1猪细小病毒AV30 VP2基因在昆虫细胞中表达及其免疫原性鉴定显示文摘本研究选用Bac-to-Bac昆虫杆状病毒表达系统构建含有猪细小病毒AV30未优化、优化后衣壳蛋白VP2基因的重组杆状病毒,感染sf9、HF细胞表达VP2蛋白。首先,扩增PPV AV30得到VP2基因,测序后由公司根据昆虫细胞嗜性进行基因优化。同时构建并收获未优化、优化重组杆状病毒r BacVP2。其感染HF细胞后,进行Western-blot及IFA实验分析表明,未优化、优化VP2蛋白在昆虫细胞中正确表达,分子质量约为64 ku,并具有良好的反应原性。电镜观察,未优化、优化VP2蛋白均自我组装成VLP,与PPV AV30病毒粒子大小形状相似。最后,用VP2-VLP免疫BALB/c小鼠,能刺激小鼠产生抗PPV的特异性抗体。免疫后的小鼠脾脏淋巴细胞在PPV AV30病毒的刺激后,出现显著的增殖情况,同时产生了Th1和Th2型相关细胞因子。本研究成功构建了VP2-VLP,并在小鼠实验中显示了良好的免疫效果,为以VP2为表位载体的嵌合型病毒样颗粒疫苗进一步研制奠定了基础。李茜 代军飞 丁耀忠 ASHENAFI Kiros Wubshe 李国秀 马炳 张杰 2020中国兽医科学2020,50,7:4
2Current and future predicting potential areas of Oxytenanthera abyssinica(A.Richard)using MaxEnt model under climate change in Northern Ethiopia显示文摘Introduction:Climate change will either improve,reduce,or shift its appropriate climatic habitat of a particular species,which could result in shifts from its geographical range.Predicting the potential distribution through MaxEnt modeling has been developed as an appropriate tool for assessing habitat distribution and resource conservation to protect bamboo species.Methods:Our objective is to model the current and future distribution of Oxytenanthera abyssinica(A.Richard)based on three representative concentration pathways(RCP)(RCP2.6,RCP4.5,and RCP8.5)for 2050s and 2070s using a maximum entropy model(MaxEnt)in Northern Ethiopia.For modeling procedure,77 occurrence records and 11 variables were retained to simulate the current and future distributions of Oxytenanthera abyssinica in Northern Ethiopia.To evaluate the performance of the model,the area under the receiver operating characteristic(ROC)curve(AUC)was used.Results:All of the AUCs(area under curves)were greater than 0.900,thereby placing these models in the“excellent”category.The jackknife test also showed that precipitation of the coldest quarter(Bio19)and precipitation of the warmest quarter(Bio18)contributed 66.8%and 54.7%to the model.From the area of current distribution,1367.51 km2(2.52%),7226.28 km2(13.29%),and 5377.26 km2(9.89%)of the study area were recognized as high,good,and moderate potential habitats of Oxytenanthera abyssinica in Northern Ethiopia,and the high potential area was mainly concentrated in Tanqua Abergele(0.70%),Kola Temben(0.65%),Tselemti(0.60%),and Tsegede(0.31%).Kafta Humera was also the largest good potential area,which accounts for 2.75%.Compared to the current distribution,the total area of the high potential regions and good potential regions for Oxytenanthera abyssinica under the three RCPs(RCP2.6,RCP4.5,and RCP8.5)would increase in the 2050s and 2070s.However,the total area of the least potential regions under the three RCPs(RCP2.6,RCP4.5,and RCP8.5)in 2050s and 2070s would decrease.Conclusion:This study can provide vital information for the protection,management,and sustainable use of Oxytenanthera abyssinica,the resource to address the global climate challenges.Yikunoamlak Gebrewahid Selemawi Abrehe Esayas Meresa Gebru Eyasu Kiros Abay Gebrehiwot Gebreab Kiros Kidanemariam Gezu Adissu Gebrekidan Abreha Girmay Darcha 2020Ecological Processes2020,9,1:4
3南非2型口蹄疫病毒结构蛋白抗原表位研究进展显示文摘口蹄疫病毒(FMDV)感染会引起偶蹄动物发生口蹄疫(FMD),产生急性系统性传染性水疱。其分为7个血清型,包括A、O、C型,亚州1型(Asia 1)及南非1、2、3型(SAT1、2、3)。其中,SAT FMDV具有明显的地域性,主要集中在撒哈拉沙漠以南,侵害水牛及野生动物,容易发生抗原变异,疫苗交叉保护效果低。近几年的报告表明,中东地区暴发了SAT2FMDV,跨越了长久以来的地域界限,也对我国的养殖业造成了潜在的威胁。我国在SAT FMDV方面的研究较少,有必要建立特异性和灵敏性高的SAT FMDV监测方法作为战略储备,以防其跨境传播。论文主要介绍了SAT2FMDV结构蛋白抗原表位国内外研究进展,以期为今后研发有关SAT2FMDV战略储备表位疫苗提供理论依据。李茜 代军飞 丁耀忠 李国秀 侯谦 Ashenafi Kiros Wubshet 马炳 张永光 张杰 2020动物医学进展2020,41,6:3

10Spatially explicit statistical modeling of random and systematic land cover transitions in the main grassland plain of Nech Sar National Park, Ethiopia显示文摘The main grassland plain of Nech Sar National Park(NSNP)is a federally managed protected area in Ethiopia designated to protect endemic and endangered species.However,like other national parks in Ethiopia,the park has experienced significant land cover change over the past few decades.Indeed,the livelihoods of local populations in such developing countries are entirely dependent upon natural resources and,as a result,both direct and indirect anthropogenic pressures have been placed on natural parks.While previous research has looked at land cover change in the region,these studies have not been spatially explicit and,as a result,knowledge gaps in identifying systematic transitions continue to exist.This study seeks to quantify the spatial extent and land cover change trends in NSNP,identify the strong signal transitions,and identify and quantify the location of determinants of change.To this end,the author classifies panchromatic aerial photographs in 1986,multispectral SPOT imagery in 2005,and Sentinel imagery in 2019.The spatial extent and trends of land cover change analysis between these time periods were conducted.The strong signal transitions were systematically identified and quantified.Then,the basic driving forces of the change were identified.The locations of these transitions were also identified and quantified using the spatially explicit statistical model.The analysis revealed that over the past three decades(1986–2019),nearly 52%of the study area experienced clear landscape change,out of which the net change and swap change attributed to 39%and 13%,respectively.The conversion of woody vegetation to grassland(~5%),subsequently grassland-to-open-overgrazed land(28.26%),and restoration of woody vegetation(0.76%)and grassland(0.72%)from riverine forest and open-overgrazed land,respectively,were found to be the fully systematic transitions whereas the rest transitions were recorded either partly systematic or random transitions.The location of these most systematic land cover transitions identified through the spatially explicit statistical modeling showed drivers due to biophysical conditions,accessibility,and urban/market expansions,coupled with successive government policies for biodiversity management,geo-politics,demographic,and socioeconomic factors.These findings provide important insights into biodiversity loss,land degradation,and ecosystem disruption.Therefore,the model for predicted probability generally suggests a 0.75 km and 0.72 km buffers which are likely to protect forest and grassland from conversion to grassland and open-overgrazed land,respectively.Kiros Tsegay Deribew 2019Ecological Processes2019,8,1:1
