| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection,histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection,tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection,and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys).In 30 of them,both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern,tumor staging,lymph node metastasis,grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma,andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. | 7 | Combinations of Hd2 and Hd4 genes determine rice adaptability to Heilongjiang Province, northern limit of China显示文摘Heading date is a key trait in rice domestication and adaption, and a number of quantitative trait loci(QTLs)have been identified. The rice(Oryza sativa L.) cultivars in the Heilongjiang Province, the northernmost region of China,have to flower extremely early to fulfill their life cycle.However, the critical genes or different gene combinations controlling early flowering in this region have not been determined. QTL and candidate gene analysis revealed that Hd2/Ghd7.1/Os PRR37 plays a major role in controlling rice distribution in Heilongjiang. Further association analysis with a collection of rice cultivars demonstrated that another three major QTL genes(Hd4/Ghd7, Hd5/DTH8/Ghd8, and Hd1)also participate in regulating heading date under natural long day(LD) conditions. Hd2/Ghd7.1/Os PRR37 and Hd4/Ghd7 are two major QTLs and function additively. With the northward rice cultivation, the Hd2/Ghd7.1/Os PRR37 and Hd4/Ghd7 haplotypes became non-functional alleles. Hd1 might be non-functional in most Heilongjiang rice varieties,implying that recessive hd1 were selected during local rice breeding. Non-functional Hd5/DTH8/Ghd8 is very rare, but constitutes a potential target for breeding extremely early flowering cultivars. | 8 | Probabilistic assessment of tunnel convergence considering spatial variability in rock mass properties using interpolated autocorrelation and response surface method显示文摘This study aims at the probabilistic assessment of tunnel convergence considering the spatial variability in rock mass properties. The method of interpolated autocorrelation combined with finite difference analysis is adopted to model the spatial variability of rock mass properties. An iterative procedure using the first-order reliability method(FORM) and response surface method(RSM) is employed to compute the reliability index and its corresponding design point. The results indicate that the spatial variability considerably affects the computed reliability index. The probability of failure could be noticeably overestimated in the case where the spatial variability is neglected. The vertical scale of fluctuation has a much higher effect on the probabilistic result with respect to the tunnel convergence than the horizontal scale of fluctuation. | 9 | Field Measurements of Influence of Sand Transport Rate on Structure of Wind-sand Flow over Coastal Transverse Ridge显示文摘The structure of wind-sand flow under different total sand transport rates was measured with field vertical anemometer and sand trap on the crest of typical coastal transverse ridge in Changli Gold Coast of Hebei Province, which is one of the most typical coastal aeolian distribution regions in China and famous for the tall and typical coastal transverse ridges. The measurement results show that, on the conditions of approximate wind velocities and same surface materials and environments, some changes happen to the structure of wind-sand flow with the increase of total sand transport rate on the crest of coastal transverse ridge. First, the sand transport rates of layers at different heights in the wind-sand flow increase, with the maximum increase at the height layer of 4-8cm. Second, the ratios of sand trans-port rates of layers at different heights to total sand transport rate decrease at the low height layer (0-4cm), but increase at the high height layer (4-60cm). Third, the distribution of the sand transport rate in the wind-sand flow can be expressed by an exponential function at the height layer of 0-40cm, but it changes from power function model to ex-ponential function model in the whole height layer (0-60cm) and changes into polynomial function model at the height layer of 40-60cm with the increase of total sand transport rate. | 10 | Peroxisome proliferator-activated receptor-delta agonist ameliorated inflammasome activation in nonalcoholic fatty liver disease显示文摘AIM: To evaluate the inflammasome activation and the effect of peroxisome proliferator-activated receptors(PPAR)-δ agonist treatment in nonalcoholic fatty liver disease(NAFLD) models.METHODS: Male C57BL/6J mice were classified according to control or high fat diet(HFD) with or without PPAR-δ agonist(GW) over period of 12 wk [control, HFD, HFD + lipopolysaccharide(LPS), HFD + LPS + GW group]. Hep G2 cells were exposed to palmitic acid(PA) and/or LPS in the absence or presence of GW.RESULTS: HFD caused glucose intolerance and hepatic steatosis. In mice fed an HFD with LPS, caspase-1 and interleukin(IL)-1β in the liver were significantly increased. Treatment with GW ameliorated