| 1 | Interaction of APT with BSA or HSA显示文摘In this work, N-n-amyl-N’-(sodium p- aminobenzenesulfonate) thiourea (APT) containing saturated fatty hydrocarbon group was synthesized. Fluorescence quenching methods in combination with UV absorption spectra and molecule modeling method were used to study the interaction between APT and bovine serum albumin (BSA) or human se-rum albumin (HSA). The binding constants of APT with BSA or HSA were determined at different tem-peratures under the optimum conditions based on the fluorescence quenching results. The binding characteristics of APT and BSA or HSA were reported and the binding sites were obtained. | 2 | Serum ficolin-2 concentrations are significantly changed in patients with hepatitis B virus infection and liver diseases显示文摘Human ficolin-2 is an important lectin complement pathway activator that is secreted from liver cells and has been implicated as an anti-infection innate immune molecule. However, the role of ficolin-2 protein and its dynamic changes over the course of and in the prognosis of chronic hepatitis B(CHB) and hepatocellular carcinoma(HCC) remain unclear. In this study, we analyzed ficolin-2 protein expression in a cohort of individuals with CHB infection, HCC and cirrhosis. A sandwich enzyme-linked immunosorbent assay(ELISA) method was used to measure serum ficolin-2 concentrations. Ficolin-2 expression in liver tissues was detected by immunohistochemical staining. Serum ficolin-2 concentrations in CHB patients were significantly higher than in healthy controls and HBV carriers. After 48 weeks of routine amelioration liver function treatment, serum ficolin-2 concentrations decreased and were positively correlated with favorable alanine aminotransferase(ALT), HBV DNA and HBe Ag-seroconversion outcomes. Interestingly, we observed much lower expression of serum and intrahepatic ficolin-2 in HCC and cirrhosis compared with healthy controls. | 5 | The genome-wide landscape of small insertion and deletion mutations in Monopterus albus显示文摘Insertion and deletion(indel) mutations, which can trigger single nucleotide substitutions on the flanking regions of genes, may generate abundant materials for disease defense, reproduction, species survival and evolution. However, genetic and evolutionary mechanisms of indels remain elusive. We establish a comparative genome-transcriptome-alignment approach for a large-scale identification of indels in Monopterus population. Over 2000 indels in 1738 indel genes, including 1-21 bp deletions and 1-15 bp insertions, were detected. Each indel gene had ~1.1 deletions/insertions, and 2-4 alleles in population. Frequencies of deletions were prominently higher than those of insertions on both genome and population levels. Most of the indels led to in frame mutations with multiples of three and majorly occurred in non-domain regions, indicating functional constraint or tolerance of the indels. All indel genes showed higher expression levels than non-indel genes during sex reversal. Slide window analysis of global expression levels in gonads showed a significant positive correlation with indel density in the genome. Moreover, indel genes were evolutionarily conserved and evolved slowly compared to nonindel genes. | 7 | Ficolin-2 binds to HIV-1 gp120 and blocks viral infection显示文摘Ficolin-2 is a lectin complement pathway activator present in normal human plasma and usually associated with infectious diseases, but little is known about the role of ficolin-2 in human immunodeficiency virus(HIV) infection. Here, we describe our novel findings that serum ficolin-2concentrations of 103 HIV-1 patients were much higher compared to those of 57 healthy donors. In vitro analysis showed that HIV-1 infection could enhance ficolin-2 expression. We further demonstrated that recombinant ficolin-2 protein could bind with HIV-1 envelope glycoprotein gp120, and subsequently induce complement dependent cytotoxicity. Moreover, ficolin-2 could block the entry of HIV-1 into target cells(TZM-b1 and MT-2 cells) and infection in a ficolin-2 dosedependent manner. | 9 | Single-cell landscape analysis reveals distinct regression trajectories and novel prognostic biomarkers in primary neuroblastoma显示文摘Neuroblastoma(NB),which is the most common pediatric extracranial solid tumor,varies widely in its clinical presentation and outcome.NB has a unique ability to spontaneously differentiate and regress,suggesting a potential direction for therapeutic intervention.However,the underlying mechanisms of regression remain largely unknown,and more reliable prognostic biomarkers are needed for predicting trajectories for NB.We performed scRNAseq analysis on 17 NB clinical samples and three peritumoral adrenal tissues.Primary NB displayed varied cell constitution,even among tumors of the same pathological subtype.Copy number variation patterns suggested that neuroendocrine cells represent the malignant cell type.Based on the differential expression of sets of related marker genes,a subgroup of neuroendocrine cells was identified and projected to differentiate into a subcluster of benign fibroblasts with highly expressed CCL2 and ZFP36,supporting a progressive pathway of spontaneous NB regression.We also identified prognostic markers(STMN2,TUBA1A,PAGE5,and ETV1)by evaluating intra-tumoral heterogeneity.Lastly,we determined that ITGB1 in M2-like macrophages was associated with favorable prognosis and may serve as a potential diagnostic marker and therapeutic target.In conclusion,our findings reveal novel mechanisms underlying regression and potential prognostic markers and therapeutic targets of NB. | Qingqing Liu Zhenni Wang Yan Jiang Fengling Shao Yue Ma Mingzhao Zhu Qing Luo Yang Bi Lijian Cao Liang Peng Jianwu Zhou Zhenzhen Zhao Xiaobin Deng Tong-Chuan He Shan Wang | 2022 | Genes & Diseases2022,9,6: | 0 |

For tumor antigens, many carbohydrate antigens belong to T-independent (TI) antigens. Carbohydrate conjugated to protein carriers can switch TI antigen to a T-dependent (TD) antigen. Attempts to add an innate immune response element (such as Toll-like receptor ligand) to carbohydrate TI-antigens have also been studied. Glycosylation inhibitors or small interfering RNA have also been used for antitumor and/or antiviral agents.
