| 2 | KRAS mutation:from undruggable to druggable in cancer显示文摘Cancer is the leading cause of death worldwide,and its treatment and outcomes have been dramatically revolutionised by targeted therapies.As the most frequently mutated oncogene,Kirsten rat sarcoma viral oncogene homologue(KRAS)has attracted substantial attention.The understanding of KRAS is constantly being updated by numerous studies on KRAS in the initiation and progression of cancer diseases.However,KRAS has been deemed a challenging therapeutic target,even“undruggable”,after drugtargeting efforts over the past four decades.Recently,there have been surprising advances in directly targeted drugs for KRAS,especially in KRAS(G12C)inhibitors,such as AMG510(sotorasib)and MRTX849(adagrasib),which have obtained encouraging results in clinical trials.Excitingly,AMG510 was the first drug-targeting KRAS(G12C)to be approved for clinical use this year.This review summarises the most recent understanding of fundamental aspects of KRAS,the relationship between the KRAS mutations and tumour immune evasion,and new progress in targeting KRAS,particularly KRAS(G12C).Moreover,the possible mechanisms of resistance to KRAS(G12C)inhibitors and possible combination therapies are summarised,with a view to providing the best regimen for individualised treatment with KRAS(G12C)inhibitors and achieving truly precise treatment. | Lamei Huang Zhixing Guo Fang Wang Liwu Fu | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 5 |
| 3 | Structural design and parameter determination for fluted-roller fertilizer applicator显示文摘The instability of the motion layer of fertilizer particle influences the precision and the accuracy of the amount of fertilizer application in the working process of fluted-roller fertilizer applicator.The characteristics of the motion layer were investigated,and a systematic scheme was proposed for designing the structures of fertilizer-filling cavity,fertilizer-filling surface and fertilizer-delivery cavity.The key parameters of the structure were studied by discrete element method.It was figured out that to ensure the smooth operation of fertilizer applicator,the fertilizer-filling angle was 105°,fertilizer-contact angle should be large than 100°,and the fertilizer-resistance angle should be smaller than 35°.Moreover,the stability of fertilizer motion morphology was investigated to verify the design.The results showed that when the roll rotational speed was larger than 120 deg/s,compared with the conventional design the CV of the amount of fertilizer application was reduced from 1.5%-3.5%to 0.5%-1.0%,and the accuracy of the fertilizer discharge was improved with the square of the correlation coefficient between the amount of fertilizer application and the roll rotational speed was larger than 0.999.It was deduced from the results that the precision and accuracy of the amount of fertilizer application are improved via eliminating the driving layer and enhancing the stable filling and conveying of the forced layer. | Shan Zeng Yipeng Tan Yu Wang Xiwen Luo Lamei Yao Dengpan Huang Zewen Mo | 2020 | International Journal of Agricultural and Biological Engineering2020,13,2: | 4 |
| 4 | Interaction between source rock and evaporite:A case study of natural gas generation in the northern Dongying Depression显示文摘It is well known that oil generated from source rocks of saline-lake facies is characteristic of low Pr/Ph ratio and high contents of gammacerane.The authors found that gas generated from the same source rocks was high in benzene and cyclohexane contents and had light carbon isotope compositions.In this study,a series of thermal simulations of gas generations on source rock with/without evaporites(such as carbonate,gypsum,and sodium chlorite,respectively),were conducted.It was found that the gypsum played an important role on the catalyzsis of gas generation in the thermal simulations.Compositions of the gaseous hydrocarbons generated from source rocks with evaporites are very similar to those of natural gases discovered from northern Dongying Depression.Meanwhile,in the thermal simulations,it was found that the carbon isotopic compositions of gaseous hydrocarbons generated from source rocks with evaporites are lighter than those of the gases from source rocks without evaporites.Therefore,it is concluded that natural gases discovered from northern Dongying Depression are the product of interaction between source rocks and evaporites(especially gypsum) of the saline facies. | JIN Qiang WANG Juan SONG Guoqi WANG Li LIN Lamei BAI Shengpin | 2010 | Chinese Journal Of Geochemistry2010,29,1: | 3 |
