1Serum level of TSGF,CA242 and CA19-9 in pancreatic cancer显示文摘AIM: To establish a method to detect the expression of the tumor specific growth factor TSGF, CA242 and CA19-9 in serum and evaluate their value in diagnosis of pancreatic cancer. METHODS: ELISA and Biochemical colorimetric assay were used to detect the serum content of TSGF, CA242 and CA19-9 in 200 normal cases, 52 pancreatitis patients and 96 pancreatic cancer patients. RESULTS: The positive likelihood ratios of TSGF, CA242 and CA19-9 were 5.4, 12.6 and 6.3, respectively, and their negative likelihood ratios were 0.10, 0.19 and 0.17, respectively. With single tumor marker diagnosed pancreatic cancer, the highest sensitivity and specificity of TSGF were 91.6% and 93.5%. In combined test with 3 markers, when all of them were positive, the sensitivity changed to 77.0% and the specificity and the positive predictive value were 100%. The levels of TSGF and CA242 were significantly higher in the patients with pancreatic cancer of head than those in the patients with pancreatic cancer of body, tail and whole pancreas, but the expression of CA19-9 had no correlation with the positions of the pancreatic cancer. The sensitivity of TSGF, CA242 and CA19-9 was increased with the progress in stages of pancreatic cancer. In stage I, the sensitivity of TSGF was markedly higher than CA242 and CA19-9. CONCLUSION: The combined use of TSGF, CA242 and CA19-9 expressions can elevate the specificity for pancreatmc cancer diagnosis. And it shows that it plays an important role to differentiate positions and tissue typing. It is a forepart diagnosis for the pancreatic cancer by combination checking. 3The culture and establishment of embryonic germ(EG)cell lines from Chinese mini swine显示文摘As a part of a basic research project on Xeno-transplantion, we have been engaged in the derivation ofembryonic stem cell lines from Chinese mini swine. Here, we reported for the first time the establishmentof two porcine EG cell lines (BPEG1 and BPEG2) from primordial germ cells of genital ridges of a 28 anda 27 d embryos respectively. Their pluripotent nature has been identified by colony morphology, markercharacterization as well as by in vitro and in vivo differentiation. 4Shake table tests of suspended ceilings to simulate the observed damage in the M_s 7.0 Lushan earthquake, China显示文摘Severe damage to suspended ceilings of metal grids and lay-in panels was observed in public buildings during the 2013 Ms7.0 Lushan earthquake in China. Over the past several years, suspended ceilings have been widely used practice in public buildings throughout China, including government offices, schools and hospitals. To investigate the damage mechanism of suspended ceilings, a series of three-dimensional shake table tests was conducted to reproduce the observed damage. A full-scale reinforced concrete frame was constructed as the testing frame for the ceiling, which was single-story and infilled with brick masonry walls to represent the local construction of low-rise buildings. In general, the ceiling in the tests exhibited similar damage phenomena as the field observations, such as higher vulnerability of perimeter elements and extensive damage to the cross runners. However, it exhibited lower fragility in terms of peak ground/roof accelerations at the initiation of damage. 5Preliminary investigation of seismic damage to two steel space structures during the 2013 Lushan earthquake显示文摘Severe damage to steel space structures is rarely reported when compared to other structural systems damaged during past major earthquakes around the world.Two gymnasiums of steel space structures in downtown Lushan County that were damaged during the 2013 M7.0 Lushan earthquake in China were investigated and the observations are summarized in this paper.Typical damage to these two steel space structures ranges from moderate to severe.Moderate damage includes global buckling and dislocation of bolted connections of truss members,and inelastic elongation of anchor bolts and sliding of pedestal plates of supports.Severe damage includes member fracture caused by local buckling,and fracture failure of anchor bolts and welds.The distribution of structural damage to these two structures is described in detail and future research opportunities are suggested.Dai Junwu Qu Zhe Zhang Chenxiao Weng Xuran 2013Earthquake Engineering and Engineering Vibration2013,12,3:11
