| 1 | SDF-1/CXCR4 axis modulates bone marrow mesenchymal stem cell apoptosis, migration and cytokine secretion显示文摘Bone marrow mesenchymal stem cells(MSCs)are considered as a promising cell source to treat the acute myocardial infarction.However,over 90%of the stem cells usually die in the first three days of transplantation.Survival potential,migration ability and paracrine capacity have been considered as the most important three factors for cell transplantation in the ischemic cardiac treatment.We hypothesized that stromal-derived factor-1(SDF-1)/CXCR4 axis plays a critical role in the regulation of these processes.In this study,apoptosis was induced by exposure of MSCs to H2O2 for 2 h.After re-oxygenation,the SDF-1 pretreated MSCs demonstrated a significant increase in survival and proliferation.SDF-1 pretreatment also enhanced the migration and increased the secretion of pro-survival and angiogenic cytokines including basic fibroblast growth factor and vascular endothelial growth factor.Western blot and RT-PCR demonstrated that SDF-1 pretreatment significantly activated the pro-survival Akt and Erk signaling pathways and up-regulated Bcl-2/Bax ratio.These protective effects were partially inhibited by AMD3100,an antagonist of CXCR4.We conclude that the SDF-1/CXCR4 axis is critical for MSC survival,migration and cytokine secretion. | Xiaolei Liu Biyan Duan Zhaokang Cheng Xiaohua Jia Lina Mao Hao Fu Yongzhe Che Lailiang Ou Lin Liu Deling Kong | 2011 | Protein & Cell2011,2,10: | 53 |

| 3 | Effects of dietary Clostridium butyricum supplementation on growth performance,intestinal development,and immune response of weaned piglets challenged with lipopolysaccharide显示文摘Background: Weanling pigs, with immature immune system and physiological function, usually experience postweaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide(LPS).Methods: In Experiment(Exp.) 1,144 weaned piglets were weaned at 21 d and randomly assigned to six groups,with six replicates per group and four pigs per replicate, receiving a control diet(CON) or diet supplemented with antibiotics(AB) or C. butyricum(CB)(0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB(0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS(50 μg/kg body weight) and the other six piglets with normal saline;animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis.Results: In Exp. 1, CB and AB diets had no effect on growth performance of piglets. In Exp. 2, 0.4% CB decreased feed-gain ratio(P < 0.1), diarrhea score(P < 0.05), and increased duodenal, jejunal, and ileal villus height and jejunal villus height/crypt depth(P < 0.05). The 0.4% CB decreased the plasma tumor necrosis factor(TNF) α(P < 0.05) but increased ileal mucosa IL-10 and TLR2 mRNA expression(P < 0.05). Furthermore, 0.4% CB altered the microbial profile, with Bacillus and Ruminococcaceae UGG-003 at genus level and Lactobacillus casei and Parasutterella secunda at species level were higher than CON in colonic content(P < 0.05).Conclusions: Dietary C. butyricum supplementation had positive effects on growth of weaned piglets with less digestible diets. There was a tendency to reduce the feed-gain ratio, which could reduce feed costs in pig production. | 5 | Review grain yield and nitrogen use efficiency in rice production regions in China显示文摘As one of the staple food crops, rice(Oryza sativa L.) is widely cultivated across China, which plays a critical role in guaranteeing national food security. Most previous studies on grain yield or/and nitrogen use efficiency(NUE) of rice in China often involved site-specific field experiments, or small regions with insufficient data, which limited the representation for the current rice production regions. In this study, a database covering a wide range of climate conditions, soil types and field managements across China, was developed to estimate rice grain yield and NUE in various rice production regions in China and to evaluate the relationships between N rates and grain yield, NUE. According to the database for rice, the values of grain yield, plant N