the steatosis and inhibited overexpression of pro-inflammatory cytokines. In Hep G2 cells, PA and LPS treatment markedly increased m RNA of several nucleotide-binding andoligomerization domain-like receptor family members(NLRP3, NLRP6, and NLRP10), caspase-1 and IL-1β. PA and LPS also exaggerated reactive oxygen species production. All of the above effects of PA and LPS were reduced by GW. GW also enhanced the phosphorylation of AMPK-α.CONCLUSION: PPAR-δ agonist reduces fatty acidinduced inflammation and steatosis by suppressing inflammasome activation. | 12 | Image formation of brain function in patients suffering from knee osteoarthritis treated with moxibustion显示文摘OBJECTIVES:Functional magnetic resonance imaging(fMRI) technology was used to study changes to the resting state blood flow in the brains of patients with knee osteoarthritis(KOA) before and after treatment with moxibustion at the acupoint of the left Dubi(ST 35) and to probe the cerebral mechanism underlying the effect of moxibustion.METHODS:The resting state brain function of 30 patients with left KOA was scanned with fMRI before and after treatment with moxibustion.The analytic methods of fractional amplitude of low frequency fluctuation(fALFF) and regional homogeneity(ReHo) were used to observe changes in resting state brain function.RESULTS:The fALFF values of the right cerebrum,extra-nucleus,left cerebellum,left cerebrum and white matter of patients after moxibustion treatment were higher than before treatment,and the fALFF values of the precentral gyrus,frontal lobe and occipital lobe were lower than before treatment(P<0.05,K≥85).The ReHo values of the thalamus,extra-nucleus and parietal lobe of patients were much higher than those before moxibustion treatment,and the ReHo values of the right cerebrum,left cerebrum and frontal lobe were lower than before treatment(P<0.05,K≥85).CONCLUSION:The influence of moxibustion on obvious changes in brain regions basically conforms to the way that pain and warmth is transmitted in the body,and the activation of sensitive systems in the body may be objective evidence of channel transmission.The regulation of brain function by moxibustion is not in a single brain region but rather in a network of many brain regions. | Hongwu Xie Fangming Xu Rixin Chen Tianyou Luo Mingren Chen Weidong Fang Fajin Lü Fei Wu Yune Song Jun Xiong | 2013 | Journal of Traditional Chinese Medicine2013,33,2: | 7 |
| 13 | The association between the genetic polymorphisms of LMP2/LMP7 and the outcomes of HCV infection among drug users显示文摘Objective:To investigate a possible association of LMP2/LMP7 genes with chronic hepatitis C virus(HCV) infection,and to assess whether LMP2/LMP7 genes could influence the outcomes of HCV infection among drug users.Methods:Genomic DNAs of 362 anti-HCV sero-positive drug users and 225 control drug users were extracted from the peripheral blood leukocytes.The sero-positive patients were divided into those who had persistent infection and those who had spontaneously cleared the infection.Polymorphisms of LMP genes were determined by PCR combined with restriction fragment length polymorphism(RFLP).Results:The distribution of LMP2 genotypes among the control,persistent infection and spontaneous clearance groups were not different.However,the LMP7 codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in control group(OR=1.75,95%CI=1.06~2.90;OR=3.16,95%CI=1.23-8.12;OR=1.94,95%CI=1.21-3.12,respectively).Similarly,the frequencies of the codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in the spontaneous clearance group(OR=1.64,95%CI=1.04-2.57;OR=2.40,95%CI=1.09-5.28;OR=1.76,95%CI=1.152.69,respectively).Stratified analysis indicated that combined genotype Gln/Lys+Lys/Lys of the LMP7 gene was related to an increasing susceptibility to HCV infection(OR=1.91,95%CI=1.02-3.55;OR=2.19,95%CI=1.243.89;OR=1.91,95%CI=1.05-3.48,OR=2.86,95%CI=1.41-5.78,respectively)and the risk of persistent HCV infection(OR=1.94,95%CI=1.12-3.34;OR=2.02,95%CI=1.21-3.38;OR=1.78,95%CI=1.12-2.85,OR=2.23,95%CI=1.09-4.58,respectively)among>30-year-old,males,the injection drug user(IDU)subjects and/or the shorter duration drug users(≤5 y).Conclusion:These results suggest that polymorphism of the LMP7 gene may have an influence on the outcomes of HCV infection,and is one of the factors accounting for the genetic susceptibility to HCV infection among drug users. | Qian Cui Yongxiang Zhang Jing Su Chao Shi Na Lei Keqin Ding Jun Li Rongbin Yu L u Wang Ning Wang | 2010 | The Journal of Biomedical Research2010,24,5: | 6 |

