1Fused-core particle technology in high-performance liquid chromatography: An overview显示文摘The advent of superfcially porous particles(SPPs)for packed HPLC columns has changed the way that many practitioners have approached the problem of developing needed separations.The very high effciency of such columns,combined with convenient operating conditions,modest back pressures and the ability to use conventional HPLC instruments has resulted in intense basic studies of SPP technology,and widespread applications in many sciences.This report contains an overview of the SPP technology frst developed in 2006 by Advanced Materials Technology,Inc.,for sub-3-mm particles,then expanded into a family of SPP products with different particle sizes,pore sizes and other physical parameters.This approach was designed so that each particle of the family could be optimized for separating a particular group of compounds,usually based on solute size.Joseph J.Kirkland Stephanie A.Schuster William L.Johnson Barry E.Boyes 2013Journal of Pharmaceutical Analysis2013,3,5:6
2Compact and robust supercontinuum generation and post-compression using multiple thin plates显示文摘We report on compact and robust supercontinuum generation and post-compression using transmission of light through multiple thin solid plates at the Swiss FEL X-ray free-electron laser facility.A single stage consisting of three thin plates followed by a chirped mirror compressor achieves compression of initially 30-fs pulses with 800-nm center wavelength to sub-10-fs duration.We also demonstrate a two-stage implementation to compress the pulses further to sub-5-fs duration.With the two-stage setup,the generated supercontinuum includes wavelengths ranging from 500 to 1100 nm.The multiplate setup is compact,robust,and stable,which makes it ideal for applications at free-electron laser facilities such as pump-probe experiments and laser-arrival timing tools.Xinhua Xie Yunpei Deng Steven L.Johnson 2021High Power Laser Science and Engineering2021,9,4:3
3Hippo signaling and epithelial cell plasticity in mammalian liver development, homeostasis, injury and disease显示文摘A traditional view of cellular differentiation is unidirectional: progenitor cells adopt specific fates in response to environmental cues resulting in deployment of cell-specific gene expression programs and acquisition of unique differentiated cellular properties such as production of structural and functional proteins that define individual cell types. In both development and in tissue repair stem and progenitor cells are thought to both self-renew to maintain the pool of precursors and to expand to give rise to transient amplifying and differentiated cell types. Recently, however, it has become appreciated that differentiated cell types can be reprogrammed to adopt progenitor and stem cell properties. In the case of epithelial cells in the mammalian liver, hepatocytes and biliary epithelial cells there is a significant degree of plasticity between these lineages that has been implicated in mechanisms of tissue repair and in liver pathologies such as cancer. Recent studies have highlighted the role of Hippo signaling, an emerging growth control and tumor suppressor pathway, in regulating epithelial cell plasticity in the mammalian liver and in this review, the role of cellular plasticity and Hippo signaling in regulating normal and abnormal tissue responses in the mammalian liver will be discussed.Randy L.Johnson 2019Science China(Life Sciences)2019,62,12:3
4Plant glycosylphosphatidylinositol anchored proteins at the plasma membrane-cell wall nexus显示文摘Approximately 1% of plant proteins are predicted to be post-translationally modified with a glycosylphosphatidylinositol(GPI) anchor that tethers the polypeptide to the outer leaflet of the plasma membrane. Whereas the synthesis and structure of GPI anchors is largely conserved across eukaryotes, the repertoire of functional domains present in the GPI-anchored proteome has diverged substantially. In plants, this includes a large fraction of the GPI-anchored proteome being further modified with plant-specific arabinogalactan(AG) O-glycans. The impor tance of the GPI-anchored proteome to plant development is underscored by the fact that GPI biosynthetic null mutants exhibit embryo lethality. Mutations in genes encoding specific GPI-anchored proteins(GAPs) further supports their contribution to diverse biological processes, occurring at the interface of the plasma membrane and cell wall including signaling, cell wall metabolism, cell wall polymer cross-linking, and plasmodesmatal transport. 5大学英语教师对课堂教学测试中道德问题的认知水平研究显示文摘本文采用定量研究方法,探究大学英语教师在课堂教学测试中对道德问题的认知水平,所用问卷包含大学英语教学测试中15个典型情景,共344名大学英语教师参加了调查。研究者用SPSS软件对调查数据进行了分析处理。研究发现,大学英语教师对6个测试情景中所涉及的道德问题认知水平较高;对另外6个情景中的道德问题认知水平一般;而对3个情景中的道德问题认知水平非常低。其中大学英语教师在测试保密性、成绩评定和测试实施三个维度的职业道德建设需要引起高度关注。研究结果启示大学英语教师应重视教学测试中的道德问题,保证教学测试的公正性与客观性。研究结果同时为高校教师职业发展提供有益借鉴。刘秀梅 樊旭梅 Robert L.Johnson 2018外语教学2018,39,4:3

