1An experiment-library least-squares method on on-line coal element contents analysis显示文摘A new experiment-library least-squares(experiment-LLs) method used in the neutron inelastic-scattering and thermal-capture analysis(NITA) technique for on-line coal analysis was developed,which has significantly decreased the non-linear radiation effects.In this method,sixty samples with preset elemental contents were made,from whose experimental spectra twenty single-element spectrum libraries corresponding to twenty kinds of coal were built using the least-squares method.The spectrum of the unknown sample was analyzed based on these twenty libraries to estimate its element contents.With the initial estimated result,the procedure of developing library and analysis was iterated to build a new library with the closest element contents to the unknown sample.Hence the experiment-LLs method can reduce non-linear radiation effects.The experiment-LLs method was performed on an improved coal analysis system which was equipped with a long-life 14 MeV pulsed-neutron generator,a bulk BGO detector with a temperature controller,a moisture meter and coal-smoothing device.The precisions of this system for ash content,water content,volatile content and calorific value have reached 1.0wt%,0.5wt%,1.0wt%,350 kJ/kg,respectively.WANG HuiDong LU JingBin LU YuPing YANG Dong MA KeYan YANG Kang LIU YuMin CHENG DaoWen 2012Science China(Physics,Mechanics & Astronomy)2012,55,11:6
2Cloning and characterization of Methoprene-tolerant(Met)and Kruppel homolog 1(Kr-h1)genes in the wheat blossom midge,Sitodiplosis mosellana显示文摘Juvenile hormone(JH),a growth regulator,inhibits ecdysteroid-induced meta-morphosis and controls insect development and diapause.Methoprene-tolerant(Met)and Krippel homolog I(Kr-h1)are two proteins involved in JH action.To gain some insight into their function in development of Sitodiplosis mosellana,an insect pest undergoing obligatory larval diapause at the mature 3rd instar stage,we cloned full-length complemen-tary DNAs of Met and Kr-h1 from this specics.SmMet encoded a putative protein,which contained three domains typical of the bHLH-PAS family and eight conserved amino acid residues important for JH binding.SmKr-h1 encoded a protein showing high sequence homology to its counterparts in other specics,and contained all eight highly conserved Zn-finger motifs for DNA-binding.Expression patterns of SmMet and SmKr hl were de-velopmentally regulated and JH III responsive as well.Their mRNA abundance increased as larvae entered carly 3rd instar,pre-diapause and maintenance stages,and peaked during post-diapause quiescence,a pattern correlated with JH titers in this species.Different from reduced expression of SmMer,SmKr-h1 mRNA increased at mid-to-late period of post-diapause development.Topical application of JH II on diapausing larvac also induced the two genes in a dose-dependent manner.Expression of SmuMer and SmKr-h1 clearly declined in the pre-pupal phase,and was significantly higher in female adults than male adults.These results suggest that JH-responsive SmMet and SmKr-h1 might play key roles in diapause induction and maintenance as well as in post-diapause quiescence and adult reproduction,whereas metamorphosis from larvae to pupac might be correlated with their reduced expression.Wei-Ning Cheng Xue-Jiao Li Jia-Jia Zhao Keyan Zhu-Salzman 2020Insect Science2020,27,2:4

5miRNA-141-5p Affects the Levels of Neutrophil Elastase in Preeclampsia by Regulating MAPK1显示文摘Objective:The objective of this study was to investigate the expression levels of microRNA-141-5p(miRNA-141-5p),MAPK1 and neutrophil elastase in patients with and without preeclampsia(PE),and the relationship between miRNA-141-5p and MAPK1 with respect to the secretion of elastase by neutrophils in patients with PE.Methods:Thirty patients with PE and 30 healthy pregnant(HP)women were recruited from The Second Hospital of Shanxi Medical University,Taiyuan,China,between February 2017 and July 2018.Neutrophils were isolated from 8 mL peripheral blood samples and cultured.We recorded neutrophil count and morphology during culture.Apoptosis was detected by flow cytometry in different groups at 0,24,and 48 h.The expression levels of elastase were detected in neutrophils by enzyme-linked immunosorbent assay,whereas the expression levels of miRNA-141-5p in peripheral blood neutrophils were detected by real-time polymerase chain reaction.We used TargetScanHuman Release 7.2 to analyze the target genes of miRNA-141-5p.The expression of MAPK1 in peripheral blood neutrophils was detected by western blotting.Data were analyzed by SPSS version 21.0 software,and comparisons between groups were carried out with the Studentt test.Results:There was no significant difference between the PE and HP groups(P>0.050)with regard to age or body mass index.The weight of newborns in the PE group(2846.00±600.00 g)was significantly lower than that in the HP group(3055.00±230.68 g).The number of neutrophilic granulocytes(NGs)in blood samples from the PE group was significantly higher than