6Effects of Zhaoyangwan on chronic hepatitis B and posthepatic cirrhosis显示文摘AIM:To study the therapeutic effects of zhaoyangwan (ZYW) on chronic hepatitis B and hepatic cirrhosis and the anti-virus, anti-fibrosis and immunoregulatory mechanisms of ZYW.METHODS: Fifty cases of chronic hepatitis B and posthepatic cirrhosis with positive serum HBsAg, HBeAg, anti-Hbc and HBV-DNA were divided randomly and single-blindly into the treatment group (treated with ZYW) and the control group (treated with interferon).After 3 month treatment,the effects of the treatment group and the control group were evaluated.RESULTS: The serum ALT normalization was 83.3%(30/36) in the treatment group and 85.7%(12/14) in the control group,with no significant difference (X^2=0.043, P>0.05).After the course, the negative expression rates of the serum HBV-DNA and HBeAg were 44.4%(16/36) and 50%(18/36) in the treatment group, and 50%(7/14) and 50%(7/14) in the control group, respectively,with no significant difference(X^2=0.125, X^2=0.00, both P>0.05). Negative HBsAg and positive HBsAb appeared in 4 cases of the treatment group and 1 case of the control group.Serum anti-HBc turned negative in 6 cases of the treatment group and 1 case of the control group, respectively. After the ZYW treatment,serum CD3^+, CD4^+, CD8^+, CD4^+/CD8^+ and NK cell activation were significantly increased. Only serum CD3^+ and NK cell activation were significantly increased in the control group with a significant difference between the two groups. The serum C4, C1q, C3, B and C9 were significantly in creased inthe treatment group. In the control group only the serum C4 was increased. The concentration of serum interferon had no change after treatment with ZYW, while it was significantly increased in the control group after treatment with interferon.The ultrastructure of the liver restored,which helped effectively to reduce the degeneration and necrosis of hepatic cells,infiltration of inflammatory cells and hepatic cirrhosis.CONCLUSION:ZYW is a pure Chinese herbal medicine. 7Experimental and analytical studies on multiple tuned mass dampers for seismic protection of porcelain electrical equipment显示文摘Porcelain electrical equipment(PEE),such as current transformers,is critical to power supply systems,but its seismic performance during past earthquakes has not been satisfactory.This paper studies the seismic performance of two typical types of PEE and proposes a damping method for PEE based on multiple tuned mass dampers(MTMD).An MTMD damping device involving three mass units,named a triple tuned mass damper(TTMD),is designed and manufactured.Through shake table tests and finite element analysis,the dynamic characteristics of the PEE are studied and the effectiveness of the MTMD damping method is verified.The adverse influence of MTMD redundant mass to damping efficiency is studied and relevant equations are derived.MTMD robustness is verified through adjusting TTMD control frequencies.The damping effectiveness of TTMD,when the peak ground acceleration far exceeds the design value,is studied.Both shake table tests and finite element analysis indicate that MTMD is effective and robust in attenuating PEE seismic responses.TTMD remains effective when the PGA far exceeds the design value and when control deviations are considered.Bai Wen Dai Junwu Zhou Huimeng Yang Yongqiang Ning Xiaoqing 2017Earthquake Engineering and Engineering Vibration2017,16,4:8

9Non-limit passive soil pressure on rigid retaining walls显示文摘This paper aims to reveal the depth distribution law of non-limit passive soil pressure on rigid retaining wall that rotates about the top of the wall(rotation around the top(RT) model). Based on Coulomb theory, the disturbance degree theory, as well as the spring-element model, by setting the rotation angle of the wall as the disturbance parameter, we establish both a depth distribution function for sand and a nonlinear depth distribution calculation method for the non-limit passive soil pressure on a rigid retaining wall under the RT model, which is then compared with experiment. The results suggest that under the RT model: the non-limit soil pressure has a nonlinear distribution; the backfill disturbance degree and the lateral soil pressure increase with an increase in the wall rotation angle; and, the points where the resultant lateral soil pressure acts on the retaining wall are less than 2/3 of the height of the wall. 10Sedimentary Evolution and Transgressions of the