accumulation, N harvest index(HIN), indigenous N supply(INS), internal N efficiency(IE_N), reciprocal internal N efficiency(RIE_N), agronomic N use efficiency(AE_N), partial N factor productivity(PEPN), physiological N efficiency(PE_N), and recover efficiency of applied N(RE_N) averaged 7.69 t ha^(–1), 152 kg ha^(–1), 0.64 kg kg^(–1), 94.1 kg kg^(–1), 53.9 kg kg^(–1), 1.98 kg kg^(–1), 12.6 kg kg^(–1), 48.6 kg kg^(–1), 33.8 kg kg^(–1), and 39.3%, respectively. However, the corresponding values all varied tremendously with large variation. Rice planting regions and N rates had significant influence on grain yield, N uptake and NUE values. Considering all observations, N rates of 200 to 250 kg ha^(–1) commonly achieved higher rice grain yield compared to less than 200 kg N ha^(–1) and more than 250 kg N ha^(–1) at most rice planting regions. At N rates of 200 to 250 kg ha^(–1), significant positive linear relationships were observed between rice grain yield and AE_N, PE_N, RE_N, IE_N, and PFPN, and 46.49, 24.64, 7.94, 17.84, and 88.24% of the variation in AE_N, PE_N, RE_N, IE_N, and PFPN could be explained by grain yield, respectively. | 6 | The roles of surfactant protein D during Aspergillus fumigatus infection in human corneal epithelial cells显示文摘AIM: To investigate roles of surfactant protein D (SP-D) and relative cytokines in human corneal epithelial(HCE) cells exposed to aspergillus fumigatus (AF) antigens.METHODS: HCE cells cultured in vitro with AF antigens and sampled at 0, 0.5, 1 hour, 2, 4, 6 and 8 hours. The expression of SP-D mRNA was evaluated by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR).The expression of SP-D protein was shown by ELISA and immunocytochemistry SP methods. The expression of NF-κB and relative downstream cytokines such as TNF-α, IL-1β, IL-8 and IL-10 in supernatant fluid were measured by ELISA.RESULTS: SP-D mRNA and protein were detected in untreated HCE cells. The expression of SP-D and the relative downstream cytokines rose after being stimulated with AF antigens. SP-D mRNA began to rise at 0.5 hour and the most significantly peak was in 2 hours. The protein of SP-D in supernatant fluid had the same trend with mRNA. Immunocytochemistry of SP-D showed positive expression and gradually increased to 6 hours, and then the expression began to decline. NF-κB was activated after treated by AF antigens and the changes had correlation with SP-D. TNF-α, IL-1β, IL-8 and IL-10 began to rise after given AF antigens 1 hour and were 1.82, 1.43, 1.12 and 1.28 times higher than the untreated HCE cells separately. The expression of TNF-α and IL-1β reached the peak at 2 hours, separately 2.80 and 2.86 times than the untreated. The expression of IL-8 and IL-10 gradually increased with a time-dependent manner.CONCLUSION: HCE cells exists SP-D and it may play a significant role in pathogenesis of keratomycosis. AF may induce human corneal epithelial cells to express inflammatory cytokines via SP-D and NF-κB pathway. | 7 | Boxb mediate BALB/c mice corneal inflammation through a TLR4/MyD88-dependent signaling pathway in Aspergillus fumigatus keratitis显示文摘AIM: To investigate whether high-mobility group box 1(HMGB1) Boxb exacerbates BALB/c mice corneal immune responses and inflammatory through the Toll-like receptor 4(TLR4)/myeloid differentiation primary response 88(My D88)-dependent signaling pathway in Aspergillus fumigatus(A. fumigatus) keratitis.METHODS: The mice corneas were pretreated with phosphate buffer saline(PBS), Boxb before A. fumigatus infection. The abdominal cavity extracted macrophages were pretreated with PBS, Boxb, TLR4 inhibitor(CLI-095), Dimethyl sulfoxide(DMSO) separately before A. fumigatus hyphae stimulation. HMGB1 was detected in normal and infected mice corneas and macrophages by real-time reverse transcriptase polymerase chain reaction(RT-PCR), the TLR4, My D88, interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) were detected by Western blot and PCR.RESULTS: In BALB/c mice corneas, the expressions of TLR4, HMGB1, IL-1β, TNF-α were increased after A. fumigatus