that in the HP group(P=0.003).There was no significant difference between the groups with regard to morphology.Apoptosis in the PE group was delayed when compared with the HP group at different time points.TheP value of apoptosis in the PE and HP groups were respectively 0.790,<0.001 and 0.030 at 0 h,24 h and 48 h.The expression levels of miRNA-141-5p in the PE group were significantly lower than those in the HP group(P<0.050).The expression levels of MAPK1 in neutrophils from the PE group were significantly higher than those in the HP group(P<0.050)by western blot.The expression levels of elastase in neutrophils from the PE group were significantly higher than those in the HP group(P<0.050).Furthermore,the number of NGs in peripheral blood from the PE group was higher than that of the HP group;however,the levels of apoptosis were lower.The expression levels of miRNA-141-5p in NGs decreased,the expression of MAPK1 increased,and the secretion of neutrophil elastase in the NG medium increased in the PE group than those in the HP group.Conclusion:Collectively,our analysis suggested that miRNA-141-5p may be involved in the pathogenesis of PE by regulating the MAPK1 signaling pathway to activate neutrophils and increase the secretion of elastase.Keyan Cheng Jialei Cui Wenli Zhou Huiqiang Liu Taotao Yang Yonghong Wang 2022Maternal-Fetal Medicine2022,4,4:0
6Deleterious effects of electron beam irradiation on development and reproduction of tomato/potato psyllids, Bactericera cockerelli显示文摘The potato/tomato psyllid Bactericera cockerelli causes serious damage to several solanaceous crops by direct feeding and vectoring Candidatus Liberibacter solanacearum,a bacterial pathogen.Electron beam(eBeam)irradiation is an environmentally friendly,chemical-free alternative method that is increasing in use for disinfestation of insect pests.We hypothesize that this irradiation technology will have detrimental effects on potato psyllid and thus impede its disease vectoring.To this end,we explored the effects of eBeam treatment ranging from 50 to 500 Gy on survival,development and reproduction of this pest.Impact on psyllids was apparently dose-dependent.When irradiated at 350 Gy,eggs could not hatch,1st instar nymphs failed to emerge,and although a small portion of irradiated 5th instar nymphs survived,the emerged adults were mostly deformed.Abnormality in eclosed adults suggests harmful effects of eBeam on metamorphosis.Reproduction was seriously impaired when female psyllids were exposed to eBeam at the 5th instar nymphal or young adult stage,presumably due to inability to form oocytes.In addition,reciprocal crosses between irradiated and untreated psyllids indicated that female psyllids were more radiosensitive than males to eBeam.Taken together,these findings indicate that eBeam negatively impacted potato psyllid development and reproduction,which would inevitably compromise its disease transmission capacity.A dose of 350 Gy can be considered as a reference dose for effective control of potato psyllids.Jiaxin Lei Jia Meng Ivy WChen Weining Cheng Andrea L.Beam Md-Sajedul Islam Woodward D.Bailey Suresh Pillai Keyan Zhu-Salzman 2020Insect Science2020,27,6:0
7Molecular and functional characterization of three odorant-binding proteins from the wheat blossom midge,Sitodiplosis mosellana显示文摘Sitodiplosis mosellana,a periodic but devastating wheat pest,relies on wheat spike volatiles as a cue in sclecing hosts for oviposition.Insect odorant-binding proteins(OBPs)are thought to play essential roles in filtering,binding and transporting hydropho-bic odorant molecules to specific receptors.To date,the molecular mechanisms underlying S.mosellana olfaction are poorly understood.Here,three S.mosellana antenna-specific OBP genes,SmosOBPII,16 and 21,were cloned and bacterially expressed.Binding properties of the recombinant proteins to 28 volatiles emitted from wheat spikes were in-vestigated using fluorescence competitive binding assays.Sequence analysis suggested that these SmosOBPs belong to the Classic OBP subfamily.Ligand-binding analysis showed that all three SmosOBPs preferentially bound alcohol,ester and ketone com-pounds,and SmosOBP11 and 16 also selectively bound terpenoid compounds.In par-ticular,the three SmosOBPs had high binding affinities(Ki<20μmol/L)to 3-hexanol and cis-3-hexenylacetate that elicited strong electroantennogram(EAG)response fromfemale antennae.In addition,SmosOBP11 displayed significantly higher binding(Ki<8μmo/L)than SmosOBP16 and 21 to l-octen-3-ol,D-panthenol,a-pinene and heptyl acetate which elicited significant EAG response,suggesting that SmosOBP11 plays a ma-jor role in recognition and transportation of these volatiles.These findings have provided important insight into the molecular mechanism by which S.mosellana specifically rec-ognizes plant volatiles for host selection,and have facilitated identification of effective volatile attractants that are potentially useful for pest monitoring and trapping.Wei-Ning Cheng Yu-Dong Zhang Wei Liu Guang-Wei Li Keyan Zhu-Salzman 2020Insect Science2020,27,4:0
