| 1 | Methylprednisolone for acute spinal cord injury: an increasingly philosophical debate显示文摘Following publication of NASCIS Ⅱ, methylprednisolone sodium succinate(MPSS) was hailed as a breakthrough for patients with acute spinal cord injury(SCI). MPSS use for SCI has since become very controversial and it is our opinion that additional evidence is unlikely to break the stalemate amongst clinicians. Patient opinion has the potential to break this stalemate and we review our recent findings which reported that spinal cord injured patients informed of the risks and benefits of MPSS reported a preference for MPSS administration. We discuss the implications of the current MPSS debate on translational research and seek to address some misconceptions which have evolved. As science has failed to resolve the MPSS debate we argue that the debate is an increasingly philosophical one. We question whether SCI might be viewed as a serious condition like cancer where serious side effects of therapeutics are tolerated even when benefits may be small. We also draw attention to the similarity between the side effects of MPSS and isotretinoin which is prescribed for the cosmetic disorder acne vulgaris. | 2 | Identification and expression profiling of Vigna mungo microRNAs from leaf small RNA transcriptome by deep sequencing显示文摘MicroRNAs(miRNAs) represent a class of small non-coding RNA molecules that play a crucial role in post-transcriptional gene regulation. Several conserved and species-specific miRNAs have been characterized to date, predominantly from the plant species whose genome is well characterized. However, information on the variability of these regulatory RNAs in economically important but genetically less characterized crop species are limited. Vigna mungo is an important grain legume, which is grown primarily for its protein-rich edible seeds. miRNAs from this species have not been identified to date due to lack of genome sequence information. To identify miRNAs from V. mungo, a small RNA library was constructed from young leaves. High-throughput Illumina sequencing technology and bioinformatic analysis of the small RNA reads led to the identification of 66 miRNA loci represented by 45 conserved miRNAs belonging to 19 families and eight non-conserved miRNAs belonging to seven families. Besides, 13 novel miRNAcandidates in V. mungo were also identified. Expression patterns of selected conserved, non-conserved, and novel miRNA candidates have been demonstrated in leaf, stem, and root tissues by quantitative polymerase chain reaction, and potential target genes were predicted for most of the conserved miRNAs. | 3 | Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers. | Feng Li Muthu K. | 4 | Wheat streak mosaic virus: incidence in field crops, potential reservoir within grass species and uptake in winter wheat cultivars显示文摘Wheat streak mosaic virus(WSMV)has become a re-emerging pathogen in cereal crops in the Czech Republic.WSMV was first reported in the former Czechoslovakia in the early 1980s,and then no record of the virus was documented until 2009.The incidence of the virus was recorded in recent years in several winter wheat fields and many grass species.Here,we surveyed the incidence of WSMV in cereal crops.The results demonstrated the existence of the virus in winter wheat and volunteer wheat during each year of the monitoring period,which spanned from 2013–2016.Although the range of infected samples was low(6.4%of the total tested samples),a high incidence of well-distributed virus was recorded.In at least six fields,the virus reached severe and potentially epidemic levels.In accordance with our previous report detailing WSMV infection of native grasses,we tested several grass species commonly grown in the Czech Republic.We found that some grass species acted as experimental hosts and possible reservoirs of the virus;these included Anthoxanthum odoratum(sweet vernal grass),Arrhenatherum elatius(false oat-grass),Lolium multiflorum(Italian rye-grass),Bromus japonicus(Japanese chess),Echinochloa crus-galli(barnyard grass),Holcus lanatus(meadow soft grass)and Holcus mollis(creeping soft grass).Some of these grass species are also important weeds of cereals,which may be the potential source of WSMV infection in cereal crops.Several widely used winter wheat cultivars were tested in the field after artificial inoculation with WSMV to evaluate virus titre by RT-qPCR.Overall,the tested cultivars had a low virus titre,which is associated with mild disease symptoms and may provide a good level of crop resistance to WSMV. | Jana Chalupniková Jiban Kumar Kundu Khushwant Singh Pavla Bartaková Eva Beoni | 2017 | Journal of Integrative Agriculture2017,16,3: | 4 |
