1Breast cancer stem-like cells can promote metastasis by activating platelets and down-regulating antitumor activity of natural killer cells显示文摘OBJECTIVE:To investigate whether cancer stem cells(CSCs) more efficiently activating platelets and evading immune surveillance than non-CSCs thus promoting metastasis.METHODS:We enriched and identified sphereforming cells(SFCs) and coincubated washed platelets with several platelet activators including collagen,4T1 and SFCs.Platelet-coating tumor cells,platelet activation and TGF-β1 release were analyzed.Then natural kell cells(NK) were incubated with supernatants of different activated platelet samples what we called sample release(SR).The degranulation assay and NKG2 D expression on NK cells were conducted by flow cytometry.Finally tissue factor(TF) expression of SFCs or 4T1 were evaluated by western blot.RESULTS:Breast cancer cell line 4T1 could form spheres in serum-free medium at low adherence.Sphere-forming cells expressed high levels of the CD24-/lowCD44 + stem cell phenotype.Both sphere-forming cells or 4T1 were coated with abundant platelets while sphere-forming cells induced significantly higher expression of platelet activating receptor CD62 p than 4T1 did(P < 0.01).And sphere-forming cells induced platelets to produce more TGF-β1 than 4T1 did(P < 0.01).Furthermore,sample releases induced by sphere-forming cells caused more vigorous inhibition of NK cells antitumor reactivity(P < 0.05) and reduced NKG2 D expression(P < 0.01).The final results showed that sphere-forming cells expressed higher levels of TF than 4T1(P < 0.05).CONCLUSION:Our findings indicate that CSCs could efficiently activate platelets,induce platelets to secrete more TGF-β1,decrease NKG2 D expression and inhibit antitumor activity of NK cell,compared with 4T1.And higher levels of TF expression of CSCs may account for this correlation of CSCs and platelets.Wang Shuo Zhang Ying Cong Weihong Liu Jie Zhang Yuren Fan Huiting XuYonggang Lin Hongsheng 2016Journal of Traditional Chinese Medicine2016,36,4:5

516.3%Efficiency binary all-polymer solar cells enabled by a novel polymer acceptor with an asymmetrical selenophene-fused backbone显示文摘Despite the significant progress made recently in all-polymer solar cells(all-PSCs),it is still quite challenging to achieve high open-circuit voltage(V_(oc))and short-circuit current density(J_(sc))simultaneously in order to further improve their performance.The recent strategy of using selenophene to replace thiophene on the Y6 based polymer acceptors has resulted in significantly improved J_(sc)s of the resulting all-PSCs.However,such modifications have also depressed V_(oc),which compromises the overall performance of the devices.Herein,we present the design and synthesis of a novel polymer acceptor,PYT-1S1Se,created by inserting an asymmetrical selenophene-fused framework to precisely manipulate optical absorption and electronic properties.Compared with the selenium-free analog,PYT-2S,and symmetrical selenium-fused analog,PYT-2Se,the PYT-1S1Se derived all-PSCs not only deliver optimized J_(sc)(24.1 mA cm^(−2))and V_(oc)(0.926 V)metrics,but also exhibit a relatively low energy loss of 0.502 eV.Consequently,these devices obtain a record-high power conversion efficiency(PCE)of 16.3%in binary all-PSCs.This work demonstrates an effective molecular design strategy for balancing the trade-off between V_(oc) and J_(sc) to achieve highefficiency all-PSCs.Huiting Fu Qunping Fan Wei Gao Jiyeon Oh Yuxiang Li Francis Lin Feng Qi Changduk Yang Tobin J.Marks Alex K.-Y.Jen 2022Science China Chemistry2022,65,2:1
6Connectome-based predictive modelling can predict follow-up craving after abstinence in individuals with opioid use disorders显示文摘Background Individual differences have been detected in individuals with opioid use disorders(OUD)in rehabilitation following protracted abstinence.Recent studies suggested that prediction models were effective for individual-level prognosis based on neuroimage data in substance use disorders(SUD).Aims This prospective cohort study aimed to assess neuroimaging biomarkers for individual response to protracted abstinence in opioid users using connectome-based predictive modelling(CPM).Methods One hundred and eight inpatients with OUD underwent structural and functional magnetic resonance imaging(fMRI)scans at baseline.The Heroin Craving Questionnaire(HCQ)was used to assess craving levels at baseline and at the 8-month follow-up of abstinence.CPM with leave-one-out cross-validation was used to identify baseline networks that could predict follow-up HCQ scores and changes in HCQ(HCQtolow V-up-HCQpa baseline).Then,the follow-up aseline predictive ability of identified networks was tested in a separate,heterogeneous sample of methamphetamine individuals who underwent MRI scanning before abstinence for SUD.Results CPM could predict craving changes induced by long-term abstinence,as shown by a significant correlation between predicted and actual HCQ fllow-up(r=0.417,p<0.001)and changes in HCQ(negative:r=0.334,p=0.002;positive:r=0.233,p=0.038).Identified craving-related prediction networks included the somato-motor network(SMN),salience network(SALN),default mode network(DMN),medial frontal network,visual network and auditory network.In addition,decreased connectivity of frontal-parietal network(FPN)-SMN,FPN-DMN and FPN-SALN and increased connectivity of subcortical network(SCN)-DMN,SCN-SALNandSCN-SMN were positively correlated with craving levels.Conclusions These findings highlight the potential applications of CPM to predict the craving level of individuals after protracted abstinence,as well as the generalisation ability;the identified brain networks might be the focus of innovative therapies in the future.Wenhan Yang Jungong Han Jing Luo Fei Tang Li Fan Yanyao Du Longtao Yang Jun Zhang Huiting Zhang Jun Liu 2023General Psychiatry2023,36,6:0