| 5 | Single-atom cobalt nanozymes promote spinal cord injury recovery by anti-oxidation and neuroprotection显示文摘Oxidative stress and inflammation are central pathophysiological processes in a traumatic spinal cord injury(SCI).Antioxidant therapies that reduce the reactive oxygen and nitrogen species(RONS)overgeneration and inflammation are proved promising for improving the outcomes.However,efficient and long-lasting antioxidant therapy to eliminate multiple RONS with effective neuroprotection remains challenging.Here,a single-atom cobalt nanozyme(Co-SAzyme)with a hollow structure was reported to reduce the RONS and inflammation in the secondary injury of SCI.Among SAzymes featuring different single metal-N sites(e.g.,Mn,Fe,Co,Ni,and Cu),this Co-SAzyme showed a versatile property to eliminate hydrogen peroxide(H_(2)O_(2)),superoxide anion(O_(2)·^(-)),hydroxyl radical(·OH),nitric oxide(·NO),and peroxynitrite(ONOO^(-))that overexpressed in the early stage of SCI.The porous hollow structure also allowed the encapsulation and sustained release of minocycline for neuroprotection in synergy.In vitro results showed that the Co-SAzyme reduced the apoptosis and pro-inflammatory cytokine levels of microglial cells under oxidative stress.In addition,the Co-SAzyme combined with minocycline achieved remarkable improved functional recovery and neural repairs in the SCI-rat model. | Yuxing Jiang Hongtao Rong Yifan Wang Shange Liu Peng Xu Zhen Luo Lamei Guo Tao Zhu(✉) Hongpan Rong Dingsheng Wang Jiatao Zhang Yu Yi Hao Wang | 2023 | Nano Research2023,16,7: | 2 |
| 6 | Human umbilical cord mesenchymal stem cells for psoriasis:a phase 1/2a,single-arm study显示文摘Psoriasis is a common,chronic immune-mediated systemic disease that had no effective and durable treatment.Mesenchymal stem cells(MSCs)have immunomodulatory properties.Therefore,we performed a phase 1/2a,single-arm clinical trial to evaluate the safety and efficacy of human umbilical cord-derived MSCs(UMSCs)in the treatment of psoriasis and to preliminarily explore the possible mechanisms.Seventeen patients with psoriasis were enrolled and received UMsC infusions.Adverse events,laboratory parameters,PASl,and PGA were analyzed.We did not observe obvious side effects during the treatment and 6-month follow-up.A total of 47.1%(8/17)of the psoriasis patients had at least 40%improvement in the PASl score,and 17.6%(3/17)had no sign of disease or minimal disease based on the PGA score.And the efficiency was 25%(2/8)for males and 66.7%(6/9)for females.After UMSC transplantation(UMSCT),the frequencies of Tregs and CD4^(+)memory T cells were significantly increased,and the frequencies of T helper(Th)17 and CD4^(+)naive T cells were significantly decreased in peripheral blood(PB)of psoriasis patients.And all responders showed significant increases in Tregs and CD4^(+)memory T cells,and significant decreases in Th17 cells and serum IL-17 level after UMsCT.And baseline level of Tregs in responders were significantly lower than those in nonresponders.In conclusion,allogeneic UMSCT is safe and partially effective in psoriasis patients,and level of Tregs may be used as a potent biomarker to predict the clinical efficacy of UMSCT.Trial registration Clinical Trials NCT03765957. | Lamei Cheng Siqi Wang Cong Peng Xiao Zou Chao Yang Hua Mei Chuang Li Xian Su Na Xiao Qi Ouyang Mi Zhang Qiaolin Wang Yan Luo Minxue Shen Qun Qin Honglin Wang Wu Zhu Guangxiu Lu Ge Lin Yehong Kuang Xiang Chen | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 2 |
