2Allyl isothiocyanate ameliorates lipid accumulation and inflammation in nonalcoholic fatty liver disease via the Sirt1/AMPK and NF-κB signaling pathways显示文摘BACKGROUND Allyl isothiocyanate(AITC),a classic anti-inflammatory and antitumorigenic agent,was recently identified as a potential treatment for obesity and insulin resistance.However,little is known about its direct impact on the liver.AIM To investigate the effect and underlying mechanism of AITC in nonalcoholic fatty liver disease(commonly referred to as NAFLD).METHODS To establish a mouse and cellular model of NAFLD,C57BL/6 mice were fed a high fat diet(HFD)for 8 wk,and AML-12 cells were treated with 200μM palmitate acid for 24 h.For AITC treatment,mice were administered AITC(100 mg/kg/d)orally and AML-12 cells were treated with AITC(20μmol/L).RESULTS AITC significantly ameliorated HFD-induced weight gain,hepatic lipid accumulation and inflammation in vivo.Furthermore,serum alanine aminotransferase and aspartate aminotransferase levels were markedly reduced in AITC-treated mice.Mechanistically,AITC significantly downregulated the protein levels of sterol regulatory elementbinding protein 1(SREBP1)and its lipogenesis target genes and upregulated the levels of proteins involved in fatty acidβ-oxidation,as well as the upstream mediators Sirtuin 1(Sirt1)and AMPactivated protein kinaseα(AMPKα),in the livers of HFD-fed mice.AITC also attenuated the nuclear factor kappa B(NF-κB)signaling pathway.Consistently,AITC relieved palmitate acid-induced lipid accumulation and inflammation in AML-12 cells in vitro through the Sirt1/AMPK and NF-κB signaling pathways.Importantly,further studies showed that the curative effect of AITC on lipid accumulation was abolished by siRNA-mediated knockdown of either Sirt1 or AMPKαin AML-12 cells.CONCLUSION AITC significantly ameliorates hepatic steatosis and inflammation by activating the Sirt1/AMPK pathway and inhibiting the NF-κB pathway.Therefore,AITC is a potential therapeutic agent for NAFLD.Chun-Xiao Li Jian-Guo Gao Xing-Yong Wan Yi Chen Cheng-Fu Xu Ze-Min Feng Hang Zeng Yi-Ming Lin Han Ma Ping Xu Chao-Hui Yu You-Ming Li 2019World Journal of Gastroenterology2019,25,34:14
3Platelet count combined with right liver volume and spleen volume measured by magnetic resonance imaging for identifying cirrhosis and esophageal varices显示文摘AIM: To determine whether the combination of platelet count(PLT) with spleen volume parameters and right liver volume(RV) measured by magnetic resonance imaging(MRI) could predict the Child-Pugh class of liver cirrhosis and esophageal varices(EV).METHODS: Two hundred and five cirrhotic patients with hepatitis B and 40 healthy volunteers underwent abdominal triphasic-enhancement MRI and laboratory examination of PLT in 109/L. Cirrhotic patients underwent endoscopy for detecting EV. Spleen maximal width(W), thickness(T) and length(L) in mm together with spleen volume(SV) and RV in mm3 were measured by MRI, and spleen volume index(SI) in mm3 was obtained by W × T × L. SV/PLT, SI/PLT and RV × PLT/SV(RVPS) were calculated and statistically analyzed to assess cirrhosis and EV.RESULTS: SV/PLT(r = 0.676) and SI/PLT(r = 0.707) increased, and PLT(r =-0.626) and RVPS(r =-0.802) decreased with the progress of Child-Pugh class(P < 0.001 for all). All parameters could determine the presence of cirrhosis, distinguish between each class of Child-Pugh class, and identify the presence of EV [the areas under the curve(AUCs) = 0.661-0.973]. 5Strength and failure characteristics of brittle jointed rock-like specimens under uniaxial compression:Digital speckle technology and a particle mechanics approach显示文摘The strength and failure characteristics of most natural rock mass are influenced by discontinues such as fissures, joints, and weak surfaces. In the present study, the strength and failure behavior of ubiquitousjoint rock-like specimens under uniaxial loading have been investigated by DIC(digital image correlation) and discrete element numerical method(PFC2 D). The results are obtained. Firstly, the UCSJof specimens with c = 15° or 30° shows similar tendency while a goes from 0° to 75°. With c = 45° or 60°, the UCSJof specimens increases when a goes from 0° to 30° and decreases after a goes beyond 30°. With c = 75°, the peak UCSJvalue is reached when a = 0°. The UCSJvalue shows an increasing trend when a goes from 60° to 75°. Secondly, the ubiquitous-joint specimens present different failure modes for various levels of a and c(b-a). 6Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. 