| 5 | 1,3-Bis(2-chloroethyl)-1-nitrosourea enhances the inhibitory effect of Resveratrol on 5-fluorouracil sensitive/resistant colon cancer cells显示文摘AIM:To study the mechanism of 5-fluorouracil(5-FU)resistance in colon cancer cells and to develop strategies for overcoming such resistance by combination treatment.METHODS:We established and characterized a 5-FU resistance(5-FU-R)cell line derived from continuous exposure(25μmol/L)to 5-FU for 20 wk in 5-FU sensitive HCT-116 cells.The proliferation and expression of different representative apoptosis and anti-apoptosis markers in 5-FU sensitive and 5-FU resistance cells were measured by the MTT assay and by Western blotting,respectively,after treatment with Resveratrol(Res)and/or 1,3-Bis(2-chloroethyl)-1-nitrosourea(BCNU).Apoptosis and cell cycle arrest was measured by 4',6'-diamidino-2-phenylindole hydrochloride staining and fluorescence-activated cell sorting analysis,respectively.The extent of DNA damage was measured by the Comet assay.We measured the visible changes in the DNA damage/repair cascade by Western blotting.RESULTS:The widely used chemotherapeutic agents BCNU and Res decreased the growth of 5-FU sensitive HCT-116 cells in a dose dependent manner.Combined application of BCNU and Res caused more apoptosis in5-FU sensitive cells in comparison to individual treatment.In addition,the combined application of BCNU and Res caused a significant decrease of major DNA base excision repair components in 5-FU sensitive cells.We established a 5-FU resistance cell line(5-FU-R)from 5-FU-sensitive HCT-116(mismatch repair deficient)cells that was not resistant to other chemotherapeutic agents(e.g.,BCNU,Res)except 5-FU.The 5-FU resistance of 5-FU-R cells was assessed by exposure to increasing concentrations of 5-FU followed by the MTT assay.There was no significant cell death noted in5-FU-R cells in comparison to 5-FU sensitive cells after5-FU treatment.This resistant cell line overexpressed anti-apoptotic[e.g.,AKT,nuclear factorκB,FLICE-like inhibitory protein),DNA repair(e.g.,DNA polymerase beta(POL-β),DNA polymerase eta(POLH),protein Flap endonuclease 1(FEN1),DNA damage-binding protein 2(DDB2)]and 5-FU-resistance proteins(thymidylate synthase)but under expressed pro-apoptotic proteins(e.g.,DAB2,CK1)in comparison to the parental cells.Increased genotoxicity and apoptosis were observed in resistant cells after combined application of BCNU and Res in comparison to untreated or parental cells.BCNU increased the sensitivity to Res of 5-FU resistant cells compared with parental cells.Fifty percent cell death were noted in parental cells when 18μmol/L of Res was associated with fixed concentration(20μmol/L)of BCNU,but a much lower concentration of Res(8μmol/L)was needed to achieve the same effect in 5-FU resistant cells.Interestingly,increased levels of adenomatous polyposis coli and decreased levels POL-β,POLH,FEN1 and DDB2 were noted after the same combined treatment in resistant cells.CONCLUSION:BCNU combined with Res exerts a synergistic effect that may prove useful for the treatment of colon cancer and to overcome drug resistance. | Dipon Das Ranjan Preet Purusottam Mohapatra Shakti Ranjan Satapathy Chanakya Nath Kundu | 2013 | World Journal of Gastroenterology2013,19,42: | 4 |
| 6 | Future changes in rainfall, temperature and reference evapotranspiration in the central India by least square support vector machine显示文摘Climate change affects the environment and natural resources immensely.Rainfall,temperature and evapotranspiration are major parameters of climate affecting changes in the environment.Evapotranspiration plays a key role in crop production and water balance of a region,one of the major parameters affected by climate change.The reference evapotranspiration or ETo is a calculated parameter used in this research.In the present study,changes in the future rainfall,minimum and maximum temperature,and ETo have been shown by downscaling the HadCM3(Hadley Centre Coupled Model version 3) model data.The selected study area is located in a part of the Narmada river basin area in Madhya Pradesh in central India.The downscaled outputs of projected rainfall,ET_0 and temperatures have been shown for the 21 st century with the HADCM3 data of A2 scenario by the Least Square Support Vector Machine(LS-SVM)model.The efficiency of the LS-SVM model was measured by different statistical methods.The selected predictors show considerable correlation with the rainfall and temperature and the application of this model has been done in a basin area which is an agriculture based region and is sensitive to the change of rainfall and temperature.Results showed an increase in the future rainfall,temperatures and ETo.The temperature increase is projected in the high rise of minimum temperature in winter time and the highest increase in maximum temperature is projected