| 1 | 人肿瘤细胞核苷酸切除修复蛋白表达与抗癌药耐药的相关性(英文)显示文摘背景与目的:核苷酸切除修复(Nucleotideexcisionrepair,NER)是真核细胞中的DNA修复多酶系统,它可能与人肿瘤细胞对抗癌药的耐药有关。本实验将探讨NER蛋白(XPA,XPB,XPD和ERCC1)的表达与人肿瘤细胞耐药的关系。方法:采用westernblot检测美国国家癌症研究所(NationalCancerInstitute,NCI)用于抗癌药筛选的60株人肿瘤细胞的ERCC1,XPA,XPBXPD表达,并与170种抗癌药物的细胞毒试验结果进行相关性分析。结果:ERCC1,XPB和XPD的蛋白表达与抗癌药物敏感性呈负相关,在170种抗癌药物中分别有13,17和32种的耐药性与上述蛋白水平相关性显著或非常显著(P<0.05)。而XPA的表达与药物敏感性无明显正/负相关。根据已确定的6种药物作用机理分析,肿瘤细胞XPD表达与其对烷化剂类抗癌药耐药相关性显著。结论:本实验结果肿瘤细胞的XPD蛋白表达与烷化剂类抗癌药的耐药相关,提示XPD在肿瘤细胞耐药过程中起重要作用。 | 陈忠平 Areti MALAPETSA Anne MONKS Timothy G. | 2 | Precore/basal core promoter mutants and hepatitis B viral DNA levels as predictors for liver deaths and hepatocellular carcinoma显示文摘AIM: To conduct a retrospective study in 400 chronic hepatitis B patients in order to identify hepatitis B viral factors associated with complications of liver disease or development of hepatocellular carcinoma.METHODS: The mean follow-up time was 83.6 ± 39.6 mo. Alpha-fetoprotein test and abdominal ultrasound were used for cancer surveillance. Hepatitis B basal core promoter mutants, precore mutants, genotypes, hepatitis B viral DNA (HBV DNA) level and hepatitis B e antigen (HBeAg) were measured. Univariate analysis and logistic regression were used to assess odds ratios for viral factors related to liver deaths and hepatocellular carcinoma development. RESULTS: During follow-up, 38 patients had liver deaths not related to hepatocellular carcinoma. On multivariate analysis, older age [odds ratio: 95.74 (12.13-891.31); P < 0.0001], male sex [odds ratio: 7.61 (2.20-47.95); P = 0.006], and higher log10 HBV DNA [odds ratio: 4.69 (1.16-20.43); P < 0.0001] were independently predictive for these liver related deaths. Also, 31 patients developed hepatocellular carcinoma. Multivariate analysis showed that older age [odds ratio: 26.51 (2.36-381.47); P = 0.007], presence of precore mutants [odds ratio: 4.23 (1.53-19.58); P = 0.02] and presence of basal core promoter mutants [odds ratio: 2.93 (1.24-7.57); P = 0.02] were independent predictors for progression to hepatocellular carcinoma.CONCLUSION: Our results show that high levels of baseline serum HBV DNA are associated with non- hepatocellular carcinoma-related deaths of liver failure, while genetic mutations in the basal core promoter and precore regions are predictive for development of hepatocellular carcinoma. | Myron J Tong Lawrence M Blatt Jia-Horng Kao Jason Tzuying Cheng William G Corey | 2006 | World Journal of Gastroenterology2006，12，41： | 11 |
