1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 22018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 8Forming limits under multi-index constraints in NC bending of aluminum alloy thin-walled tubes with large diameters显示文摘With increasing diameters of aluminum alloy thin-walled tubes (AATTs), the tube forming limits, i.e. the minimum bending factors, and their predictions under multi-index constraints including wrinkling, thinning and flattening have been being a key problem to be urgently solved for improving tube forming potential in numerical control (NC) bending processes of AATTs with large diameters. Thus in this paper, a search algorithm of the forming limits is put forward based on a 3D elastic-plastic finite element (FE) model and a wrinkling energy prediction model for the bending processes under axial compression loading (ACL) or not. This algorithm enables to be considered the effects of process parameter combinations including die, friction parameters on the multi-indices. Based on this algorithm, the forming limits of the different size tubes are obtained, and the roles of the process parameter combinations in enabling the limit bending processes are also revealed. The followings are found: the first, within the appropriate ranges of friction and clearances between the different dies and the tubes enabling the bending processes with smaller bending factors, the ACL enables the tube limit bending processes after a decrease of the mandrel ball thickness and diameters; then, without considering the effects of the tube geometry sizes on the tube constitutive equations, the forming limits will be decided by the limit thinning values for the tubes with diameters smaller than 80 mm, while the wrinkling for the tubes with diameters no less than 80 mm. 9Numerical modeling and dynamic analysis of the 2017 Xinmo landslide in Maoxian County, China显示文摘A catastrophic landslide occurred at Xinmo village in Maoxian County, Sichuan Province,China, on June 24, 2017. A 2.87×106 m3 rock mass collapsed and entrained the surface soil layer along the landslide path. Eighty-three people were killed or went missing and more than 103 houses were destroyed. In this paper, the geological conditions of the landslide are analyzed via field investigation and high-resolution imagery. The dynamic process and runout characteristics of the landslide are numerically analyzed using a depth-integrated continuum method and Mac Cormack-TVD finite difference algorithm.Computational results show that the evaluated area of the danger zone matchs well with the results of field investigation. It is worth noting that soil sprayed by the high-speed blast needs to be taken into account for such kind of large high-locality landslide. The maximum velocity is about 55 m/s, which is consistent with most cases. In addition, the potential danger zone of an unstable block is evaluated. 13Repurposing of clinically approved drugs for treatment of coronavirus disease 2019 in a 2019-novel coronavirus-related coronavirus model显示文摘Background:Medicines for the treatment of 2019-novel coronavirus(2019-nCoV)infections are urgently needed.However,drug screening using live 2019-nCoV requires high-level biosafety facilities,which imposes an obstacle for those institutions without such facilities or 2019-nCoV.This study aims to repurpose the clinically approved drugs for the treatment of coronavirus disease 2019(COVID-19)in a 2019-nCoV-related coronavirus model.Methods:A 2019-nCoV-related pangolin coronavirus GX_P2V/pangolin/2017/Guangxi was described.Whether GX_P2V uses angiotensin-converting enzyme 2(ACE2)as the cell receptor was investigated by using small interfering RNA(siRNA)-mediated silencing of ACE2.The pangolin coronavirus model was used to identify drug candidates for treating 2019-nCoV infection.Two libraries of 2406 clinically approved drugs were screened for their ability to inhibit cytopathic effects on Vero E6 cells by GX_P2V infection.The anti-viral activities and anti-viral mechanisms of potential drugs were further investigated.Viral yields of RNAs and infectious particles were quantified by quantitative real-time polymerase chain reaction(qRT-PCR)and plaque assay,respectively.Results:The spike protein of coronavirus GX_P2V shares 92.2%amino acid identity with that of 2019-nCoV isolate Wuhanhu-1,and uses ACE2 as the receptor for infection just like 2019-nCoV.Three drugs,including cepharanthine(CEP),selamectin,and mefloquine hydrochloride,exhibited complete inhibition of cytopathic effects in cell culture at 10μmol/L.CEP demonstrated the most potent inhibition of GX_P2V infection,with a concentration for 50%of maximal effect[EC50]of 0.98μmol/L.The viral RNA yield in cells treated with 10μmol/L CEP was 15,393-fold lower than in cells without CEP treatment([6.48±0.02]×10-4vs.1.00±0.12,t=150.38,P<0.001)at 72 h post-infection(p.i.).Plaque assays found no production of live viruses in media containing 10μmol/L CEP at 48 h p.i.Furthermore,we found CEP had potent anti-viral activities against both viral entry(0.46±0.12,vs.1.00±0.37,t=2.42,P<0.05)and viral replication([6.18±0.95]×10-4vs.1.00±0.43,t=3.98,P<0.05).Conclusions:Our pangolin coronavirus GX_P2V is a workable model for 2019-nCoV research.CEP,selamectin,and mefloquine hydrochloride are potential drugs for treating 2019-nCoV infection.Our results strongly suggest that CEP is a wide-spectrum inhibitor of pan-betacoronavirus,and further study of CEP for treatment of 2019-nCoV infection is warranted.Hua-Hao Fan Li-Qin Wang Wen-Li Liu Xiao-Ping An Zhen-Dong Liu Xiao-Qi He Li-Hua Song Yi-Gang Tong 2020Chinese Medical Journal2020,,9:15
