1A human circulating immune cell landscape in aging and COVID-19显示文摘Age-associated changes in immune cells have been linked to an increased risk for infection.However,a global and detailed characterization of the changes that human circulating immune cells undergo with age is lacking.Here,we combined scRNA-seq,mass cytometry and sCATAC-seq to compare immune cell types in peripheral blood collected from young and old subjects and patients with COVID-19.We found that the immune cell landscape was reprogrammed with age and was characterized by T cell polarization from naive and memory cells to effector,cytotoxic,exhausted and reg-ulatory cells,along with increased late natural killer cells,age-associated B cells,inflammatory monocytes and age-associated dendritic cells.In addition,the expression of genes,which were implicated in coron-avirus susceptibility,was upregulated in a cell subtype-specific manner with age.Notably,COVID-19 promoted age-induced immune cell polarization and gene expression related to inflammation and cellular senes-cence.Therefore,these findings suggest that a dysreg-ulated immune system and increased gene expression associated with SARS-CoV-2 susceptibility may at least partially account for COVID-19 vulnerability in the elderly.Yingfeng Zheng Xiuxing Liu Wenqing Le Lihui Xie He Li Wen Wen Si Wang Shuai Ma Zhaohao Huang Jinguo Ye Wen Shi Yanxia Ye Zunpeng Liu Moshi Song Weiqi Zhang Jing-Dong J.Han Juan Carlos lzpisua Belmonte Chuanle Xiao Jing Qu Hongyang Wang Guang-Hui Liu Wenru Su 2020Protein & Cell2020,11,10:10

3Temporal-spatial distributions of road silt loadings and fugitive road dust emissions in Beijing from 2019 to 2020显示文摘Road silt loading(sL)is an important parameter in the fugitive road dust(FRD)emissions.In this study,the improved Testing Re-entrained Aerosol Kinetic Emissions from Roads(TRAKER)combined with the AP-42 method was firstly developed to quickly measure and estimate the sLs of paved roads in Beijing,China.The annual average sLs in Beijing was 0.59±0.31 g/m^(2)in 2020,and decreased by 22.4%compared with that in 2019.The seasonal variations of sLs followed the order of spring>winter>summer>autumn in the two years.The seasonal mean road sLs on the same type road in the four seasons presented a decline trend from^(2)019 to 2020,especially on the Expressway,decreasing 47.4%-72.7%.The road sLs on the different type roads in the same season followed the order of Major arterial∼Minor arterial∼Branch road>Express road,and Township road∼Country highway>Provincial highway∼National highway.The emission intensities of PM10 and PM_(2.5)from FRD in Beijing in 2020 were lower than those in 2019.The PM10 and PM_(2.5)emission intensities at the four planning areas in the two years all presented the order of the capital functional core area>the urban functional expansion area>the urban development new area>the ecological conservation and development area.The annual emissions of PM10 and PM_(2.5)from FRD in Beijing in 2020 were 74,886 ton and 18,118 ton,respectively,decreasing by∼33.3%compared with those in 2019.Haibin Wang Lihui Han Tingting Li Song Qu Yuncheng Zhao Shoubin Fan Tong Chen Haoran Cui Junfang Liu 2023Journal of Environmental Sciences2023,,10:1
4Aging weakens Th17 cell pathogenicity and ameliorates experimental autoimmune uveitis in mice显示文摘Aging-induced changes in the immune system are associated with a higher incidence of infection and vaccination failure.Lymph nodes,which filter the lymph to identify and fight infections,play a central role in this process.However,careful characterization of the impact of aging on lymph nodes and associated autoimmune diseases is lacking.We combined single-cell RNA sequencing(scRNA-seq)with flow cytometry to delineate the immune cell atlas of cervical draining lymph nodes(CDLNs)of both young and old mice with or without experimental autoimmune uveitis(EAU).We found extensive and complicated changes in the cellular constituents of CDLNs during aging.When confronted with autoimmune challenges,old mice developed milder EAU compared to young mice.Within this EAU process,we highlighted that the pathogenicity of T helper 17 cells(Th17)was dampened,as shown by reduced GM-CSF secretion in old mice.The mitigated secretion of GMCSF contributed to alleviation of IL-23 secretion by antigen-presenting cells(APCs)and may,in turn,weaken APCs’effects on facilitating the pathogenicity of Th17 cells.Meanwhile,our study further unveiled that aging downregulated GM-CSF secretion through reducing both the transcript and protein levels of IL-23R in Th17 cells from CDLNs.Overall,aging altered immune cell responses,especially through toning down Th17 cells,counteracting EAU challenge in old mice.He Li Lei Zhu Rong Wang Lihui Xie Jie Ren Shuai Ma Weiqi Zhang Xiuxing Liu Zhaohao Huang Binyao Chen Zhaohuai Li Huyi Feng Guang-Hui Liu Si Wang Jing Qu Wenru Su 2022Protein & Cell2022,13,6:1

