1Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157
2Effect of cholecystokinin octapeptide on tumor necrosis factor α transcription and nuclear factor-κB activity induced by lipopolysaccharide in rat pulmonary interstitial macrophages显示文摘AIM: To elucidate the anti-inflammatory mechanism ofan intestinal neuropeptide, sulfated cholecystokininoctapeptide (sCCK-8), the effects of sCCK-8 onlipopolysaccharide (LPS)-induced tumor necrosis factorpulmonary interstitial macrophages (PIMs) werestudied.METHODS: PIMs from rat were stimulated with LPS(:1mg @ L-1) in the presence or absence of sCCK-8 (10-8-:10-6mol@ L-1) or/and CCK receptor antagonistproglumide (2 mg @ L-1). The expression of TNF-α mRNAwas assayed by reverse transcription polymerase chainreaction (RT-PCR) at 3h of the stimulation, and nuclearelectrophoretic mobility shift assay (EMSA) at 1 h ofat 30 min of the stimulation was detected by Westernblot.RESULTS: sCCK-8, at concentrations from 10-8 mol @ L-1to 10-6 mol @ L-1 obviously inhibited LPS-induced TNF-αdependent manner, P<0.05, P<0.01. Stimulation PIMsP<0.01, which was elevated by sCCK-8, P<0.05. 3Pyroptosis: mechanisms and diseases显示文摘Currently,pyroptosis has received more and more attention because of its association with innate immunity and disease.The research scope of pyroptosis has expanded with the discovery of the gasdermin family.A great deal of evidence shows that pyroptosis can affect the development of tumors.The relationship between pyroptosis and tumors is diverse in different tissues and genetic backgrounds.In this review,we provide basic knowledge of pyroptosis,explain the relationship between pyroptosis and tumors,and focus on the significance of pyroptosis in tumor treatment.In addition,we further summarize the possibility of pyroptosis as a potential tumor treatment strategy and describe the side effects of radiotherapy and chemotherapy caused by pyroptosis.In brief,pyroptosis is a double-edged sword for tumors.The rational use of this dual effect will help us further explore the formation and development of tumors,and provide ideas for patients to develop new drugs based on pyroptosis.Pian Yu Xu Zhang Nian Liu Ling Tang Cong Peng Xiang Chen 2021Signal Transduction and Targeted Therapy2021,6,4:26

5Research on in-situ condition preserved coring and testing systems显示文摘As shallow resources are increasingly depleted,the mechanics'theory and testing technology of deep insitu rock has become urgent.Traditional coring technologies obtain rock samples without retaining the in-situ environmental conditions,leading to distortion of the measured parameters.Herein,a coring and testing systems retaining in-situ geological conditions is presented:the coring system that obtains in-situ rock samples,and the transfer and testing system that stores and analyzes the rocks under a reconstructed environment.The ICP-Coring system mainly consists of the pressure controller,active insulated core reactor and insulation layer and sealing film.The ultimate bearing strength of 100 MPa for pressurepreservation,temperature control accuracy of 0.97%for temperature-retained are realized.CH_(4)and CO permeability of the optimized sealing film are as low as 3.85 and 0.33 ppm/min.The average tensile elongation of the film is 152.4%and the light transmittance is reduced to 0%.Additionally,the pressure and steady-state temperature accuracy for reconstructing the in-situ environment of transfer and storage system up to 1%and±0.2 is achieved.The error recorded of the noncontact sensor ring made of lowdensity polymer is less than 6%than that of the contact test.The system can provide technical support for the deep in-situ rock mechanics research,improving deep resource acquisition capabilities and further clarifying deep-earth processes.He-Ping Xie Tao Liu Ming-Zhong Gao Ling Chen Hong-Wei Zhou Yang Ju Feng Gao Xiao-Bo Peng Xiong-Jun Li Rui-Dong Peng Ya-Nan Gao Cong Li Zhi-Qiang He Ming-Qing Yang Zhi-Yu Zhao 2021Petroleum Science2021,18,6:19
6Genetic variation of yellow pigment and its components in foxtail millet(Setaria italica(L.) P.Beauv.) from different eco-regions in China显示文摘Kernel color is an important trait for assessing the commercial and nutritional quality of foxtail millet.Yellow pigment content(YPC)and carotenoid components(lutein and zeaxanthin)of 270 foxtail millet accessions,including 50 landraces and 220improved cultivars,from four different eco-regions in China were surveyed using spectrophotometry and high performance liquid chromatography methods.Results indicated that YPC had rich variance,ranging from 1.91 to 28.54 mg kg^(–1),with an average value of 17.80 mg kg^(–1).The average YPC of improved cultivars(18.31 mg kg^(–1))was significantly higher than that of landraces(15.51 mg kg^(–1)).The YPC in cultivars from the Loess Plateau spring sowing region(LPSSR)was the highest(20.59 mg kg^(–1)),followed by the North China summer sowing region(NCSSR,18.25 mg kg^(–1)),the northeast spring sowing region(NSSR,17.25 mg