| 1 | 我国首次分离到基因Ⅰ型乙型脑炎病毒显示文摘目的 对新分离乙型脑炎病毒鉴定 ,了解病毒E基因区段的氨基酸序列特征及基因分型。方法 2 0 0 1年在上海市采集蚊虫标本 ,用组织培养的方法分离到 7株病毒。进行病毒一般生物学鉴定 ,对病毒PrM和E基因区段约 2 0 0 0nt进行了扩增、克隆、测序。应用ClustalX软件做碱基配对和比较分析 ,种系发生采用PHYLIP软件包分析。结果 7株病毒可以引起BHK 2 1细胞规律病变 ,病变时间为 6 0h。乳鼠脑内接种病毒后 72h死亡。所有新分离病毒与标准乙型脑炎病毒抗体阳性反应。用病毒PrM区段 (4 5 6~ 6 95位核苷酸 )进行的基因分型分析 ,新分离的 7株病毒属于基因Ⅰ型乙型脑炎病毒。以减毒活疫苗株SA14 14 2为标准 ,对 7株病毒的E基因区段进行分析 ,7株病毒之间核苷酸同源性在 97.7%以上 ,氨基酸同源性在 99.2 %以上 ;7株病毒与疫苗株SA14 14 2核苷酸差异在 12 .2 %左右 ,氨基酸差异在 2 .8%~ 3.2 %之间 ,共有 14个共同的氨基酸位点差异。结论 2 0 0 1年在上海采集的蚊虫中新分离的 7株病毒属于基因Ⅰ型乙型脑炎病毒 。 | 王环宇 付士红 李晓宇 宋宏 闵继光 邓娟 杨益良 Ichiro Kurane 梁国栋 | 2004 | 中华微生物学和免疫学杂志2004,24,11: | 79 |
| 2 | 中国流行性乙型脑炎病毒分子生物学特性研究显示文摘为了从分子水平了解中国流行性乙型脑炎(乙脑)病毒与国外分离株的差异,对1949年以来在中国乙脑主要流行地区分离的19株乙脑病毒的PrM C区及E蛋白基因区的核苷酸序列进行了对比研究,以期了解不同时间不同地区在中国流行的乙脑病毒的分子差异。结果显示:病毒生物学初步鉴定显示,病毒感染BHK细胞后56h所有细胞均发生病变,约50%以上细胞脱落(CPE );3日龄乳鼠接种病毒72h之内死亡;所有病毒均与乙脑病毒(A2株)抗血清发生阳性反应,但反应强度不同,提示所有病毒均为乙脑病毒。病毒PrM C(456~695)区核苷酸序列与各个国家及地区、各个基因型以及各个年代的73株乙脑病毒相应序列,用Woan RuChen建立的方法进行基因分型分析,结果显示,所有19株中国分离的病毒均属于基因型Ⅲ型,与国外相关文献所报道的乙型脑炎病毒中国分离株的基因型相符。以乙型脑炎病毒疫苗株P3株为标准,对各病毒E蛋白500个氨基酸序列进行分析。各毒株核苷酸差异在0和4 2%之间,氨基酸差异在0和4 0%之间。所有核苷酸差异均为碱基的替换,无论核苷酸或氨基酸均无插入或丢失。大多数核苷酸变异在氨基酸编码的第三位,属于沉默突变,不引起氨基酸变化。18株病毒E蛋白活性结构域与疫苗株P3株相比共有247个氨基酸的差异,平均每株病毒有12 35个氨基酸的差异,? | 李晓宇 宋宏 付士红 王环宇 俞永新 董关木 陶三菊 陈端 Ichiro Kurane 梁国栋 | 2004 | 病毒学报2004,20,3: | 31 |
| 3 | 新疆出血热病毒分子流行病学研究显示文摘目的研究新疆出血热病毒(XHF)分子流行病学,揭示其与相关病毒之间关系,分析XHF的流行来源和分布特点。方法RTPCR检测20012002年XHF患者和蜱标本中XHF病毒S基因,阳性标本直接进行核苷酸序列测定。计算机软件进行S基因部分片段和S全基因序列同源性比较和S基因、M基因的种系发生树分析。结果不同年份人和蜱来源的病毒S基因部分片段核苷酸序列均显示较高同源性(97.3%~100%)。S基因进化树分析将病毒分成了欧洲、非洲和亚洲3组,其中亚洲毒株由中亚分离株和中国分离株组成,中国所有的XHF病毒S基因(同源性93.0%~99.5%)均集中在一个分枝下形成独立的一组,明显区别于世界上其他地区分离株(同源性81.40%~96.4%)。M基因进化树分析表明序列间差异不完全与病毒分离的地理区域相关。结论S基因分析显示XHF病毒蜱分离株与人分离株遗传背景接近,我国XHF病毒有共同的进化途径和基因结构特点,并具明显的地域性。 | 唐青 赵秀芹 王环宇 司马义巴吾东 张玉贞 Masayuki Saijo Shigeru Morikawa 梁国栋 Ichiro Kurane | 2005 | 中华实验和临床病毒学杂志2005,19,4: | 11 |
| 4 | Manometric findings in patients with isolated distal gastroesophageal reflux显示文摘AIM:To analyze manometric abnormalities in patients with isolated distal reflux and compare these findings in patients with erosive and non-erosive disease. METHODS:Five hundred and fifty patients who presented to the outpatient clinic of Turkiye Yuksek Ihtisas Hospital with gastroesophageal reflux disease-like symptoms were enrolled.Each individual was evaluated with esophageal manometry,24-h ambulatory pH monitoring,and upper gastrointestinal endoscopy. Manometric findings for the patients with isolated distal reflux were compared to findings in controls who were free of reflux disorders or hypersensitive esopha- gus.Findings for isolated distal reflux patients with and without erosive reflux disease were also compared. RESULTS:Of the 550 subjects enrolled,97(17.6%, mean age 48 years)had isolated distal reflux and 100 had no abnormalities on ambulatory pH monitoring (control group,mean age 45 years).There were no significant differences between the isolated distal reflux group and control group with respect to age,body mass index,and esophageal body contraction amplitude (EBCA).Mean lower esophageal sphincter pressure was significantly higher in the control group(12.7±10.3 mmHg vs 9.6±7.4 mmHg,P=0.01).Fifty-five(56.7%) of the 97 patients with isolated distal reflux had erosive reflux disease.There were no statistical differences be- tween the erosive reflux disease and non-erosive reflux disease subgroups with respect to mean EBCA,lower esophageal sphincter pressure,or DeMeester score.However,13%of patients with gastroesophageal reflux disease had distal wave amplitudes≤30 mmHg, whereas none of the patients with non-erosive reflux disease had distal wave amplitudes in this low category. CONCLUSION:Patients with erosive and non-erosive disease present with similar manometric abnormalities. | 5 | 我国新分离乙脑病毒02-76株的全基因序列特征显示文摘目的对我国新分离乙脑病毒02-76株进行全基因序列测定和分析,了解乙脑病毒基因组结构及毒力特性。