Western Subei Basin in Eastern China since the Late Pliocene显示文摘Sedimentary sequences in the Subei Basin are complex and have been affected by interactions between the ocean and rivers since the Late Pliocene, including the Yellow River, Huaihe River, and the Yangtze River. This sedimentary evolution, in particular the timing of Pleistocene transgressions, has long been a matter of controversy owing to the lack of precise chronological evidence. The aim of this study is to explore the evolution of the sedimentary environment throughout the past 3.00 Ma in this region on the basis of a comprehensive analysis of particle size and foraminifera and ostracods collected in the TZK9 core from the Subei Basin combined with geochronological studies of magnetostratigraphy, AMS^(14)C and optically stimulated luminescence(OSL). The results show that fluvial facies in the sedimentary environment from 3.00 to 1.01 Ma. There were fluvial facies and reflects six sea-level high stands from 1.01 to 0.25 Ma. The study area was affected by four large-scale transgressions since 0.25 Ma. The four marine sedimentary layers known as DU7(buried at 48–52 m), DU5(buried at 35–41 m), DU3(buried at 16–23 m), and DU1(buried at 2–4 m) are recorded in the MIS7(210–250 ka), MIS5, MIS3, and Holocene, respectively. The magnitude of the DU5 transgression was identical to that of the DU3 transgression, both were larger than the DU7 transgression, and the DU1 transgression was the weakest. The variation of transgression strength reflects the influence of global changes in sea level, tectonic subsidence, shell ridges, and sand dams. In the TZK9 core, we found evidence of seven sea-level high stands from the Early–Middle Pleistocene, and the first one caused by regional rapid subsidence and could be traced back to 0.83-0.84 Ma. The sea-level high stands and the age of the first one recorded above was different from other cores in eastern China, this was caused by the lack of absolute age control and the differences in paleotopography during this period. This study reconstructs sedimentary evolution, determines the transgression and its age, establishes the chronology since the Late Pliocene, and provides a scientific framework for further paleoenvironmental and tectonic studies. 12Flow structures inside a large-scale turbulent fluidized bed of FCC particles:Eulerian simulation with an EMMS-based sub-grid scale model显示文摘Turbulent fluidized bed reactors are widely used in industry. However, CFD simulations of the hydrodynamic characteristics of these reactors are relatively sparse, despite the urgent demand from industry. To address this problem, Eulerian simulations with an EMMS-based sub-grid scale model, accounting for the effect of sub-grid scale structures on the inter-phase friction, are performed to study the hydrodynamics inside a large-scale turbulent fluidized bed of FCC particles. It is shown that the simulated axial and radial solid concentration profiles, entrained solid fluxes and standard deviation of the solid concentration fluc- tuation agreed well with experimental data available in the literature. In-depth analysis of time-averaged particle velocity and solid concentration shows that a dense-suspension upflow regime coexists with fast fluidization regime in this bed, which is reminiscent of the hydrodynamic characteristics in high-density circulating fluidized bed (CFB) risers, even though they are operated in different fluidization regimes. The Reynolds stresses in turbulent fluidized beds are anisotropic, but the degree of anisotropy is far less pronounced than the reported values in CFB risers. 13A novel key pre-distribution scheme for wireless distributed sensor networks显示文摘A novel key pre-distribution scheme for sensor networks is proposed, which enables sensor nodes to communicate securely with each other using cryptographic techniques. The approach uses the rational normal curves in the projective space with the dimension n over the finite field F q . Both secure connectivity and resilience of the resulting sensor networks are analyzed. By choosing the parameters q and n properly, this key pre-distribution scheme has some advantages over the previous known schemes. In addition, if the number of the rational normal curves in the scheme becomes too large for an application, the size may be reduced by choosing a part of the curves randomly. 