6Preparation and photocatalytic performance study of dual Z-scheme Bi_(2)Zr_(2)O_(7)/g-C_(3)N4/Ag_(3)PO_(4) for removal of antibiotics by visible-light显示文摘At present,the high re-combination rate of photogenerated carriers and the low redox capability of the photocatalyst are two factors that severely limit the improvement of photocatalytic performance.Herein,a dual Z-scheme photocatalyst bismuthzirconate/graphitic carbon nitride/silver phosphate (Bi_(2)Zr_(2)O_(7)/g-C_(3)N4/Ag_(3)PO_(4)(BCA)) was synthesized using a co-precipitation method,and a dual Z-scheme heterojunction photocatalytic system was established to decrease the high re-combination rate of photogenerated carriers and consequently improve the photocatalytic performance.The re-combination of electron-hole pairs(e-and h+) in the valence band (VB) of g-C_(3)N4increases the redox potential of e-and h+,leading to significant improvements in the redox capability of the photocatalyst and the efficiency of e--h+separation.As a photosensitizer,Ag_(3)PO_(4)can enhance the visible light absorption capacity of the photocatalyst.The prepared photocatalyst showed strong stability,which was attributed to the efficient suppression of photo-corrosion of Ag_(3)PO_(4)by transferring the e-to the VB of g-C_(3)N4.Tetracycline was degraded efficiently by BCA-10%(the BCA with 10 wt.%of AgPO_(4)) under visible light,and the degradation efficiency was up to 86.2%.This study experimentally suggested that the BCA photocatalyst has broad application prospects in removing antibiotic pollution.Zhengjun Qu Zhenyang Jing Xiaoming Chen Zexiang Wang Hongfei Ren Lihui Huang 2023Journal of Environmental Sciences2023,,3:0
7Effect of orphanin FQ and morphine on sodium channel current in somatosensory area of rat cerebral cortex显示文摘BACKGROUND:Some experiments have demonstrated that injecting orphanin FQ (OFQ) into lateral ventricle, which can obviously decrease the pain threshold. It is indicated that OFQ is an anti-opiate substance. However, whether OFQ has effects on sensory neuron ion channel in cerebral cortex needs to be further studied. OBJECTIVE:To investigate the effects of OFQ, morphine or their combination on sodium channel current of somatosensory neurons in rat cerebral cortex. DESIGN:Repeated measurement trial. SETTING:Department of Physiology, Harbin Medical University. MATERIALS:Fifty healthy Wistar rats, aged 12-16 days, of either gender, were provided by the Experimental Animal Center, Second Hospital Affiliated to Harbin Medical University. OFQ was purchased from Sigma-Aldrich Company, and morphine was provided by the Shenyang First Pharmaceutical Factory. PC2C patch clamp amplifier and LabmasterTL1were purchased from Yibo Life Science Instrument Co.,Ltd. of Huazhong University of Science and Techgnology. METHODS:This experiment was carried out in the Department of Physiology (provincial laboratory), Harbin Medical University between January 2005 and May 2006. Cortical neurons were acutely isolated from rats, and prepared into cell suspension following culture. ①Sodium channel current of somatosensory neurons in rat cerebral cortex was recorded before and after administration by whole-cell Patch clamp technique after 50 nmol/L OFQ being added to extracellular fluid. ②The amplitude of sodium channel current of somatosensory neurons in rat cerebral cortex was recorded before and after administration by the same method after 20 μmol/L morphine being added to extracellular fluid, and then the change of sodium channel current was recorded after 50 nmol/L OFQ being added. MAIN OUTCOME MEASURES:The amplitude of sodium channel current of somatosensory neurons in rat cerebral cortex following the administration of OFQ, morphine separately or their combination.. RESULTS:①The amplitude of sodium channel current of somatosensory neurons in rat cerebral cortex was significantly lower after administration of 50 nmol/L OFQ than before at the clampe of the voltage of -30 mV (P < 0.05). ②The amplitude of sodium channel current of somatosensory neurons in rat cerebral cortex was significantly lower after administration of 20 μmol/L morphine than before at the clampe of the voltage of -30 mV (P < 0.05). The sodium channel current recovered to -(2 345.24±174.18)pA after 50 nmol/L OFQ was administrated. There were significant differences in the amplitude of Na+ channel current between two interventions (P < 0.05). CONCLUSION:Morphine and OFQ can respectively reduce the amplitude of sodium channel current of somatosensory neurons in rat cerebral cortex, and OFQ can reverse the effect of morphine partly. 