方法设计乙脑病毒全基因组扩增引物,RT-PCR扩增片段,PCR产物直接测序,拼接后获得全基因序列。通过Clustal X(1.8)、DNASTAR、GENEDOC(3.2)等生物学软件进行核苷酸序列及氨基酸序列分析和病毒的系统进化分析。结果新分离乙脑病毒02.76株全基因组全长10977个核苷酸,从96位到10391位,共10296个核苷酸编码一个开放阅读框,编码3432个氨基酸。与我国1949年分离的Beijing-1株相比较共存在248个核苷酸差异,16个氨基酸差异。与GenBank中选择的29株乙脑病毒全基因序列比较发现,其核苷酸总体差异率为0.6%-15.1%,氨基酸总体差异率为0.2%-4.6%。通过PrM/C区段、E区段、3’NTR区段及全基因序列进行系统进化分析均显示该毒株属于基因3型乙脑病毒。结论新分离的乙脑病毒02-76株属于基因3型,与中国分离株SA-14进化关系最接近。 | 王环宇 潘晓玲 付士红 王雷 李铭华 Ichiro Kurane 梁国栋 | 2006 | 中华实验和临床病毒学杂志2006,20,3: | 6 |
| 6 | Cerium dioxide as a new reactive sorbent for fast degradation of parathion methyl and some other organophosphates显示文摘Cerium dioxide was used for the first time as reactive sorbent for the degradation of the organophosphate pesticides parathion methyl,chlorpyrifos,dichlofenthion,fenchlorphos,and prothiofos,as well as of some chemical warfare agents—nerve gases soman and O-ethyl S-[2-(diisopropylamino) ethyl] methylphosphonothioate(VX).CeO2 specimens were prepared by calcination of basic cerous carbonate obtained by precipitation from an aqueous solution.The CeO2 samples containing certain amounts(1 wt.%–5 wt.%) of the neighboring lanthanides(La,Pr,Nd) were prepared in a similar way from pure lanthanide salts.It was shown that ceria accelerated markedly the decomposition of parathion methyl causing the cleavage of the P-O-aryl bond in the pesticide molecule.A similar reaction mechanism was proposed for the degradation of other organophosphate pesticides and nerve agents.The degradation times(reaction half-times) were in an order of minutes in the presence of CeO2,compared to hours or days under common environmental conditions.The reaction in suitable organic solvents allowed conversions of about 90% for parathion methyl loading of 20 mg pesticide/g CeO2 within 2 h with a reactant half-life in the order of 0.1 min.The key parameter governing the degradation efficiency of CeO2 was the temperature during calcination.At optimum calcination temperature(about 773.15 K),the produced ceria retained a sufficiently high surface area,and attained an optimum degree of crystallinity(related to a number of crystal defects,and thus potential reactive sites).The presence of other lanthanides somewhat decreased the reaction rate,but this effect was not detrimental and permitted the possible use of chemically impure ceria as a reactive sorbent.A fast organophosphate degradation was demonstrated not only in non-polar solvents(such as heptane),but also in polar aprotic solvents(acetonitrile,acetone) that are miscible with water.This opens new possibilities for designing more versatile decontamination strategies.The cleavage of phosphate ester bonds is of a great importance not only for the degradation of dangerous chemicals(chemical weapons,pesticides),but also for interactions of ceria(especially the nano-sized one) in biologically relevant systems. | Pavel Janos Pavel Kuran Martin Kormunda Vaclav Stengl Tomas Matys Grygar Marek Dosek Martin Stastny Jakub Ederer Vera Pilarova Lubos Vrtoch | 2014 | Journal of Rare Earths2014,32,4: | 4 |