infection. While pretreatment with Boxb significantly increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α compared with PBS control after infection. In BALB/c mice abdominal cavity extracted macrophages, pretreatment with Boxb increased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α, while pretreatment with CLI-095 and Boxb significantly decreased the expressions of TLR4, HMGB1, My D88, IL-1β, TNF-α. | 8 | β-elemene enhances the radiosensitivity of gastric cancer cells by inhibiting Pak1 activation显示文摘AIM:To explore the potential of β-elemene as a radiosensitizer for gastric cancer cells and the underlying mechanisms.METHODS:SGC7901,MKN45,MKN28,N87,and AGS human gastric cancer cell lines were used to screen for radioresistant gastric cancer cell lines. A 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium(MTT) assay was used to determine the effects of β-elemene and IPA-3 on cell viability in MKN45 and SGC7901 gastric cancer cell lines. A clonogenic survival assay and annexin V-FITC/PI apoptosis detection assay were used to evaluate cellular radiosensitivity and radiation-induced cell death,respectively. A proteomic method,isobaric tags for relative and absolute quantitation(i TRAQ),was employed to screen the proteins regulated by β-elemene pretreatment prior to ionizing radiation(IR) in SGC7901 gastric cancer cell line. IPA-3 was used as a specific small molecule inhibitor of p21-activated protein kinase 1(Pak1) to target Pak1 signaling. Protein levels of PAK1IP1(p21-activated protein kinase-interacting protein 1),total Pak1(t-Pak1),phospho-Pak1(T423),phospho-ERK1/2( Thr202/Tyr204),and cleaved caspase-3(17 k Da) were assessed by western blotting.RESULTS:MKN45 and SGC7901 gastric cancer cell lines were relatively more resistant to IR. β-elemene pretreatment decreased clonogenic survival following IR in MKN45 and SGC7901 gastric cancer cell lines. Additionally,β-elemene pretreatment prior to IR increased radiation-induced cell death compared with IR alone in MKN45(10.4% ± 0.9% vs 34.8% ± 2.8%,P < 0.05) and SGC7901(11.6% ± 0.9% vs 46.7% ± 5.2%,P < 0.05) human gastric cancer cell lines,respectively,consistent with the level of cleaved caspase-3(17 k Da). Through i TRAQ analysis and western blot validation,we found that β-elemene upregulated PAK1IP1 and downregulated phospho-Pak1(T423) and phosphoERK1/2 in SGC7901 gastric cancer cells. IR increased the level of phospho-Pak1(T423). Pretreatment with β-elemene decreased radiation-induced Pak1 and ERK1/2 phosphorylation. Inhibition of Pak1 using IPA-3 decreased clonogenic survival following IR. In addition,IPA-3 increased radiation-induced cell death in MKN45(13.4% ± 0.3% vs 26.6% ± 1.0%,P < 0.05) and SGC7901(16.0% ± 0.6% vs 37.3% ± 1.7%,P < 0.05) gastric cancer cell lines,respectively,consistent with the level of cleaved caspase-3(17 k Da). | 10 | Effects of dietary supplementation with essential oils and protease on growth performance,antioxidation,inflammation and intestinal function of weaned pigs显示文摘This experiment evaluated the impacts of essential oils(EO)and protease(PRO),independently or in combination,on growth performance,antioxidation,inflammation and intestinal function of weaned pigs.One hundred and sixty weaned pigs(21 d of age,BW of 6.74±0.20 kg)were randomly divided into 4 treatments with 8 replicate pens of 5 pigs per pen.Dietary treatments included the following:1)control diet(CON),2)CON with 300 mg/kg essential oils(EO),3)CON with 500 mg/kg protease(PRO),4)CON with 300 mg/kg essential oil and 500 mg/kg protease(EO + PRO).On d 8,one pig from each penwas selected for sampling.The remaining pigs were fed for an additional week and growth performance was monitored during this period.Dietary treatments had no marked effects(P>0.05)on the growth performance of pigs.However,pigs receiving EO diet had higher(P<0.05)serumglutathione peroxidase(GSH-Px)activity,and tended to decrease(P=0.063)serum concentration of tumor necrosis factor-a(TNF-a).In addition,pigs receiving EO diet had higher(P<0.05)abundances of phylum Actinobacteria,and genera