| 1 | Multiparametric analysis of colorectal cancer immune responses显示文摘Colorectal cancer(CRC) is a heterogeneous disease, with a diverse and plastic immune cell infiltrate. These immune cells play an important role in regulating tumour growth-progression or elimination. Some populations of cells have a strong correlation with disease-free survival, making them useful prognostic markers. In particular, the infiltrate of CD3^+ and CD8^+ T cells into CRC tumours has been validated worldwide as a valuable indicator of patient prognosis. However, the heterogeneity of the immune response, both between patients with tumours of different molecular subtypes, and within the tumour itself, necessitates the use of multiparametric analysis in the investigation of tumour-specific immune responses. This review will outline the multiparametric analysis techniques that have been developed and applied to studying the role of immune cells in the tumour, with a focus on colorectal cancer. Because much of the data in this disease relates to T cell subsets and heterogeneity, we have used T cell populations as examples throughout. Flow and mass cytometry give a detailed representation of the cells within the tumour in a single-cell suspension on a per-cell basis. Imaging technologies, such as imaging mass cytometry, are used to investigate increasing numbers of markers whilst retaining the spatial and structural information of the tumour section and the infiltrating immune cells. | 3 | Keratinocyte 生长 factor-2 和自体同源的浆液加强处于在老鼠的角膜损坏的间充质的干细胞的再生效果显示文摘AIM:To investigate the healing process after severe corneal epithelial damage in rats treated with mesenchymal stem cells(MSCs)cultured with or without keratinocyte growth factor(KGF-2)and autologous serum(AS)on amniotic membrane(AM).Many patients are blind and devastated by severe ocular surface diseases due to limbal stem cell deficiency.Bone marrow-derived MSCs are potential sources for cellbased tissue engineering to repair or replace the corneal tissue,having the potential to differentiate to epithelial cells.METHODS:The study included 5 groups each including 10 female'Sprague Dawley'rats in addition to20 male rats used as bone marrow donors.Group I rats received AM+MSCs,Group II rats AM+MSCs cultured with KGF-2,Group III rats AM+MSCs cultured with KGF-2+AS,Group IV rats only AM and Group V rats,none.AS was derived from blood drawn from male rats and bone marrow was obtained from the femur and tibia bones of the same animals.Therapeutic effect was evaluated with clinical,histopathological and immunohistochemical assessment.MSC engraftment was demonstrated via detection of donor genotype(Y+)in the recipient tissue(X)with polymerase chain reaction.RESULTS:Corneal healing was significantly better in Groups I-III rats treated with MSC transplantation compared to Group IV and Group V rats with supportive treatment only.The best results were obtained in Group III rats with 90%transparency,70%lack of neovascularization,and 100%epithelium damage limited to less than 1/4 of cornea.CONCLUSION:We suggest that culture of MSCs with KGF-2 and AS on AM is effective in corneal repair in case of irreversible damage to limbal stem cells. | Ferda Alpaslan Pinarli Gülsen Okten Umit Beden Tunc Fisgin Mehmet Kefeli Nurten Kara Feride Duru Leman Tomak | 2014 | International Journal of Ophthalmology(English edition)2014,7,2: | 4 |

| 5 | Microfluidic actuators based on temperature-responsive hydrogels显示文摘The concept of using stimuli-responsive hydrogels to actuate fluids in microfluidic devices is particularly attractive,but limitations,in terms of spatial resolution,speed,reliability and integration,have hindered its development during the past two decades.By patterning and grafting poly(N-isopropylacrylamide)PNIPAM hydrogel films on plane substrates with a 2μm horizontal resolution and closing the system afterward,we have succeeded in unblocking bottlenecks that thermo-sensitive hydrogel technology has been challenged with until now.In this paper,we demonstrate,for the first time with this technology,devices with up to 7800 actuated micro-cages that sequester and release solutes,along with valves actuated individually with closing and opening switching times of 0.6±0.1 and 0.25±0.15 s,respectively.Two applications of this technology are illustrated in the domain of single cell handling and the nuclear acid amplification test(NAAT)for the Human Synaptojanin 1 gene,which is suspected to be involved in several neurodegenerative diseases such as Parkinson’s disease.The performance of the temperature-responsive hydrogels we demonstrate here suggests that in association with their moderate costs,hydrogels may represent an alternative to the actuation or handling techniques currently used in microfluidics,that are,pressure actuated polydimethylsiloxane(PDMS)valves and droplets. | Loïc D'Eramo Benjamin Chollet Marie Leman Ekkachai Martwong Mengxing Li Hubert Geisler Jules Dupire Margaux Kerdraon Clémence Vergne Fabrice Monti Yvette Tran Patrick Tabeling | 2018 | Microsystems & Nanoengineering2018,4,1: | 3 |
