| 1 | Long-term trend and abrupt events of the Holocene Asian monsoon inferred from a stalagmiteδ^(18)O record from Shennongjia in Central China显示文摘A high-resolution oxygen-isotope record from a thorium-uranium-dated stalagmite from Shanbao Cave at Shennongjia reflects variations in the amount of monsoon precipitation for the period from 11.5 to 2.1 ka (1 ka = 1000 cal aBP). Between 11.5 and 9.3 ka, a sharp decrease in δ18O indicates a rapid increase in monsoon precipitation. An interval of generally high monsoon precipitation is observed between 9.3 and 4.4 ka. An arid period has prevailed between 4.4 and 2.1 ka. The long-term trend of Shanbao record appears to follow summer insolation at 33°N latitude. An abrupt decrease in monsoon precipitation around 4.3 ka is synchronous with the collapse of Neolithic culture in central China. This abrupt change could have resulted from the amplifi- cation of the gradually decreased summer insolation by the positive vegetation-atmosphere-aerosol feed- back. The weakened Asian monsoon events were in concert with decreased Greenland temperature dur- ing the early Holocene, centered at 8.2, 8.6, 9.3, 10.2 and 11.0 ka. | 2 | Effects of Transgenic Tobacco Plants Expressing ACA Gene from Amaranthus caudatus on the Population Development of Myzus persicae显示文摘To investigate the possible function of the agglutinin from Amaranthus caudatus L. (ACA) in plant defending against insect pests, ACA cDNA was cloned by RT-PCR and the 5' and 3' sequences were confirmed by rapid amplification of cDNA ends (RACE). The phloem-specific expression vector of ACA gene, pBCACAc, was constructed based on the plant binary vector pBC438 and transfered into tobacco plants via Agrobacterium-mediated transformation method. Results from PCR and Southern blotting analysis showed that AOA gene was integrated into the genomes of transformed plants and the transgene integration varied from one to four estimated copies per genome. Western blotting analysis indicated that ACA gene was transcribed and translated in the transgenic plants. The bioassay of Myzus persicae Sulzer on detached leaves demonstrated that the 78% transgenic tobacco plants displayed an average aphid-resistant rate of more than 75%. Some apterous progeny of M. persicae were found dead on the resistant plants. | 4 | A high-resolution stalagmite O-C isotope record from Nanjing and its rapid response to climatic events显示文摘Here we discussed rapid response of the cave temperature and vegetation to the four Dansgaard-Oeschger cold and warm cycles during 50-40 kaBP based on results of oxygen and carbon stable isotopic compositions from a stalagmite in Tangshan, Nanjing. It is found that the amount of C3 vegetation relative to C4-type declines during the D-O warm events, indicating the decrease of the effective mete-oric precipitation. Compared with O-isotope records of the Greenland ice core, the stalagmite record displays a very similar pattern to Greenland ice core record over the dec-ade-century time scale, suggesting that the changes of the East Asian monsoon climate are in accordance with the high-latitude polar climate in the short-term time scale. The age of the ice-rafted H5 event in the stalagmite record, how-ever, preceded that of Greenland ice cores by 2 ka. | 5 | IMPRINT OF SOLAR ACTIVITY ON NANJING STALAGMITE ANNUAL LAYER THICKNESS SEQUENCE DURING THE LAST GLACIAL MAXIMUM显示文摘A 3000-year-long stalagmite chronology from Hulu Cave near Nanjing was established by counting annual layers under microscope. Based on the 230Th age, this chronology covers the period 24-21 kaBP, within the Last Glacial Maximum (LGM). Two proxies, annual layer thickness and gray level were measured along the growth axis of the stalag-mite profile in order to establish a high-resolution East Asian monsoon history during the LGM. The high correlation coefficient (r = 0.55) between the two proxies suggests that both of them were controlled by a common factor, possibly reflecting changes in the strength of summer monsoon circulation and its pre-cipitation. Low frequency variations of the annual layer thickness, ranging from centennial to millennial scales, are approximately in agreement with the 10Be-flux recorded in the Greenland ice core, indi-cating that changes in East Asian monsoon strength might be forced by solar outputs during the LGM pe-riods. | 7 | Roles of pre-formed martensite in below-Ms bainite formation,microstructure,strain partitioning and impact absorption