| 3 | Spontaneous movement of a retrotransposon generated genic dominant male sterility providing a useful tool for rice breeding显示文摘Male sterility in plants provides valuable breeding tools in germplasm innovation and hybrid crop production.However,genetic resources for dominant genic male sterility,which hold great promise to facilitate breeding processes,are extremely rare in natural germplasm.Here we characterized the Sanming Dominant Genic Male Sterility in rice and identified the gene SDGMS using a map-based cloning approach.We found that spontaneous movement of a 1978-bp long terminal repeat(LTR)retrotransposon into the promoter region of the SDGMS gene activates its expression in anther tapetum,which causes abnormal programmed cell death of tapetal cells resulting in dominant male sterility.SDGMS encodes a ribosome inactivating protein showing N-glycosidase activity.The activation of SDGMS triggers transcription reprogramming of genes responsive to biotic stress leading to a hypersensitive response which causes sterility.The results demonstrate that an ectopic gene activation by transposon movement can give birth to a novel trait which enriches phenotypic diversity with practical utility. | Conghao Xu Yifeng Xu Zhengji Wang Xiaoyu Zhang Yuying Wu Xinyan Lu Hongwei Sun Lei Wang Qinglu Zhang Qinghua Zhang Xianghua Li Jinghua Xiao Xu Li Mingfu Zhao Yidan Ouyang Xianbo Huang Qifa Zhang | 2023 | National Science Review2023,10,9: | 1 |

| 8 | Inhibitory Effects of Cordyceps militaris( Bombyx mori )Extract against Respiratory Syncytial Virus显示文摘[Objective] This study aimed to investigate the inhibitory effects of 95%,70% ethanol extracts and water extract of Cordyceps militaris( Bombyx mori). on cells infected with respiratory syncytial virus( RSV).[Method] Human embryonic kidney cells( HEK293,RSV virus-sensitive cells) were infected with respiratory syncytial virus and incubated with tissue cell culture method,to analyze the inhibitory effects of different extracts of Cordyceps militaris( Bombyx mori) on proliferation of RSV-infected cells by cytopathic effect( CPE) observation and tetrazolium salt reduction assay( MTT method).[Result]Different extracts of Cordyceps militaris( Bombyx mori) were non-toxic to HEK293 cells. CPE occurred successively in various sample groups after 48 h,and the degree of CPE was reduced gradually with the increasing concentration. To be specific,95% ethanol extract(100 μg / ml),70% ethanol extract(100,25 μg / ml) and water extract(100,25 μg/ml) of Cordyceps militaris(Bombyx mori) exhibited inhibitory effects against RSV subtype A; 70% ethanol extract(100,25 μg/ml) and water extract(100,25 μg / ml) of Cordyceps militaris( Bombyx mori) exhibited inhibitory effects against RSV subtype B; the effects of water extract(100 μg / ml) of Cordyceps militaris( Bombyx mori) on RSV subtypes A and B were similar to positive control group. However,different extracts of Cordyceps militaris( Bombyx mori) with low concentrations(6. 25,1. 56 μg / ml) exhibited no inhibitory effects against respiratory syncytial virus. | 9 | Activation of NLRP3 Inflammasome via Drp1 Overexpression in Kupffer Cells Aggravates Ischemia-reperfusion Injury in Hepatic Steatosis显示文摘Background and Aims:Donors with fatty livers are considered to address the shortage of livers for transplantation,but those livers are particularly sensitive to ischemia-reperfusion injury(IRI),and an increased incidence of graft failure is observed.Kupffer cells account for 20–35%of liver nonparenchymal cells,and have been shown to participate in the process of IRI and inflammatory reactions of hepatic steatosis.NOD-like receptor thermal protein domain-associated protein 3(NLRP3)is an intracellular sensor activated by Kupffer cells to promote generation and participates in IRI.Dynamics-associated protein 1(Drp1)is one of the main proteins regulating mitochondrial division and exacerbates IRI by affecting mitochondrial dynamics.The mechanism of interaction of Kupffer cells with Drp1 and NLRP3 to aggravate IRI has not been clarified.Methods:A mouse model of hepatic steatosis was established by feeding the mice with a high-fat diet.In vitro experiments were performed using AML12 normal mouse liver cells and RAW264.7 mononuclear macrophage cells cultured in medium with palmitate and oleic acid.Western blotting and immunohistochemical(IHC)staining were used to detect the expression of NLRPP3 and Drp1 in IRI in the control and high-fat diet groups.The expression of F4/80+cells during IRI in hepatic steatosis was verified by IHC staining,and the role of NLRPP3 and Drp1 in Kupffer-cell mediated IRI was investigated by targeting Drp-1 inhibition.Results:Drp1 and NLRP3 expression was increased during IRI in hepatic steatosis,and the expression of Drp1 and NLRP3 were decreased after the elimination of Kupffer cells.That indicated Kupffer cells were involved in the process of IRI in hepatic steatosis through the action of Drp1 and NLRP3.After Drp1 inhibition,liver function was restored and NLRP3 expression level was reduced.Conclusions:Kupffer cells aggravated IRI in hepatic steatosis via NLRP3 and Drp1.Drp1 inhibitors might be useful as specific therapeutics to alleviate IRI in hepatic steatosis and may have promise in case of liver donor shortage. | Lu Zhang Mingfu Wang Ran An Jun Dai Shujun Liu Ming Chen Haoran Ding | 2023 | Journal of Clinical and Translational Hepatology2023,11,5: | 0 |