7Support technology of deep roadway under high stress and its application显示文摘Roadway instability has been a major concern in the fields of mining engineering. This paper aims to provide practical and efficient strategy to support the roadways under high in-situ stress. A case study on the stability of deep roadways was carried out in an underground mine in Gansu province, China. Currently,the surrounding rock strata is extremely fractured, which results in a series of engineering disasters, such as side wall collapse and severe floor heave in the past decades. Aiming to solve these problems, an improved support method was proposed, which includes optimal bolt parameters and arrangement, floor beam layout by grooving, and full length grouting. Based on the modeling results by FLAC3D, the new support method is much better than the current one in terms of preventing the large deformation of surrounding rock and restricting the development of plastic zones. For implementation and verification, field experiments, along with deformation monitoring, were conducted in the 958 level roadway of Mining II areas. The results show that the improved support can significantly reduce surrounding rock deformation, avoid frequent repair, and maintain the long-term stability of the roadway. 8The preoperative serum cystatin-C as an independent prognostic factor for survival in upper tract urothelial carcinoma显示文摘Cystatin-C(Cys-C)has been reported as a valuable prognostic biomarker in various malignancies.However,its effect on upper tract urothelial carci noma(UTUC)patie nts has not bee n investigated before.Thus,to explore the impact of Cys-C on survival outcomes in patients undergoing radical nephroureterectomy(RNU),a total of 538 patients with UTUC who underwent RNU between 2005 and 2014 in our center(West China Hospital,Chengdu,China)were included in this study.Kaplan-Meier method and Cox regressi on an a lyses were performed to assess the relationship betwee n Cys-C and survival outcomes using SPSS versi on 22.0.The cutoff value of Cys-C was set as 1.4 mg I1 using the receiver operating characteristic(ROC)curves and Youden index.The mean age of patients included was 66.1±11.1 years,and the media n follow-up durati on was 38(interquartile ran ge:19-56)mon ths.Overall,162(30.1%)patients had elevated Cys-C,and they were much older and had worse renal function than those with Cys-C<1.4 mg I^-1(both P<0.001).Meanwhile,Kaplan-Meier analysis revealed that the group with elevated Cys-C had worse cancer-specific survival(CSS,P=0.001),disease recurrence-free survival(RFS,P=0.003),and overall survival(OS,P<0.001).Multivariable Cox analysis suggested that the elevated Cys-C was identified as an independent prognostic predictor of CSS(hazard ratio[HR]:1.997,95%confidential interval[CI]:1.331-2.996),RFS(HR:1.429,95%CI:1.009-2.023),and OS(HR:1.989,95%CI:1.366-2.896).In conclusion,our result revealed that the elevated preoperative serum Cys-C was significantly associated with worse outcomes in UTUC patients undergoing RNU.Ping Tan Ming Shi Jie Chen Hang Xu Nan Xie Huan Xu Yong Jiang Jian-Zhong Ai Liang-Ren Liu Lu Yang Qiang Wei 2019Asian Journal of Andrology2019,21,2:7
9Brief inhalation of sevoflurane can reduce glial scar formation after hypoxic-ischemic brain injury in neonatal rats显示文摘Previous studies have demonstrated that sevoflurane postconditioning can provide neuroprotection after hypoxic-ischemic injury and improve learning and memory function in developing rodent brains.The classical Rice-Vannucci model was used to induce hypoxic-ischemic injury,and newborn(postnatal day 7)rats were treated with 2.4%sevoflurane for 30 minutes after hypoxic-ischemic injury.Our results showed that sevoflurane postconditioning significantly improved the learning and memory function of rats,decreased astrogliosis and glial scar formation,increased numbers of dendritic spines,and protected the histomorphology of the hippocampus.Mechanistically,sevoflurane postconditioning decreased expression of von Hippel-Lindau of hypoxia-inducible factor-1α and increased expression of DJ-1.Injection of 1.52μg of the hypoxia-inducible factor-1αinhibitor YC-1(Lificiguat)into the left lateral ventricle 30 minutes before hypoxic-ischemic injury reversed the neuroprotection induced by sevoflurane.This finding suggests that sevoflurane can effectively alleviate astrogliosis in the hippocampus and reduce learning and memory impairments caused by glial scar formation after hypoxic-ischemic injury.The underlying mechanism may be related to upregulated DJ-1 expression,reduced ubiquitination of hypoxia-inducible factor-1α,and stabilized hypoxiainducible factor-1αexpression.This study was approved by the Laboratory Animal Care Committee of China Medical University,China(approval No.2016PS337K)on November 9,2016.Qiu-Shi Gao Ya-Han Zhang Hang Xue Zi-Yi Wu Chang Li Ping Zhao 2021Neural Regeneration Research2021,16,6:5