| 7 | A20 promotes colorectal cancer immune evasion by upregulating STC1 expression to block “eat-me” signal显示文摘Immune checkpoint inhibitors(ICIs)have induced durable clinical responses in a subset of patients with colorectal cancer(CRC).However,the dis-satisfactory response rate and the lack of appropriate biomarkers for selecting suitable patients to be treated with ICIs pose a major challenge to current immunotherapies.Inflammation-related molecule A20 is closely related to cancer immune response,but the effect of A20 on“eat-me”signal and immunotherapy efficacy remains elusive.We found that A20 downregulation prominently improved the antitumor immune response and the efficacy of PD-1 inhibitor in CRC in vitro and in vivo.Higher A20 expression was associated with less infiltration of immune cells including CD3(+),CD8(+)T cells and macrophages in CRC tissues and also poorer prognosis.Gain-and loss-A20 functional studies proved that A20 could decrease the“eat-me”signal calreticulin(CRT)protein on cell membrane translocation via upregulating stanniocalcin 1(STC1),binding to CRT and detaining in mitochondria.Mechanistically,A20 inhibited GSK3βphosphorylating STC1 at Thr86 to slow down the degradation of STC1 protein.Our findings reveal a new crosstalk between inflammatory molecule A20 and“eat-me”signal in CRC,which may represent a novel predictive biomarker for selecting CRC patients most likely to benefit from ICI therapy. | Min Luo Xueping Wang Shaocong Wu Chuan Yang Qiao Su Lamei Huang Kai Fu Sainan An Fachao Xie Kenneth Kin Wah To Fang Wang Liwu Fu | 2023 | Signal Transduction and Targeted Therapy2023,8,9: | 1 |

| 10 | Yak rumen microbiome elevates fiber degradation ability and alters rumen fermentation pattern to increase feed efficiency显示文摘Rumen microbes play an important role in ruminant energy supply and animal performance.Previous studies showed that yak(Bos grunniens)rumen microbiome and fermentation differ from other ruminants.However,little is understood about the features of the rumen microbiome that make yak adapted to their unique environmental and dietary conditions.This study was to investigate the rumen microbiome and metabolome to understand how yak adapt to the coarse forage and harsh environment in the Qinghai-Tibetan plateau.Nine female Qaidam yellow cattle(Bos taurus),9 dzomo(hybrids of cattle and yak)and 9 female plateau yak(B.grunniens),about 5 to 6 years old,were used in this study.Rumen fermentation parameters,fibrolytic enzyme activities,and rumen metataxonomic were determined.Then 18(6 samples per group)were selected for rumen metagenomic and metabolome analysis.Metataxonomic analysis revealed that the rumen microbiota was significantly different among plateau yak,Qaidam yellow cattle,and dzomo(P<0.05).Metagenomic analysis displayed a larger gene pool encoding a richer repertoire of carbohydrate-active enzymes in the rumen microbiome of plateau yak and dzomo than Qaidam yellow cattle(P<0.05).Some of the genes encoding glycoside hydrolases that mediate the digestion of cellulose and hemicellulose were significantly enriched in the rumen of plateau yak than Qaidam yellow cattle,but glycoside hydrolase 57 that primarily includes amylases was abundant in Qaidam yellow cattle(P<0.05).The rumen fermentation profile differed also,Qaidam yellow cattle having a higher molar proportion of acetate but a lower molar proportion of propionate than dzomo and plateau yak(P<0.05).Based on metabolomic analysis,rumen microbial metabolic pathways and metabolites were different.Differential metabolites are mainly amino acids,carboxylic acids,sugars,and bile acids.Changes in rumen microbial composition could explain the above results.The present study showed that the rumen microbiome of plateau yak helps its host to adapt to the Qinghai-Tibetan plateau.In particular,the plateau yak rumen microbiome has more enzymes genes involved in cellulase and hemicellulase than that of cattle,resulting higher fibrolytic enzyme activities inyak,further providing stronger fiber degradation function. | Congcong Zhao Lamei Wang Shanlin Ke Xinhua Chen Akos Kenez Wei Xu Dangdang Wang Fan Zhang Yong Li Zhanhong Cui Yu Qiao Jing Wang Wenjuan Sun Jianhua Zhao Junhu Yao Zhongtang Yu Yangchun Cao | 2022 | Animal Nutrition2022,,4: | 1 |
