1Genetic diversity and construction of core collection in Chinese wheat genetic resources显示文摘Genetic diversity among 5029 accessions representing a proposed Chinese wheat core collection was analyzed using 78 pairs of fluorescent microsatellite (SSR) primers mapped to 21 chromosomes. A stepwise hierarchical sampling strategy with priority based on 4×105 SSR data-points was used to construct a core collection from the 23090 initial collections. The core collection consisted of 1160 accessions, including 762 landraces, 348 modern varieties and 50 introduced varieties. The core ac-counts for 23.1% of the 5029 candidate core accessions and 5% of the 23090 initial collections, but retains 94.9% of alleles from the candidate collections and captures 91.5% of the genetic variation in the initial collections. These data indicate that it is possible to maintain genetic diversity in a core collection while retaining fewer accessions than the accepted standard, i.e., 10% of the initial collections captured more than 70% of their genetic diversity. Estimated genetic representation of the core con-structed by preferred sampling (91.5%) is much higher than that by random sampling (79.8%). Both mean genetic richness and genetic diversity indices of the landraces were higher than those of the modern varieties in the core. Structure and principal coordinate analysis revealed that the landraces and the modern varieties were two relatively independent subpopulations. Strong genetic differentiation associated with ecological environments has occurred in the landraces, but was relatively weak in the modern cultivars. In addition, a mini-core collection was constructed, which consisted of 231 accessions with an estimated 70% representation of the genetic variation from the initial collections. 4Anti-wear and friction performance of ZrO_2 nanoparticles as lubricant additive显示文摘The surface of ZrO2 nanoparticles was modified with silane coupling agent KH-560 by dispersing the nanoparticles in 20#machine oil.The tribological performance of the 20#machine oil with ZrO2 nanoparticles was evaluated by a four-ball tester and a thrust-ring machine.The loss of the thrust-ring weight was measured by an electronic balance with a precision of 0.0001 g.The worn surface was examined under a metallographic microscope with 400-times magnification.The results showed that the average friction coefficient decreased by 27.34%,and that the weight loss of the thrust-ring indicated no wear or negative wear,the wear loss after six loads tests being-0.0163 g.The anti-wear(AW)and reducing friction(RF)abilities of the oil with ZrO2 nanoparticles addition were considerably improved,maximizing at an additive concentration of 0.5wt%.Shiyu Ma Shaohua Zheng Duxia Cao Hongna Guo 2010Particuology2010,8,5:11

6Analysis of coherent structures in drag-reducing polymer solution flow based on proper orthogonal decomposition显示文摘Direct numerical simulation(DNS) of forcing homogeneous isotropic turbulence with polymers was performed.In order to understand the polymers effect on turbulent coherent structures,proper orthogonal decomposition was performed to identify coherent structures based on DNS data,so as to analyze the remarkable difference due to the addition of polymers.The results showed that the numbers for eigenmodes required for capturing coherent structures were 32 and 24 for the Newtonian fluid and polymer solution flows,respectively,which means the decrease of the complexity in polymer solution flow.Through the POD energy spectrum,it was found that the turbulent kinetic energy is distributed onto a large number of eigenmodes whether in the Newtonian fluid flow or polymer solution flow,suggesting that polymer solution flow is still turbulent in one aspect.Besides,the POD eigenmodes were investigated,which found that the small-scale structures are inhibited in polymer solution flow.CAI WeiHua LI FengChen ZHANG HongNa WANG Yue WANG Lu 2012Science China(Physics,Mechanics & Astronomy)2012,55,5:5