8Acetylcholine modulates transient outward potassium channel in acutely isolated cerebral cortical neurons of rats显示文摘BACKGROUND:The neuronal transient outward potassium channel has been shown to be highly associated with acetylcholine.However,the influence of acetylcholine on the transient outward potassium current in cerebral cortical neurons remains poorly understood. OBJECTIVE:To investigate acetylcholine modulation on transient outward potassium current in rat parietal cortical neurons using the whole-cell patch-clamp technique. DESIGN,TIME AND SETTING:A neuroelectrophysiology study was performed at the Department of Physiology,Harbin Medical University between January 2005 and January 2006. MATERIALS:Wistar rats were provided by the Animal Research Center,the Second Hospital of Harbin Medical University;PC-IIC patch-clamp amplifier and IBBClamp data collection analysis system were provided by Huazhong University for Science and Technology,Wuhan,China;PP-83 microelectrode puller was purchased from Narrishage,Japan. METHODS:The parietal somatosensory cortical neurons were acutely dissociated,and the modulation of acetylcholine(0.1,1,10,100μmol/L) on transient outward potassium channel was recorded using the whole-cell patch-clamp technique. MAIN OUTCOME MEASURES:Influence of acetylcholine on transient outward potassium current, potassium channel activation,and inactivation. RESULTS:The inhibitory effect of acetylcholine on transient outward potassium current was doseand voltage-dependent(P<0.01).Acetylcholine was found to significantly affect the activation process of transient outward potassium current,i.e.,the activation curve of transient outward potassium current was left-shifted,while the inactivation curve was shifted to hyperpolarization. Acetylcholine significantly prolonged the time constant of recovery from inactivation of transient outward potassium current(P<0.01). 9Multi-proxy reconstructions of hydrological changes from continental shelf sediments in the northern South China Sea during the interval 9200–6200 cal a BP显示文摘Past hydroclimatic conditions in southern China are poorly constrained owing to the lack of high-resolution marine-sediment records. In this study, we present high-resolution geochemical and grain-size records of marine sediments from the coastal shelf of the northern South China Sea to investigate regional hydrological variations.Results suggest a warm and humid climate during the interval 9 200–7 600 cal a BP, followed by a cold and dry climate from 7 600 cal a BP to 6 500 cal a BP, and progressive humidification during the period 6 500–6 200 cal a BP.A prominent hydrological anomaly occurred during 7 600–6 500 cal a BP. 10Hypothalamic-Pituitary-Gonadal(HPG)Axis and Transcriptional Regulatory Elements Regulate piwil2 Gene Expression During Gametogenesis and Gonadal Development in Japanese Flounder(Paralichthys olivaceus)显示文摘The P-element induced wimpy testis(Piwi)proteins,which are associated with PIWI-interacting RNAs(piRNAs),play important roles in meiosis,germ cell division,and germline maintenance.In this study,we identified and characterized the Paralichthys olivaceus piwil2 gene,a constituent factor of the piRNA pathways involved in the biogenesis of reproductive development.The biological analysis indicated that piwil2,which contains PAZ and PIWI domains,was highly conserved between teleosts and tetrapods.The piwil2 distribution profile in different tissues confirmed a sexually dimorphic expression pattern,with a higher expression level in testis.In situ hybridization demonstrated that piwil2 was expressed in the oogonia and oocytes of the ovaries as well as in the Sertoli cells and spermatocytes of the testes.Gene piwil2 showed a maternally inherited expression pattern during embryonic development,and was highly expressed during the early embryonic development.Different luciferase reporters were constructed to determine the transcriptional regulatory mechanisms of piwil2.The piwil2 core promoter region was located at−360 bp to−60 bp.Furthermore,some representative sex hormones,including human chorionic gonadotropin,17α-methyltestosterone,and estradiol-17βhad distinct regulatory effects on piwil2.In a summery,these results indicate that piwil2,regulated by sex hormones and transcriptional elements,has vital functions in the reproductive cycle and gonadal development.NI Feifei YU Haiyang QU Jiangbo MENG Lihui LIU Xiumei YAN Weijie CHANG Jing ZHANG Quanqi WANG Xubo YU Haiyang 2020Journal of Ocean University of China2020,19,6:0
