| 6 | Solute-homogenization model and its experimental verification in Mg-Gd-based alloys显示文摘Composition homogenization in solid solution is important for industrial alloys. In the present work, a solute homogenization model is proposed based on the chemical short-range-order tendency in Mg-Gdbased alloys. After a calculation using the cluster-plus-glue-atom model, the stable Mg-Gd structural unit is derived, [Gd-Mg12]Mg6, where one solute Gd is nearest-neighbored with twelve Mg atoms to form the characteristic hcp cluster [Gd-Mg12] and this cluster is matched with six Mg glue atoms. Such a local unit is then mixed with [Mg-Mg12]Mg3, the stable unit for pure Mg. Assuming that the Gd-containing units are arranged in fcc-or bcc-like lattice points and the Mg units in their octahedral interstices, three proportions between the two units are obtained, 1:1, 2:3, and 1:3, which constitute three solute homogenization modes. The prevailing Mg-Gd-based alloys are consequently classified into three groups, respectively exemplified by GW103 K(Mg-10 Gd-3 Y-0.4 Zr, wt%), GW83 K(Mg-8 Gd-3 Y-0.4 Zr), and GW63 K(Mg-6 Gd-3 Y-0.4 Zr). Mg-Gd-Y-Zr alloys were designed following the model(where Y and Zr were also added in substitution for Gd) and prepared by permanent-mould casting. | 7 | Age and menopausal status are important factors influencing the serum human epididymis secretory protein 4 level:a prospective cross-sectional study in healthy Chinese people显示文摘Background::Human epididymis secretory protein 4(HE4)is a new ovarian cancer biomarker.The factors influencing HE4 levels are not clear,and the reference data in China are limited.Here,we aim to evaluate the effects of menopause and age on HE4 levels and to provide a possible reference value for HE4 in healthy Chinese people.Methods::A total of 2493 healthy females aged 40 years or older were recruited from March 2013 to March 2017 with the cooperation of four medical institutions across Beijing,China.The serum levels of HE4 and cancer antigen 125(CA125)were measured by enzyme-linked immunosorbent assay.The Wilcoxon rank-sum test of variance and a stratified analysis were used to analyze the relationships among age,menopausal status,and levels of HE4 or CA125.Confidence intervals(5%-95%)were determined for reference ranges in different populations.Results::There was a statistically significant difference in median HE4 levels between the post-menopausal(n=2168)and premenopausal groups(n=325)(36.46 vs.24.04 pmol/L,Z=-14.41,P<0.001).HE4 increased significantly with age in the post-menopausal groups(H=408.18,P<0.001)but not in the pre-menopausal subjects(Z=-0.43,P=0.67).The upper 95th percentile of HE4 levels were 44.63 pmol/L for pre-menopausal women,78.17 pmol/L for post-menopausal women,and 73.3 pmol/L for all women.In the post-menopausal population,the HE4 reference ranges were 13.15 to 47.31,14.31 to 58.04,17.06 to 73.51,24.50 to 115.25,and 35.71 to 212.37 pmol/L for different age groups from forty divided by decade.The CA125 level was affected mainly by menopausal status and not age.Conclusions::Menopausal status and age were both important factors influencing the level of HE4,and age affected HE4 levels mainly in post-menopausal women.The HE4 level was higher in the post-menopausal population than in the pre-menopausal population and increased with age. | Hong-Yan Cheng Lin Zeng Xue Ye Rui-Qiong M a Zhi-Jian Tang Hong-Ling Chu Yi-M ing Zhao Li-Rong Zhu Yu-Nong Gao Xiao-Hong Chang Heng Cui | 2020 | Chinese Medical Journal2020,,11: | 6 |
| 8 | Spatio-temporal variation in rock exhumation linked to large-scale shear zones in the southeastern Tibetan Plateau显示文摘Crustal-scale shear zones are believed to have played an important role in the tectonic and landscape evolution of orogens. However, the variation of long-term rock exhumation between the interior of shear zones and adjacent regions has not been documented in detail. In this study, we obtained new zircon U-Pb, biotite ^(40)Ar/^(39)Ar, zircon and apatite(U-Th)/He data, and conducted inverse thermal history modeling from two age-elevation profiles(the Pianma and Tu'er profiles) in the southeastern Tibetan Plateau. Our goal is to constrain the exhumation history of the Gaoligong and Chongshan shear zones and adjacent regions, so as to explore the effect of the shear zones on exhumation and their thermal effect on cooling that should not be ignored. Our results suggest that during the interval of 18–11 Ma the exhumation rates of rocks within the Gaoligong shear zone are anomalously high compared with those outside of. The rapid cooling during 18–11 Ma appears to be restricted to the shear zone, likely due to localized thermal effects of shearing and exhumation. After 11 Ma, both the areas within and outside of the shear zones experienced a similar two-stage exhumation history: slower cooling until the early Pliocene, and then a rapid increase in cooling rate since the early Pliocene. Our