7Optimization of Extraction of Total Flavonoids from HERBA BLUMEAE RIPARIAE Using Response Surface Methodology显示文摘[Objectives]This study aimed to optimize the ultrasonic-assisted extraction process of total flavonoids from a Zhuang medicine HERBA BLUMEAE RIPARIAE using response surface methodology.[Methods]Using rutin as a reference substance,a standard curve was drawn at a wavelength of 292 nm.Content determination was performed through absorbance under different conditions.The effects of ultrasonic time,ethanol volume fraction and liquid-to-solid ratio on the yield of total flavonoids from HERBA BLUMEAE RIPARIAE were investigated by single-factor tests.By using Box-Behnken test,the ultrasonic-assisted extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was optimized.[Results]The optimal extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was as follows:ultrasonic time of 30 min,ethanol volume fraction of 50%and liquid-to-solid ratio of 40∶1(mL/g).Under the optimal extraction conditions,the average yield of total flavonoids from HERBA BLUMEAE RIPARIAE was 85.47 mg/g.[Conclusions]The optimized extraction process was convenient and feasible.This study will provide a reference for the extraction of total flavonoids from HERBA BLUMEAE RIPARIAE,and provide certain guidance and basis for further development and utilization of the plan resource of Blumea riparia(Bl.)DC.Hongna SU Yining ZHANG Xuexue LI Xuelian ZHAO Yisong LI Wenbing LI Yuan LIU 2020Medicinal Plant2020,11,2:5
8Simultaneous extraction of DNA and RNA from Escherichia coli BL 21 based on silica-coated magnetic nanoparticles显示文摘The extraction of nucleic acid is recognized as one of the most essential steps in molecular biology for initiating other downstream applications such as sequencing, amplification, hybridization, and cloning. Many commercial kits and methods are currently available that allow the extraction of only one type of nucleic acids-DNA or RNA. However, in parallel clinical detection of several diseases, a method for simultaneous extraction of both DNA and RNA from the same source is needed in such cases. In this study, a method for simultaneous extraction of DNA and RNA from bacteria based on magnetic nanoparticles(MNPs) was described. Lysis buffers were prepared to help the nucleic acid released and adsorbed to MNPs. Then, two washing buffers were used to remove the contamination of proteins and carbohydrates. The nucleic acids were finally eluted by Deoxyribonuclease(DNase) and Ribonucleases(RNase) free water. Different factors which might affect the purification of the nucleic acid were investigated, and the quantity and quality parameters of the nucleic acid were also recorded. The DNA and RNA extracted from bacteria were then respectively subjected to polymerase chain reaction(PCR) and reverse transcription PCR(RT-PCR) to further confirm its quality. 9Overexpressed Cyclin D1 and CDK4 proteins are responsible for the resistance to CDK4/6 inhibitor in breast cancer that can be reversed by PI3K/mTOR inhibitors显示文摘CDK4/6 inhibitors are the standard treatment in advanced HR+/HER2-breast cancer patients.Nevertheless,the resistance to CDK4/6 inhibitors is inevitable and the strategies to overcome resistance are of great interest.Here,we show that the palbociclibresistant breast cancer cells expressed significantly higher levels of Cyclin D1 and CDK4 proteins because of upregulated protein synthesis.Silencing Cyclin D1 or CDK4 led to cell cycle arrest while silencing Cyclin E1 or CDK2 restored the sensitivity to palbociclib.Furthermore,PI3K/mTOR pathway was hyper-activated in palbociclib-resistant cells,leading to more phosphorylated 4E-BP1 and higher levels of Cyclin D1 and CDK4 translation.Targeting PI3K/mTOR pathway with a specific PI3Kαinhibitor(BYL719)or an mTOR inhibitor(everolimus)reduced the protein levels of Cyclin D1 and CDK4,and restored the sensitivity to palbociclib.The tumor samples expressed significantly higher levels of Cyclin D1,CDK4,p-AKT and p-4E-BP1 after progression on palbociclib treatment.In conclusion,our findings suggest that overexpressed Cyclin D1 and CDK4 proteins lead to the resistance to CDK4/6 inhibitor and PI3K/mTOR inhibitors are able to restore the sensitivity to CDK4/6 inhibitors,which provides the biomarker and rationale for the combinational use of CDK4/6 inhibitors and PI3K/mTOR inhibitors after CDK4/6 inhibitor resistance in breast cancer.Zijie Cai Jingru Wang Yudong Li Qianfeng Shi Liang Jin Shunying Li Mengdi Zhu Qi Wang Lok Lam Wong Wang Yang Hongna Lai Chang Gong Yandan Yao Yujie Liu Jun Zhang Herui Yao Qiang Liu 2023Science China(Life Sciences)2023,66,1:4

