1Cyclin A2:At the crossroads of cell cycle and cell invasion显示文摘Cyclin A2 is an essential regulator of the cell division cycle through the activation of kinases that participate to the regulation of S phase as well as the mitotic entry. However,whereas its degradation by the proteasome in mid mitosis was thought to be essential for mitosis to proceed,recent observations show that a small fraction of cyclin A2 persists beyond metaphase and is degraded by autophagy. Its implication in the control of cytoskeletal dynamics and cell movement has unveiled its role in the modulation of Rho A activity. Since this GTPase is involved in both cell rounding early in mitosis and later,in the formation of the cleavage furrow,this suggests that cyclin A2 is a novel actor in cytokinesis. 3N-acetylcysteine and glycyrrhizin combination:Benefit outcome in a murine model of acetaminophen-induced liver failure显示文摘BACKGROUND Acetaminophen overdose is the most frequent cause of drug-induced liver failure in developed countries.Substantial progress has been made in understanding the mechanism of hepatocellular injury,but N-acetylcysteine remains the only effective treatment despite its short therapeutic window.Thus,other hepatoprotective drugs are needed for the delayed treatment of acetaminopheninduced hepatotoxicity.Our interest focused on glycyrrhizin for its role as an inhibitor of high mobility group box 1(HMGB1)protein,a member of the family of damage-associated molecular pattern,known to play an important pathological role in various diseases.AIM To investigate the efficacy of the N-acetylcysteine/glycyrrhizin combination compared to N-acetylcysteine alone in the prevention of liver toxicity.METHODS Eight-week-old C57BL/6J wild-type female mice were used for all our experiments.Mice fasted for 15 h were treated with acetaminophen(500 mg/kg)or vehicle(phosphate-buffered saline)by intraperitoneal injection and separated into the following groups:Glycyrrhizin(200 mg/kg);N-acetylcysteine(150 mg/kg);and N-acetylcysteine/glycyrrhizin.In all groups,mice were sacrificed 12 h following acetaminophen administration.The assessment of hepatotoxicity was performed by measuring plasma levels of alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase.Hepatotoxicity was also evaluated by histological examination of hematoxylin and eosin-stained tissues sections.Survival rates were compared between various groups using Kaplan-Meier curves.RESULTS Consistent with data published in the literature,we confirmed that intraperitoneal administration of acetaminophen(500 mg/kg)in mice induced severe liver injury as evidenced by increases in alanine aminotransferase,aspartate aminotransferase and lactate dehydrogenase but also by liver necrosis score.Glycyrrhizin administration was shown to reduce the release of HMGB1 and significantly decreased the severity of liver injury.Thus,the co-administration of glycyrrhizin and N-acetylcysteine was investigated.Administered concomitantly with acetaminophen,the combination significantly reduced the severity of liver injury.Delayed administration of the combination of drugs,2 h or 6 h after acetaminophen,also induced a significant decrease in hepatocyte necrosis compared to mice treated with N-acetylcysteine alone.In addition,administration of N-acetylcysteine/glycyrrhizin combination was associated with an improved survival rate compared to mice treated with only N-acetylcysteine.CONCLUSION We demonstrate that,compared to N-acetylcysteine alone,co-administration of glycyrrhizin decreases the liver necrosis score and improves survival in a murine model of acetaminophen-induced liver injury.Our study opens a potential new therapeutic pathway in the prevention of acetaminophen hepatotoxicity.Charlotte Minsart Sandrine Rorive Arnaud Lemmers Eric Quertinmont Thierry Gustot 2020World Journal of Hepatology2020,12,9:4
4Effects of temperature,pH and O_(2) on the removal of hydrogen sulfide from biogas by external biological desulfurization in a full scale fixed-bed trickling bioreactor(FBTB)显示文摘Hydrogen sulfide(H_(2)S)is a critical component of biogas formed under anaerobic conditions by sulfur and sulfate reducing bacteria from animal manure and renewable energy crops.H_(2)S causes high corrosion in equipment,has a negative environmental impact,inhibits the biogas formation process and is furthermore odorous and toxic.Although several methods for internal and external desulfurization found their way into practice and had been explored at laboratory scale,no data were available on the performance of such methods in full scale practice,especially for an external fixed-bed trickling bioreactor(FBTB).The effects of temperature,pH and air ratio on H_(2)S removal efficiency(RE)were studied.The study was conducted at a research biogas plant with a given output of 96 m^(3) biogas per hour,and an H_(2)S concentration ranging between 500 ppm and 600 ppm(1 ppm=1 cm^(3)/m^(3))on average.The FBTB column has been designed to hold a packing volume of 2.21 m^(3) at a gas retention time of 84 seconds being loaded at an average of 32.88 g H_(2)S/(m^(3)·h).The highest H_(2)S RE of 98% was found at temperatures between 30℃ and 40℃.A major decline in RE to 21%-45%was observed at temperatures from 5℃ to 25℃.The results clearly showed a temperature optimum range for sulfate reducing bacteria.The results reveal that RE is little affected by different pH values and air ratios.During the experimental period,the practical suitability of the FBTB system could be proved while avoiding the disadvantages of internal biological desulfurization methods.Hans-Joachim Naegele Jonas Lindner Wolfgang Merkle Andreas Lemmer Thomas Jungbluth Claus Bogenrieder 2013International Journal of Agricultural and Biological Engineering2013,6,1:3
5Pyrolysis-induced shrinking of threedimensional structures fabricated by twophoton polymerization:experiment and theoretical model显示文摘The introduction of two-photon polymerization(TPP)into the area of Carbon Micro Electromechanical Systems(CMEMS)has enabled the fabrication of three-dimensional glassy carbon nanostructures with geometries previously unattainable through conventional UV lithography.Pyrolysis of TPP structures conveys a characteristic reduction of feature size—one that should be properly estimated in order to produce carbon microdevices with accuracy.In this work,we studied the volumetric shrinkage of TPP-derived microwires upon pyrolysis at 900℃.Through this process,photoresist microwires thermally decompose and shrink by as much as 75%,resulting in glassy carbon nanowires with linewidths between 300 and 550 nm.Even after the thermal decomposition induced by the pyrolysis step,the linewidth of the carbon nanowires was found to be dependent on the TPP exposure parameters.We have also found that the thermal stress induced during the pyrolysis step not only results in axial elongation of the nanowires,but also in buckling in the case of slender carbon nanowires(for aspect ratios greater than 30).Furthermore,we show that the calculated residual mass fraction that remains after pyrolysis depends on the characteristic dimensions of the photoresist microwires,a trend that is consistent with several works found in the literature.This phenomenon is explained through a semi-empirical model that estimates the feature size of the carbon structures,serving as a simple guideline for shrinkage evaluation in other designs.Braulio Cardenas-Benitez Carsten Eschenbaum Dario Mager Jan G.Korvink Marc JMadou Uli Lemmer Israel De Leon Sergio O.Martinez-Chapa 2019Microsystems & Nanoengineering2019,5,1:2