results indicate a synchronized exhumation but with spatially varied exhumation rates. | 13 | Electrochemical properties of carbon-coated LiFePO_4 and LiFe_(0.98)Mn_(0.02)PO_4 cathode materials synthesized by solid-state reaction显示文摘Olivine structured LiFePO 4 /C (lithium iron phosphate) and Mn 2+ -doped LiFe 0.98 Mn 0.02 PO 4 /C powders were synthesized by the solid-state reaction. The effects of manganese partial substitution and different carbon content coating on the surface of LiFePO 4 were considered. The structures and electrochemical properties of the samples were measured by X-ray diffraction (XRD), cyclic voltammetry (CV), charge/discharge tests at different current densities, and electrochemical impedance spectroscopy (EIS). The electrochemical properties of LiFePO 4 cathodes with x wt.% carbon coating (x= 3, 7, 11, 15) at =0.2C, 2C (1C= 170 mAh·g 1 ) between 2.5 and 4.3 V were investigated. The measured results mean that the LiFePO 4 with 7 wt.% carbon coating shows the best rate performance. The discharge capacity of LiFe 0.98 Mn 0.02 PO 4 /C composite is found to be 165 mAh·g 1 at a discharge rate, = 0.2C, and 105 mAh·g 1 at =2C, respectively. After 10 cycles, the discharge capacity has rarely fallen, while that of the pristine LiFePO 4 /C cathode is 150 mAh·g 1 and 98 mAh·g 1 at =0.2 and 2C, respectively. | 17 | Multi-omics approaches identify SF3B3 and SIRT3 as candidate autophagic regulators and druggable targets in invasive breast carcinoma显示文摘Autophagy is a critical cellular homeostatic mechanism,and its dysfunction is linked to invasive breast carcinoma(BRCA).Recently,several omics methods have been applied to explore autophagic regulators in BRCA;however,more reliable and robust approaches for identifying crucial regulators and druggable targets remain to be discovered.Thus,we report here the results of multi-omics approaches to identify potential autophagic regulators in BRCA,including gene expression(EXP),DNA methylation(MET)and copy number alterations(CNAs)from The Cancer Genome Atlas(TCGA).Newly identified candidate genes,such as SF3 B3,TRAPPC10,SIRT3,MTERFD1,and FBXO5,were confirmed to be involved in the positive or negative regulation of autophagy in BRCA.SF3 B3 was identified firstly as a negative autophagic regulator,and siRNA/shRNA-SF3 B3 were shown to induce autophagyassociated cell death in in vitro and in vivo breast cancer models.Moreover,a novel small-molecule activator of SIRT3,1-methylbenzylamino amiodarone,was discovered to induce autophagy in vitro and in vivo.Together,these results provide multi-omics approaches to identify some key candidate autophagic regulators,such as the negative regulator SF3 B3 and positive regulator SIRT3 in BRCA,and highlight SF3 B3 and SIRT3 as new druggable targets that could be used to fill the gap between autophagy and cancer drug development. | Shouyue Zhang Jin Zhang Yang An Xiaoxi Zeng Ziyi Qin Yuqian Zhao Heng Xu Bo Liu | 2021 | Acta Pharmaceutica Sinica B2021,11,5: | 3 |

| 19 | An enrichment model using regular health examination data for early detection of colorectal cancer显示文摘Objective: Challenges remain in current practices of colorectal cancer(CRC) screening, such as low compliance,low specificities and expensive cost. This study aimed to identify high-risk groups for CRC from the general population using regular health examination data.Methods: The study population consist of more than 7,000 CRC cases and more than 140,000 controls. Using regular health examination data, a model detecting CRC cases was derived by the classification and regression trees(CART) algorithm. Receiver operating characteristic(ROC) curve was applied to evaluate the performance of models. The robustness and generalization of the CART model were validated by independent datasets. In addition, the effectiveness of CART-based screening was compared with stool-based screening.Results: After data quality control, 4,647 CRC cases and 133,898 controls free of colorectal neoplasms were used for downstream analysis. The final CART model based on four biomarkers(age, albumin, hematocrit and percent lymphocytes) was constructed. In the test set, the area under ROC curve(AUC) of the CART model was 0.88 [95%confidence interval(95% CI), 0.87-0.90] for detecting CRC. At the cutoff yielding 99.0% specificity, this model’s sensitivity was 62.2%(95% CI, 58.1%-66.2%), thereby achieving a 63-fold enrichment of CRC cases. We validated the robustness of the method across subsets of test set with diverse CRC incidences, aging rates, genders ratio, distributions of tumor stages and locations, and data sources. Importantly, CART-based screening had the higher positive predictive value(1.6%) than fecal immunochemical test(0.3%).Conclusions: As an alternative approach for the early detection of CRC, this study provides a low-cost method using regular health examination data to identify high-risk individuals for CRC for further examinations.
