| 2 | Dietary advanced glycation end-products aggravate non-alcoholic fatty liver disease显示文摘AIM To determine if manipulation of dietary advanced glycation end product(AGE), intake affects nonalcoholic fatty liver disease(NAFLD) progression and whether these effects are mediated via RAGE. METHODS Male C57Bl6 mice were fed a high fat, high fructose, high cholesterol(HFHC) diet for 33 wk and compared with animals on normal chow. A third group were given a HFHC diet that was high in AGEs. Another group was given a HFHC diet that was marinated in vinegar to prevent the formation of AGEs. In a second experiment, RAGE KO animals were fed a HFHC diet or a high AGE HFHC diet and compared with wildtype controls. Hepatic biochemistry, histology, picrosirius red morphometry and hepatic mR NA were determined. RESULTS Long-term consumption of the HFHC diet generated significant steatohepatitis and fibrosis after 33 wk. In this model, hepatic 4-hydroxynonenal content(a marker of chronic oxidative stress), hepatocyte ballooning, picrosirius red staining, α-smooth muscle actin and collagen type 1A gene expression were all significantly increased. Increasing the AGE content of the HFHC diet by baking further increased these markers of liver damage, but this was abrogated by pre-marination in acetic acid. In response to the HFHC diet, RAGE-/-animals developed NASH of similar severity to RAGE+/+ animals but were protected from the additional harmful effects of the high AGE containing diet. Studies in isolated Kupffer cells showed that AGEs increase cell proliferation and oxidative stress, providing a likely mechanism through which these compounds contribute to liver injury. CONCLUSION In the HFHC model of NAFLD, manipulation of dietary AGEs modulates liver injury, inflammation, and liver fibrosis via a RAGE dependent pathway. | 3 | Hematopoietic cell transplantation for Crohn’s disease;is it time?显示文摘AIM: To review all studies in the literature that have assessed Hematopoietic cell transplantation (HCT) and Crohn’s disease (CD) with the ultimate aims of determining if this is a viable treatment option for those with CD. A secondary aim was to review the above literature and determine if the studies shed further light on the mechanisms involved in the pathogenesis of CD. METHODS: An extensive Medline search was performed on all articles from 1970 to 2005 using the ; bone marrow transplant, stem cell, hematopoietic cell, Crohn’s disease and inflammatory bowel disease. RESULTS: We identified one case in which a patient developed CD following an allogeneic HCT from a sibling suffering with CD. Evidence for transfer of the genetic predisposition to develop CD was also identified with report of a patient that developed severe CD following an allogeneic HCT. Following HCT it was found that the donor (that had no signs or symptoms of CD) and the recipient had several haplotype mismatches in HLA class Ⅲ genes in the IBD3 locus including a polymorphism of NOD2/CARD15 that has been associated with CD. Thirty three published cases of patients with CD who underwent either autologous or allogeneic HCT were identified. At the time of publication 29 of these 33 patients were considered to be in remission. The median follow-up time was seven years, and twenty months for allogeneic and autologous HCT respectively.For patients who underwent HCT primarily for treatment of their CD there have been no mortalities related to transplant complications. CONCLUSION: Overall these preliminary data suggest that both allogeneic and autologous HCT may be effective in inducing remission in refractory CD. | 4 | 罗格列酮与全反式维甲酸联合对骨髓瘤细胞增殖抑制作用及其机制探讨显示文摘目的探讨罗格列酮(RGZ)与全反式维甲酸(ATRA)在体内外对骨髓瘤细胞生长的影响及其作用途径。方法用RG2和ATRA单独或联合处理多发性骨髓瘤细胞系U266和RPMI8226细胞,采用^1H—TdR掺入法检测细胞增殖变化;Annexin—V和PI双染后采用流式细胞术检测细胞凋亡;用半定量RT-PCR方法检测凋亡抑制基因FLIP、XIAP及survivin mRNA表达变化;采用荧光CaspACE试剂盒检测caspase-3活性变化;采用裸鼠体内成瘤试验观察骨髓瘤细胞在动物体内生长情况。结果RGZ对骨髓瘤细胞具有增殖抑制作用,且呈剂量依赖关系(U266组r=0.991,P〈0.01;RPMI8226组r=0.961,P〈0.01);与RGZ单用组比较,ATRA与RGZ联合对骨髓瘤细胞的增殖抑制作用更明显(U266组P〈0.001;RPMI8226组P〈0.01);10μmol/L的RGZ处理组U266细胞凋亡率为(9.8±1.7)%,RPMI8226细胞凋亡率为(10.7±3.3)%,且诱导凋亡作用呈时间、剂量依赖性,ATRA与RGZ联合应用时,凋亡细胞的比例较相应的RGZ单独处理组显著升高(P值均〈0.01);RGZ能抑制FLIP、XIAP及survivin mRNA的表达,ATRA能协同RGZ进一步抑制FLIP、XIAP及survivin mRNA的表达;经RGZ处理的骨髓瘤细胞caspase-3活性显著增加,RGZ与ATRA联合处理后caspase-3活性较RGZ处理组显著增加(P〈0.01);RGZ可明显抑制骨髓瘤细胞在裸鼠体内的生长(P〈0.001),并且ATRA与RGZ联合能增强RGZ在裸鼠体内对骨髓瘤细胞生长的抑制作用(P〈0.01)。结论RGZ能通过抑制FLIP、XIAP及survivin的表达、促进caspase-3的活化从而诱导细胞凋亡,抑制细胞增殖。ATRA能增强RGZ对骨髓瘤细胞的上述作用,两者联合具有协同效应。 | 黄海雯 吴德沛 陈广华 常惠荣 Chow HCH Leung AYH Liang R | 2009 | 中华血液学杂志2009,30,4: | 5 |

| 8 | The effects of Spirulina on anemia and immune function in senior citizens显示文摘Anemia and immunological dysfunction(i.e.immunosenescence)are commonly found in older subjects and nutritional approaches are sought to counteract these phenomena.Spirulina is a filamentous and multicellular bule-green alga capable of reducing inflammation and also manifesting antioxidant effects.We hypothesized that Spirulina may ameliorate anemia and immunosenescence in senior citizens with a history of anemia.We enrolled 40 volunteers of both sexes with an age of 50 years or older who had no history of major chronic diseases.Participants took a Spirulina supplementation for 12 weeks and were administered comprehensive dietary questionnaires to determine their nutritional regimen during the study.Complete cell count(CCC)and indoleamine 2,3-dioxygenase(IDO)enzyme activity,as a sign of immune function,were determined at baseline and weeks 6 and 12 of supplementation.Thirty study participants completed the entire study and the data obtained were analyzed.Over the 12-week study period,there was a steady increase in average values of mean corpuscular hemoglobin in subjects of both sexes.In addition,mean corpuscular volume and mean corpuscular hemoglobin concentration also increased in male participants.Older women appeared to benefit more rapidly from Spirulina supplements.Similarly,the majority of subjects manifested increased IDO activity and white blood cell count at 6 and 12 weeks of Spirulina supplementation.Spirulina may ameliorate anemia and immunosenescence in older subjects.We encourage large human studies to determine whether this safe supplement could prove beneficial in randomized clinical trials. | Carlo Selmi Patrick SC Leung Laura Fischer Bruce German Chen-Yen Yang Thomas P Kenny Gerry R Cysewski M Eric Gershwin | 2011 | Cellular & Molecular Immunology2011,8,3: | 2 |