Bifidobacterium,and lower(P<0.05)phylum Bacteroidetes and genera Alloprevotella in colonic digesta.Pigs receiving PRO diet decreased(P<0.05)the serum concentration of malondialdehyde(MDA)and diamine oxidase activity,increased(P<0.05)the villus height and the ratio of villus height to crypt depth in duodenum,increased sucrase activity in jejunal mucosa,and also increased the abundance of phylum Actinobacteria in colonic digesta.Furthermore,the synergistic effects of EO and PROwas observed(P<0.05)for pigs with decreasing serum TNF-a concentration and increasing serum GSH-Px activity.Collectively,the results indicated that dietary supplementation of EO and PRO had no significant effects on growth performance of weaned pigs.EO diet appeared to improve antioxidant activity and intestinal microbiota,while PRO diet improved intestinal morphology and digestive enzyme activity,and there was a synergistic effect of EO and PRO on reducing inflammatory parameters in weaned pigs. | Xie Peng Qiang Zhou Cheng Wu Jian Zhao Quan Tan Ying He Liang Hu Zhengfeng Fang Yan Lin Shengyu Xu Bin Feng Jian Li Yong Zhuo Chris Van Ginneken Xuemei Jiang De Wu Lianqiang Che | 2022 | Animal Nutrition2022,,2: | 8 |

| 12 | Effects of COX-2 inhibitor NS-398 on IL-10 expression in rat fungal keratitis显示文摘·AIM:To investigate the expression of interleukin-10 (IL-10) and the effect of NS-398(COX-2 inhibitor) on the expression of IL-10 in fungal keratitis in rats,and analyze its effects on anti-fungus immunity.·METHODS:Ninety Wister rats were randomly divided into 3 groups.Group A was blank control group (10 eyes).Group B was fungal keratitis group (40 eyes).Group C was fungal keratitis group treated with NS-398 (40 eyes).PAS staining,100g/L potassium hydroxide (KOH) smear and fungal culture confirmed the successful establishment of fungal keratitis model.After the central epithelium was scraped,Fusarium solani colonies were applied and contact lens was put on the right cornea of group B and C,and plane contact lens was put on the left cornea of control eyes.Phosphate buffered saline (PBS) eyedrops were given for group B and NS-398 eyedrops for group C.The expression of IL-10 on corneas of group B and C on the 1st day,3rd days,7th days,and 14th days were detected by immunohistochemistry and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR).·RESULTS:Histopathologic examination showed neutrophil infiltration and severe tissue necrosis in ulcer cornea.PAS staining confirmed the existence of hyphae and spores in the superficial layer of stroma.In the blank and control groups almost no expression of IL-10 was detected at any observing points.In group B the expression of IL-10 increased at first and decreased thereafter.Its expression also showed significant difference at any observing points (P£?0.01).Compared with group B,the expression of IL-10 in group C showed no difference on the 1stday,decrease on the 3rd day,but a significant increase on the 7th day and 14th day.·CONCLUSION:IL-10 takes part in the occurrence and development of fungal keratitis.NS-398 can upgrade the expression of IL-10 in fungal keratitis in the later period of the ulcer.Meanwhile,pathologic observation showed a slightly corneal opacity.IL-10 may play an important role in the process of cornea anti-damage repair.· | Na Li Cheng-Ye Che Li-Ting Hu Jing Lin Qing Wang and Gui-Qiu Zhao | 2011 | International Journal of Ophthalmology(English edition)2011,4,2: | 7 |

| 14 | Expression of dectin-1 during fungus infection in human corneal epithelial cells显示文摘AIM: To evaluate the expression of dendritic cell-associated C-type lectin-1(dectin-1) in human corneal epithelial(HCE) cells infected by fungus. · METHODS: A total of 20 cases of healthy donor corneas were group A,and 20 patients(20 eyes) suffered from fungal keratitis(FK) composed group B. Real-time qPCR and immunohistochemistry were applied to detect dectin-1 expression in corneal epithelium of both groups. HCE cells were cultured with aspergillus fumigatus(AF) antigens in vitro. The expression of dectin-1 mRNA was measured by real-time qPCR