1Chaiqinchengqi decoction regulates necrosis-apoptosis via regulating the release of mitochondrial cytochrome c and caspase-3 in rats with acute necrotizing pancreatitis显示文摘OBJECTIVE: To explore the effect and the mechanism of Chaiqinchengqi decoction(CQCQD) on the apoptosis-necrosis switch of pancreatic acinar cells in acute necrotizing pancreatitis(ANP) in rats.METHODS: Sixty Sprague-Dawley rats were randomized into the control group, the ANP group and the CQCQD group. The acute pancreatitis(AP)model was induced by intraperitoneal injections of4 g/kg 8% L-Arginine(PH 7.0) twice with a 1 h interval. Rats in the CQCQD group were intragastrically administered CQCQD(20 mL/kg every 2 h, 3 times,then 20 mL/kg every 6 h, 3 times). Rats were killed at the 6 and 24 h after the induction of AP.The pancreatic tissues were collected for pathology and to isolate pancreatic acinar cells and mitochondria.RESULTS: CQCQD significantly ameliorated the severity of ANP by reducing the pancreatic histopathology score, indicated by lactate dehydrogenase levels at the 6 and 24 h. The CQCQD group promoted the apoptosis of pancreatic acinar cells by raising the apoptosis index compared with the ANP group and the control group. Mitochondrial cytochrome c at the 6 and 24 h in the ANP group were lower than that in the control group or the CQCQD group(0.67±0.13 vs 1.54±0.03 vs 0.81±0.09; 0.71±0.08 vs 1.55±0.09 vs 0.89±0.16, P<0.01). The cytochrome c levels in the cytoplasm at the 6 and 2 h in the CQCQD group were higher than in the control group(1.36±0.15 vs 0.67±0.04, 1.46±0.08 vs 0.59±0.09, P<0.01), or the ANP group(0.96±0.13, P>0.05;0.97±0.09, P<0.05). CQCQD increased caspase-3 activity over the ANP group at the 6 h.CONCLUSION: CQCQD can induce apoptosis and relieve the necrosis of pancreatic acinar cells via promoting the release of mitochondrial cytochrome c and increasing pancreatic caspase-3 activity in ANP rats.Ziqi Lin Jia Guo Ping Xue Lei Huang Lihui Deng Xiaonan Yang Qing Xia 2014Journal of Traditional Chinese Medicine2014,34,2:10
2Genetic diversity and DNA fingerprinting in jute(Corchorus spp.) based on SSR markers显示文摘Genetic diversity analysis and DNA finger printing are very useful in breeding programs,seed conservation and management. Jute(Corchorus spp.) is the second most important natural fiber crop after cotton. DNA fingerprinting studies in jute using SSR markers are limited. In this study, 58 jute accessions, including two control varieties(Huangma 179 and Kuanyechangguo) from the official variety registry in China were evaluated with 28 pairs of SSR primers. A total of 184 polymorphic loci were identified. Each primer detected 3 to 15 polymorphic loci, with an average of 6.6. The 58 jute accessions were DNA-fingerprinted with 67 SSR markers from the 28 primer pairs. These markers differentiated the 58 jute accessions from one another, with Co SSR305-120 and Co SSR174-195 differentiating Huangma 179 and Kuanyechangguo, respectively. NTSYS-pc2.10 software was used to analyze the genetic diversity in the 58 jute accessions. Their genetic similarity coefficients ranged from 0.520 to 0.910 with an average of 0.749, indicating relatively great genetic diversity among them. The 58 jute accessions were divided into four groups with the coefficient 0.710 used as a value for classification, consistent with their species and pedigrees. 7Study on rolling process optimization of high carbon steel wire显示文摘The existing problems in the manufacture of SWRH82B high carbon steel wire were discussed by sampling and testing the microstructure and properties of the steel from the workshop. To solve the problems, the experimental parameters for thermal simula- tion were optimized, and the thermal simulating experiments were carried out on a Gleeble1500 thermal simulator. The process pa- rameters for the manufacture were optimized after analysis of the data, and the productive experiments were performed after the wa- ter box in front of the no-twist blocks was reconstructed, to control the temperature of the loop layer. The results from the productive experiments showed that the cooling rate of 10-15°C/s was reasonable before phase transformation, about 5°C/s during phase trans- formation, and 600-620°C was the suitable starting temperature for phase transformation. 15Test on dynamic characteristics of subgrade of heavy-haul railway in cold regions显示文摘Dynamic characteristics of heavy-haul railway subgrade under vibratory loading in cold regions are investigated via low-temperature dynamic triaxial tests with multi-stage cyclic loading process. The relationship between dynamic shear stress and dynamic shear strain of frozen soil of subgrade under train loading and the influence of freezing temperatures on dynamic constitutive relation, dynamic shear modulus and damping ratio are observed in this study. Test results show that the dynamic constitutive relations of the frozen soils with different freezing temperatures comply with the hyperbolic model, in which model parameters a and b decrease with increasing freezing temperature. The dynamic shear modulus of the frozen soils decreases with increasing dynamic shear strains initially, followed by a relatively smooth attenuation tendency, whereas increases with decreasing freezing temperatures. The damping ratios decrease with decreasing freezing temperatures.