11Mesoscale SST perturbation-induced impacts on climatological precipitation in the Kuroshio-Oyashio extension region,as revealed by the WRF simulations显示文摘Mesoscale coupling between perturbations of mesoscale sea surface temperature (SST) and lowlevel winds has been extensively studied using available high-resolution satellite observations. However, the climatological impacts of mesoscale SST perturbations (SST meso ) on the free atmosphere have not been fully understood. In this study, the rectified eff ect of SSTmeso on local climatological precipitation in the Kuroshio- Oyashio Extension (KOE) region is investigated using the Weather Research and Forecasting (WRF) Model;two runs are performed, one forced by low-resolution SST fields (almost no mesoscale signals) and another by additional high-resolution SSTmeso fields extracted from satellite observations. Climatological precipitation response to SST meso is characterized mainly by enhanced precipitation on the warmer flank of three oceanic SST fronts in this region. The results show that the positive correlation between the 10-m wind speed perturbations and SSTmeso is well captured by the WRF model with a reasonable spatial pattern but relatively weak strength. The addition of SSTmeso improves the climatological precipitation simulated by WRF with a better representation of fine-scale structures compared with satellite observations. A closer examination on the underlying mechanism suggests that while the pressure adjustment mechanism can explain the climatological precipitation enhancement along the fronts and the relatively high contribution of the convective precipitation, other factors such as synoptic events should also be taken into consideration to account for the seasonality of the precipitation response.GAO Jiaxiang ZHANG Rong-Hua WANG Hongna 2019Journal of Oceanology and Limnology2019,37,2:3

13LIN9 confers paclitaxel resistance in triple negative breast cancer cells by upregulating CCSAP显示文摘LIN9 functions to regulate cell mitotic process.Dysregulation of LIN9 expression is associated with development of human cancers.In this study we assessed the association of LIN9 expression with paclitaxel resistance and clarified the underlying mechanisms for the first time.LIN9 expression in breast cancer tissues was retrieved from publicly available online databases and statistically analyzed.Human TNBC cell lines MDA-MB-231 and MDA-MB-468 and their corresponding paclitaxelresistant sublines 231PTX and 468PTX were used to assess the expression of LIN9 by qRT-PCR and Western blot,cell growth by cell counting,cell viability by MTS assay,and cell apoptosis by flow cytometry.The data showed that high LIN9 expression in breast cancer patients receiving chemotherapy was related to poor overall survival(OS).LIN9 expression was upregulated in paclitaxel-resistant TNBC cells compared to their parental cells.Knockdown of LIN9 or treatment of paclitaxel-resistant TNBC cells with a bromo-and extra-terminal domain inhibitor(BETi)JQ1 which also decreased LIN9 expression enhanced the sensitivity of paclitaxel-resistant TNBC cells to paclitaxel.Mechanistically,decreased LIN9 in resistant cell lines reduced tumor cell viability,promoted multinucleated cells formation and induced tumor cell apoptosis,potentially by directly regulating microtubule-binding protein CCSAP.In conclusion,high LIN9 expression contributed to poor clinical outcomes and paclitaxel resistance in TNBC and BETi,targeting LIN9 expression,could be a reversible drug for PTX-resistant TNBC patients.Hongna Lai Rui Wang Shunying Li Qianfeng Shi Zijie Cai Yudong Li Yujie Liu 2020Science China(Life Sciences)2020,63,3:2
14Representing surface wind stress response to mesoscale SST perturbations in western coast of South America using Tikhonov regularization method显示文摘Interaction between mesoscale perturbations of sea surface temperature(SSTmeso)and wind stress(WSmeso)has great influences on the ocean upwelling system and turbulent mixing in the atmospheric boundary layer.Using daily Quik-SCAT wind speed data and AMSR-E SST data,SSTmeso and WSmeso fields in the western coast of South America are extracted by using a locally weighted regression method(LOESS).The spatial patterns of SSTmeso and WSmeso indicate strong mesoscale SST-wind stress coupling in the region.The coupling coefficient between SSTmeso and WSmeso is about 0.0095 N/(m^2·℃)in winter and 0.0082 N/(m^2·℃)in summer.Based on mesoscale coupling relationships,the mesoscale perturbations of wind stress divergence(Div(WSmeso))and curl(Curl(WSmeso))can be obtained from the SST gradient perturbations,which can be further used to derive wind stress vector perturbations using the Tikhonov regularization method.The computational examples are presented in the western coast of South America and the patterns of the reconstructed WS meso are highly consistent with SSTmeso,but the amplitude can be underestimated significantly.By matching the spatially averaged maximum standard deviations of reconstructed WSmeso magnitude and observations,a reasonable magnitude of WSmeso can be obtained when a rescaling factor of 2.2 is used.As current ocean models forced by prescribed wind cannot adequately capture the mesoscale wind stress response,the empirical wind stress perturbation model developed in this study can be used to take into account the feedback effects of the mesoscale wind stress-SST coupling in ocean modeling.Further applications are discussed for taking into account the feedback effects of the mesoscale coupling in largescale climate models and the uncoupled ocean models.CUI Chaoran ZHANG Rong-Hua WANG Hongna WEI Yanzhou 2020Journal of Oceanology and Limnology2020,38,3:2