8大学英语课堂教学测试中教师道德观念影响因素研究显示文摘本文用质性和量化相结合的研究方法研究大学英语课堂教学测试中影响教师道德观念的因素。来自全国143所高校的344名大学英语教师参加了问卷调查。问卷包括15个大学英语课堂教学测试中的典型情景,受访者对情景中教师做法是否符合职业道德规范做出判断,并就判断理由进行文字说明。研究采用专家对各情景的判断意见作为权威参照,根据受访者与专家意见的一致程度得出受访者对课堂教学测试中职业道德的整体认知水平。通过多元回归计算揭示教学测试中教师道德观念认知水平与教师性别、教龄、职称、对教学测试的态度以及是否对职业培训感兴趣之间的关系。同时,该研究还对受访者所给出的判断理由进行主题编码,用质性分析法剖析课堂教学测试中影响教师职业道德认知水平的主要因素。研究结论有助于提高教学测试的公正性,为高校教师职业发展提供借鉴,更好地发挥测试对课堂教学和学生发展的积极作用。刘秀梅 樊旭梅 Robert L.Johnson 2018外语电化教学2018,,1:2
9Social-ecological correlates of fundamental movement skills in young children显示文摘Purpose: To identify the social-ecological correlates associated with fundamental movement skills at the child, family, and environment levels in young children.Methods: Preschool children from 4 Colorado Head Start/preschool centers were recruited from 2010 to 2012. Two hundred twenty-eight children(128 girls; age = 56.08 § 4.09 months; body mass index(BMI) z-score = 0.53 § 1.12(mean § SD); 42.1% Hispanic/Latino) and 159 families were included in the final analysis. Children's perceived competence and fundamental movement skills were assessed via the Pictorial Scale of Perceived Competence and Social Acceptance and the Bruininks-Oseretsky Test of Motor Proficiency, 2 nd edition. Data on the number of children in the family, parent age, BMI, education, employment status, family income, perception of child coordination, and home physical activity environment were collected via a questionnaire. Linear regressions adjusted for child BMI, age, sex, and school site were performed at each level.Results: Child perceived cognitive competence was positively associated with locomotor skills(p = 0.04; adjusted R2= 0.035) and object-control skills(p = 0.003; adjusted R2= 0.083) at the child level. Parent education, BMI, and perception of child coordination were positively associated with locomotor skills and explained 8.8% of variance, but only parent education was significant(p = 0.04) at the family level. 13Characterization of a novel murine Sost ERT2 Cre model targeting osteocytes显示文摘Transgenic mice are widely used to delete or overexpress genes in a cell specific manner to advance knowledge of bone biology,function and disease.While numerous Cre models exist to target gene recombination in osteoblasts and osteoclasts,few target osteocytes specifically,particularly mature osteocytes.Our goal was to create a spatial and temporal conditional Cre model using tamoxifen to induce Cre activity in mature osteocytes using a Bac construct containing the 5’and 3’regions of the Sost gene(Sost ERT2 Cre).Four founder lines were crossed with the Ai9 Cre reporter mice.One founder line showed high and specific activity in mature osteocytes.Bones and organs were imaged and fluorescent signal quantitated.While no activity was observed in 2 day old pups,by 2 months of age some osteocytes were positive as osteocyte Cre activity became spontaneous or‘leaky’with age.The percentage of positive osteocytes increased following tamoxifen injection,especially in males,with 43%to 95%positive cells compared to 19%to 32%in females.No signal was observed in any bone surface cell,bone marrow,nor in muscle with or without tamoxifen injection.No spontaneous signal was observed in any other organ.However,with tamoxifen injection,a few positive cells were observed in kidney,eye,lung,heart and brain.All other organs,28 in total,were negative with tamoxifen injection.However,with age,a muscle phenotype was apparent in the Sost-ERT2 Cre mice.Therefore,although this mouse model may be useful for targeting gene deletion or expression to mature osteocytes,the muscle phenotype may restrict the use of this model to specific applications and should be considered when interpreting data.Delphine B.Maurel Tsutomu Matsumoto Julian A.Vallejo Mark L.Johnson Sarah L.Dallas Yukiko Kitase Marco Brotto Michael J.Wacker Marie A.Harris Stephen E.Harris Lynda F.Bonewald 2019Bone Research2019,7,1:1