energies of low-carbon bainitic steel显示文摘The roles of pre-formed martensite(PM)in below-Ms bainite formation,microstructure,crystallography,strain partitioning and mechanical properties of a low-carbon bainitic steel were investigated using electron-backscattered diffraction,transmission electron microscopy,micro digital image correlation technique and mechanical tests.It is demonstrated that the pre-formation of martensite eliminates the incubation time for bainite transformation at various austempering temperatures below Ms,indicative of its acceleration effect at the early stage of transformation.This effect is mainly attributed to the surfaces or tips of the PM acting as the nuclei of subsequently-formed bainite,with initial bainite tending to form around the PM.However,the finishing time for below-Ms bainite transformation,especially at even lower temperatures,is retarded,owing to the dividing effect of PM on parent austenite grains,the decreasing effect of lowered isothermal temperature on the diffusion rate of carbon atoms and the strengthening effect of lowered isothermal temperature on supercooled austenite.PM and its adjacent bainitic laths have nearly the same crystallographic orientation and belong to the same block.The pre-formation of martensite largely refines the bainitic blocks/laths and retained austenite.The specimens with PM show relatively uniform strain partitioning among various phases,contrasting with the specimens without PM,for which strains are highly concentrated in the bainite region nearby fresh martensite/austenite(M/A)blocks or between adjacent M/A blocks.The impact absorption energies of the specimens with PM,when austempered at 30-60℃ below Ms,are more than twice higher than those of the specimens without PM,at no expense of tensile properties. | Lihe Qian Zhi Li Tongliang Wang Dongdong Li Fucheng Zhang Jiangying Meng | 2022 | Journal of Materials Science & Technology2022,,1: | 4 |
| 8 | A new loss-of-function allele 28y reveals a role of ARGONAUTE1 in limiting asymmetric division of stomatal lineage ground cell显示文摘In Arabidopsis thaliana L., stomata are produced through a series of divisions including asymmetric and symmetric divisions. Asymmetric entry division of meristemoid mother cell produces two daughter cells, the smaller meristemoid and the larger sister cell, a stomatal lineage ground cell(SLGC). Stomatal lineage ground cells can differentiate into epidermal pavement cells but have the potential to divide asymmetrically, spacing divisions, to create satellite meristemoids. Peptide ligands and TOO MANY MOUTHS(TMM) and ERECTA family receptors regulate the initiation of stomatal lineages, activity, and orientation of spacing divisions. Here, we reported that a natural mutant 28 y displayed an increased stomatal density and index. Using map-based cloning, we identified mutation in ARGONAUTE1(AGO1)as the cause of 28 y phenotypes. Time-lapse tracing of stomatal lineage cells reveals that stomatal overproduction in 28 y is caused by the excessive asymmetric spacing division of SLGCs.Further genetic results demonstrated that AGO1 acts downstream of TMM and negatively regulates the SPCH transcripts,but in a brassinosteroid-independent manner. | 14 | Disease-tolerance of transgenic tobacco plants expressing Ah-AMP gene of Amaranthus hypochondriacus显示文摘An antimicrobial peptide gene from Amaranthus hypochondriacus, Ah-AMP, was amplified by PCR and cloned. Sequence analysis results revealed that this gene is 261 bp in length encoding a precursor polypeptide of 87 amino acid residues. Ah-AMP gene was inserted in the binary vector pBin438 to construct a plant expression vector pBinAH916. Leave explants of Nicotiana tabacum var. SR1 were transformed with Agrobacterium tumefaciens LBA4404 harboring the above expression vector. Results from PCR, Southern and Northern blot analyses confirmed that the Ah-AMP gene had been integrated into the tobacco genome and was transcribed at mRNA level. Two bacterial-resistant transgenic plants were selected by inoculating the plants with Pseudomonas solanacearum and statistic analysis of two T1 lines showed that the resistance increased by 2.24 and 1.62 grade and the disease index decreased by 49.6% and 37.3% respectively when compared with the non-transformed control plants SR1. The results from challenging the plants with inoculums of Phytophthora parasitica showed that the symptom development was delayed and disease index was significantly reduced.