14Infective viruses produced from full-length complementary DNA of swine vesicular disease viruses HK/70 strain显示文摘The full-length cDNA clone of swine vesicular disease virus HK/70 strain named pSVOK12 was constructed in order to study the antigenicity, replication, maturation and pathogenicity of swine vesicular disease virus. In vitro transcription RNA from pSVOK12 transfected IBRS-2 cells and the re- covered virus RNA were isolated and sequenced, then indirect hemagglutination test, indirect im- munofluorescence assays, eleectron microscope test, 50% tissue culture infecting dose (TCID50) assays and mouse virulence studies were performed to study the antigenicity and virulence of the recovered virus. The result showed that the infectious clones we ob- tained and the virus derived from pSVOK12 had the same biological properties as the parental strain HK/70. 17Subcutaneous and intramuscular fat transcriptomes show large differences in network organization and associations with adipose traits in pigs显示文摘Subcutaneous fat(SCF)and intramuscular fat(IMF)deposition is relevant to health in humans,as well as meat production and quality in pigs.In this study,we generated RNA sequence data for 122 SCF,120 IMF,and 87 longissimus dorsi muscle(LDM)samples using 155 F6 pigs from a specially designed heterogeneous population generated by intercrossing four highly selected European commercial breeds and four indigenous Chinese pig breeds.The phenotypes including waist back fat thickness and intramuscular fat content were also measured in the 155 F6 pigs.We found that the genes in SCF and IMF differed largely in both expression levels and network connectivity,and highlighted network modules that exhibited strongest gain of connectivity in SCF and IMF,containing genes that were associated with the immune process and DNA double-strand repair,respectively.We identified 215 SCF genes related to kinase inhibitor activity,mitochondrial fission,and angiogenesis,and 90 IMF genes related to lipolysis and fat cell differentiation,displayed a tissue-specific association with back fat thickness and IMF content,respectively.We found that cis-expression QTL for trait-associated genes in the two adipose tissues tended to have tissuedependent predictability for the two adipose traits.Alternative splicing of genes was also found to be associated with SCF or IMF deposition,but the association was much less extensive than that based on expression levels.This study provides a better understanding of SCF and IMF gene transcription and network organization and identified critical genes and network modules that displayed tissue-specific associations with subcutaneous and intramuscular fat deposition.These features are helpful for designing breeding programs to genetically improve the two adipose traits in a balanced way.Yifeng Zhang Yingchun Sun Zhongzi Wu Xinwei Xiong Junjie Zhang Junwu Ma Shijun Xiao Lusheng Huang Bin Yang 2021Science China(Life Sciences)2021,64,10:5

19Coupling control on pillar stress concentration and surface cracks in shallow multi-seam mining显示文摘In order to ensure safe mining and reduce surface damage in shallow multi-seam mining,the failure characteristics of interburden strata with different coal pillars offset distances between pillars in the upper and lower seams,the distribution characteristics of stress concentration in coal pillars,and the development characteristics of stratum cracks and subsidence were investigated by physical and UDEC2D simulation.Meanwhile,the effect of different coal pillar offset distances on stress concentration of coal pillar and development of stratum cracks were studied.Based on those results,a formula for safe mining and reducing surface damage was established,which provided a theoretical basis for safe and environmentally friendly mining in shallow multi-seam.According to the results,the optimal coal pillar offset distance(the side to side horizontal distance of the upper and lower coal pillars)between the upper and lower coal seams was developed to reduce the stress concentration of coal pillars and surface damage.The results of this study have been applied in Ningtiaota coal mine and have achieved good results in safe and environmentally friendly mining.Qingxiang Huang Junwu Du Jie Chen Yanpeng He 2021International Journal of Mining Science and Technology2021,31,1:4
20LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy.Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang 2022Science China(Life Sciences)2022,65,6:4
