1在心血管的疾病的先进 glycation 结束产品的角色显示文摘 Advanced glycation end products(AGEs) are produced through the non enzymatic glycation and oxidation of proteins,lipids and nucleic acids.Enhanced formation of AGEs occurs particularly in conditions associated with hyperglycaemia such as diabetes mellitus(DM).AGEs are believed to have a key role in the development and progression of cardiovascular disease in patients with DM through the modif ication of the structure,function and mechanical properties of tissues through crosslinking intracellular as well as extracellular matrix proteins and through modulating cellular processes through binding to cell surface receptors [receptor for AGEs(RAGE)].A number of studies have shown a correlation between serum AGE levels and the development and severity of heart failure(HF).Moreover,some studies have suggested that therapies targeted against AGEs may have therapeutic potential in patients with HF.The purpose of this review is to discuss the role of AGEs in cardiovascular disease and in particular in heart failure,focussing on both cellular mechanisms of action as well as highlighting how targeting AGEs may represent a novel therapeutic strategy in the treatment of HF.Zeinab Hegab Stephen Gibbons Ludwig Neyses Mamas A Mamas 2012World Journal of Cardiology2012,4,4:58
2小鼠肺上皮干细胞体外增殖和分化特性及其影响因素的初步研究显示文摘目的 初探几种细胞组分、细胞因子、信号转导分子等在肺上皮干细胞体外增殖和分化过程中的调控作用.方法 采用细胞外基质替代物Matrigel三维体外培养体系,辅以免疫组织化学分析,观察几种常见的对干细胞增殖分化发挥重要作用的因素对肺上皮干细胞体外集落形成数量、集落大小及分化潜能的影响.结果 小鼠肺上皮干细胞需与成纤维细胞共培养才能在体外形成集落,包括A型、B型和C型3种,它们各具代表性的形态特征和分化谱特点,成纤维细胞通过密切接触与肺上皮干细胞发挥相互作用.肝内皮细胞和肺内皮细胞均可以支持肺上皮干细胞集落生长.成纤维细胞生长因子(FGF)显著增加所有类型集落的数目和体积,其作用下各型集落基底细胞标志消失,而肺泡上皮细胞标志表达增加,其中FGF-9的效应不被其受体抑制剂消除;人白血病抑制因子(LIF)、活化素受体样激酶5抑制剂(ALK5-I)或Rho相关蛋白激酶抑制剂(ROCK-I)处理后,肺上皮干细胞形成的集落数量和体积均得以增加,但对集落类型和分化谱影响各异.结论 小鼠肺上皮干细胞生长依赖成纤维细胞;内皮细胞对肺上皮干细胞增殖潜能作用有限;FGF促进肺上皮干细胞增殖和分化;LIF、ALK5-I或ROCK-I亦促进肺上皮干细胞的增殖和分化.研究结果为小鼠肺上皮干细胞的生物学特性及其影响因素的深入研究奠定基础,为未来肺上皮干细胞修复性治疗相关疾病和损伤提供了实验学依据.高静韬 Ahmed Hegab 黑田葵 荒井大辅 刘宇红 李琦 2018国际呼吸杂志2018,38,8:3

15几种影响小鼠气管上皮基底干细胞体外增殖及分化潜能因素的研究显示文摘目的探讨多种细胞组分、细胞因子、信号转导分子等在气管上皮基底干细胞(airway basal stem cells,ABSCs)体外增殖和分化过程中的调控作用。方法采用细胞外基质替代物Matrigel三维体外培养体系,辅以免疫组化分析,观察了几种常见的对干细胞增殖分化发挥重要作用的因素对ABSCs体外集落形成数量、集落大小及分化潜能的影响。结果成纤维细胞较内皮细胞能显著提高ABSCs集落增殖和分化潜能。成纤维细胞生长因子(FGFs)能增加ABSCs体外集落形成数量与直径,并诱导其向分泌性细胞分化。同时给予FGFs及其受体阻滞剂处理,FGFs诱导的集落增殖效应消失。LIF、ALK5-I或ROCK—I处理后,ABSCs形成的集落数量和直径也显著增加,并诱导其向纤毛上皮细胞或分泌性细胞分化。结论小鼠ABSCs不依赖成纤维细胞,但共培养能增强其增殖和分化能力;内皮细胞对ABSCs增殖和分化能力促进作用有限;FGFs促进ABSCs的增殖和分化,其效应能被其受体阻滞剂消除;LIF、ALK5-I或ROCK—I亦促进ABSCs的增殖和分化。研究结果将为小鼠ABSCs的生物学特性及其影响因素的深入研究奠定基础,为未来ABSCs修复性治疗相关疾病和损伤提供了实验学依据。高静韬 Ahmed Hegab 黑田葵 荒井大辅 刘宇红 李琦 2017国际呼吸杂志2017,37,15:1
