two structures,we proposed the dual-channel pooling layer.Combining the two structures with ResNet50,we proposed an improved ResNet50 CNN for the applications in multi-category brain tumor classification.We used data enhancement before training to avoid model over fitting and used five-fold cross-validation in experiments.Finally,the experimental results show that the network proposed in this paper can effectively classify healthy brain,meningioma,diffuse astrocytoma,anaplastic oligodendroglioma and glioblastoma.Linguo Li Shujing Li Jian Su 2021Computers, Materials & Continua2021,,11:0
17Traffic signal control in mixed traffic environment based on advance decision and reinforcement learning显示文摘Reinforcement learning-based traffic signal control systems (RLTSC) can enhance dynamic adaptability, save vehicle travelling timeand promote intersection capacity. However, the existing RLTSC methods do not consider the driver’s response time requirement, sothe systems often face efficiency limitations and implementation difficulties.We propose the advance decision-making reinforcementlearning traffic signal control (AD-RLTSC) algorithm to improve traffic efficiency while ensuring safety in mixed traffic environment.First, the relationship between the intersection perception range and the signal control period is established and the trust region state(TRS) is proposed. Then, the scalable state matrix is dynamically adjusted to decide the future signal light status. The decision will bedisplayed to the human-driven vehicles (HDVs) through the bi-countdown timer mechanism and sent to the nearby connected automatedvehicles (CAVs) using the wireless network rather than be executed immediately. HDVs and CAVs optimize the driving speedbased on the remaining green (or red) time. Besides, the Double Dueling Deep Q-learning Network algorithm is used for reinforcementlearning training;a standardized reward is proposed to enhance the performance of intersection control and prioritized experiencereplay is adopted to improve sample utilization. 18Identification of Crop Diseases Based on Improved Genetic Algorithm and Extreme Learning Machine显示文摘As an indispensable task in crop protection,the detection of crop diseases directly impacts the income of farmers.To address the problems of low crop-disease identification precision and detection abilities,a new method of detection is proposed based on improved genetic algorithm and extreme learning machine.Taking five different typical diseases with common crops as the objects,this method first preprocesses the images of crops and selects the optimal features for fusion.Then,it builds a model of crop disease identification for extreme learning machine,introduces the hill-climbing algorithm to improve the traditional genetic algorithm,optimizes the initial weights and thresholds of the machine,and acquires the approximately optimal solution.And finally,a data set of crop diseases is used for verification,demonstrating that,compared with several other common machine learning methods,this method can effectively improve the crop-disease identification precision and detection abilities and provide a basis for the identification of other crop diseases.Linguo Li Lijuan Sun Jian Guo Shujing Li Ping Jiang 2020Computers, Materials & Continua2020,,10:0