10A novel design of 3D carbon host for stable lithium metal anode显示文摘Rational design of porous conductive hosts with high electrical conductivity,large surface area,and adequate interior space is desirable to suppressing dendritic lithium growth and accommodating large volume change of lithium metal anode during the Li plating/stripping process.However,due to the conductive nature of the conductive hosts,Li is easily deposited directly on the top of the hosts,which hinders it from fully functioning.To circumvent the issue,in this study,we designed a novel porous carbon host with a gradient-pore-size structure based on one-dimensional(1D)carbon with different diameters.With this kind of host,stable cycling with high and stable Coulombic efficiency of~98%is achieved at 0.5 mA cm^(−2) with an areal capacity of 1 mAh cm^(−2) over 320 cycles.In contrast,the normal three-dimensional(3D)carbon nanotube host presents a moss-like Li morphology with wildly fluctuating Coulombic efficiency after 100 cycles.The results reveal that the unique gradient-pore-size structure of the 3D conductive host greatly improves the performance of lithium metal batteries.Hang Liu Jie Di Ping Wang Rui Gao Han Tian Pengfei Ren Qingxi Yuan Wanxia Huang Ruiping Liu Qiang Liu Ming Feng 2022Carbon Energy2022,4,4:5
11Investigation of Portevin–Le Chatelier effect in rolled α-phase Mg-Li alloy during tensile and compressive deformation显示文摘Avoiding the Portevin-Le Chatelier(PLC)effect is very important concern for wrought Mg-Li alloys.In this study,the special PLC effect was found in rolled Mg-5Li-3Al-2Zn(LAZ532)alloy during tensile and compressive deformation.By observing microstructure evolution of the alloy during tensile and compressive deformation,it was found that prismaticand pyramidalslips were activated during tensile deformation,resulting in plenty of dislocation accumulation.In the deformation process after compressive yielding,the deformations in coarse grains and fine grains were dominated by{1012}extension twinning and grain boundary slip,respectively.Based on experimental result analysis,the sudden appearance of PLC effect in the later stage of axial tensile deformation(along rolled direction)was caused by interaction between solute atoms and dislocations.In the process of axial compressive deformation(along rolled direction),PLC effect presented the complex and changeable phenomenon of appeared-disappeared-appeared,which was mainly caused by the continuous nucleation of twin in the material,the activation of grain boundary slip and the shear deformation of twin,respectively.Xiaoqiang Li Chunlong Cheng Qichi Le Lei Bao Peipeng Jin Ping Wang Liang Ren Hang Wang Xiong Zhou Chenglu Hu 2020Journal of Materials Science & Technology2020,52,17:3
12Two factors affecting the success rate of the second non-invasive prenatal screening after initial no-call result: experience from a single tertiary center in China显示文摘Background:One inevitable shortcoming of non-invasive prenatal screening(NIPS)/cell-free DNA(cfDNA)sequencing is the uninterpretable(“no-call”)result,which is mainly caused by an insufficient fetal fraction.This study was performed to investigate the factors associated with a successful second NIPS in these cases and determine the optimal management for women with initial no-call results.Methods:We retrospectively analyzed the data of women who underwent NIPS with initial no-call results due to an insufficient fetal fraction from 2017 to 2019 in our center.We compared these women's maternal and pregnancy information with the data of women who had attained a successful second NIPS result and women who had received no-call results for a second time.Results:Among the 33,684 women who underwent NIPS,137 with a no-call result underwent a retest.Comparison between the 87(63.50%)women with a successful retest and the other 50(36.50%)women showed a significant difference in both the initial fetal fraction and maternal body mass index(BMI),whereas the other factors showed no significant differences.In addition,with an initial fetal fraction of<2.00%,the retest success rate was very limited.Conclusions:We identified two major factors associated with a successful NIPS retest:the initial fetal fraction and the maternal BMI.These findings suggest the need for specialized management for this subset of women and would be instructional for the counseling for these women.Ying Lin Dong Liang Hang Li Chun-Yu Luo Ping Hu Zheng-Feng Xu 2021Chinese Medical Journal2021,,12:2

