2Risk Management for International Portfolios with Basket Options:A Multi-Stage Stochastic Programming Approach显示文摘The authors consider the problem of active international portfolio management with basket options to achieve optimal asset allocation and combined market risk and currency risk management via multi-stage stochastic programming(MSSP). The authors note particularly the novel consideration and signi?cant bene?t of basket options in the context of portfolio optimization and risk management.Extensive empirical tests strongly demonstrate that basket options consistently have more clearly improvement on portfolio performances than a portfolio of vanilla options written on the same underlying assets. 7Machine Learning‑Enhanced Flexible Mechanical Sensing显示文摘To realize a hyperconnected smart society with high productivity,advances in flexible sensing technology are highly needed.Nowadays,flexible sensing technology has witnessed improvements in both the hardware performances of sensor devices and the data processing capabilities of the device’s software.Significant research efforts have been devoted to improving materials,sensing mechanism,and configurations of flexible sensing systems in a quest to fulfill the requirements of future technology.Meanwhile,advanced data analysis methods are being developed to extract useful information from increasingly complicated data collected by a single sensor or network of sensors.Machine learning(ML)as an important branch of artificial intelligence can efficiently handle such complex data,which can be multi-dimensional and multi-faceted,thus providing a powerful tool for easy interpretation of sensing data.In this review,the fundamental working mechanisms and common types of flexible mechanical sensors are firstly presented.Then how ML-assisted data interpretation improves the applications of flexible mechanical sensors and other closely-related sensors in various areas is elaborated,which includes health monitoring,human-machine interfaces,object/surface recognition,pressure prediction,and human posture/motion identification.Finally,the advantages,challenges,and future perspectives associated with the fusion of flexible mechanical sensing technology and ML algorithms are discussed.These will give significant insights to enable the advancement of next-generation artificial flexible mechanical sensing.Yuejiao Wang Mukhtar Lawan Adam Yunlong Zhao Weihao Zheng Libo Gao Zongyou Yin Haitao Zhao 2023Nano-Micro Letters2023,15,4:1

9A moment-based criterion for determining the number of components in a normal mixture model显示文摘Determining the number of components is a crucial issue in a mixture model. A moment-based criterion is considered to estimate the number of components arising from a normal mixture model. This criterion is derived from an omnibus statistic involving the skewness and kurtosis of each component. The proposed criterion additionally provides a measurement for the model fit in an absolute sense. 12Two types of coumarins-specific enzymes complete the last missing steps in pyran- and furanocoumarins biosynthesis显示文摘Pyran-and furanocoumarins are key representatives of tetrahydropyrans and tetrahydrofurans,respectively,exhibiting diverse physiological and medical bioactivities.However,the biosynthetic mechanisms for their core structures remain poorly understood.Here we combined multiomics analyses of biosynthetic enzymes in Peucedanum praeruptorum and in vitro functional verification and identified two types of key enzymes critical for pyran and furan ring biosynthesis in plants.These included three distinct P.praeruptorum prenyltransferases(PpPT1e3)responsible for the prenylation of the simple coumarin skeleton 7 into linear or angular precursors,and two novel CYP450 cyclases(PpDC and PpOC)crucial for the cyclization of the linear/angular precursors into either tetrahydropyran or tetrahydrofuran scaffolds.Biochemical analyses of cyclases indicated that acid/base-assisted epoxide ring opening contributed to the enzyme-catalyzed tetrahydropyran and tetrahydrofuran ring refactoring.The possible acid/base-assisted catalytic mechanisms of the identified cyclases were theoretically investigated and assessed using site-specific mutagenesis.We identified two possible acidic amino acids Glu303 in PpDC and Asp301 in PpOC as vital in the catalytic process.This study provides new enzymatic tools in the epoxide formation/epoxide-opening mediated cascade reaction and exemplifies how plants become chemically diverse in terms of enzyme function and catalytic process.Yucheng Zhao Yuedong He Liangliang Han Libo Zhang Yuanzheng Xia Fucheng Yin Xiaobing Wang Deqing Zhao Sheng Xu Fei Qiao Yibei Xiao Lingyi Kong 2024Acta Pharmaceutica Sinica B2024,14,2:0

