| 1 | Transient receptor potential vanilloid 4-dependent calcium influx and ATP release in mouse and rat gastric epithelia显示文摘AIM: To explore the expression of transient receptor potential vanilloid 4(TRPV4) and its physiological meaning in mouse and rat gastric epithelia. METHODS: RT-PCR and immunochemistry were used to detect TRPV4 m RNA and protein expression in mouse stomach and a rat normal gastric epithelial cell line(RGE1-01), while Ca2+-imaging and electrophysiology were used to evaluate TRPV4 channel activity. ATP release was measured by a luciferin-luciferase assay. Gastric emptying was also compared between WT and TRPV4 knockout mice. RESULTS: TRPV4 m RNA and protein were detected in mouse tissues and RGE1-01 cells. A TRPV4-specific agonist(GSK1016790A) increased intracellular Ca2+ concentrations and/or evoked TRPV4-like current activities in WT mouse gastric epithelial cells andRGE1-01 cells, but not TRPV4 KO cells. GSK1016790 A or mechanical stimuli induced ATP release from RGE1-01 cells while TRPV4 knockout mice displayed delayed gastric emptying in vivo. | 2 | Chromosome-scale scaffolding of the black raspberry (Rubus occidentalis L.) genome based on chromatin interaction data显示文摘Black raspberry(Rubus occidentalis L.)is a niche fruit crop valued for its flavor and potential health benefits.The improvement of fruit and cane characteristics via molecular breeding technologies has been hindered by the lack of a high-quality reference genome.The recently released draft genome for black raspberry(ORUS 4115-3)lacks assembly of scaffolds to chromosome scale.We used high-throughput chromatin conformation capture(Hi-C)and Proximity-Guided Assembly(PGA)to cluster and order 9650 out of 11,936 contigs of this draft genome assembly into seven pseudo-chromosomes.The seven pseudo-chromosomes cover~97.2%of the total contig length(~223.8 Mb).Locating existing genetic markers on the physical map resolved multiple discrepancies in marker order on the genetic map.Centromeric regions were inferred from recombination frequencies of genetic markers,alignment of 303 bp centromeric sequence with the PGA,and heat map showing the physical contact matrix over the entire genome.We demonstrate a high degree of synteny between each of the seven chromosomes of black raspberry and a high-quality reference genome for strawberry(Fragaria vesca L.)assembled using only PacBio long-read sequences.We conclude that PGA is a cost-effective and rapid method of generating chromosome-scale assemblies from Illumina short-read sequencing data. | Rubina Jibran Helge Dzierzon Nahla Bassil Jill M.Bushakra Patrick P.Edger Shawn Sullivan Chad E.Finn Michael Dossett Kelly J.Vining Robert VanBuren Todd C.Mockler Ivan Liachko Kevin M.Davies Toshi M.Foster David Chagné | 2018 | Horticulture Research2018,5,1: | 4 |

| 6 | Multi-Level Knowledge Engineering Approach for Mapping Implicit Aspects to Explicit Aspects显示文摘Aspect’s extraction is a critical task in aspect-based sentiment analysis,including explicit and implicit aspects identification.While extensive research has identified explicit aspects,little effort has been put forward on implicit aspects extraction due to the complexity of the problem.Moreover,existing research on implicit aspect identification is widely carried out on product reviews targeting specific aspects while neglecting sentences’dependency problems.Therefore,in this paper,a multi-level knowledge engineering approach for identifying implicit movie aspects is proposed.The proposed method first identifies explicit aspects using a variant of BiLSTM and CRF(Bidirectional Long Short Memory-Conditional Random Field),which serve as a memory to process dependent sentences to infer implicit aspects.It can identify implicit aspects from four types of sentences,including independent and three types of dependent sentences.The study is evaluated on a largemovie reviews dataset with 50k examples.The experimental results showed that the explicit aspect identification method achieved 89%F1-score and implicit aspect extraction methods achieved 76%F1-score.In addition,the proposed approach also performs better than the state-of-the-art techniques(NMFIAD andML-KB+)on the product review dataset,where it achieved 93%precision,92%recall,and 93%F1-score. | Jibran Mir Azhar Mahmood Shaheen Khatoon | 2022 | Computers, Materials & Continua2022,,2: | 1 |

Detection of these illegal additives is very meaningful and the developed method meets this objective in a simple, rapid and cost effective way. Detection limits for chrysoidin and malachite green in aqueous solutions reached0.001 mg/L and 0.0001 mg/L respectively. Linear relationship between the Raman intensities and chrysoidin concentrations in range of 0.001-0.05 mg/L was found with regression coefficient(R) of 0.994.To investigate the practical application performance of the method, the substrate was applied to detect chrysoidin in Sprite at 0.01 mg/L and malachite green in fish pond water at 0.0001 mg/L.