at the stimulation of 0,4,8 and 24h separately. Dectin-1 protein was detected by immunocytochemistry at 0 and 24h separately. ·RESULTS: Dectin-1 expressed in corneal epithelium of normal persons and FK patients. Vitro cellular experiment showed that the expression of dectin-1 mRNA in HCE cells began to increase after stimulation of AF antigens at 4h,and dectin-1 protein expression increased after stimulation at 24h. · CONCLUSION: Dectin-1 expressed in corneal epithelium of normal persons. Eyelids, lateral canthus, medial canthus, palpebral conjunctiva, sclera and cornea, anterior chamber, lens, anterior vitreous were examined by slit lamp to find whether there were deposition of uric acid crystals, ocular vascular tortuosity, redness and subconjunctival hemorrhage. The ophthalmic exams of visual acuity, intraocular pressure, fundus were used to assess any gout-related eye disease. ·RESULTS: Uric acid crystals were found in 3 patients and the positions of the deposite were in corneal stroma, corneal epithelium and superficial stroma, and sclera respectively. The incidence was 0.79% . Dilatated and tortuous blood vessels in conjunctiva and sclera surface were found in 38 (23.8% ), 40 (44.0% ), 58 (45.0% ), 9 (18.0% ) patients in groups A, B, C and D, respectively. The differences between group B and D, group C and D were statistically significant(P <0.01, P <0.01).Transparent vesicles with metal-like reflected light in subconjunctiva were seen in 26 (16.2%), 29 (31.9%), 41 (31.8%), 2 (4.00%) patients in groups A, B, C and D, respectively. The differences between A and D, B and D, C and D were statistically significant (P <0.05, P <0.01, P <0.01). Subconjunctival hemorrhage was found in all groups, the difference among the four groups showed no statistically significance. · CONCLUSION: Gout can cause ocular surface abnormalities, such as tophi deposition, subconjunctival transparent vesicles and hemorrhage, and vascularchanges. | 16 | Effect of corneal graft diameter on therapeutic penetrating keratoplasty for fungal keratitis显示文摘AIM: To evaluate the effect of corneal graft diameter on therapeutic penetrating keratoplasty(PKP) for fungal keratitis. METHODS: A total of 116 patients (116 eyes) suffered from fungal keratitis underwent PKP at the Affiliated Hospital of Medical College Qingdao University from May 2006 to May 2010. They were divided into two groups according to the corneal graft diameter. 64 eyes' corneal graft diameter was 8.00mm or larger and 52 eyes' graft diameter was smaller than 8.00mm. The follow-up time was 2 years. The postoperative visual acuity and complications were documented and compared. RESULTS: Sixty-two (96.88%) eyes and fifty (96.15%) eyes preserved eyeballs respectively in two groups. There was no statistical difference in postoperative visual acuity (P =0.961), corneal graft clear rate (P =0.132) or the incidence of recurred fungal infection (P =0.770) between two groups. But there was a higher incidence of graft rejection (P =0.020) and secondary glaucoma (P =0.039) in group with corneal graft diameter 8.00mm or larger. CONCLUSION: PKP is an effective treatment approach for fungal keratitis. | 17 | Kinase domain insert containing receptor promoter controlled suicide gene system selectively kills human umbilical vein endothelial cells显示文摘AIM: To study the selective killing of human umbilical vein endothelial cells (HUVECs) by a double suicide gene under the regulation of a kinase domain insert containing receptor (KDR) promoter and mediated by an adenoviral gene vector. METHODS: Human KDR promoter was cloned by polymerase chain reaction (PCR), and two recombinant adenoviral plasmids pAdKDR-CdglyTK, pAdCMV-CDglyTK were constructed according to a two-step transformation protocol. These two newly constructed plasmids were then transfected into 293 packaging cells to grow adenovirus, which were further multiplied and purifi ed. HUVECs and LoVo cells were infected with either of the two resultant recombinant adenoviruses (AdKDR-CDglyTK and AdCMV-CDglyTK) respectively, and the infection rates