| 1 | Theropod assemblages and a new ichnotaxon Gigandipus chiappei ichnosp.nov.from the Jiaguan Formation,Lower Cretaceous of Guizhou Province,China显示文摘A newly discovered Jiaguan Formation(Lower Cretaceous) tracksite from the Linjiang region of Guizhou Province, China, reveals the first example of a Cretaceous track morphotype attributable to the non-avian theropod ichnogenus Gigandipus, here named Gigandipus chiappei ichnosp nov. The theropod dominated locality also reveals the second report of the avian theropod ichnogenus Wupus, one of the largest avian traces currently known from the Lower Cretaceous. The Linjiang site provides evidence to support previous interpretations of a distinctive Lower Cretaceous theropod-dominated ichnofauna that was widespread in China and East Asia and highlights the similarity between Lower Cretaceous theropod ichnotaxa in East Asia and those found in the Lower Jurassic both in East Asia and elsewhere. | 2 | Intra-strain Elicitation and Suppression of Plant Immunity by Ralstonia solanacearum Type-III Effectors in Nicotiana benthamiana显示文摘Effector proteins delivered inside plant cells are powerful weapons for bacterial pathogens,but this exposes the pathogen to potential recognition by the plant immune system.Therefore,the effector repertoire of a given pathogen must be balanced for a successful infection.Ralstonia solanacearum is an aggressive pathogen with a large repertoire of secreted effectors.One of these effectors,RipE1,is conserved in most R.solanacearum strains sequenced to date.In this work,we found that RipE1 triggers immunity in N.benthamiana,which requires the immune regulator SGT1,but not EDS1 or NRCs.Interestingly,RipE1-triggered immunity induces the accumulation of salicylic acid(SA)and the overexpression of several genes encoding phenylalanine-ammonia lyases(PALs),suggesting that the unconventional PALmediated pathway is responsible for the observed SA biosynthesis.Surprisingly,RipE1 recognition also induces the expression of jasmonic acid(JA)-responsive genes and JA biosynthesis,suggesting that both SA and JA may act cooperatively in response to RipE1.We further found that RipE1 expression leads to the accumulation of glutathione in plant cells,which precedes the activation of immune responses.R.solanacearum secretes another effector,RipAY,which is known to inhibit immune responses by degrading cellular glutathione.Accordingly,RipAY inhibits RipE1-triggered immune responses.This work shows a strategy employed by R.solanacearum to counteract the perception of its effector proteins by plant immune system. | Yuying Sang Wenjia Yu Haiyan Zhuang Yali Wei Lida Derevnina Gang Yu Jiamin Luo Alberto P.Macho | 2020 | Plant Communications2020,1,4: | 2 |
| 3 | Enhanced chemoselectivity of a plant cytochrome P450 through protein engineering of surface and catalytic residues显示文摘Cytochrome P450s(P450s)are the most versatile catalysts utilized by plants to produce structurally and functionally diverse metabolites.Given the high degree of gene redundancy and challenge to functionally characterize plant P450s,protein engineering is used as a complementarystrategy to study the mechanisms of P450-mediated reactions,or to alter their functions.We previously proposed an approach of engineering plant P450s based on combining high accuracy homology models generated by Rosetta combined with data-driven design using evoluti onary information of these enzymes.With this strategy,we repurposed a multi-functional P450(CYP87D20)into a monooxygenase after red esigning its active site.Since most plant P450s are membrane-anchored proteins that are adapted to the micro-environments of plant cells,expressing them in heterologous hosts usually results in problems of expression or activity.Here,we applied compu-tational design to tackle these issues by simultaneous optimization of the protein surface and active site.After screening 17 variants,effective su bstitutions of surface residues were observed to improve both expression and activity of CYP87D20.In addition,the identified substitutions were additive and by com-bining them a highly eficient C11 hydroxylase of cucurbitadienol was created to participate in the mogrol biosynthesis.This study shows the importance of considering the interplay between surface and active site residues for P450 engineering.Our integrated strategy also opens an avenue to create more tai loring enzymes with desired functions for the metabolic engineering of high-valued compounds like mogrol,the precursor of natural sweetener mogrosides. | Xiaopeng Zhang Wei Luo Yinying Yao Xuming Luo Chao Han Yang Zhong Bo Zhang Dawei Li Lida Han Sanwen Huang Per Greisen Yi Shang | 2021 | aBIOTECH2021,2,3: | 1 |
| 4 | Precise tuning of low-crystalline Sb@Sb_(2)O_(3) confined in 3D porous carbon network for fast and stable potassium ion storage显示文摘Metal antimony(Sb)is a promising anode material of potassium-ion batteries(PIBs)for its high theoretical capacity but limited by its inferior cycle stability due to the serious volume expansion during cycling.Herein,we design and construct a kind of low-crystalline Sb nanoparticles coated with amorphous Sb2O3 and dispersed into three-dimensional porous carbon via a strategy involving NaCl template-assisted insitu pyrolysis and subsequent low-temperature heat-treated in air.Significantly,the crystallinity and ratio of Sb/Sb_(2)O_(3) have been precisely tuned and controlled,and the optimized sample of HTSb@Sb_(2)O_(3)@C-4 displays a high reversible specific capacity of 543.9 m Ah g^(-1) at 0.1 A g^(-1),superior rate capability and excellent cycle stability(~273 m Ah g^(-1) at 2 A g^(-1) after 2000 cycles)as an anode of PIBs.The outstanding potassium-ion storage performance can be ascribed to the appropriate crystallinity and the multiplebuffer-matrix structure comprising an interconnected porous conductive carbon to relieve the volume changes and suppress the aggregation of Sb,a Sb nanoparticle core to shorten the ion transport pathways and decrease the mechanical stress,and a low-crystalline Sb_(2)O_(3) as the shell to consolidate the interface between Sb and carbon as well as facilitate the rapid electron transport.The dynamic analysis shows that the composite is mainly controlled by pseudocapacitance mechanism.This work provides a novel thought to design high-performance composite electrode in energy storage devices. | Qun Ma Lida Song Yuan Wan Kangze Dong Zhiyuan Wang Dan Wang Hongyu Sun Shaohua Luo Yanguo Liu | 2021 | Journal of Materials Science & Technology2021,,35: | 1 |