| 15 | Extended multiscale finite element method for mechanical analysis of heterogeneous materials显示文摘An extended multiscale finite element method (EMsFEM) is developed for solving the mechanical problems of heterogeneous materials in elasticity.The underlying idea of the method is to construct numerically the multiscale base functions to capture the small-scale features of the coarse elements in the multiscale finite element analysis.On the basis of our existing work for periodic truss materials, the construction methods of the base functions for continuum heterogeneous materials are systematically introduced. Numerical experiments show that the choice of boundary conditions for the construction of the base functions has a big influence on the accuracy of the multiscale solutions, thus,different kinds of boundary conditions are proposed. The efficiency and accuracy of the developed method are validated and the results with different boundary conditions are verified through extensive numerical examples with both periodic and random heterogeneous micro-structures.Also, a consistency test of the method is performed numerically. | 17 | Influence of Copper on Transport and Dissipation of Lambda-Cyhalothrin and Cypermethrin in Soils显示文摘Repeated applications of bordeaux mixture (a blend of copper sulfate and calcium hydroxide) and pyrethroid insecticides (Pys) have led to elevated copper (Cu) and Pys concentrations in vineyard surface soils. To understand the potential influence of Cu on the fate of Pys in the soil environment, we selected two Pys, cypermethrin (CPM) and lambda-cyhalothrin (λ-CHT), and two typical Chinese vineyard soils, Haplic Acrisol and Luvic Phaeozem, as experimental samples. The dissipation experiment was conducted at room temperature in the dark, and the transport of both Pys through the soils was investigated using soil thin-layer chromatography. The results showed that the transport of Pys in both soils increased as the Cu^(2+) concentration increased from 0 to 100 mg L^(-1) , and Pys were more transportable in Haplic Acrisol (HA) than in Luvic Phaeozem (LP) under the same experimental conditions. For CPM, only 100 mg L^(-1) of Cu^(2+) significantly (P<0.05) increased Pys transport through both soils relative to water. Lambda-CHT was significantly (P<0.05) transported through HA by all the Cu^(2+) concentrations compared to water, and all but the 1 mg L^(-1) of Cu^(2+) significantly (P<0.05) increased the transport of λ-CHT through LP relative to water. However, the dissipation rates of CPM and λ-CHT decreased with the addition of Cu to soils. | 18 | Construction of Prokaryotic Expression Vector of Mouse Nanog Gene and Its Expression显示文摘The aim of this study is to construct a prokaryotic expression vector of mouse Nanog gene and to express it in E.coli.A pair of primers was designed according to digestion sites in plasmid pGEX-KG and the Nanog gene sequence published by GenBank.The DNA fragment of 918 bp was amplified by polymerase chain reaction(PCR)from the pNA992 recombinant plasmid with Nanog gene,then cloned into pGEX-KG and transformed into the host E.coli strain TGⅠ.The sequence of the fragment was matched with the original sequence of pNA992.It indicated that fusion expression vector,pGEX-KG- Nanog,was constructed successfully.The pGEX-KG-Nanog plasmid was extracted from E.coli strain TGⅠand was transformed into BL21(DE3)for expression.After induction by isopropy1-β-D-thiogalactoside(IPTG)at 37°C,the expression product of Nanog gene was identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE),and the expression condition was optimized.Nanog fusion protein was successfully expressed in the form of inclusion bodies. | 19 | Establishment and validation of a model for brain injury state evaluation and prognosis prediction显示文摘Purpose:Traumatic brain injury(TBI)is one of the leading causes of disability and death in modern times,whose evaluation and prognosis prediction have been one of the most critical issues in TBI management.However,the existed models for the abovementioned purposes were defective to varying degrees.This study aims to establish an ideal brain injury state clinical prediction model(BISCPM).Methods:This study was a retrospective design.The six-month outcomes of patients were selected as the end point event.BISCPM was established by using the split-sample technology,and externally validated via different tests of comparison between the observed and predicted six-month mortality in validating group.TBI patients admitted from July 2006 to June 2012 were recruited and randomly divided into establishing model group and validating model group.Twenty-one scoring indicators were included in BISCPM and divided into three parts,A,B,and C.Part A included movement,pupillary reflex and diameter,CT parameters,and secondary brain insult factors,etc.Part B was age and part C was medical history of the patients.The total score of part A,B and C was final score of BISCPM.Results:Altogether 1156 TBI patients were included with 578 cases in each group.The score of BISCPM from validating group ranged from 2.75 to 31.94,averaging 13.64±5.59.There was not statistical difference between observed and predicted mortality for validating group.The discrimination validation showed that the BISCPM is superior to international mission for prognosis and analysis of clinical trials(IMPACT)lab model.Conclusion:BISCPM is an effective model for state evaluation and prognosis prediction of TBI patients.The use of BISCPM could be of great significance for decision-making in management of TBI. | Xia Li Chao Lü Jun Wang Yi Wan Shu-Hui Dai Lei Zhang Xue-An Hu Xiao-Fan Jiang Zhou Fei | 2020 | Chinese Journal of Traumatology2020,23,5: | 3 |