14Geochronological evidence of Indosinian(high-pressure) metamorphic event and its tectonic significance in Taxkorgan area of the Western Kunlun Mountains,NW China显示文摘The CL images,LA-ICP-MS in situ trace elements analysis,and U-Pb dating for zircons indicate that the metamorphic ages of the sillimanite-garnet-biotite gneiss and the garnet-amphibole gneiss from eastern Taxkorgan of the Western Kunlun Mountains are 220±2 and 220±3 Ma respectively,and their protolith ages are younger than 253±2 and 480±8 Ma respectively.Two samples were collected at the same outcrops with HP mafic granulite and HP pelitic granulite.Mineral assemblage of the sillimanite-garnet-biotite gneiss(Grt+Sill+Per+Q) is consistent with that of HP pelitic granulite at early high amphibolite-granulite facies stage.Mineral assemblage of the garnet-amphibole gneiss(Grt+Amp+Pl+Q) is consistent with retro-metamorphic assemblage of HP mafic granulite at amphibolite facies stage.The dating results suggest that these HP granulites underwent peak metamorphism at 220±2 to 253±2 Ma.Thus,the Kangxiwar tectonic zone was probably formed by subduction and collision of the Paleo-Tethys Ocean during Indosinian.Protolith ages of the two samples,together with previously published U-Pb zircon dating age,suggest that the sillimanite-garnet schist-quartzite unit is a late Paleozoic unit,not a part of the Paleoproterozoic Bulunkuole Group.YANG WenQiang1,LIU Liang1,CAO YuTing1,WANG Chao1,2,HE ShiPing2,LI RongShe2 & ZHU XiaoHui1 1 State Key Laboratory of Continental Dynamics,Department of Geology,Northwest University,Xi’an 710069,China 2 Xi’an Institute of Geology and Mineral Resource,Xi’an 710054,China 2010Science China Earth Sciences2010,53,10:13
15Effect of dilution holes on the performance of a triple swirler combustor显示文摘A triple swirler combustor is considered to be a promising solution for future high temperature rise combustors. The present paper aims to study dilution holes including primary dilution holes and secondary dilution holes on the performance of a triple swirler combustor. Experimental investigations are conducted at different inlet airflow velocities(40–70 m/s) and combustor overall fuel–air ratio with fixed inlet airflow temperature(473 K) and atmospheric pressure. The experimental results show that the ignition is very difficult with specific performance of high ignition fuel–air ratio when the primary dilution holes are located 0.6H(where H is the liner dome height)downstream the dome, while the other four cases have almost the same ignition performance. The position of primary dilution holes has an effect on lean blowout stability and has a large influence on combustion efficiency. 16Expression and identification of recombinant soluble single-chain variable fragment of monoclonal antibody MC3显示文摘AIM: To generate soluble single chain variable fragments (ScFv) of monoclonal antibody MC3 recognizing colorectal and gastric carcinomas.METHODS: mRNA was isolated from the hybridoma cell lineproducing MC3 and the DNAs encoding variable domains ofheavy and light chains(VH and VL) oftthe antibody wereamplified separately byRT-PCR and assembled into ScFvDNA with a linker DNAThe ScFv DNA was iigated into thephagemid vector pCANTAB5E and the ligated sample wastransformed into E. coil TG1. After two rounds of panning with gastric carcinomacell line AGS highly expressing MC3-binding antigen, thephage clones displaying ScFv fragments of the antibodywere selected by ELISA. 4 phage clones showing strongsignal in ELISA were used to infect E. coil HB2151 toexpress soluble ScFvs. The soluble ScFve were identified byDot blot and Western blot, and their antigen-binding activitywas assayed by ELISA. The VH and VL DNAs of the ScFvDNA derived from phage clone 19 were sequenced.RESULTS: The VH, VL and ScFv DNAs were about 340 bp,320 bp and 750 bp respectively. After two rounds of panningto the recombinant phages, 18 antigen-positive phageclones were selected from 30 preselected phage clones byELISA. All the soluble ScFvs derived from the 4 out of the 18antigen-positive phage clones were about Mr 32 000 andconcentrated in periplasmatic space under the given culturecondition. The soluble ScFvs could bind the antigen, andthey shared the same binding site with MC3. 19Pb isotope geochemistry of lead, zinc, gold and silver deposit clustered region, Liaodong rift zone,northeastern China显示文摘Pb isotopic analyses were reported for sulfide and hydrothermal carbonate minerals and marble of the Xiquegou lead-zinc, the Zhenzigou zinc-lead and the Gaojiapuzi silver deposits from the Qingchengzi ore field and the Beiwagou zinc-lead deposit in the west, Proterozoic Liaodong rift zone. Pb isotopic ratios of the marble from the Qingchengzi ore field range from 18.24 to 30.63 for 206 Pb/204Pb, 15.59 to 17.05 for207Pb/204Pb and 37.43 to 38.63 for 208Pb/204Pb. The marble gives a Pb-Pb isochron age of 1822±92 Ma, which is interpreted as the age of the metamorphism of the marble. Ore Pb, including Pb of sulfide and hydrothermal car- bonate minerals, from the Qingchengzi ore field shows limited variation with 206 Pb/204Pb=17.66- 17.96, 207 Pb/204Pb=15.60-15.74 and 208Pb/204Pb=37.94-38.60. In contrast, ore Pb from the Beiwagou deposit gives different Pb isotopic ratios with 206Pb/204Pb=15.68-15.81, 207 Pb/204Pb= 15.34-15.45 and 208Pb/204Pb=35.30-35.68. Pb of all deposits from the Liaodong rift zone is derived from the upper crust. Ore Pb of the Qingchengzi deposits is derived from a young upper crust. The model Th/U ratios of 4.40 to 4.74 for ore Pb are significantly different from that of 1.7 to 4.4 given by the marble of the Qingchengzi ore field, suggesting that marble is not the source of the ore Pb. Ore Pb of the Beiwagou deposit is extracted from its source and the deposit is formed at the Paleoproterozoic era.