| 11 | Genomic insights into the origin, adaptive evolution, and herbicide resistance of Leptochloa chinensis, a devastating tetraploid weedy grass in rice fields显示文摘Chinese sprangletop (Leptochloa chinensis), belonging to the grass subfamily Chloridoideae, is one of the most notorious weeds in rice ecosystems. Here, we report a chromosome-scale reference genome assembly and a genomic variation map of the tetraploid L. chinensis. The L. chinensis genome is derived from two diploid progenitors that diverged ∼10.9 million years ago, and its two subgenomes display neither fractionation bias nor overall gene expression dominance. Comparative genomic analyses reveal substantial genome rearrangements in L. chinensis after its divergence from the common ancestor of Chloridoideae and, together with transcriptome profiling, demonstrate the important contribution of tetraploidization to the gene sources for the herbicide resistance of L. chinensis. Population genomic analyses of 89 accessions from China reveal that L. chinensis accessions collected from southern/southwestern provinces have substantially higher nucleotide diversity than those from the middle and lower reaches of the Yangtze River, suggesting that L. chinensis spread in China from the southern/southwestern provinces to the middle and lower reaches of the Yangtze River. During this spread, L. chinensis developed significantly increased herbicide resistance, accompanied by the selection of numerous genes involved in herbicide resistance. | 13 | FY-4 Meteorological Satellite显示文摘FY-4 is the second generation of Chinese geostationary satellite for quantitative remote sensing meteorological application. The detection efficiency, spectral bands, spatial and time resolution have been greatly improved with respect to those of first generation, as well as the radiometric calibration and sensitivity. The combination of multichannel detection and vertical sounding was first realized on FY-4, because both the Advanced Geostationary Radiation Imager(AGRI) and Geostationary Interferometric Infrared Sounder(GIIRS) are on the same spacecraft. The main performance of the payloads including AGRI, GIIRS and Lightning Mapping Imager, and the spacecraft bus are presented, the performance being equivalent to the level of the third generation meteorological satellites in Europe and USA. The acquiring methods of remote sensing data including multichannel and high precision quantitative observing, imaging collection of the ground and cloud, vertical observation of atmospheric temperature and moisture, lightning imaging observation and space environment detection are shown. | 15 | Frequency reconfigurable antenna actuated by inflated triangular structure:Design and analysis显示文摘This paper proposes a frequency reconfigurable triangular antenna actuated by an inflated triangular structure.The open path antenna is transformed from an open type to a closed structure by inflating.Inflatable structures are easy to manufacture by fusing 2 inextensible membranes together along a defined pattern of lines.However,the prediction of their deployed shape remains a challenge.To solve the pattern changed problem,guided by geometric analyses and local buckle characteristics,the inflated triangular structure has been designed and verified by experiment and simulation.In the process of transformation of the antenna,the resonant frequency of the antenna is changed because this frequency is determined by the conformational change.The resonant frequency changes from GHz to kHz when the design of initial structure sizes is from millimeter to meter.The measured peak gains,the frequency,and the radiation direction are also reconfigurable by the initial size.Finally,the reconfigurable resonator array is presented,which is coupled to electric fields to absorb all incident radiation.In this work,the changed pattern design by inflating is applied to the antenna design,and its frequency reconfigurability is achieved.Through the electricity performance analysis of the reconfigurable antenna,precise manufacturing will be possible and provide guidance for manufacturing frequency reconfigurable antennas. | Jiaming GUO Xinghan QIU Jiawei ZHANG Lamei ZHANG Changguo WANG | 2023 | Chinese Journal of Aeronautics2023,36,5: | 0 |