14Submarine Landslide Identified in DLW3102 Core of the Northern Continental Slope, South China Sea显示文摘In this paper, we take DLW3101 core obtained at the top of the canyon(no landslide area) and DLW3102 core obtained at the bottom of the canyon(landslide area) on the northern continental slope of the South China Sea as research objects. The chronostratigraphic framework of the DLW3101 core and elemental strata of the DLW3101 core and the DLW3102 core since MIS5 are established by analyzing oxygen isotope, calcium carbonate content, and X-Ray Fluorescence(XRF) scanning elements. On the basis of the information obtained by analyzing the sedimentary structure and chemical elements in the landslide deposition, we found that the DLW3102 core shows four layers of submarine landslides, and each landslide layer is characterized by high Si, K, Ti, and Fe contents, thereby indicating terrigenous clastic sources. L1(2.15–2.44 m) occurred in MIS2, which is a slump sedimentary layer with a small sliding distance and scale. L2(15.48–16.00 m) occurred in MIS5 and is a debris flow-deposited layer with a scale and sliding distance that are greater than those of L1. L3(19.00–20.90 m) occurred in MIS5; its upper part(19.00–20.00 m) is a debris flow-deposited layer, and its lower part(20.00–20.90 m) is a sliding deposition layer. The landslide scale of L3 is large. L4(22.93–24.27 m) occurred in MIS5; its upper part(22.93–23.50 m) is a turbid sedimentary layer, and its lower part(23.50–24.27m) is a slump sedimentary layer. 17Clinical and dermoscopic features of surgically treated melanocytic nevi: a retrospective study of 1046 cases显示文摘Background: Compared with Caucasians, unique demographic and clinical features have been reported in Chinese patients with malignant melanoma, but similar comparative studies of melanocytic nevi (MN) are lacking. This study examined the clinical and dermoscopic features of MN in surgically treated Chinese cases. Methods: Clinical data and dermoscopic findings from 1046 cases of MN were collected and analyzed. Cases were treated from January 1 to December 31, 2014 at the Department of Dermatology and Venerology, Peking University First Hospital. The association between nevi location and histologic subtypes was examined with Chi-squared test and univariate logistic regression. Chi-squared test was also used to analyze the proportion of globular patterns across different body sites, and proportion of parallel furrow patterns across different histologic subtypes. Results: The majority of the nevi were from female patients, irrespective of location. The range of age at the time of nevi onset was from 0 (birth) to 79 years. There were 381 (36.4%, 381/1046) congenital nevi;of these 81.6%(311/381) were present at birth. Nevi appeared before 30 years of age in 83.2%(870/1046) of the cases. Median values of length growth rate in congenital and acquired MN were 2.0 and 1.6, respectively. Median values of length growth rates in four age groups (0–9, 10–19, 20–29, and ≥30 years) of congenital nevi were 2.2, 2.0, 2.4, and 2.0, respectively. In acral nevi, which often need to be differentiated from acral lentiginous melanoma, 50.2%(109/217) were junctional (odds ratio [OR];95% confidence interval [CI]: 91.572 [52.210–160.959], P < 0.05). Acral location was also associated with a higher likelihood of compound nevi subtype (OR [95% CI]: 14.468 [8.981–23.306], P < 0.05). The globular (59.4%, 354/596) and pseudonetwork (48.8%, 291/596) dermoscopic patterns were often seen in the head and neck region. In areas other than head and neck and acral regions, the globular pattern was the commonest pattern (34.8%, 71/204) regardless of age. Parallel furrow pattern occurred in 46.0%(87/189) of acral MN, followed by fibrillar pattern (21.7%, 41/189). 18Influence of site conditions on ground motion at far field loess sites during strong earthquake显示文摘Loess is widely distributed in China and the Loess Plateau is one of the major areas where strong earthquakes often take place. The seismic amplification effects were discovered in the Plateau during the Wenchuan Ms8.0 earthquake and some other strong events. Based on earth tremor observation, borehole exploration and site seismic response analysis, the site effects of topography of Loess Yuan on ground motion were investigated in details. The earth tremor investigation shows that predominant frequencies at the bottom sites of Loess Yuan are greater than those at the top obviously. The sites seismic response analysis shows that the Loess Yuan may amplify peak ground acceleration (PGA) by 1.44 2.0 times.