14Spectroscopic studies of the interaction between phosphorus heterocycles and cytochrome P450显示文摘P450 fatty acid decarboxylase Ole T from Staphylococcus aureus(Ole TSA)is a novel cytochrome P450 enzyme that catalyzes the oxidative decarboxylation of fatty acids to yield primarily terminal alkenes and CO_(2) or minor a-andβ-hydroxylated fatty acids as side-products.In this work,the interactions between a series of cycloalkyl phosphorus heterocycles(CPHs)and Ole TSAwere investigated in detail by fluorescence titration experiment,ultraviolet-visible(UV-vis)and31 P NMR spectroscopies.Fluorescence titration experiment results clearly showed that a dynamic quenching occurred when CPH-6,a representative CPHs,interacted with Ole TSAwith a binding constant value of 15.2×104 M-1 at 293 K.The thermodynamic parameters(ΔH,ΔS andΔG)showed that the hydrogen bond and van der Waals force played major roles in the interaction between Ole TSAand CPHs.The UV-vis and31 P NMR studies indicated the penetration of CPH-6 into the interior environment of Ole TSA,which greatly affects the enzymatic activity of Ole TSA.Therefore,our study revealed an effective way to use phosphorus heterocyclic compounds to modulate the activity of cytochrome P450 enzymes.Dumei Ma Libo Zhang Yingwu Yin Yuxing Gao Qian Wang 2021Journal of Pharmaceutical Analysis2021,11,6:0
15Protein phosphatase 6 (Pp6) is crucial for regulatory T cell function and stability in autoimmunity显示文摘Regulatory T(T_(reg))cells constitute a dynamic population that is critical in autoimmunity.T_(reg) cell therapies for autoimmune diseases are mainly focused on enhancing their suppressive activities.However,recent studies demonstrated that certain inflammatory conditions induce T_(reg) cell instability with diminished FoxP3 expression and convert them into pathogenic effector cells.Therefore,the identification of novel targets crucial to both T_(reg) cell function and plasticity is of vital importance to the development of therapeutic approaches in autoimmunity.In this study,we found that conditional Pp6 knockout(cKO)in T_(reg) cells led to spontaneous autoinflammation,immune cell activation,and diminished levels of FoxP3 in CD4^(+)T cells in mice.Loss of Pp6 in T_(reg) cells exacerbated two classical mouse models of T_(reg)-related autoinflammation.Mechanistically,Pp6 deficiency increased CpG motif methylation of the FoxP3 locus by dephosphorylating Dnmt1 and enhancing Akt phosphorylation at Ser473/Thr308,leading to impaired FoxP3 expression in T_(reg) cells.In summary,our study proposes Pp6 as a critical positive regulator of FoxP3 that acts by decreasing DNA methylation of the FoxP3 gene enhancer and inhibiting Akt signaling,thus maintaining T_(reg) cell stability and preventing autoimmune diseases.Wei Cai Junxun Zhang Hong Zhou Xiangxiao Li Fangzhou Lou Yang Sun Zhenyao Xu Jing Bai Qianqian Yin Zhikai Wang Libo Sun Xiaojie Cai Sibei Tang Yue Wu Li Fan Hong Wang Honglin Wang Qun Li 2022Genes & Diseases2022,9,2:0
16Structure-based design,synthesis of novel probes for cytochrome P450 OleT显示文摘Cytochrome P450 OleT_(SA),a new cytochrome P450 enzyme from Staphylococcus aureus,catalyzes the oxidative decarboxylation and hydroxylation of fatty acids to generate terminal alkenes and fatty alcohols.The mechanism of this bifurcative chemistry remains largely unknown.Herein,a class of derivatized fatty acids were synthesized as probes to investigate the effects of substrate structure on the product type of P450 OleT_(SA).The results demonstrate that the fine-tuned structure of substrates,even in a remote distance from the carboxyl group,significantly regulates OleT catalyzed decarboxylation/hydroxylation reactions.Molecular docking analysis indicated the potential interactions between the carboxylate groups of different probes and the enzyme active center which was attributed to the bifurcative chemistry.Dumei Ma Libo Zhang Yingwu Yin Qian Wang 2021Chinese Chemical Letters2021,32,4:0