15Electromyography and Fos immunostaining study establish a possible functional link between trigeminal proprioception and the oculomotor system in rats显示文摘The objective of this study was to explore whether there was a functional link between trigeminal proprioception and the oculomotor system mediated through jaw muscle afferents.Electromyography(EMG) was undertaken of the levator palpebrae(LP) and superior rectus(SR),and Fos expression was detected in the brainstem following consecutive down-stretching of the lower jaw at 2-4 Hz in rats.Retrograde tracing was undertaken of the interstitial nucleus of Cajal and Darkschewitsch nucleus(INC/DN) pre-oculomotor neurons.FMG-like responses were recorded from the LP/SR during down-stretching of the lower jaw at 2-4 Hz in 3 out of 11 rats.Fos expression was induced by consecutive down-stretching of the lower jaw at 2-4 Hz for 20-30 seconds.Interestingly,Fos expression was distributed mainly in the bilateral INC/DN area.We also examined Fos-like immunoreactivity in central mesencephalic and paramedian pontine reticular formation that harbors premotor neurons controlling horizontal eye movement,but no Fos-like staining was observed therein.By injection of retrograde tracers into the oculomotor nucleus combined with Fos immunostaining,double labeled pre-oculomotor neurons were visualized to distribute in the INC/DN.In conclusions,there may exist a trigeminal proprioceptive-oculomotor system neural circuit through jaw muscle afferents in rats.Judging from Fos distribution pattern,this pathway might be related to vertical and torsional eye movements.Houcheng Liang Jingdong Zhang Pifu Luo Hongna Zhu Ying Qiao Anle Su Ting Zhang 2017The Journal of Biomedical Research2017,31,3:2
16Optimization of Ultrasonic-enzymatic Extraction of Total Flavonoids from Cardamine tangutorum O.E.Schulz by Response Surface Methodology and Content Determination显示文摘[Objectives]This study aimed to optimize the extraction process of total flavonoids in Cardamine tangutorum O.E.Schulz.[Methods]On the basis of single-factor experiments,the ultrasonic-enzymatic extraction process of total flavonoids in C.tangutorum O.E.Schulz was optimized by Box-Behnken design from aspects of ethanol volume fraction,extraction time,extraction temperature and enzyme addition,with yield of total flavonoids as the evaluation index.[Results]The optimal extraction process of total flavonoids in C.tangutorum O.E.Schulz was as follows:ethanol volume fraction of 70%,ultrasonic extraction time of 43 min,ultrasonic extraction temperature of 37℃and cellulose addition of 0.07 mg.Under the optimal conditions,the yield of total flavonoids from C.tangutorum O.E.Schulz reached 75.90 mg/g.[Conclusions]The optimized extraction process was reasonable and feasible,and it could provide a reference for the extraction of total flavonoids in C.tangutorum O.E.Schulz.This study provided a foundation for the quality control of food/medicinal materials and the development and research of big health products of C.tangutorum O.E.Schulz.Xiaoxia CAI Bochao LI Xuexue LI Hongna SU Wenbing LI Yuan LIU 2020Medicinal Plant2020,11,6:2
17Radiation pre-vulcanization effect on properties of the truck tyre's transition layer and the truck tyre显示文摘In this paper,the natural rubber is chosen as the main constituents for the transition layer of all-steel load radial tyre,which is pre-vulcanized by 500-keV E-beam irradiation of up to 60 kGy.The results show that the green strength of transitional layer increases with the dose,reaching four times as much as the control(without irradiation) at 60 kGy.The final mechanical properties do not differ significantly from those of the control except that the aging and fatigue performance increased.However,thickness of the natural rubber transitional layer for an average single tyre can be reduced by 1 mm(or 1.5 kg) without obvious adverse effect on tyre performance.YANG Mingcheng ZHANG Benshang ZHU Jun LI Zhaopeng LI Kunhao GUO Dongquan ZHANG Hongna 2013Nuclear Science and Techniques2013,24,2:2