in the pre-monsoon season or from March to May.Highest increase is projected in the 2080 s in 2081-2091 and 2091-2099 in maximum temperature and 2091-2099 in minimum temperature in all the stations.Winter maximum temperature has been observed to have increased in the future.High rainfall is also observed with higher ETo in some decades.Two peaks of the increase are observed in ET_0 in the April-May and in the October.Variation in these parameters due to climate change might have an impact on the future water resource of the study area,which is mainly an agricultural based region,and will help in proper planning and management. | Sananda Kundu Deepak Khare Arun Mondal | 2017 | Geoscience Frontiers2017,8,3: | 4 |

| 14 | Apoptotic and autophagic death union by Thuja occidentalis homeopathic drug in cervical cancer cells with thujone as the bioactive principle显示文摘Objective‘Multi-targeting’drugs can prove fruitful to combat drug-resistance of multifactorial disease—cervical cancer.This study envisioned to reveal if Thuja homeopathic mother tincture(MT)and its bioactive component could combat human papillomavirus(HPV)-16-infected SiHa cervical cancer cells since it is globally acclaimed for HPV-mediated warts.Methods Thuja MT was studied for its antiproliferative and antimigratory properties in SiHa cells followed by microscopic determination of reactive oxygen species(ROS)generation by 2’,7’-dichlorodihydrofluorescein diacetate(DCFDA)staining and loss in mitochondrial membrane potential(MMP)by rhodamine(Rh)123 staining.Apoptosis and autophagy inductions were studied by acridine orange(AO)ethidium bromide(EB)staining and immunoblot analyses of marker proteins.The bioactive component of Thuja MT detected by gas chromatography-mass spectrometry was studied for antiproliferative and antimigratory properties along with in silico prediction of its cellular targets by molecular docking and oral drug forming competency.Results Thuja MT showed significant antiproliferative and antimigratory potential in SiHa cells at a 50%inhibitory concentration(IC50)dosage of 17.3μL/mL.An increase in DCFDA fluorescence and loss in Rh123 fluorescence prove that Thuja MT acted through the burst of ROS and loss in MMP respectively.AO/EB-stained cells under the microscope and immunoblot analyses supported Thuja-induced cellular demise via dual pathways—apoptosis and autophagy.Immunoblots showed cleavage of caspase-3 and poly adenosine diphosphate-ribose polymerase-1(PARP-1)along with upregulation of Beclin-1,microtubule-associated protein 1 light chain 3B(LC3B)-II,and p62 proteins.Hence,the apoptotic cascade followed a caspase-3-dependent pathway supported by PARP-1 cleavage,while autophagic death was Beclin-1-dependent and mediated by accumulation of LC3BII and p62 proteins.Thujone,detected as the bioactive principle of Thuja MT showed greater anti-proliferative and anti-migratory potential at an IC5077μg/mL along with excellent oral drug competency with the ability for gastrointestinal absorption and blood-brain-barrier permeation with nil toxicity.Molecular docking depicted thujone with the strongest affinity for mammalian target of rapamycin,phosphoinositide 3-kinase,and protein kinase B followed by B-cell lymphoma 2,murine double minute 2 and adenosine monophosphate-activated protein kinase,which might act as upstream triggers of apoptotic-autophagic crosstalk.Conclusion Robust‘multi-targeting’anticancer potential of Thuja drug and thujone for HPV-infected cervical cancer ascertain its therapeutic efficacy for HPV infections. | Asmita Pal Sucharita Das Soumalee Basu Rita Kundu | 2022 | Journal of Integrative Medicine2022,20,5: | 2 |

| 20 | Seismoelectric wave propagation modeling in a borehole in water-saturated porous medium having an electrochemical interface显示文摘Water-saturated porous media often exhibit a seismoelectric effect due to the existence of an electrical double layer and a relative flow of pore fluid. Here we consider the seismoelectric waves in an open borehole surrounded by water-saturated porous formation which exhibits discontinuity of electrochemical properties at a cylindrical interface. We carefully analyze the seismoelectric interface response since these signals show sensitivity to contrasts in electrochemical properties across an interface. Both coupled and approximate methods are used to compute borehole seismoelectric fields. The simulation results show that the radiated electromagnetic wave from the electrochemical interface is generated due to the change of salinity in pore fluid in the porous formation. However, the elastic properties of the formation remain unchanged across such an electrochemical interface. As a result it is difficult to recognize such a change in electrochemical properties using only elastic waves.