kg^(–1)),and the Inner Mongolia Plateau spring sowing region(IMPSSR,13.92 mg kg^(–1)).The variation coefficients of YPC in cultivars from NSSR,LPSSR,and IMPSSR were higher than that from NCSSR.A similar carotenoid profile was also obtained for 270 foxtail millet cultivars.Lutein and zeaxanthin accounted for approximately 55–65%of YPC in accessions.The lutein content was higher than zeaxanthin content in all cultivars.The ratio of lutein to zeaxanthin ranged from 1.51 to 6.06 with an average of 3.34.YPC was positively correlated with lutein(r=0.935,P<0.01),zeaxanthin(r=0.808,P<0.01),and growth duration(r=0.488,P<0.01),whereas it was negatively correlated with grain protein(r=-0.332,P<0.01)and 1 000-kernel weight(r=-0.153,P<0.05).Our study is useful for screening and selecting cultivars with high levels of yellow pigment and for enhancing phytochemical concentrations in breeding programs.YANG Yan-bing JIA Guan-qing DENG Li-gang QIN Ling CHEN Er-ying CONG Xin-jun ZOU Ren-feng WANG Hai-lian ZHANG Hua-wen LIU Bin GUAN Yan-an DIAO Xian-min YIN Yan-ping 2017Journal of Integrative Agriculture2017,16,11:15

8Clinical and muscle magnetic resonance image findings in patients with late-onset multiple acyl-CoA dehydrogenase deficiency显示文摘Background:Late-onset multiple acyl-coA dehydrogenase deficiency (MADD) is an autosomal recessive inherited metabolic disorder. It is still unclear about the muscle magnetic resonance image (MRI) pattern of the distal lower limb pre- and post-treatment in patients with late-onset MADD. This study described the clinical and genetic findings in a cohort of patients with late-onset MADD, and aimed to characterize the MRI pattern of the lower limbs.Methods:Clinical data were retrospectively collected from clinic centers of Peking University People's Hospital between February 2014 and February 2018. Muscle biopsy, blood acylcarnitines, and urine organic acids profiles, and genetic analysis were conducted to establish the diagnosis of MADD in 25 patients. Muscle MRI of the thigh and leg were performed in all patients before treatment. Eight patients received MRI re-examinations after treatment.Results:All patients presented with muscle weakness or exercise intolerance associated with variants in the electron transfer flavoprotein dehydrogenase gene. Muscle MRI showed a sign of both edema-like change and fat infiltration selectively involving in the soleus (SO) but sparing of the gastrocnemius (GA) in the leg. Similar sign of selective involvement of the biceps femoris longus (BFL) but sparing of the semitendinosus (ST) was observed in the thigh. The sensitivity and specificity of the combination of either 'SO+/GA-' sign or 'BFL+/ST-' sign for the diagnosis of late-onset MADD were 80.0% and 83.5%, respectively. Logistic regression model supported the findings. The edema-like change in the SO and BFL muscles were quickly recovered at 1 month after treatment, and the clinical symptom was also relieved.Conclusions:This study expands the clinical and genetic spectrums of late-onset MADD. Muscle MRI shows a distinct pattern in the lower limb of patients with late-onset MADD. 11Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 13Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang 2020National Science Review2020,7,6:6
14Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5

While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 17Identification of microRNAs involved in crosstalk between nitrogen, phosphorus and potassium under multiple nutrient deficiency in sorghum显示文摘Nitrogen(N),phosphorus(P),and potassium(K)are important for plant growth and development.MicroRNAs(miRNAs)play important roles in regulating plant response to nutrient(N,P,and K)deficiencies.Several miRNAs have been identified under nutrient deficiency conditions in many plant species.However,the manner in which miRNAs regulate the interaction between NPK signaling pathways under multiple nutrient deficiency remains largely unknown.We systematically compared and identified microRNAs involved in both single and triple NPK nutrient deficiency responses.We identified 32 shoot and 17 root miRNAs differentially expressed under potassium deficiency.Several NP starvation-associated miRNAs including miR169s and miR399s,were also regulated by K deficiency.Several identified miRNAs including miR5565c,miR5564,and miR1432 have not previously been associated with respectively N,P,and K deficiency(−N,−P,and−K).Expression correlation analysis between miRNAs and their predicted targets showed that miR169,miR172,and miR160 displayed expression trends exactly opposite to those of their corresponding predicted targets.Of 550 predicted novel miRNAs,novel_mir_42 was upregulated in shoots under−K but was downregulated under−N and−P.The effects of combined NPK starvation were not a simple addition of the individual stresses on sorghum seedlings.The identified common and specific differentially expressed miRNAs were observed under single and combined NPK deficiencies.These findings will help to further elucidate the functions of microRNAs and their interactions under multiple nutrient deficiency.Zhenxing Zhu Dan Li Ling Cong Xiaochun Lu 2021The Crop Journal2021,9,2:4