| 3 | Juvenile hominoid cranium from the terminal Miocene of Yunnan,China显示文摘Fossil apes are known from several late Miocene localities in Yunnan Province,southwestern China,principally from Shihuiba(Lufeng)and the Yuanmou Basin,and represent three species of Lufengpithecus.They mostly comprise large samples of isolated teeth,but there are also several partial or complete adult crania from Shihuiba and a single juvenile cranium from Yuanmou.Here we describe a new,relatively complete and largely undistorted juvenile cranium from the terminal Miocene locality of Shuitangba,also in Yunnan.It is only the second ape juvenile cranium recovered from the Miocene of Eurasia and it is provisionally assigned to the species present at Shihuiba,Lufengpithecus lufengensis.Lufengpithecus has most often been linked to the extant orangutan,Pongo pygmaeus,but recent studies of the crania from Shihuiba and Yuanmou have demonstrated that this is unlikely.The new cranium reinforces the view that Lufengpithecus represents a distinct,late surviving lineage of large apes in the late Miocene of East Asia that does not appear to be closely affiliated with any extant ape lineage.It substantially increases knowledge of cranial morphology in Lufengpithecus and demonstrates that species of this genus represent a morphologically diverse radiation of apes,which is consistent with the dynamic tectonic and biotic milieu of southwestern China in the late Miocene. | JI XuePing JABLONSKI Nina G SU Denise F DENG ChengLong FLYNN Lawrence J YOU YouShan KELLEY Jay | 2013 | Chinese Science Bulletin2013，58，31： | 8 |
| 4 | ERCC2诱导表达在人脑胶质瘤细胞对BCNU耐药中的作用显示文摘目的 探讨BCNU诱导ERCC2表达在胶质瘤细胞耐药中的作用。方法 选用ERCC2基础表达在蛋白水平相同 ,而mRNA水平不同的 2株人脑胶质瘤细胞T98 G和SK MG 4(在mRNA水平 ,T98 G比SK MG 4高 2 5倍 ) ,在体外培养时用BCNU处理 ,随后采用WesternBlot测定肿瘤细胞的ERCC2蛋白水平变化。结果 对BCNU耐药的T98 G在BCNU处理后 6h ,ERCC2蛋白水平上升 6倍 ,随后逐渐下降 ,48h回复到基础水平。而对BCNU敏感的SK MG 4,BCNU处理后ERCC2蛋白水平没有增加。结论 本研究提示 ,mRNA基础水平与胶质瘤耐药密切相关 ,在mRNA基础水平较高的肿瘤细胞 ,BCNU能诱导ERCC2蛋白表达 ,并直接与肿瘤耐药相关。 | 陈忠平 Grard Mohr Lawrence C Panasci | 2003 | 中华神经外科杂志2003，19，1： | 5 |

| 7 | 转染ERCC2的人脑胶质瘤细胞增加对烷化剂类抗癌药耐药性显示文摘目的 探讨切割修复交叉互补基因 2 (excisionrepaircross complementingrodentrepairdeficiencygene 2 ,ERCC2 )在人脑胶质瘤耐药中的作用。方法 采用来自p2E ER2的 2 4kbEcoRⅠ片段和pcDNA3载体组建成pcDNA3ERCC2质粒。再用脂质体转染技术 ,将pcDNA3ERCC2质粒转入SKMG 4细胞。转染ERCC2的细胞用G41 8连续筛选 2周。再从存活的细胞集落挑取单细胞进行克隆。克隆细胞进一步增殖并经RT PCR和Westernblot证实为稳定转染细胞用于下一步试验。转染不带ERCC2的pcDNA3载体作为对照。然后 ,采用改良的SulforhodamineB(SRB)抗癌药筛选法 ,对稳定转染和亲代SKMG 4细胞进行细胞毒试验。结果 细胞毒性试验结果显示转染ERCC2的SKMG 4胶质瘤细胞对顺铂和麻法林的耐药性明显增加。顺铂的IC90 ( μmol/L)在亲代SKMG 4、转染pcDNA3载体的对照SKMG 4和转染ERCC2的SKMG 4细胞分别为 1 1 75、1和 1 75 ;麻法林的IC90 ( μmol/L)分别为 5、5 6和 9。转染ERCC2的SKMG 4细胞与转染空载体的对照细胞相比较 ,顺铂和麻法林的IC90 分别增加了 75 %和 61 %。结论 转染ERCC2基因的胶质瘤细胞对顺铂和麻法林的耐药性明显增加 ,证明ERCC2在烷化剂类抗癌药物耐药中起重要作用。 | 陈忠平 许志元 Grard mohr Lawrence C.Panasci | 2002 | 中国神经精神疾病杂志2002，28，5： | 2 |
