| 3 | Prognostic assessment of apoptotic gene polymorphisms in non-small cell lung cancer in Chinese显示文摘Apoptosis plays a key role in inhibiting tumor growth, progression and resistance to anti-tumor therapy. We hypothesized that genetic variants in apoptotic genes may affect the prognosis of lung cancer. To test this hypothesis, we selected 38 potentially functional single nucleotide polymorphisms (SNPs) from 12 genes (BAX, BCL2, BID, CASP3, CASP6, CASP7, CASP8, CASP9, CASP10, FAS, FASLG and MCL1) involved in apoptosis to assess their prognostic significance in lung cancer in a Chinese case cohort with 568 non-small cell lung cancer (NSCLC) patients. Thirty-five SNPs passing quality control underwent association analyses, 11 of which were shown to be significantly associated with NSCLC survival (P<0.05). After Cox stepwise regression analyses, 3 SNPs were independently associated with the outcome of NSCLC (BID rs8190315: P=0.003; CASP9 rs4645981: P=0.007 and FAS rs1800682: P=0.016). A favorable survival of NSCLC was significantly associated with the genotypes of BID rs8190315 AG/GG (adjusted HR=0.65, 95% CI: 0.49-0.88), CASP9 rs4645981 AA (HR=0.22, 95% CI: 0.07-0.69) and FAS rs1800682 GG (adjusted HR=0.67, 95% CI: 0.46-0.97). Time-dependent receptor operation curve (ROC) analysis revealed that the area under curve (AUC) at year 5 was significantly increased from 0.762 to 0.819 after adding the risk score of these 3 SNPs to the clinical risk score. The remaining 32 SNPs were not significantly associated with NSCLC prognosis after adjustment for these 3 SNPs. | 5 | Rotation-based finite elements:reference-configuration geometry and motion description显示文摘Infinitesimal-rotation finite elements allow creating a linear problem that can be exploited to systematically reduce the number of coordinates and obtain efficient solutions for a wide range of applications,including those governed by nonlinear equations.This paper discusses the limitations of conventional infinitesimal-rotation finite elements(FE)in capturing correctly the initial stress-free reference-configuration geometry,and explains the effect of these limitations on the definition of the inertia used in the motion description.An alternative to conventional infinitesimal-rotation finite elements is a new class of elements that allow developing inertia expressions written explicitly in terms of constant coefficients that define accurately the reference-configuration geometry.It is shown that using a geometrically inconsistent(GI)approach that introduces the infinitesimal-rotation coordinates from the outset to replace the interpolation-polynomial coefficients is the main source of the failure to capture correctly the reference-configuration geometry.On the other hand,by using a geometrically consistent(GC)approach that employs the position gradients of the absolute nodal coordinate formulation(ANCF)to define the infinitesimal-rotation coordinates,the reference-configuration geometry can be preserved.Two simple examples of straight and tapered beams are used to demonstrate the basic differences between the two fundamentally different approaches used to introduce the infinitesimal-rotation coordinates.The analysis presented in this study sheds light on the differences between the incremental co-rotational solution procedure,widely used in computational structural mechanics,and the non-incremental floating frame of reference formulation(FFR),widely used in multibody system(MBS)dynamics. | Ahmed A.Shabana Lingmin Xu | 2021 | Acta Mechanica Sinica2021,37,1: | 4 |

Methods:The structure,function,drug target and vaccine target of CPR from Plasmodium falciparum were analyzed and predicted by bioinformatics methods.Results:PfCPR,which was older CPR,had close relationship with the CPR from other Plasmodium species,but it was distant from its hosts,such as Homo sapiens and Anopheles.PfCPR was located in the cellular nucleus of Plasmodium falciparum.335aa-352aa and 591aa - 608aa were inserted the interior side of the nuclear membrane,while 151aa-265aa was located in the nucleolus organizer regions.PfCPR had 40 function sites and 44 protein-protein binding sites in amino acid sequence.The teriary structure of laa-700aa was forcep-shaped with wings.15 segments of PfCPR had no homology with Homo sapien CPR and most were exposed on the surface of the protein.These segments had 25 protein-protein binding sites.While 13 other segments all possessed function sites. | 9 | Association of long non-coding RNA HOTAIR and MALAT1 variants with cervical cancer risk in Han Chinese women显示文摘Long noncoding RNA(lncRNA) HOTAIR and MALAT1 are implicated in the development of multiple cancers. Genetic variants within HOTAIR and MALAT1 may affect the gene expression, thereby modifying genetic susceptibility to cervical cancer. A case-control study was designed, including 1 486 cervical cancer patients and 1 536 healthy controls. Based on RegulomeDB database, 11 SNPs were selected and genotyped by using Sequenom’s Mass ARRAY. Univariate and multivariate logistic regression models were used to calculate the odds ratio(OR) and 95% confidence interval(CI). We found that the A allele of rs35643724 in HOTAIR was associated with increased risk of cervical cancer, while the C allele of rs1787666 in MALAT1 was associated with decreased risk. Compared to individuals with 0–1 unfavorable allele, those with 3–4 unfavorable alleles showed18% increased odds of having cervical cancer. Drug-resistant cellular models are essential for understanding the drug resistance and developing new therapeutics. The low efficiency and long time required in creating these models are major obstacles hindering drug resistance research and drug screening. Herein, we report an approach that can accelerate(shortening the time from years to 3 weeks) the establishment of cancer cell line-based, inheritable drug resistance models by specific knockout of MED12 gene using CRISPR/Cas9 system. The resultant MED12^(KO) A375(melanoma)cell line was resistant to inhibitors of B-Raf proto-oncogene, serine/threonine kinase(BRAF), whereas the resultant MED12^(KO) PC9(non-small cell lung cancer) cell line was resistant to inhibitors of epidermal growth factor receptor(EGFR). Evaluation of anti-cancer drugs and their combinations shows that certain combinations of BRAF inhibitors and TGF-β receptor(TGF-βR) inhibitors are active in suppressing the growth of MED12^(KO) A375 cells, and a few combinations of EGFR inhibitors and TGF-βR inhibitors were active in suppressing the growth of MED12^(KO) PC9 cells.