were estimated by detection of green ? uorescent protein (GFP) expression. Infected cells were cultured in culture media containing different concentrations of 5-? uorocytosine (5-FC) and ganciclovir (GCV), and the killing effects were measured. RESULTS: The two recombinant adenoviral plasmids pAdKDR-CdglyTK, pAdCMV-CDglyTK were successfullyconstructed and transfected into 293 cells. The resultant recombinant adenoviruses infected cells caused similar infection rates; and the infected cells exhibited different sensitivity to the prodrugs: HUVECs infected with AdCMV-CDglyTK and LoVo cells infected with AdCMV-CDglyTK were highly sensitive to the prodrugs, and HUVECs infected with AdKDR-CDglyTK were similarly sensitive but signifi cantly more sensitive than the LoVo cells infected with AdKDR-CdglyTK (P < 0.001). CONCLUSION: Selective killing of HUVECs may be achieved by gene transfer of double suicide gene under the regulation of the KDR promoter. | 19 | Expression of S100B during the innate immune of corneal epithelium against fungi invasion显示文摘AIM: To explore the expression of S100 B in corneal epithelial cells under Aspergillus stimulation both in vivo and in vitro.·METHODS: Immortalized human corneal epithelial cells(HCECs) were exposed to inactive Aspergillus fumigatus(A. fumigatus) conidia at 0, 4, 8, 12, 16, and 24 h respectively. The corneas of Wistar rats were exposed to active A. fumigatus at 0, 12, 24, 48 h and the normal rat corneas were used for normal control. The m RNA level of S100 B was evaluated by real time quantitative reverse transcription-polymerase chain reaction(q RT-PCR).S100 B protein expression in cornea epithelium was detected by immunohistochemical/immunocytochemical staining(IHC/ICC).· RESULTS: Histopathology revealed a significant inflammatory cell infiltration in fungal keratitis human and rat cornea. Corneal epithelial cells didn't express or rarely express S100 B at baseline. A. fumigatus significantly induced S100 B m RNA expression in cultured corneal epithelial cells in a time depended manner in vitro, the m RNA began to rise significantly at8 h in vitro(P <0.05) and continue to rise as time prolonged(P <0.01). In vivo, S100 B m RNA level was low in the normal corneas. However, it was increased in keratitis corneas from 12 h after infection(P <0.05) and reached to a peak at 24h(P <0.001). | 20 | Dectin-1 expression at early period of Aspergillus fumigatus infection in rat's corneal epithelium显示文摘AIM:To investigate the expression of dendritic cell-associated C-type lectin-1 (dectin-1) at the early period of Aspergillus fumigatus infection in rat's corneal epithelium. ·METHODS:A total of 72 Wistar rats were randomly divided into three groups:A, B and C. The right eyes were chosen as experimental eyes. Group A was control group. Rats in group B were not inoculated with Aspergillus fumigatus. Group C was taken as Aspergillus fumigatus keratitis model. Rats in group B and C (six from each group) were executed randomly at 4, 8, 16 and 24 hours after experimental model being established to assess the expression of dectin-1 mRNA through real-time PCR. Another six rats in group B and C were executed randomly at 24 hours to assess the expression of dectin-1 protein through immunohistochemistry. ·RESULTS:The results of real-time PCR indicated that dectin-1 mRNA expression was low in corneal epithelium of normal rats'. There was no significantly difference of dectin-1 mRNA expression in group A and B (P >0.05). The expression of Aspergillus fumigatus infected corneal epithelium increased gradually after 8 hours in group C. The synchronous expression of group A and C had significant difference (P <0.01). Immunohistochemisty discovered that dectin-1 receptor existed in normal rat's corneal epithelium . Dectin-1 protein increased after 24 hours in group C. There was a significant difference of synchronous expression in group B and C(P<0.01). · CONCLUSION:Dectin-1 exists in rat's cornealepithelium and its expression significantly increases at the early period of Aspergillus fumigatus infection.