18Utility of Droplet Digital PCR Assay for Quantitative Detection of Norovirus in Shellfish, from Production to Consumption in Guangxi, China显示文摘Objective Shellfish are recognized as important vehicles of norovirus-associated gastroenteritis. The present study aimed to monitor norovirus contamination in oysters along the farm-to-fork continuum in Guangxi, a major oyster production area in Southwestern China. Methods Oyster samples were collected monthly from farms, markets, and restaurants, from January to December 2016. Norovirus was detected and quantified by one-step reverse transcription-droplet digital polymerase chain reaction(RT-ddPCR). Results A total of 480 oyster samples were collected and tested for norovirus genogroups I and II. Norovirus was detected in 20.7% of samples, with genogroup II predominating. No significant difference was observed in norovirus prevalence among different sampling sites. The norovirus levels varied widely, with a geometric mean of 19,300 copies/g in digestive glands. Both norovirus prevalence and viral loads showed obvious seasonality, with a strong winter bias. Conclusion This study provides a systematic analysis of norovirus contamination ‘from the farm to the fork' in Guangxi. RT-ddPCR can be a useful tool for detection and quantification of low amounts of norovirus in the presence of inhibitors found particularly in foodstuffs. 19Dissection of the genetic architecture for tassel branch number by QTL analysis in two related populations in maize显示文摘Tassel branch number(TBN) is the principal component of maize tassel inflorescence architecture and is a typical quantitative trait controlled by multiple genes. The main objective of this research was to detect quantitative trait loci(QTLs) for TBN. The maize inbred line SICAU1212 was used as the common parent to develop BC1S1 and recombinant inbred line(RIL) populations with inbred lines 3237 and B73, respectively. The two related populations consisted of 123 and 238 lines, respectively. Each population was grown and phenotyped for TBN in two environments. Eleven QTLs were detected in the BC1S1 population, located on chromosomes 2, 3, 5, and 7, accounted for 4.45–26.58% of the phenotypic variation. Two QTLs(qB11Jtbn2-1, qB12Ctbn2-1, qBJtbn2-1; q11JBtbn5-1, qB12Ctbn5-1, qBJtbn5-1) that accounted for more than 10% of the phenotypic variation were identified. Three QTLs located on chromosomes 2, 3 and 5, exhibited stable expression in the two environments. Ten QTLs were detected in the RIL population, located on chromosomes 2, 3, 5, 8, and 10, accounted for 2.69–13.58% of the TBN variation. One QTL(qR14Dtbn2-2) explained >10% of the phenotypic variation. One common QTL(qB12Ctbn2-2, qR14Dtbn2-2, q RJtbn2-2) was detected between the two related populations. Three pairs of epistatic effects were identified between two loci with or without additive effects and accounted for 1.19–4.26% of the phenotypic variance. These results demonstrated that TBN variation was mainly caused by major effects, minor effects and slightly modified by epistatic effects. 20The boundary conditions for simulations of a shake-table experiment on the seismic response of 3D slope显示文摘Boundary conditions can significantly affect a slope's behavior under strong earthquakes.To evaluate the importance of boundary conditions for finite element(FE) simulations of a shake-table experiment on the slope response, a validated three-dimensional(3D) nonlinear FE model is presented, and the numerical and experimental results are compared.For that purpose, the robust graphical user-interface 'Slope SAR', based on the open-source computational platform Open Sees, is employed, which simplifies the effort-intensive pre-and post-processing phases.The mesh resolution effect is also addressed.A parametric study is performed to evaluate the influence of boundary conditions on the FE model involving the boundary extent and three types of boundary conditions at the end faces.Generally, variations in the boundary extent produce inconsistent slope deformations.For the two end faces, fixing the y-direction displacement is not appropriate to simulate the shake-table experiment, in which the end walls are rigid and rough.In addition, the influence of the length of the 3D slope's top face and the width of the slope play an important role in the difference between two types of boundary conditions at the end faces(fixing the y-direction displacement and fixing the(y, z) direction displacement).Overall, this study highlights that the assessment of a comparison between a simulation and an experimental result should be performed with due consideration to the effect of the boundary conditions.Tang Liang Cong Shengyi Ling Xianzhang Ju Nengpan 2017Earthquake Engineering and Engineering Vibration2017,16,1:3
