35-Hydroxymethylome in Circulating Cell-free DNA as A Potential Biomarker for Non-small-cell Lung Cancer显示文摘Non-small-cell lung cancer(NSCLC), the most common type of lung cancer accounting for 85% of the cases, is often diagnosed at advanced stages owing to the lack of efficient early diagnostic tools. 5-Hydroxymethylcytosine(5 hmC) signatures in circulating cell-free DNA(cfDNA) that carries the cancer-specific epigenetic patterns may represent the valuable biomarkers for discriminating tumor and healthy individuals, and thus could be potentially useful for NSCLC diagnosis. Here,we employed a sensitive and reliable method to map genome-wide 5 hmC in the cfDNA of Chinese NSCLC patients and detected a significant 5 hmC gain in both the gene bodies and promoter regions in the blood samples from tumor patients compared with healthy controls. Specifically, we identified six potential biomarkers from 66 patients and 67 healthy controls(mean decrease accuracy>3.2, P < 3.68E-19) using machine-learning-based tumor classifiers with high accuracy. 5CMHX008,a PPAR γ partial agonist,enhances insulin sensitivity with minor influences on bone loss显示文摘Traditional thiazolidinediones(TZDs),such as rosiglitazone,are peroxisome proliferator-activated receptor g(PPARg)potent agonists that can be used to treat type 2 diabetes but carry unwanted effects,including increased risk for fracture.The present work aimed to compare the insulin-sensitizing efficacies and bone-loss side effects of CMHX008,a novel TZDs-like PPARg partial agonist,with those of rosiglitazone.A TR-FRET PPARg competitive binding assay was used to compare the binding affinity between CMHX008 and rosiglitazone.Mice were administered vehicle,CMHX008 or rosiglitazone for 16 weeks.Mesenchymal stem cells(MSCs)were used to examine differences in differentiation into osteoblasts after compounds treatment.TR-FRET showed lower affinity to PPARg by CMHX008 compared with rosiglitazone.Mice treated with CMHX008 showed insulin sensitization similar to that of mice treated with rosiglitazone,which was related to the significant inhibition of PPARg Ser273 phosphorylation and improved insulin sensitivity by facilitating the phosphorylation of insulin receptor and Akt in adipose tissues.Micro-CT and histomorphometric analyses demonstrated that the degree of trabecular bone loss after treatment with CMHX008 was weaker than that observed with rosiglitazone,as evidenced by consistent changes in BV/TV,Tb.N,Tb.Th,Tb.Sp,and the mineral apposition rate.MSCs treated with CMHX008 showed higher ALP activity and mRNA levels of bone formation markers than did cells treated with rosiglitazone in the osteoblast differentiation test.Thus,CMHX008 showed insulin-sensitizing effects similar to those of rosiglitazone with a lower risk of bone loss,suggesting that PPARg sparing eliminates the skeletal side effects of TZDs while maintaining their insulin-sensitizing properties.Yi Hou Xuemei Cao Xiangnan Hu Xinyu Li Xiaoqin Shi Hongying Wang Chuan Peng Jiayu Li Jibin Li Qifu Li Chaodong Wu Xiaoqiu Xiao 2018Genes & Diseases2018,5,3:6

13In-situ observation and modeling approach to evolution of pore-fracture structure in coal显示文摘The characterisation of the pore-fracture structure(PFS)and its evolution in coal during mining are essential for preventing gas outbursts and improving gas extraction efficiency.In this study,the evolution of the PFS in coal samples under the condition of mining stress was directly captured in situ by combination of the mechanical testing system with high-precision visualisation nuclear magnetic resonance equipment.A fractional derivative model was established to describe the relationship between stress and porosity based on experimental results of the PFS under different stress states.The results showed that with an increase in the deviatoric stress,the adsorption pore content increases rapidly initially and then increases slowly or remains unchanged;the seepage pore and fracture(SPF)content decreases initially and then increases.The SPF compressibility coefficient decreases with an increase in the deviatoric stress.The fractional derivative model can accurately describe the stress sensitivity of the SPFs at the pre-peak stage,thus providing a new approach for accurately characterising the seepage characteristics of coal reservoirs.Hongwei Zhou Zelin Liu Jiawei Zhao Bocen Chen Xiangnan Li Jiangcheng Zhong 2023International Journal of Mining Science and Technology2023,33,3:1