| 5 | Study on Quality Standard of Lujing Yiqi Shengxue Pills显示文摘[Objectives]This study was conducted to establish the quality standard for Lujing Yiqi Shengxue Pills to provide a basis for its quality control.[Methods]According to the quality requirements in the 2020 edition of Chinese Pharmacopoeia,the quality of Lujing Yiqi Shengxue Pills was evaluated from the aspects of properties,identification,inspection and content determination.Radix Astragali,Radix Angelicae Sinensis,Macrocephalae Rhizoma stir-fried with bran and Radix Paeoniae Alba in the preparation were identified and studied by thin layer chromatography(TLC).A high performance liquid chromatography(HPLC)method was established to determine the content of paeoniflorin in the preparation.[Results]The thin layer spots of Radix Astragali,Radix Angelicae Sinensis,Macrocephalae Rhizoma and Radix Paeoniae Alba were clear and round,and the negative preparations had no interference.The properties,moisture,weight difference,loading difference,dissolution time limit and microbial limit all met the requirements.The regression formula for the quantitative analysis of paeoniflorin in the concentration range of 4.86-194.40μg/ml was:Y=12.0241X+0.8797,R^(2)=0.9999;and the average recovery was 98.9%,and the RSD was 1.7%.[Conclusions]The method has good repeatability,and is stable,feasible,simple and convenient in operation and suitable for quality control of Lujing Yiqi Shengxue Pills. | Lida CHEN Xianmei XU Yuchuan LI Yuanfeng YANG Yuling LUO | 2022 | Agricultural Biotechnology2022,11,3: | 0 |
| 6 | A study of the lipid-mediated dimerization of the RAGE TM+JM domains by molecular dynamic simulations显示文摘Receptor for Advanced Glycation End-products(RAGE) binds to a number of ligand families to display important roles in hyperglycemia, senescence, inflammation, neurodegeneration and cancer. It is reported that RAGE regulates the related biological processes via homo-dimerization by the transmembrane(TM) domain, and evidence further shows that the intracellular domain of RAGE has an influence on the dimerization activity of RAGE. In this study, we explored the underlying interaction mechanism of RAGE TM domains by multiscale coarse-grained(CG) dynamic simulations. Two switching packing modes of the TM dimeric conformations were observed. Through a series of site-directed mutations, we further emphasized the key roles of the A^(342)xxxG^(346)xxG^(349)xxxT^(353)xxL^(356)xxxV^(360) motif in the left-handed configuration and the L^(345)xxxG^(349)xxG^(352)xxxL^(356) motif in the right-handed configuration. In addition, we revealed that the juxtamembrane(JM) domain within JM-A375 can determine the RAGE TM dimeric structure. | 7 | Study on the Extraction Techniques of Impatiens balsamina(Tougucao)显示文摘[Objectives]This study was conducted to investigate the extraction method of Impatiens balsamina L.(Tougucao),and to optimize the extraction process conditions and parameters.[Methods]Different ethanol concentrations,ethanol amounts and extraction time were investigated by an orthogonal test L_(9)(3^(4))and high performance liquid chromatography(HPLC),and the extraction process parameters were evaluated by the yield of extract and the content of quercetin.[Results]Under the three optimal extraction conditions of impregnation,heating reflux and ultrasound,the extract yield and quercetin contents of I.balsamina were 8.27%,9.50%,7.09%and 1.60,3.05,3.03μg/g,respectively.Heating reflux extraction and ultrasonic extraction showed higher extract yields and quercetin contents of I.balsamina,while the values were lower under the impregnation extraction conditions.The primary and secondary factors were:ethanol amount(C)>ethanol concentration(A)>extraction time(B).The optimal extraction process of I.balsamina was as follows:adding 10 times of 60%ethanol and ultrasonically extracting for 30 min.[Conclusions]The method is simple,convenient,accurate and reliable,and provides a scientific basis for the research on the extraction process of I.balsamina and its preparations. | Lida CHEN Yuanzhe ZHANG Yuling LUO Xianmei XU Hai ZHAO Yan WU | 2022 | Agricultural Biotechnology2022,11,4: | 0 |
| 8 | Preparation and Characterization of Glass-Ceramic Basalt Fiber显示文摘Basalt fiber(BF)is widely applied in the construction industry to improve the mechanical properties of construction materials.Recent studies show that BF has the potential to further enhance its performance via a crystallization approach.In this work,the glass-ceramic basalt fibers(GCBFs)were prepared through nucleation and crystallization treatments according to the crystallization kinetics calculations.Results from XRD and SEM show that GCBFs have main crystalline phases of Diopside and Augite reach crystallinity of around 46%±10%.In particular,the GCBFs sample with the largest mean crystal size maintains the lowest tensile strength of~197 MPa(compared with the pristine BFs of~737 MPa).Moreover,the weight loss and ion dissolution of GCBFs were explored in seawater environments and it was investigated that,GCBFs have better anti-seawater corrosion than the pristine BFs and have the potential to apply in the marine industry. | Lida Luo Tan Yan Jin Liu Linfeng Ding Qingwei Wang Wan Jiang | 2022 | Journal of Harbin Institute of Technology(New Series)2022,29,6: | 0 |