16Hydrogel-elastomer-based stretchable strain sensor fabricated by a simple projection lithography method显示文摘Stretchable strain sensor detects a wide range of strain variation and is therefore a key component in various applications.Unlike traditional ones made of elastomers doped with conductive components or fabricated with liquid conductors,ionically conductive hydrogel-based strain sensors remain conductive under large deformations and are biocompatible.However,dehydration is a challenging issue for the latter.Researchers have developed hydrogel-elastomer-based strain sensors where an elastomer matrix encapsulates a hydrogel circuit to prevent its dehydration.However,the reported multistep approaches are generally time-consuming.Our group recently reported a multimaterial 3D printing approach that enables fast fabrication of such sensors,yet requires a self-built digital-light-processing-based multimaterial 3D printer.Here,we report a simple projection lithography method to fabricate hydrogel-elastomer-based stretchable strain sensors within 5 minutes.This method only requires a UV projector/lamp with photomasks;the chemicals are commercially available;the protocols for preparing the polymer precursors are friendly to users without chemistry background.Moreover,the manufacturing flexibility allows users to readily pattern the sensor circuit and attach the sensor to a 3D printed soft pneumatic actuator to enable strain sensing on the latter.The proposed approach paves a simple and versatile way to fabricate hydrogel-elastomer-based stretchable strain sensors and flexible electronic devices.Zhenqing Li Xiangnan He Jianxiang Cheng Honggeng Li Yuan-Fang Zhang Xiaojuan Shi Kai Yu Hui Ying Yang Qi Ge 2021International Journal of Smart and Nano Materials2021,12,3:1

18Computer Aided Design Technology for Convex Faceted Gem Cuts Based on the Half-Edge Data Structure显示文摘Aiming to increase the efficiency of gem design and manufacturing, a new method in computer-aided-design (CAD) of convex faceted gem cuts (CFGC) based on Half-edge data structure (HDS), including the algorithms for the implementation is presented in this work. By using object-oriented methods, geometrical elements of CFGC are classified and responding geometrical feature classes are established. Each class is implemented and embedded based on the gem process. Matrix arithmetic and analytical geometry are used to derive the affine transformation and the cutting algorithm. Based on the demand for a diversity of gem cuts, CAD functions both for free-style faceted cuts and parametric designs of typical cuts and visualization and human-computer interactions of the CAD system including two-dimensional and three-dimensional interactions have been realized which enhances the flexibility and universality of the CAD system. 19Molecular characterization of secaloindoline genes in introduced CIMMYT primary hexaploid triticale显示文摘In order to widen gene germplasm for kernel hardness in triticale, 60 synthetic hexaploid triticales were tested by the single kernel characterization system(SKCS) and secaloindoline alleles were identified by sequencing. Phenot^ing showed that frequencies of soft, mixed and hard genotypes were 43.3%, 48.3%, and 8.4%, respectively. Genotyping identified three known secaloindoline-a alleles and four known secaloindoline-b alleles. Three new Sina-Rl alleles were designated Sina-Rld, Sina-Rle and Sina-Rlf. Compared to Sina-Dlc, Sina-Rld showed four point mutations causing changes in four amino acids, Sina-Rle had one point mutation causing an alanine to glycine substitution, and Sina-Rlf possessed five point mutations but produced the same amino add sequence as Sina-Rld. Two new Sinb-Rl alleles were discovered and designated Sinb-Rle and Sinb-Rl/. Compared to Sinb-Rla, Sinb-Rle possessed a triplet-code insertion and four point mutations causing a cysteine insertion and two amino acid substitutions, and Sinb-Rl/possessed three point mutations causing a cysteine insertion and a change of arginine to glycine.Association of hardness index with secaloindoline alleles indicated t±iat SKCS of the Sina-Rld genotype was significantly lower than that of Sina-Rle, and Sinb-Rle was significantly lower than that of the Sinb-Rld genotype. A total of eight allelic combinations of secaloindoline genes were identified; Sma-Rld/Sinb-Rle and Sina-Rle/Sinb-Rld were relatively prevalent in the triticales surveyed. 20ABA-mediated regulation of leaf and root hydraulic conductance in tomato grown at elevated CO2 is associated with altered gene expression of aquaporins显示文摘Elevated CO_(2)concentration in the air(e[CO_(2)])decreases stomatal density(SD)and stomatal conductance(gs)where abscisic acid(ABA)may play a role,yet the underlying mechanism remains largely elusive.We investigated the effects of e[CO_(2)](800 ppm)on leaf gas exchange and water relations of two tomato(Solanum lycopersicum)genotypes,Ailsa Craig(WT)and its ABA-deficient mutant(flacca).Compared to plants grown at ambient CO_(2)(400 ppm),e[CO_(2)]stimulated photosynthetic rate in both genotypes,while depressed the gs only in WT.SD showed a similar response to e[CO_(2)]as gs,although the change was not significant.e[CO_(2)]increased leaf and xylem ABA concentrations and xylem sap pH,where the increases were larger in WT than in flacca.Although leaf water potential was unaffected by CO_(2)growth environment,e[CO_(2)]lowered osmotic potential,hence tended to increase turgor pressure particularly for WT.e[CO_(2)]reduced hydraulic conductance of leaf and root in WT but not in flacca,which was associated with downregulation of gene expression of aquaporins.It is concluded that ABA-mediated regulation of gs,SD,and gene expression of aquaporins coordinates the whole-plant hydraulics of tomato grown at different CO_(2)environments.Liang Fang Lamis Osama Anwar Abdelhakim Josefine Nymark Hegelund Shenglan Li Jie Liu Xiaoying Peng Xiangnan Li Zhenhua Wei Fulai Liu 2019Horticulture Research2019,6,1:1
